Abstract

CIDR is a device to stimulate and controled estrous cycle in livestock. This study aim to investigate the estrus characteristics and responses of ewes CIDR treated for 12 and 13 days interval. CIDR were treated into 16 ewes for 12 days or 13 days interval. The evaluation of clinical sign of estrus, ferning image, vaginal smear sample, and measurement of vaginal mucus resistance with estrus detector, were performed before treatment of CIDR, at 0, 12, 24, 36, 48, and 54 hour after CIDR removal. Ferning image for both group started from 24 hour after CIDR removal with few ferning pattern. 36 hour after CIDR removal, ferning image already clear. 48 hour after CIDR removal, ferning showed the optimum image, and cover all view. 54 hour after removal, ferning started to decrease. The composition of vaginal epithel dominated by superficial cell started at 24 hours after CIDR removal followed by appearance of estrus sign. The mean of estrus mucus resistance in both groups showed high values before estrus then decreased during estrus. The means of electrical resistance value for both group approximately 310Ù to 700Ù before estrus and approximately 210Ù to 290Ù during estrus. The number of ewes shown the sign of estrus were 75% in group of 12 days interval and 100% in group 13 days. The mean of onset estrus for 12 and 13 days interval groups are 28 and 30 hours after CIDR removal respectively. In conclution, the results of this study showed that CIDR treatment for 13 days show better intensity of estrus sign than 12 days interval group.

Abstract

Meat is one of the livestock products producing from abattoir, which in its implementation must be able to maintain good quality including of cleanliness, health, and halal for consuming by coommunity. This study aims to determine the community’s perception on the existence of abattoirs in the Tahoa Village, Kolaka District. This research is quantitative descriptive using a Likert scale. The samples used in this study ware 34 people. The results of the study showed that of the odor variable has a value of 357 and categorized as in the disturbed category. As well as for, water pollution with a value of 207 was categorized as quite disturbed and also for waste with a value of 233 was categorized as quite disturbed. The total of all three sub-variable values (odor, water pollution and waste) was 797, and it was categorized as moderately disturbed. In conclusion of this study shows that the community’s perception of the existence of abattoirs in the Tahoa Village, Kolaka District, is quite disturbed.

Abstract

The data available on the female reproductive organ of mouse deer (Tragulus javanicus) is still very limited. A study was therefore conducted to investigate the distribution and the concentration of carbohydrate residues during the development of ovary follicles. An ovary at luteal phase was used in this study. Thin sections of the ovary were prepared occording to the standard methods and they were then histochemically stained with flourecnece-labelled lectins such as peanut agglutinin (PNA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), Winged bean agglutinin (WGA) and Ulex europaeus agglutinin (UEA). The result showed that changes in the distribution and the concentration of carbohydrate occured during the development of the follicle. During the preantral stage, the cytoplasm of oosit contained carbohydrate with the residues of glucosa dan mannosa. Zona pelusida contained carbohydrates with residues of glucosa, mannosa, galactosa dan N-asetylgalactosamine, whereas extracellular matrix contained carbohydrate with the residues of glucosa dan mannosa. In the antral follicle, the cyitoplasm of oocytes contained carbohydarte with the residues of galactosa dan N-asetylgalactosamine, whereas its zona pelusida, extracellular matrix and follicular fluid contained carbohydarte with the residues of fucosa, N-asetylglucosamin and cyalic acid. Diffrences in the types and the distribution pattern of carbohydrates were observed in this study, both in preantral and antral follicles.

