Abstract

Penelitian ini bertujuan mengamati pola pertumbuhan folikel dan keberhasilan inseminasi buatan dengan semen cair pada induk kuda warmblood yang disinkronisasi estrus dan ovulasi (ovsynch). Induk kuda berjumlah lima ekor berumur 6-18 tahun digunakan dalam penelitian ini. Sinkronisasi estrus dilakukan pada induk kuda yang memiliki korpus luteum berdiameter minimal 3,0 cm dengan injeksi prostaglandin 7,5 mg secara intramuskular. Induksi ovulasi dilakukan dengan memberikan hCG 1500 IU secara intravena 48 jam setelah sinkronisasi estrus dan diulang setiap 24 jam sampai terjadinya ovulasi folikel (dosis jamak) yang diamati dengan ultrasound. Inseminasi buatan dilakukan berulang mengikuti setiap pemberian hCG sampai terjadinya ovulasi dengan dosis inseminasi 1,5x109 spermatozoa. Sinkronisasi estrus dan ovulasi dengan menggunakan hCG dosis jamak menghasilkan ovulatori dominan folikel berdiameter 4,810,92 cm dan korpus rubrum berdiameter 3,820,45 cm serta menghasilkan 60% kebuntingan. Kesimpulan sinkronisasi ovulasi dengan pemberian hCG dosis jamak pada kuda warmblood yang diinseminasi buatan dengan semen cair efektif menghasilkan kebuntingan yang tinggi.

Abstract

The objective of this study was to evaluate the effect of exercise levels on the hematological and metabolical status, and carcass quality of Garut sheep. In total 24 of Garut sheep used in this study. The levels of exercise contain three treatments namely non exercise, semi exercise and exercise. Blood collection through the jugular vein of sheep samples was carried out at the end of the study, using a 5 mL EDTA tube to measure the haematological condition of the animal sample. Likewise, collecting muscle samples to determine the physical quality of meat.The data was analyzed by analysis of variance and continued by Duncan test for a post hoc test. The results showed that the models of exercise were not significantly (P0.05) affected to hematological level and also carccas quality including juiceness, tenderness, drip loss and water holding capacity. Organoleptic sensory using semiexercise was more favourable meat compare to exercise and non exercise. While for the colour of meat, non exercise treatment was significantly (P0.05) affected to the colour of meat compare to exercise and semi exercise. Furthermore the semi exercise was the best treatment to produce fresh taste meat with good sensory characteristic.

Abstract

The aim of this study was to evaluate the ability of rats Leydig cells collected with Nycodenz gradient in producing testosterone in vitro. Leydig cells were collected using 5 column of Nycodenz gradient (4, 8, 10, 12, and 15%) and cells were evaluated regarding its concentration, viability, and purity of Leydig cells. Media used to cultured Leydig cells were Dulbeccos Modified Eagles Medium (DMEM)+10% newborn calf serum (NBCS); DMEM+10% NBCS+2,5 IU/mL human chorionic gonadotrophin (hCG); DMEM+10% NBCS+ 5 g/mL insulin, 10 g/mL transferrin, and 5 g/mL Se (ITS); DMEM+10% NBCS+hCG+ITS at 5% CO2 incubator with temperature of 37.5 C for 3 days. Culture medium was collected every day for testosterone analysis with enzyme-linked immunosorbent assay (ELISA). By adding ITS to the medium, Leydig cells concentration was significantly increased (8.92x106 cells/mL) compared to medium with serum (7.74x106 cells/mL) or hCG (7.68x106 cells/mL) (P0.05). ITS and hCG in medium significantly increased Leydig cells concentration (10.40x106 cells/mL) at day 3 of culture (P0.05). The result of parallelism test showed that the assay obtained good validity to measure testosterone concentration in culture medium. Testosterone in medium was detected at 1.80-2.60 ng/mL at day 1 of culture. In conclusion, Leydig cells collected with Nycodenz gradient had no effect to testosterone secretion from Leydig cells in vitro.

Abstract

Penelitian ini bertujuan mengetahui kadar kolagen kulit (matriks ekstraseluler), jumlah sel (kadar DNA), aktivitas sintetik (kadar RNA) padajaringan kulit, serta kadar hormon progesteron pada berbagai tingkatan umur tikus, dan memperoleh umur tikus ovariektomi yang cocok digunakan sebagai hewan model penuaan. Rancangan percobaan yang digunakan adalah rancangan acak lengkap (RAL) yang terdiri atas sembilan kelompok percobaan masing-masing terdiri atas tiga ekor tikus, yaitu umur 12 bulan (K1), 18 bulan (K2), 24 bulan (K3), 30 bulan (K4), 36 bulan (K5), umur 12 bulan dalam kondisi 1 bulan pascaovariektomi (K6), 12 bulan dalam kondisi 3 bulan pascaovariektomi (K7 ), 18 bulan dalam kondisi 1 bulan pascaovariektomi (K8), dan umur 24 bulan dalam kondisi 1 bulan pascaovariektomi (K9). Data yang diperoleh dianalisis dengan menggunakan analisis varian dan dilanjutkan dengan uji Duncan. Komponen matriks ekstraseluler dan matriks seluler menurun seiring dengan bertambahnya usia. Umur tikus normal yang cocok digunakan sebagai hewan model penuaan adalah tikus umur 24-36 bulan. Tikus ovariektomi yang cocok digunakan sebagai hewan model penuaan adalah tikus umur 12 bulan pascaovariektomi 3 bulan.

