Abstract

This study was conducted to know the influence of cattle fat and coconut oil in affecting bloodLDL cholesterol. Eighteen male local chickens (Gallus gallus) at the age of 2 months were used. Thechickens were allotted into 3 groups of 6 each. The chickens in each groups were fed with 521 standardration (control groups); 9% cattle fat in standard ration 521 (group II); 9% coconut oil in standardration 521 (group III) for 45 days. All chickens were fed ration and drunk ad libitum every day. At theend of the experiment, the blood samples were collected for analysis blood LDL cholesterol withenzymatic methods using spectrophotometre. Analysis of variance with completely randomized designindicated that treatment significantly effect on blood LDL cholesterol (P0.01). From the results could beconcluded that cattle fat 9% supplementation could be increase blood LDL cholesterol levels. While coconut oil supplementation significantly effect on decrease blood LDL cholesterol levels. Keywords: cattle fat, coconut oil, LDL cholesterol

Abstract

Penelitian ini bertujuan mengkaji aktivitas enzim alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), dan creatinine kinase (CK) pada kambing peranakan Ettawa. Sebanyak 43 ekor kambing peranakan Ettawa dari berbagai kelompok status fisiologis (jantan muda, betina muda, jantan dewasa, betina dewasa, betina bunting, dan betina laktasi) yang secara klinis sehat dan tidak cacat digunakan dalam penelitian ini. Pemeriksaan klinis dan pengambilan sampel darah dilakukan sesuai dengan skedul pemeriksaan dan pengambilan sampel sesuai dengan standard klinik yang diuraikan Baumgartner (1999). Pengambilan sampel darah dilakukan pada jam 7.00-8.00 wib. Pemeriksaan enzim di dalam serum diperiksa dengan menggunakan metode standar sebagaimana diuraikan oleh Kraft dan Duer (1999). Aktivitas AST, ALT, ALP di dalam serum memiliki level yang sebanding dengan level pada ruminansia lain. Sementara itu, aktivitas CK lebih tinggi dibanding nilai yang dinyatakan di dalam literatur untuk ruminansia kecil lainnya.

Abstract

The aim of this research was to determine if the substitution of rosella filtrate (Hibiscus sabdarifa L.) in citrate-based diluent and egg yolks with the ratio 4:1 (16 mL citrate + 4 mL yolk) would help maintain the progressive motility of Kacang goat spermatozoa at room temperature. This study used a male Kacang goat with an average sperm quality of approximately 205 x 107 spermatozoa/mL and motility of 81.6%. An electro-ejaculator was used once a week to collect the semen. The treatments were labelled as follows: P0 was the negative control group, treatment P1 was given 5% rosella filtrate, treatment P2 was given 10% rosella filtrate, and treatment P3 was given 15% rosella filtrate. Examination of the motility of the spermatozoa was carried out at room temperature. The examination was carried out once every four hours. It was concluded that the rosella filtrate could maintain the progressive motility of spermatozoa of the Kacang goat at optimal concentrations. The optimal motility percentage was found at a concentration of 15% at a storage life of 0-4 hours.

Abstract

This study aimed to find out the optimum incubation time to complete mouse oocyte maturation at Metaphase II (MII) stage and determine the optimum sucrose concentration enabling to induce nuclear swelling for visualization that is important for enucleating process at the initial procedure of somatic cell nuclear transfer (SCNT). In this current study, mice were used as animal model. Completely randomized design was arranged, consists of 2 trials with 4 treatments and 7 replications. In the first trial, the oocytes were cultured at 0-2, 4-6, 8-10, and 12-14 h in 5% CO2 incubator at 37 C. Second, the MII oocytes obtained from previous trial were cultured in M199 medium containing different concentrations of sucrose (0, 1.5, 3, and 6%). The parameters measured were the oocyte viability at various stages, i.e germinal vesicle (GV), metaphase I (MI), anaphase/telophase I (A/T I), and metaphase II (MII), and the viability of swollen nuclear oocytes using Hoechst/PI staining. The results showed that the optimum incubation time required by oocytes to reach MII stage was 12-14 h with a percentage of 57.1412.67%, while the optimum sucrose concentration for nuclear swelling was found at 3% with a percentage of 1000.00%. Our findings provided preliminary results related to the maturation process of the mouse oocyte nucleus, which is meaningful for the initial procedure of SCNT.

