Abstract

Tujuan dari penelitian ini adalah memperoleh informasi infeksi dan sifat-sifat biologis virus Newcastle disease (ND) pada ayam ras maupunayam buras. Sejumlah 529 sampel usap kloaka dikoleksi dari pasar-pasar tradisional dan peternakan rakyat di daerah Bogor dan Tangerang. Sebanyak 20 sampel terdeteksi positif melalui real time reverse transcription-polymerase chain reaction (RRT-PCR) sedangkan dengan isolasi virus terdeteksi positif sebanyak 11 sampel. Selanjutnya empat isolat representasi lokasi dipilih untuk karakterisasi patogenisitas dengan metode mean death time (MDT) pada telur ayam berembrio (TAB) specific pathogen free (SPF), dan karakterisasi antigenisitas menggunakan metode haemagglutination inhibition (HI) dan virus netralisasi (VN) untuk melihat keragaman antar isolat virus. Uji MDT memperlihatkan dua isolat(kode isolat II dan XIII) adalah mesogenik, satu isolat (kode isolate VIII) termasuk lentogenik, dan satu isolat (kode isolat TW) bersifat velogenic. Tiga dari empat isolat menunjukkan hubungan antigenik dengan virus ND strain Komarov dan G7 (genotipe 7) tetapi tidak dengan strain referensi Lasota, sementara isolat velogenic (kode TW) yang berasal dari ayam kampung menunjukkan reaksi silang tinggi dengan antiserum terhadap tiga strain referensi di atas. Sampel-sampel yang diambil di lapang baik itu berasal dari pasar dan peternakan rakyat meskipun secara klinis tidak memperlihatkan gejala sakit dapat diisolasi virus ND dengan karakteristik yang beragam.

Abstract

This study aimed to explore the histomorphology of the pancreatic tissue of medaka fish to develop medaka fish as a diabetic animal model. Adult medaka fish were obtained with a body length of 2.530.22 cm and a weight of 265.825 g. Medaka fish were fixed using Boiun's fixative solution. Macro anatomical observations of the pancreas were conducted using a stereomicroscope. Histological preparations were made according to the Hematoxylin-eosin staining standard procedure. Pancreatic histomorphology of medaka fish was carried out using a light microscope with 100x and 400x magnification. The macro anatomy observations of the pancreas showed the interconnection with several tissues, such as spleen, gallbladder, liver, fat, and intestines. The pancreas was located toward the liver, oversees the intestines, and covered with fat. Pancreatic histomorphological observations showed exocrine cells composed of serous pyramidal acinar cells and endocrine cells (islets of Langerhans). Intralobular and intercalated ducts (ducts of Boll) were composed of simple cuboidal epithelium. This study concluded that the pancreas organ in medaka fish is a single organ, and histomorphologically has both of exocrine and endocrine cells.

Abstract

The objective of this research is to know the influence of vitrification method toward thecondition of bovine oocytes cytoskeleton based on microtubules using the immunohistochemistrytechnique. Oocytes classified in two treatments such as fresh oocytes (control) and oocytes withvitrification treatments of 10, 20, 30, 40, and 50% of ethylene glycol (EG). Data were analyzed bydescriptive analyzing. The results of this research indicated that vitrification treatments were affected tomicrotubules organization of oocytes. Concentration of EG 10, 20, 40, and 50% of EG could not repaircytoskeleton after vitrification, but cytoskeleton structure could be protected by EG 30%.