Abstract

Differential display technique was used to identify differentially expressed gene ofswordtail fish (Xiphophorus spp.) that induced by 1 g/ml copper for 24 hours. From eightprimers that have been used for differential display, seven of them showed the differentialgenes which were not present in the control. The reamplification of these fragments rangingfrom below 100 to 200 bp. But only two fragments were successfully cloned and sequenced,those are H-AP10 and H-AP11. Sequence analysis showed that both of the sequences revealedhigh similarity with Homo sapiens stress-associated endoplasmic reticulum protein, a signalgenerated that induces apoptosis.Keywords: differential display, swordtail fish, copper pollution, mRNA expression, biomarker

Abstract

Penelitian ini bertujuan mengembangkan teknik enzyme-linked immunosorbent assay (ELISA) untuk mendeteksi antibodi terhadap virus bovine ephemeral fever (BEF). Pada penelitian ini dikembangkan uji ELISA langsung (direct ELISA) dan tidak langsung (indirect ELISA) dengan menggunakan antibodi monoklonal (blocking ELISA). Hasil penelitian menunjukkan bahwa uji direct ELISA tidak dapat digunakan dengan baik karena terjadi positif palsu. Uji blocking ELISA bereaksi lebih baik dan dapat dikembangkan lebih lanjut untuk mendeteksi antibodi terhadap penyakit BEF. Dapat disimpulkan bahwa pengembangan teknik deteksi dini terhadap BEF dengan mempergunakan antibodi monoklonal dapat diterapkan dalam upaya pengawasan penyakit dan surveilans.

Abstract

Genetic diversity of three Iraqi local goat breeds (Black, Hybrid, and Meriz) was studied by using random amplified polymorphic DNA(RAPD) markers. Fifteen primers were used, they generated a total of 259 amplification products, from which 142 bands were polymorphic,revealing 55.29% polymorphism. It was found that the mean value of Neis gene diversity (h) and Shannon's nformation index (I) were 0.1778 and 0.2546, respectively. According to the analysis of genetic distances among breeds, the highest genetic distance recorded between Black and Hybrid 0.5108, while the lowest genetic distance recorded between Hybrid and Meriz 0.1431. The phylogenetic dendrogram based on Neis genetic distance analysis was in agreement with their phenotypic similarities in which the tree separated the surveyed breeds into two clusters, the first one including Black goat breed, and the second cluster includes Hybrid and Meriz breeds. These results indicated that Black goat breed is most genetically distant from both Hybrid and Meriz breeds. Finally, the moderate value for both of polymorphism and genetic diversity which obtained from the present study indicate that these breeds are genetically different, which provide a good background about these breeds forgenetic improvement and conservation of our local goat breeds in the future.

Abstract

This study was conducted to determine the prevalence rate of mares GTCT in Indonesia. Ultrasound examination of ovaries were performed on mares in Java and Madura Island of Indonesia. Thirteen mares had been affected by GTCT from 2913 number of examinations. The prevalence rate of GTCT during year 2006 and 2007 were 4.71% and 5.56%, respectively followed by a decrease in prevalence rate of GTCT during 2008-2014. The reproductive cycle in a mare with GTCT had returned to normal 6 months after ovariectomy.

Abstract

This study aims to delineate the tissue structure of male cat external reproductive organs using ultrasound with different types of consoles and transducers frequencies through indirect method. External organ of 5 tom weighing 2-3 kg was evaluated using ultrasound with stationary console (linear transducer, 10 MHz) and portable console (linear transducer, 6.5 MHz), transcutaneously. The results showed that both console and transducer could not visualize the structure of organs sized less than 10 mm through direct method. However, the indirect method using stationary console with a 10 MHz transducer enable to visualize the organ and depicts clearly the internal organ structure such as penis and its parts, scrotum and its constituent layers, caput and cauda of epididymis, and testicles. Furthermore, portable console with 6.5 MHz transducer was still able to provide an adequate image of those organs. In general, fibrous tissues such as tunica vaginalis, tunica Dartos, funiculusspermaticus, and urethra were visible in ultrasonogram as hyperechoic structures, while soft tissues such as the parts of testicles and penis were visible as hypoechoic structures. Tissue containing fluid such as urethral lumen was visible as anechoic structure. In conclusion, indirect ultrasound imaging method was sufficient to visualize the organs sized less than 10 mm using 3-15 MHz transducer in which was currently widely used in animal health care clinics.

Abstract

Penelitian ini dilakukan untuk mengaji respons metafisis tulang femur distalis tikus ovariektomi yang mengonsumsi suplemen kalsitriolselama enam minggu. Lima belas tikus Wistar umur delapan minggu, secara acak dibagi tiga kelompok (normal/N, ovariektomi/Ov, dan ovariektomi + kalsitriol/OvD) masing-masing 5 tikus. Tikus kelompok N dan Ov diberi pakan standar, sedangkan tikus kelompok OvD diberi pakan standar + kalsitriol. Pada umur 14 minggu, tikus dimasukkan kandang metabolik individu untuk studi balan. Pada hari 4-7 studi balan,setiap hari sisa pakan, urin, dan feses dikoleksi untuk pemeriksaan Ca. Pada akhir penelitian, tikus dieutanasia, tulang femur kanan diambil pemeriksaan histopatologis dengan pengecatan hematoksilin dan eosin. Hasil penelitian menunjukkan bahwa konsumsi Ca dan retensi Ca pada tikus OvD tidak berbeda signifikan meskipun cenderung meningkat, sedangkan ekskresi Ca dalam feses dan urin meningkat sangat signifikan (P0,01). Metafisis tulang femur distalis tikus kelompok OvD terlihat zona spikulum tulang trabekula lebih pendek dan berbentuk irreguler, dan rongga sumsum tulang didominasi jaringan adiposit. Dari hasil penelitian dapat disimpulkan bahwa suplementasi kalsitriol selama enam minggu pada tikus ovariektomi menyebabkan osteoporosis pada metafisis tulang femur distalis.
