Abstract

Aloe vera mengandung lignin, asam folat, vitamin A,dan 20 jenis asam amino ( Jatnika, 2009) dan akar manis (Glycyrrhiza glabra L.)mengandung glycyrrhizin, steroid, dan asam beta –glycyrrhetinik yang bermanfaat bagi kesehatan rambut(Dep, 2014). Tujuan dari penelitian ini adalah untuk mendapatkan sediaan hair tonic dengan aktivitas terbaik dari kombinasi ekstrak air lidah buaya dan akar manis. Skrining fitokimia dari ekstrak air akar manis dan Aloe vera dilakukan, selanjutnya sediaan hair tonic dibuat dalam 3 formula dengan zat aktif ekstrak lidah buaya 7,5% dan ekstrak air akar manis 2,5% dengan variasi etanol 96% sebanyak 5%, 10% dan 15% selanjutnya diuji aktivitasnya pada kelinci jantan putih sebanyak 4 ekor selama 28 hari . Evaluasi sediaan hair tonic meliputi uji organoleptik, pH, berat jenis, viskositas dan uji hedonik. Pengujian aktivitasnya dilakukan dengan mengukur laju pertumbuhan rambut, percepatan pertumbuhan rambut  dan bobot rambut. Hasil penelitian menunjukkan Sediaan  hair tonic yang mengandung campuran ekstrak air lidah buaya 7,5% dan ekstrak air akar manis 2,5% dengan etanol 96% sebanyak 5%, 10% dan 15% dapat dibuat sediaan larutan penyubur rambut yang baik, dan yang memiliki aktivitas optimum adalah sediaan  hair tonic yang menggunakan etanol 96% sebanyak 15 % dengan karakteristik warna coklat, bau khas, homogen, bobot jenis 1,0577 g/ml,viskositas 2,8479 cp terhadap aquades, pH 5,5 dan efektif sebagai penyubur rambut dengan laju pertumbuhan rambut 2,6617 cm dalam 28 hari, percepatan pertumbuhan rambut 0,1635 cm/hari dan bobot rambut 0,1854 gram dalam 28 hari.Kata Kunci : Aloe vera L., Glycyrrhiza glabra L., hair tonic, laju pertumbuhan rambut, bobot rambut.

Abstract

One of the plants which contain the alkaloid is a plant jembirit (Tabernaemontana sphaerocarpa BL. ).Sap and leaves from this plants have been used to treat skin diseases and sprain. Alkaloid from jembirit plants showed potent cytotoxicity against various human cancer cell. The goal of this research is to find out the influence of the extraction method against the level of total alkaloid jembirit leaves.Jembirit leaves were extracted by maceration method and extraction with Soxhlet apparatus used  ethanol 70 % as a solvent . Standardization of extracts conducted by test of ash content, test of drying shrinkage, and extract yield. Qualitative analysis conducted by alkaloid test. Determination of total alkaloid was analyzed with visible spectrophotometry method using Bromocresol green as complexing agent. The results showed that jembirit leaves contained alkaloid compounds. The determination resulted the levels of total alkaloid of maceration was 0.727% ± 0.0032, levels of total alkaloid extraction with Soxhlet apparatus was 0.666% ± 0.0022. The stastitical analysis showed significance differences of total alkaloid levels between maceration method and extraction with Soxhlet apparatus viewed from siginificancy value (0.001<0.005).