Abstract

Sixty four samples from female and male local chickens in high and low attitude areas of Bali wereexamined for Leucocytozoon infection. Erytrocyte count, haemoglobin level and packed cell volume (PCV)were examined. Data obtained from this study was analyzed by Wilcoxon Mann Whitney Test. The resultshowed that no significant difference was observed on erytrocyte, haemoglobin and PCV values betweeninfected (2,48 x 106/mm; 9,36g/dl; and 33,62% respectively) and uninfected (2,56x106/mm3; 9,47g/dl and34,28% respectively). A significant difference was however observed on the erythrocyte count between malechickens in higher area (2,60x106/mm3) and female in lower area (2,59x106/mm3). No significant differencewas observed on haemoglobin and PCV between male chickens in higher area (9,35g/dl & 34,62% and9,56g/dl & 34,75% respectively) and female chickens in lower area (9,38g/dl & 32,63% and 9,16g/dl &32,5% rspectively).

Abstract

A study on effect of various concentrations of turmeric juice on pathological changes mice liver hasbeen carried out. As many as 25 mice were used in this study and they were divided into 5 groups, i.e.: groupK-: 0.2 ml vegetable oil without CCl4; group K+: CCl4 without turmeric extract; group T1: CCl4 with 10%turmeric extract; group T2: CCl4 with 20% turmeric extract and group T3: CCl4 with 30% turmeric extract.All mice, except the K- group, were treated with 0.2 ml CCl4 orally for 7 days. On day-8 to day-29, the CCl4treated mice were given various concentrations of turmeric extract, i.e.: 10%, 20% and 30%; whereas, the KandK+ groups were given drinking water adlibitum. On day-30, all mice were sacrificed and their livertissues were collected and then processed to histopathological staining with haematoxylin-eosin. Theresult showed a clear evidence to suggest that turmeric extract can induce a better recovery compared tonon-treated mice. The improvement of pathological condition was characterized by reduction of pathologicalchanges found in the liver cells of mice, such as: hidrophic degeneration. fatty degeneration, and necrosis.More importantly, treatment with 30% of turmeric juice induced a better recovery than those with 10%and 20%.

Abstract

The success of individual in keeping healthy from infectious disease is closely related to the ability oftheir body immune responses againts infectious agents. It is reported that one way to increase the immuneresponses is by system to antioxidant. Red fruit oil contains carotenoid and tocoferol that function asantioxidant. This study was aimed at investigating the effect of red fruit oil on spleen lymphocyteproliferation in mice infected with Listeria monocytogenes. Sixty female Balb/c mice were used in thisstudy. The animals were randomly selected and divided into five groups, each group were 12 mice. The firstand second groups received aquadest only, whereas the third, fourth, and fifth groups received red fruit oilwith different doses, ie. 0.3, 0.6, and 1.2 mL/kbBW/day, respectively. Lymphocyte proliferation activitieswere tested by using 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay method.Lymphocyte proliferation assay demonstrated that there was significant difference between the groupswith red fruit oil and aquadest, started at day 2 post-infection. The optical density (OD) values recorded atday 3 in groups I, II, III, IV, and V were 0.769 ± 0.025, 0.904 ± 0.048, 1.110 ± 0.020, 1.021 ± 0.033, 0.979 ±0.002, respectively. The highest optical density (OD), ie. 1.194 ± 0.032, occurred at day 6 after receiving 0,3mL/kgBW/day. In conclusion, red fruit oil could increase lymphocyte proliferation.

Abstract

The objective of this study was to identify the proliferation and differentiation of mice cerebrum cellsand its protein product. Research has been conducted on in vitro growth of three days old rat cerebrum cellsin Dulbecco’s Modified Eagle Medium (DMEM) containing 10% Amino Acid Non Essential (AANE), 10%Fetal Bovine Serum (FBS), 3mM NaHCO3, 50 mg/ml gentamycin, and supplemented with and without 5mg/ml insulin, 10 mg/ml transferin, 5 mg/ml selenium (ITS). Culture was done in 5% CO2, then incubatedat 37oC until 90% confluent. Identification were done on cell proliferation and population doubling time(PDT), number of neuron and glial cells, length of axon and dendrit, and protein secretion. The number andlength of neuronal cells were calculated by using hemocytometer and eyepiece micrometer, respectivelly.Protein secreted into culture medium, designed as conditioned medium (CM) was analysized using sodiumdodecyl sulfate–polyacrilamide gel electrophoresis (SDS-PAGE) method. Quantitative data were analyzedusing statistical T-test on Minitab program. In vitro culture of rat cerebrum cells showed two types of cellsincluding nerve cells of bipolar and multipolar neurons and glial cells including astrocyte (the fibrous andprotoplasmic), oligodendrocyte, and microglia. Supplementation of ITS into the culture medium increasedthe cell proliferation rate (P<0.05) with lower PDT, and quantitatively, the 30 kDa secreted protein asindicated by the higher intensity of the protein band.

