Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37 C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory

Abstract

Analisis genetika menggunakan random amplified polymorphic DNA (RAPD) untuk mengetahui kemiripan genetika antara kambing peranakan Ettawa (PE) hasil inseminasi buatan (IB) dan kawin alam (KA). Hasil penelitian menunjukkan bahwa dari 20 primer OPA hanya 10 primer yang bisa digunakan untuk mengamplifikasi fragmen deoxyribonucleic acid (DNA) dalam kelompok kambing PE, yang terdiri atas 21 kambing hasil KA dan 19 kambing hasil IB. Berdasarkan profil DNA yang diperoleh dan hasil analisis dengan software numerical taxonomy and multivariate analysis system (NTSYS) disimpulkan bahwa hubungan genetika antara kambing PE hasil kawin alam dengan inseminasi buatan adalah sebesar 48% dan dikategorikan rendah.

Abstract

The purpose of this study was to analyze the success of the artificial insemination (AI) program in local cattle in Pante Bidari District, EastAceh Regency, Aceh Province. It was conducted in five villages: Alue Ie Mirah, Buket Bata, Grong-Grong, Meunasah Tunong, and Paya DemamEmpat. The survey method was employed in this study. The respondents were selected by using purposive sampling method. There were 40breeders and 3 inseminators included as the respondents. To be involved in the study, the breeders had to meet certain criteria, such as having anexperience of breeding local cows, at least twice using the AI system. As for the inseminators, they were required to have a permit from the EastAceh Livestock Service. The results showed that the conception rate (CR) was 76.45%; the service per conception (S/C) was 1.22; the calvinginterval (CI) was 12.70 months, and the calving rate (CvR) was 89.92%. This study concluded that the implementation of the artificialinsemination program in Pante Bidari District, East Aceh Regency, has been running effectively.

Abstract

Dogs and cats are often kept as pets, thus they stay close to humans. Parasitic/zoonotic diseases in humans were often caused by ectoparasite in cats and dogs. This study aimed to determine the morphological differences between cat fleas and dog fleas and the prevalence of cats and dogs infested by fleas around the home environment in Ilir Barat 1 District, Palembang City. The method used was descriptive exploratory method with field observation techniques and specimen collection. The study was conducted in November - December 2018 in five Sub-districts, (Bukit Lama, Bukit Baru, Lorok Pakjo, Demang Lebar Daun, and 26 Ilir 1). The results showed that cat fleas (Ctenocephalides felis) and dog fleas (Ctenocephalides canis) had different color and some morphological differences, such as the shape of the head, the number of spines in the genal comb, the number of notches in the tibia, and the number of seta in the metapisternum. The highest prevalence of fleas infestation in cats was in Bukit Lama and 26 Ilir 1 (33.3%), while the highest prevalence of infestation in dogs was in Demang Lebar Daun Village (36.6%). Only in Lorok Pakjo Sub-district that no dog fleas infesting dogs are found. __________________________________________________________

Abstract

virus was isolated from suspected Newcastle disease (ND) chicken in backyard farm at Sibang Village, Badung Regency, Bali Province. The isolates were then propagated and confirmed for NDV serologically. RNA isolation was performed by standard Trizol method. Phylogenetic tree analysis of NP gene (nt1020-1561) and HN gene (nt7019-7754) were performed using sequence of Badung-02/AK/14 and selected NDV strains from GenBank. Based on the NP gene sequence, the newly isolate closely related with other NDV strains belong to genotype VII that previously isolated such as Banjarmasin-010/10, Bali-1/07, and Cockatoo/90 with genetic distance 0.2%, 12.6%, and 18.0% respectively. The genetic distance with LaSota/46 virus (genotype II) is 19.6%. Based on HN gene sequence, genetic distance of the Badung-02/AK/14 with other viruses belong to genotype VII such as Banjarmasin-010/10, Bali-1/07, and Cockatoo/90 are 0.4%, 3.7%, and 4.2% respectively. The genetic distance with LaSota/46 virus (genotype II) is 9.0%. There were no difference between the result of nucleotide and amino acid sequence analysis, both in NP and HN gene (P0.05).