Abstract

Penelitian ini bertujuan mengetahui ekspresi insulin pada pankreas mencit (Mus musculus) yang diinduksi streptozotocin berulang dengan pewarnaan imunohistokimia yang berguna sebagai hewan model diabetes melitus. Tiga puluh ekor mencit jantan galur Balb-C, umur 12-14 minggu dengan bobot badan 30-40 g dikelompokkan menjadi 2 kelompok perlakuan, masing-masing kelompok terdiri atas 15 ekor. Kelompok 1 (K1) diberikan pelarut streptozotocin, sedangkan kelompok 2 (K2) diberikan streptozotocin dengan dosis 40 mg/kg bobot badan dalam 50 mM natrium sitrat bufer pH 4,5 secara intraperitoneal sebanyak 0,5 ml selama 5 hari berturut-turut. Hewan percobaan dari masing-masing kelompok dieutanasia sebanyak 2 ekor pada hari ke-0, 7, 14, 21, dan 28 setelah perlakuan, selanjutnya mencit diperfusi dan dinekropsi untuk mengambil jaringan pankreas sebagai sampel pemeriksaan imunohistokimia dengan metode streptavidin peroksidase menggunakan antibodi mouse anti-insulin (1:300). Berdasarkan uji statistik menggunakan analisis varian, ekspresi insulin pada sel beta Langerhans pankreas K1 lebih tinggi dibandingkan K2 (P0,05). Waktu pengamatan dan interaksi antara kelompok dan waktu pengamatan menunjukkan perbedaan yang signifikan (P0,05). Induksi dosis rendah streptozotocin secara berulang dapat menurunkan jumlah ekspresi sel beta Langerhans pankreas yang imunoreaktif terhadap insulin.

Abstract

The objective of this experiment was to determine the addition of sodium dodecyl sulfate (SDS) to Ringer Lactate-Egg Yolk (RL-EY) extender on pelung chicken semen preservation. Semen was collected three times a week from three pelung chickens. Collected semen was evaluated macroscopically and microscopically. Only ejaculates of at least 70% sperm motility was then divided into three equal microtubes. Each of them diluted with RL-EY with ratio 90%:10%, then added with 0.00, 0.025, and 0.05% SDS respectively. The liquid semen then stored at 5 C for 72 hours. Sperm motility and viability were observed every 12 hours. The addition of 0.025% SDS showed higher spermatozoa motility and viability (72.081.44% and 80.821.30%) which were significantly higher (P0.05) than 0.00 and 0.05% SDS addition at 24 hours of storage. There was no differences on the spermatozoa motility and viability between 0.00 and 0.05% SDS addition. The decrease of spermatozoa motility and viability was observed in 0.025% SDS addition (4.170.56% and 4.650.59%, respectively) that significantly lower compared to 0% and 0.05% SDS addition at 24 hours of storage. In conclusion, 0.025% SDS addition in a Ringer Lactate -Egg Yolk extender maintained pelung spermatozoa motility and viability better than 0.00 and 5% SDS addition.

Abstract

Penelitian ini bertujuan mengetahui pengaruh suhu dan waktu penyimpanan ovarium terhadap tingkat fertilisasi oosit secara in vitro pada domba. Ovarium dibawa dari rumah potong hewan (RPH) dalam medium NaCl fisiologis pada suhu yang berbeda yaitu 27-28 C, 36-37 C, dan 4 C. Oosit kemudian dikoleksi dari setiap kelompok berdasarkan waktu penyimpanan yang berbeda yaitu 2-4, 5-7, dan 8-10 jam setelah domba dipotong. Oosit dikoleksi dan dimaturasi secara in vitro dalam inkubator 5% CO2, 38,5C selama 28 jam. Oosit kemudian difertilisasi ke dalam drop spermatozoa selama 14 jam dalam inkubator CO2 5%, 38,5 C. Tingkat fertilisasi dievaluasi berdasarkan jumlah pronukleus yang terbentuk. Tingkat fertilisasi oosit yang dikoleksi dari ovarium yang disimpan pada suhu 27-28 C tidak berbeda dengan tingkat fertilisasi oosit yang disimpan pada suhu 36-37 C dan pada suhu 4 C, 2-4 jam setelah kematian hewan (masing-masing 53; 66,66; dan 63%) (P0,05). Tingkat fertilisasi oosit mulai mengalami penurunan pada tiga kelompok perlakuan setelah ovarium disimpan selama 8-10 jam, tingkat fertilisasi oosit yang disimpan pada suhu 27-28 C; 36-37 C; dan suhu 4 C masing-masing berturut-turut sebesar 9,8; 22,22; dan 12,24%. Dari hasil penelitian disimpulkan bahwa penyimpanan ovarium pada suhu 27-28 C dan 36-37 C selama 5-7 jam dapat mempertahankan kompetensi oosit dibandingkan dengan penyimpanan pada suhu 4 C.