Abstract

Studies have been conducted to determine the potential antiproliferative effect of Nigella sativa seed(NSS) extract on 7,12- dimetilbenza[a]antrasena (DMBA)-induced female rats. The chloroform NSS extracteffect was observed on rat gatric cells. Sprague Dawley  female rats were divided into five groups, eachgroup consisted of 12 individuals. A control group was DMBA, Groups B, C and D were the groups treatedwith NSS extract with ranked dose 250 mg / kg, 500 mg / kg and 750 mg / kg and group E is the corn oilcontrol group. Histopathological observation of cells using H&E staining and cell proliferation activity wasobserved with the mean AgNOR (mAgNOR). mAgNOR values   were analyzed using Kolmogorov-Smirnovtest, than followed with ANOVA one way, and Tukey test HSD. The results showed H&E staining and  thechloroform exctract showed mAgNOR N.sativa semen were able to reduced gastric cell damage and lowergastric cell proliferation in DMBA- induced rat. The results showed that N.sativa potential to develop asa chemopreventive agent in gastric cancer.

Abstract

The effectiveness of atherogenic diet in raising plasma cholesterol level of cynomolgus macaques(Macaca fascicularis) can be predicted for population, but not for individuals.This study aimed to evaluatethe individual plasma cholesterol of cynomolgus macaques in response to IPB-1 atherogenic diet. Theanimals under study were 22 adult malemonkeys from the animal facility of Primate Research CenterBogor Agricultural University (PSSP IPB). All animals were intervened with the IPB-1 atherogenic diet forthree months and individual plasma cholesterol wasevaluated in a monthly basis.The results showedthat the monkeys’ plasma cholesterol had increased significantly (P <0.05) after one month of interventionperiod and the increases were very significantly different (P <0.01) after three months. Individually, increasesin plasma cholesterol varied among animals. Based on these variations, 2 animals were categorised ashypo-response, 18 animals as hyper-response and 2 animals as extreme.This study showed the variationof individual plasma cholesterol of cynomolgus macaques in response tothe IPB-1 atherogenic diet so thatthe animals might be selected based on the their responsiveness.

Abstract

Mycobacterial culture provides definitive diagnosis of tuberculosis (TB), but commercially readyto-use culture media for Mycobacterium bovis are rarely available. The aims of this study were todevelop and to evaluate the ability of M. Bovis to grow in Modified Ogawa Agar (MOA) in comparisonwith the available culture media, such as Löwenstein Jensen (LJ) and Modified Ogawa (MO). Eachmedia were inculation with 0.1 ml suspension of 105 CFU/mL M. bovis and M. phlei in PhosphateBuffer Saline (PBS) and each media was replicated in five tubes. Mycobacterium phlei grew in everymedium since day 4. M. bovis grew in media LJ and MO since day 17, but failed to grow in mediumMOA. The recovery rate of M. phlei in LJ and MOA were significantly different. The ability of MOA tocultivate M. phlei was different from LJ. Colonies of M. phlei in MOA were easier to be harvested, muchsimpler to prepare, and more feasible than medium LJ. The recovery rate of M. bovis in media LJ andMO were not significantly different, but medium MO were much simpler to prepare and more feasiblethan medium LJ. Media MOA were able to cultivate M. phlei, but proven unable to cultivate M. bovisin this research.
