Abstract

The study was carried out to observe the effect of ethanolic extract of estrogenic purwoceng given 13-21 days age of pregnancy on reproductive performance such as the weight of ovarium, uterus, placenta, andpups, correlation ratio of corpus luteum number, implantation site number, and litter size in rats. Thepregnant rats were divided in to two groups. One groups cosisted of rats treated with purwoceng (83.33 mg/kg body weight) and the other groups was used as control (no treatment). The ovary, uterus, placenta, andpups were collected and weighed on day 21 of pregnancy. The result showed that the rats given 83,33 mg/kg BW ethanol extract of purwoceng tended to increase the weight of ovarium, uterus, and pups but it didnot increase the weight of plasenta. Number of site of implantation, ratio of implantation site to numberof corpus luteum, and ratio of number of pups to implantation site were higher on rat given 83,33 mg/kgBW ethanol extract of purwoceng. Conclusion of this study is ethanol extract of Pimpinella alpina rootsgiven at postplasentation improve reproductive performance.

Abstract

Dengue Haemorrhagic fever (DHF) is one of fearsome diseases in society. Incidence of the disease isincreasing. Dengue fever is caused by dengue virus and transmitted by Aedes aegypti mosquito vector.Various chemical controls have been conducted to prevent the spread of the disease, but active contents ofthe chemical controlling substances are suspected causing many negative effect, in environment, such asvector resistance, death of non target living creatures, and environmental contamination.This researchobjective was to find an alternative solution in order to control the dengue vector by using entomopathogenicfungi as biological control agent. This research was conducted by isolation and identification of fungiinfecting mosquito larvae. Macroscopic observation revealed that one of the nine isolation products wasLagenidium giganteum. The effectiveness test in laboratory showed the zoospore LD50 to Ae.aegypti larvaeof instar 2nd was 2,35 x 106 zoospore/ml, while the LD95 value was 1,35 x 107 zoospore/ml. The oosporeeffectiveness test showed LD50 was 6,7 x 102 oospore/ml and LD95 was 1,94 x 103 oospore/ml. Using LPCBdye and blue tolouidin 2,5%, the infection mechanism of L.giganteum fungi in Ae.aegypti mosquito larvawas detected. The research is concluded that the entomophatogen fungi L. giganteum was very prospectiveto be used as a biological control agent against vector of DHF.

Abstract

Groupers are bio-indicators of the health of coral ecosystems and high economic value. Various speciesof grouper fish sold in markets in Indonesia is not known. The purpose of this study was to identify thespecies of grouper fish market located in Karangasem and Kedonganan. The method used in this studywas polymerase chain reaction(PCR) techniqueand mitochondrial DNA 16S rRNA gene as a molecularmarker. Grouper samples were collected from the fish market Karangasem total of 11 samples and 42samples collected from Kedonganan. Sequences from each sample was obtained from the analysis inBerkeley Sequencing Facility, California. Sequencing samples were then analyzed by BLAST and matchedwith the sequences found in GenBank. The analysis showed that six species can be identified from the fishmarket Karangasem namely Cephalopholis leopardus, C.cyanostigma, C. miniata, C. sonnerati, Variolaalbimarginata, and Epinephelus fasciatus. Seven species can be identified in the fish market Kedonganannamely E. fuscoguttatus, E. coeruleopunctatus, E. merra, E. polyphekadion, C. cyanostigma, C. miniata andV.albimarginata.

Abstract

Controlling SepticemiaEpizooticae (SE) through vaccination program has been undertaken in KupangDistrict. However, numbers of fatal cases is still being reported. A cross sectional study have been undertakenin order to determine the prevalence and factors affecting herd immunity at farm level in Kupang District.A total of 434 cow’s sera from 87 farmers were collected and further analyzed using Enzyme-linkedImmunosorbent Assay (ELISA). An antibody titer of e”200 ELISA Unit was used as the indicator ofprotective immunity. The prevalence of herd immunity was 73.7% and vaccination coverage was 90.8%.Based on unweighted logistic regression analysis it was found that factors affecting the animals protectiveimmunity were: animal >2 years of age (r = +1.45601; OR = 4.3); housing system ( r = +1.03958; OR = 2.8);reservoir animals (r = +0.090147; OR = 2.5), the sex of the animal (female) (r = + 0.080138; OR =2.2); oncefrequency of vaccination (r = +0.61015; OR = 1.8); and 6-12 months post vaccination period (r = + 0.58968;OR = 1.8). Based on linier regression analysis, the prevalence of herd immunity was increased during rainyseason, whereas it was decreased when cows werebought from animal market or from other district.