Abstract

Catechin and their derivatives have been studied to have antiviral potential against COVID-19 through in silico method pharmacokinetics screening and molecular docking. Pharmacokinetics and toxicity profiles were obtained through the ADMETSAR server and SwissADME server. Then proceed with the prediction of affinity through the method molecular docking using the software application MOE 2007.09. The testmaterial is in the form of a 3D catechin structure and its derivatives as well as several control ligands downloaded via Pubmed. While templatethe Receptor M pro protein and the Spike glycoprotein are downloaded from pdb.org (PDB ID: 6LU7 and 6LXT). The pharmacokinetic profile of catechins is relatively better than all control ligands with the lowest toxicity. Molecular docking results also show that catechins and theirderivatives have a stronger affinity than control ligands. This research proves that catechin has antiviral potential through inhibition of Mpro protein and Spike glycoprotein COVID-19 virus.

Abstract

This study aimed to verify the effect of ethanol extract of jaloh leaves at various doses on parasitemia percentage of in mice (Mus musculus), wich inoculated Plasmodium berghei. This study used 12 male mice (Mus musculus) strains Balb / C with average body weight of 30.2 grams of 2-month-old. This research using Completely Randomized Design (CRD) with four treatments and three replications. All the mice were infected with Plasmodium berghei prior to treatment on three days post infected the mice in group 2,3 and 4 were given with extract ethanol jaloh leaves at dose 50, 100, 150 mg/kgbw inspectively. The mice in group 1 (K0) were not given anything and consident as control.Observations parasitemia percentage using microscopy on preparations stained thin blood smear Giemsa. The parasitemia percentage data were analyzed using one way ANOVA. On the average (SD) percentage of parasitemia during the three days in four treatments (K0, K1, K2, and K3) on the first day were 24.871.86; 24.331.53; 24.201.06; and 24.931.01. On the second day were 22.672.52; 23.001.00; 22.801.20; and 24.132.01; and on the third day were 24.771.66; 22.973.76; 21.131.90; and 24.271.42. Statistical analysis showed that ethanol extract of the jaloh leaves for KO, KI, KII, and KIV respectively treatment did not affect the percentage of parasitemia in mice (Mus musculus) inoculated Plasmodium berghei.

Abstract

Penelitian ini bertujuan mengetahui efek inhibin B terhadap ekspresi protamine P2 di dalam kepala spermatozoa pada kauda epididimis. Penelitian ini menggunakan 24 ekor tikus jantan berumur 4 bulan yang dikelompokkan secaraacak ke dalam 4 kelompok (KO, KI, KII, dan KIII), setiap kelompok terdiri atas 6 ekor tikus. Kelompok KO merupakan kelompok kontrol hanya diinjeksi dengan PBS. Kelompok KI, KII, dan KIII diinjeksi dengan inhibin B dengan dosis masing-masing adalah 25, 50, dan 100 pg/ekor. Tikus diinjeksi sebanyak 5 kali secara intra peritoneal dengan interval waktu pemberian 12 hari selama 48 hari. Injeksi pertama, inhibin B dicampur dengan 0,05 ml PBS dan 0,05 ml CFA. Injeksi kedua hingga kelima, inhibin B dicampur dengan 0,05 ml PBS dan 0,05 ml IFA. Pada hari ke-6 setelah injeksi inhibin B terakhir, hewan coba dikorbankan secara dislocatio cervicalis lalu jaringan kauda epididimis dikoleksi dan difiksasi dengan paraformaldehid 4%. Setelah melalui proses dehidrasi, jaringan blok di dalam parafin dipotong dengan ketebalan 6 mikron dan diwarnai secara imunohistokimia dengan menggunakan antibodi anti protamine P2. Pengamatan secara imunohistokimia menunjukkan adanya ekspresi protamine P2 di dalam kepala spermatozoa pada semua kelompok perlakuan. Akan tetapi, seiring dengan penambahan dosis inhibin B menyebabkan terjadinya penurunan tingkat ekspresi protamine P2 di dalam kepala spermatozoa pada kauda epididimis. Hasil penelitian menunjukkan bahwa injeksi inhibin B dengan dosis 100 pg/ekor menurunkan secara nyata jumlah ekspresi protamine P2 di dalam kepala spermatozoa pada epididimis (P0,05) dibanding KO.