Abstract

Virus avian influenza (AI) merupakan virus dengan materi genetik RNA single-stranded sense negatif, beramplop yang termasuk dalamfamili Orthomyxoviridae. Onestep simplex reverse transcriptase-polymerase chain reaction (RT-PCR) merupakan salah satu metode diagnosis yang dapat diandalkan untuk mendeteksi virus AI. Tujuan dari penelitian ini adalah untuk menerapkan metode diagnosis cepat virus AI pada spesimen lapangan secara langsung dari pasar unggas berdasarkan amplifikasi RT-PCR gen M dan H5 dengan metode yang berbasis onestep simplex RT-PCR tanpa melalui proses inokulasi dan propagasi virus AI dalam telur ayam berembrio (TAB). Sebanyak 35 sampel spesimen lapangan dari swab trakea unggas yang berasal dari pasar unggas di Terban, Kotamadya Yogyakarta digunakan dalam penelitian ini. Amplifikasi DNA secara onestep simplex RT-PCR pada gen matriks (M) dilakukan terhadap seluruh sampel. Pada sampel-sampel yang menunjukkan hasilpositif pada amplifikasi gen M kemudian dilakukan amplifikasi RT-PCR secara lebih lanjut untuk gen H5 virus AI. Produk hasil amplifikasi RT-PCR gen M dan H5 divisualisasikan menggunakan elektroforesis gel agarose konsentrasi 1% dengan pewarnaan SYBRSafe DNA Gel Staining. Hasil amplifikasi RT-PCR gen M menunjukkan bahwa dari 35 sampel diperoleh 8 sampel positif terinfeksi virus AI tipe A yang ditunjukkan dengan adanya fragmen DNA sebesar 200 bp, sedangkan hasil amplifikasi gen H5 sebanyak 5 dari 8 sampel-sampel tersebut merupakan virus AI tipe A subtipe H5 yang ditunjukkan dengan adanya fragmen DNA sebesar 545 bp. Diagnosis cepat virus AI tipe A subtipe H5 secara langsung dari spesimen lapangan di pasar unggas dapat dilakukan dengan metode onestep simplex RT-PCR, namun metode diagnosis tersebut tidak dapat mendeteksi keberadaan virus AI dalam sampel yang virusnya terlalu sedikit.

Abstract

The aimed of this research is inventoried the parasites of carp in floating cage net at Laut TawarLake. This study used 100 fishes consisted of 50 fries and 50 reproductive stadia. The result of thisresearch indicates the parasites found in the fishes are Trichodina sp., Ichtyophthirius multifiliis,Dactylogyrus sp., Gyrodactylus sp., Epistylis sp. and Lernaea sp. Protozoan parasites were found in allfishes and infected fish organs, while metazoan parasites infected fins and gills and copepods only infectedskin.Keywords: parasites, carps, floating cage net, Laut Tawar Lake

Abstract

Penelitian ini bertujuan mengkaji keragaman genetik gen penyandi ND4 pada Tarsius bancanus, T. b. borneanus, T. dianae dan T. spectrum dan untuk penegakan taksonominya. Deoxyribonucleic acid (DNA) diisolasi dari biopsi jaringan masing-masing spesies Tarsius dengan cara diekstraksi untuk digunakan sebagai DNA cetakan dalam proses amplifikasi dengan metode polymerase chain reaction (PCR). Primer yang digunakan dalam penelitian ini didesain untuk mengamplifikasi gen ND4 dan dilanjutkan dengan elektroforesis. Produk PCR hasil amplifikasi yang telah dimurnikan, selanjutnya dipergunakan sebagai DNA cetakan untuk reaksi penentuan runutan nukleotida. Runutan nukleotida gen ND4 hasil pengurutan dilakukan penjajaran berganda dengan primata lain yang diambil dari Genbank menggunakan Clustal W. Selain berdasarkan runutan nukleotida, gen ND4 dianalisis berdasarkan runutan asam amino dari basa-basa yang diterjemahkan mengikuti vertebrate mitochondrial translation code yang ada pada program MEGA versi 4.1. Konstruksi pohon filogenetika menggunakan metode neighbor joining. Hasil penelitian menunjukkan dari 1378 nukleotida ditemukan 119 situs yang bersifat beragam. Jarak genetika berdasarkan nukleotida gen ND4 yang dihitung menggunakan model dua parameter Kimura, terdapat nilai paling kecil 0,6%, nilai terbesar 13%, dan nilai rata-rata sebesar 6,1%. Filogram berdasarkan hasil runutan nukleotida gen ND4 yang menggunakan metode neighbor joining, dapat mengidentifikasi dan membedakan percabangan antar spesies Tarsius.

Abstract

This study aimed to determine the effect of age on bulls productivity and fresh semen characteristics of Simmental bull in Indonesia. A total of 1071 data of semen collection and production from four age groups (two years old (yo), four yo, 10 yo with high semen rejection (10 HR), and 10 yo with low semen rejection (10 LR) were used in this study to evaluate the productivity and characteristics of fresh semen. The results showed that the pre-freezing and post-freezing semen rejection rate of 10 HR group was higher (P0.05) than the other groups. The four yo group had the percentage of second semen ejaculation each collection was higher (P0.05) than the other groups. Furthermore, semen volume, sperm concentration, and total sperm concentration significantly increased (P0.05) until four yo and then decreased (P0.05) in 10 yo groups. The 10 HR group had the volume and total sperm concentration significantly different (P0.05) with a group of 10 LR. Total sperm motility, individual motility, and mass movement were lower (P0.05) in 10 HR than the other groups. In conclusion, age differences of bulls can affect the productivity and characteristics of fresh semen.