Abstract

Until recently, two clades of of avian influenza viruses (AIVs) designated as 2.3.2 and 2.2.3 havebeen circulating in Indonesia. Mutations of AIV genes have cretaed many more variants of the virus. It istherefore important to evaluate the appropriate methods used for the detection and diagnosis of AI virusin the field. Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) have been used as a standardmethod for detection of AIV in many laboratories in Indonesia. The success of RT-PCR for detection ofAIV virus is dependent on the nucleotide sequences of primer that match with the circulating of AIVs. Theaims of this study was to develop RT-PCR by designing primers for H5 subtype specific to the circulatingAIVs in the field. The primers were designed using Primer Design software, and optimization andvalidation of the primer were conducted using AIVs that have been characterized in the previous study.The primers were then used RT-PCR using AIV isolates from field samples and their sensitivity andspecificity were then determined. The results showed that the H5 primers designed in this study, H5-IDand H5-NLP, was able to detect the AIVs in field samples better than the H5-specific primers have beenused previously. In conclusion, H5 primers designed based on recent viruses in the field showed betterresults in the detection of AI virus as compared to the previous primers. As AIV-H5N1 subtype in the fieldwill continue to change and evolve, the use of primers designed in this study is recommended for diagnosisof H5 AIV.

Abstract

A study to detect the presence of bovine major histocompatibility complex (Bo-MHC) in maduracattle has been carried out. Lymphocyte samples were collected from 25 cattle. The protein of lymphocytesamples was analysed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) andtransferred onto nitrocellulose membrane. Bo-MHC proteins on the membrane were detected by monoclonalantibodies (anti-Bo-MHC class I) and BAQ150A (anti-Bo-MHC class II). The proteins of 48 kDa and 11kDa were detected by MAb B5C which appeared to be the alpha and beta chain of Bo-MHC class I. MAbBAQ150A detected protein band of 26 KDa which was likely to be the beta chain of Bo-MHC class II. Bo-MHC class I dan class II were clearly detected in madura cattle by MAbs which provides a basis for furtherinvestigation on the role of the MHC in the susceptibility the animals to many diseases.

Abstract

Dugong dugon yang merupakan mamalia laut, terdaftar sebagai spesies langka yang dilindungi. Sampai saat ini, belum banyak laporan yang tersedia tentang nilai fisiologis normal dari mamalia ini, sehingga menyulitkan dokter hewan dalam memeriksa dan mendiagnosis penyakit pada hewan ini. Oleh karena itu dilakukan studi tentang nilai normal elektrokardiogram (EKG), frekuensi jantung, respirasi, dan suhu tubuh dari seekor Dugong dugon dengan menggunakan alat pemantau pasien (Patient monitor). Parameter yang diukur adalah frekuensi jantung, respirasi, suhu tubuh, dan nilai EKG: gelombang P, komplek QRS, interval PR, interval QT, dan mean electricity axis (MEA). Hasil perekaman menunjukkan bahwa nilai normal frekuensi jantung, respirasi dan suhu tubuh berturut-turut adalah (45,73+4,10) denyut/menit, (10,11+3,44) inspirasi/menit, dan (31,42+0,41)0C. Tidak ada perbedaan yang nyata antara nilai EKG normal pada hantaran I (dada kanan) dan hantaran II. Sementara itu, pada hantaran III menunjukkan nilai voltase yang isoelektris (0 mV). Pada Lead II, gelombang P menunjukkan durasi (0,05+0,02) detik dengan voltase (0,03+0,01) mV, gelombang R menunjukkan voltase (0,07+0,02) mv dengan durasi QRS komplek (0,06+0,01) detik, interval PR (0,26+0,01) detik, interval QT (0,21+0,11) detik, axis jantung (MEA: +90 sampai -135 ) condong ke kanan, dengan irama jantung reguler. Nilai normal Dugong dugon ini menunjukkan karakteristik yang spesifik dibanding nilai normal pada anjing, hal ini mungkin karena adana perbedaan ekologis, struktur anatomi, dan ketebalan dinding thoraks pada kedua hewan tersebut.