Abstract

Diabetes melitus (DM) merupakan  penyakit metabolik  yang menyebabkan kerusakan sel termasuk pankreas. Kuersetin danomega-3 telah diteliti sebagai terapi tambahan pada DM karena dapat menurunkan stres oksidatif dan mencegah kerusakan sel. Tujuan penelitian ini adalah mengetahui efek kombinasi kuersetin dan omega 3 dalam melindungi sel β pankreas dari kerusakan pada tikus DM. Subyek terdiri dari 28 ekor tikus, dibagi menjadi 1 kelompok sehat dan 6 kelompok DM tipe 2 (4 ekor perkelompok). Bahan uji adalah kuersetin dosis 5, 20 dan 80 mg/kgBB/hari dan omega-3 dosis 25, 100 dan 400  mg/kgBB/hari peroral selama 4 minggu. Setelah perlakuan, jaringan pankreas diambil untuk pengecatan hematoksilin-eosin dan dihitung persentasekerusakannya.Pada penelitian ini, kombinasi kuersetin 80 mg/kgbb/hari dan omega-3 400 mg/kgbb/hari menurunkan tingkat kerusakan sel Β pankreas paling baik secara bermakna (p<0,05) dari pada kombinasi dosis lebih kecil maupun kuersetin saja atau omega-3 saja. Jadi kombinasi kuersetin 80 mg/kgbb/hari dan omega-3 400 mg/kgbb/hari menurunkan tingkat kerusakan sel β pankreas paling baik secara bermakna jika dibandingkan kombinasi dosis lebih kecil maupun tanpa kombinasi.Kata Kunci: Kuersetin, omega-3, diabetes melitus tipe-2, histologi sel β pankreas, hematoksilin-eosin.

Abstract

Some species from Piper genus were known for traditional medicine and ornamental plant. Previous studies showed that Piper betle and Piper acre had antioxidant activities. But, the study of antioxidant activity from other species of Piper was still limited. Because of that, the purposes of this study were to determine the antioxidant activity, and total flavonoid content of Piper nigrum, Piper aduncum, Piper retrofractum, and Piper crocatum. Extraction was done by gradient maceration using n-hexane, ethyl acetate, and ethanol as solvents. The extracts were concentrated by rotary evaporator. Antioxidant activity was evaluated by DPPH (1,1-diphenyl-2-picrylhydrazyl) free scavenging method. Total flavonoid determination was done by colorimetry method using aluminum chloride (AlCl3) as a reagent. The result showed that the IC50 values of Piper nigrum, Piper aduncum, Piper retrofractum, and Piper crocatum were ranged between 58-1249 μg/mL. The total flavonoid values of all extracts were between 1.4-8.3 mg QE/100 mg. The extract which had the strongest antioxidant activity was the ethanol extract of Piper nigrum with an IC50 value was  57.72 μg/mL. The highest values of total flavonoid was showed by the ethanol extract of Piper aduncum which was 8.3 mg QE/100 mg extract.

Abstract

The used of Gentamicin is still regarded for overcoming infectious diseases. Several preparations of the gentamicin are available in the market as injection, eye or ear drops, and topical dosage forms. Based on the standard method, determination of the gentamicin is carried out by microbiological assay. Several instrumental method have been reported. In this research, Thin Layer Chromatography-Densitometry (TLC-Densitometry) has been validated and used for determining gentamicin in cream dosage form. The TLC was carried out on a Silica gel GF254 using KH2PO4 20% and ninhidrin 2% solution (in ethanol 96%) as eluent and spots visualization respectively. Three spots appeared on the chromatogram having Rf value 0.51, 0.47, and 0.36. All the spots gave maximum absorption at 400 nm. The spot with Rf value of 0.51 was the highest intensity. Limit of Detection (LoD) and Limit of quantitation (LoQ) of the major component was 0.019 and 0.064 respectively. The other validation characteristics met the requirement for determination of gentamicin in the cream dosage form.

Abstract

Probiotics are living microorganisms that can have a positive impact on health when consumed in adequate amounts. This research aimed to determine the effect of different binders on the viability of Lactobacillus acidophilus, a member of Lactic Acid Bacteria (LAB). The probiotic granules were prepared with wet granulation method using mannitol as the filling ingredient. Formulas I-III used acacia as a binder (3%, 4%, and 5%), while Formulas IV-VI used sodium alginate (1%, 1.5%, and 2%). The probiotic granules produced from all formulas were evaluated for flow time, the angle of repose, compressibility, and LAB viability. The LAB viability test results showed that all of the six formulas met the probiotic requirements, i.e., at least 107 CFUs (Colony Forming Units) per gram. The viable LAB in Formulas I-VI were 3.94x107, 4.4x107, 2.7x107, 2.6x107, 3.5x107, and 2.3x107 CFU/g, respectively. The data were analyzed by comparing the average values of the One-Sample T-Test results, followed by Mann-Whitney test. The results revealed that each formula had different capacity in maintaining the viability of Lactobacillus acidophilus with different binders (i.e., acacia and sodium alginate) as a probiotic product. Formula II (4% of acacia) produced probiotic granules that met the requirements of compressibility and the angle of repose. However, its granular flow time exceeded the standard.