Abstract

This study examined the correlation between the concentration of blood metabolites (triglyceride, albumin, phosphorous) on the reproductive performances (S/C and PPM) of lactating Holstein Friesian crossbred (HFC) cows. Blood samples were collected through the caudal vein 8 hours after feeding and the plasma was analyzed at the Integrated Research and Testing Laboratory of Universitas Gadjah Mada (LPPT UGM). Samples were stored at 5 C. Triglyceride, albumin, and phosphorous were measured using glucose oxidase-phenol 4-aminoantipyrine (GOD-PAP) method, photometric bromocresol green method, and cypress diagnostic, respectively. Blood metabolites data were analyzed using Pearson correlation model. The results showed that the correlation coefficients of triglycerides, albumin, and phosphorus levels on PPM and S/C were -0.521 and -0.650; -0.447 and -0.612; -0.513 and -0.700, respectively. In conclusion, there are significant negative correlation between triglyceride, albumin and phosphorus levels on the PPM, and very significant negative correlation between triglyceride, albumin and phosphorus levels on the S/C of lactating HFC cows.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak etanol daun pegagan terhadap gambaran darah dan peningkatan fungsi kognitif pada tikus. Penelitian ini menggunakan 16 ekor tikus Wistar jantan, dibagi secara acak ke dalam 4 kelompok dan masing-masing diberi ekstrak etanol daun pegagan setiap hari dengan level 0, 100, 300, dan 600 mg ekstrak/kg bobot badan selama 8 minggu. Hasil penelitian menunjukkan bahwa pemberian ekstrak pada level 300 mg/kg bobot badan meningkatkan nilai packed cell volume (PCV), sel darah merah (SDM), dan nilai hemoglobin (Hb). Pengamatan aktivitas pada kelompok dengan level 300 dan 600 mg/kg bobot badan menunjukkan adanya peningkatan dibandingkan kelompok kontrol (P0,05). Pemberian ekstrak etanol daun pegagan juga berperan dalam meningkatkan keberhasilan pengenalan lorong target. Hasil di atas mengindikasikan dengan kuat bahwa ekstrak pegagan mempunyai peran penting dalam perbaikan profil hematologi dan meningkatkan fungsi kognitif.

Abstract

This research was aimed to study the effect of administration of glycerol and dimetilacetamida (DMA) in BTS and MIIIextender supplemented with sodium dodecyl sulphate (SDS) on boar frozen semen. A number of four boars were used in this study for semen collection (n=20). The collected semen was evaluated both macroscopically and microscopically. In this study, only the semen that demonstrated70% sperm motility, 200.106/mL sperm concentration, and20% sperm abnormalities were used and divided into eight tubes. A number of 4 tubes were diluted with 5 mL of BTS, and the rest with 5 mL MIII. The sampel was stored at 20-22 C for 2 hours, followed by centrifugation for 15 minutes (at 2000 rpm), and taken of pellet with 1 ml supernatant. The pellet that was resulted from centrifugation using BTS, then re-diluted with BTS-glycerol 5% (BTSG), BTS DMA 5% (BTSD), BTS-glycerol 5% and SDS (BTSG-S), BTS-DMA 5% and SDS (BTSD-S). Four other pellets that were centrifuged with MIII also re-diluted with MIII-glycerol 5% (MIIIG), MIII-DMA 5% (MIIID), MIII-glycerol 5% and SDS (MIIIG-S), MIII-DMA 5% and SDS (MIIID-S). Next, all of diluted semen were inserted into 0.5 mL straw and equilibrated for 2 hours (4 C), then frozen and stored in liquid nitrogen. The evaluation of frozen semen quality was conducted at 24 hours after frozen. The result of this study showed that post-thawing motility of spermatozoa in BTSD-S (40.170.2%) was found higher (P0.05) compared to seven other dilution processes. Therefore, it is concluded that the concentration of 5% DMA that supplemented with SDS in BTS dilution much better for maintaining boars frozen semen quality.