Abstract

Adhesi merupakan tahap inisiasi dari proses kolonisasi bakteri. Kemampuan adhesi ini diperantarai oleh keberadaan hemaglutinin pada permukaan bakteri. Tujuan penelitian ini adalah melihat kemampuan hemaglutinin pada Escherichia coli dalam proses adhesi. Uji adhesi pada sel epitel pipi dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian menunjukkan bahwa Escherichia coli yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik yakni rata-rata 67,6 bakteri/sel dibandingkan dengan yang tidak memiliki hemaglutinin rata-rata 7,2 bakteri/sel. Kemampuan adhesi ini berkaitan dengan keberadaan hemaglutinin di permukaan bakteri.

Abstract

Penelitian ini bertujuan mengetahui ekspresi protein AdhF36 strain Salmonella Typhi, sebagai kajian kandidat vaksin berbasis molekul adesin. Strain yang digunakan pada penelitian ini bersumber dari Rumah Sakit Hasan Sadikin Bandung, Laboratorium Kesehatan Sulawesi Tengah, Palu, Rumah Sakit Udayana Denpasar, dan Rumah Sakit Saiful Anwar, Malang. Hasil penelitian menunjukkan bahwa sekitar 12 strain Salmonella Typhi tersebut mengekspresikan dan memiliki protein AdhF36. Hal ini memberikan gambaran umum bahwa protein AdhF36 potensial dikembangkan sebagai kandidat vaksin.

Abstract

The purpose of this study was to evaluate the effect of age on wound healing and physical development of local cats (Felis catus) after ovariohysterectomy. A total of 15 female cats were divided into three age groups (2-4, 5-9, and 10 months old) with each group consisting of five cats. Thetreatment group was sterilized using the lateral flank ovariohysterectomy method. Observations of surgical procedures and physical conditions were carriedout during anesthesia, including onset, duration of anesthesia and surgery, body temperature, heart rate, and respiratory rate. Post-operative observationswere made to evaluate wound healing, hematological profile (RBC, Hb, PCV, WBC, PLT), body weight, Body Condition Score (BCS), bone density, andgrowth plate closure. The results showed that there was no significant difference (P0.05) during anesthesia, wound healing, or an increase in body weight,BCS, and bone density in all age groups. Ovariohysterectomy at 2-4 months old delayed the closure of the bone growth plates (P0.05).Ovariohysterectomy in pubertal and post-pubertal cats is safe to perform as long as good surgical preparation is provided.

Abstract

The aim of this study was to identify proteolytic bacteria from the ruminal and reticulum fluids of aceh cattle based on the 16S rRNA gene. Samples used were ruminal and reticulum fluids of aceh cattle slaughtered in Abattoir of Aceh Besar. Samples were diluted and cultured into Skim Milk Agar medium at 39 C for 48 hours. The morphology of bacterial colonies growth in the medium was observed. Colonies resulted in the largest clear zone were isolated and used for Deoxyribonucleic Acid (DNA) isolation, 16S rRNA gene amplification and sequencing. Theresults showed that morphology of dominant colonies was yellowish white color, round shape, position on the agar surface. The results ofphylogenetic analysis of RS1 and ReS2 isolates isolated from rumen and reticulum fluids of aceh cattle respectively had a close familialrelationship and belonged to the bacterial group of Kosakonia. Sequence homology showed isolate RS1 and ReS2 are probably either newEnterobacteriaceae species or unconfirmed species. Halo zone produced by ruminal bacteria had a wider diameter (25 mm vs 20 mm) than thatcaused by reticulum bacteria). Based on the results, RS1 (bacterium in the rumen) and ReS2 (bacterium in the reticulum) belong to similar type, namely Kosakonia sp. with a proteolytic activity. Presumably, these bacteria originate from the rumen that enters the reticulum with degraded feed.

Abstract

The study was done to investigate the glycogen distribution in the stomach of long tailed macaques (Macaca fascicularis). The study used eight individuals of long tailed macaques consist of 6 fetuses (55, 70, 85, 100, 120, and 150 days pregnancy) and 2 infants (10 and 105 days). The gastrict tract were histologically stained using Hematoxyllin-Eosin (HE), and histochemically stained using Periodic Acid Schiff (PAS), to detect glycogen. The results revealed that the histogenesis pattern of stomach glandullar cells are developed in line with the age of the fetuses and infant. We also found that glycogen existance are increase in line with the age in the glandullar stomach. From the present study we concluded that the glycogen distribution in gastrict glandular cells were influenced by the age in the fetal period, while in the infant was under influence of the digestion activities, and the activities of the gastrict glandular cells was already seen at 55 days gestation.