Abstract

Elephantopus scaber Linn had been used as traditional medicine. It contain some lacton sesquiterpene with antiproliferative activity. The study aim was to explore the effect of chloroform fraction of ethanolic extract of E.scaber on the expression of p53 on T47D cell line. The ethanol extract was found by maceration method. The extract was then fractionated using petroleum ether and chloroform respectively to get the chloroform fraction. The expression of p53 was observed by immunocytochemistry method with antip53 antibody. The p53 cell line was cultured and treated with fraction with concentration of 7.06 and 3.53 µg/ml. The expression of p53 was showed by brown colour in the nucleus of the cell. The result showed that chloroform fraction treatment with concentration of 7.06 µg/ml induced apoptotic cell death in the end stage (necrosis). While the treatment with lower concentration 3.53 µg/ml could increase the p53 expression.

Abstract

The different health problems in elderly patients require complex management. However, if the use of medicines is uncontrolled, it will lead to polypharmacy. This study aimed to analyze the effect of health education on family caregivers’ knowledge of polypharmacy particularly to prevent drug therapy problem in elderly patients. This quasi-experimental study used one-group pre-test post-test design. The population was family caregivers who had elderly members in their care and lived within the working area of Bengkuring Public Health Center. The population members were selected according to the inclusion criteria until a minimum sample size was reached. Having met the criteria, the respondents were asked to fill in a pre-test questionnaire before they partook in the health education for polypharmacy and a post-test questionnaire after their participation. The pre-test and post-test information were then analyzed using a paired t-test. This study revealed that the family caregivers’ knowledge of polypharmacy improved by 32.5% with a probability value (p= 0.000< 0.001) less than the level of significance (5%). The contribution of the health education to the family caregivers’ knowledge of polypharmacy was 74.3%. There was an improvement in the family caregivers’ knowledge of polypharmacy after participating in the health education.

Abstract

Endophytic fungi used as a new resource to produce bioactive compounds such as antibacterial, antioxidant and anticancer. This study aimed to analyze the antibacterial, antioxidant, cytotoxic fraction of the separation ethyl acetate extract of mycelium of endophytic fungi genus Fusarium sp. Diethyl ether fraction showed high antibacterial activity in E.coli (20,75 µg/mL), S.typhi (35,08 µg/mL), and S.aureus (51,96 µg/mL). Test showed that the antioxidant activity of ethanol 96% fraction has highest antioxidant activity and total phenolic content (75,85 ± 0.87 mg GAE). Cytotoxic test on T47D breast cancer cells showed that the fraction of diethyl ether have highest activity with IC50 of 10,16 ± 0,88 µg /mL.

Abstract

Dexamethasone as glucocorticoids has been widely used to reduce inflammation and tissue damage in a variety of conditions. It was commonly prescribed in combination with the other compounds such as chlorpheniramine maleate for the relief of asthma. It was important to observe the quality and safety of the combination dosage form, called pulveres or puyer in order to serve the patient oriented medication. This research aimed to develop analytical method validation and determine the content uniformity of the divided powder containing dexamethasone compounded by pharmacy in Yogyakarta and hospital in Central Java, Indonesia. A validated high performance liquid chromatography (HPLC) method was chosen to determine dexamethasone in the samples. The column of ACE 5 C18 (250 x 4.6 mm) was used in this research and the methanol:water (65:35) was used as the mobile phase at the flow rate of 1 mL/min. The UV detection was performed at 239 nm. Analytical method validation was performed on the parameters of system suitability, limit of detection, limit of quantification, accuracy, precision, and linearity. The results showed that dexamethasone contains in divided powder compounded by pharmacy in Yogyakarta and hospital in Central Java were met the acceptance requirement.
