Abstract

Listeria monocytogenes is included in the foodborne pathogen, which has been associated with severaloutbreaks of human listeriosis especially in high risk groups. Listeria monocytogenes could be found inGouda cheeses because of poor hygienic and sanitation practices. In addition, this bacteria could surviveduring the making of cheese and cheese ripening process. The purpose of this study was to identify thepresence of L. monocytogenes in local and imported Gouda cheeses and how the safety level of the Goudacheese against contamination of L. monocytogenes. This study used the conventional method in accordancewith the Bacteriological Analytical Manual, US Food and Drug Administration and Bergey’s Manual ofDeterminative Bacteriology to detect the presence of L. monocytogenes at 15 samples of local Gouda cheeseand 15 samples of imported Gouda cheese sold in supermarkets in Jakarta and Bogor. The results of thisstudy showed that was not found L. monocytogenes in local and imported Gouda cheese. It could be concludedthat is Gouda cheese relatively safe from L. monocytogenes and meets Indonesian National Standard.

Abstract

The aim of this study was to measure the effect of corn oil in feed on estrogen hormone level in blood, hierarchy follicular performance and productivity of Japanese quail. Thirty female Japanese quail were housed in caged and fed ad libitum, the birds were randomly divided into three grouped namely: Po, P1 and the (P0) was fed with commercial feed without corn oil, whereas the first test treatment (P1) was fed with commercial feed mixture containing 3% corn oil, and the second test treatment (P2) was feed with commercial feed mixture containing 6% corn oil along the study. Every day the egg production was recorded. After eight weeks, blood samples were collected from jugular vein to isolate the serum. Then all samples were sacrificed to observe the hierarchy follicular performance. Based on this study it was found that 3% and 6 % corn oil supplementation on feed (P1 and P2) did not cause any significant changes in serum estrogen concentration, and productivity, but had significant changes on hierarchy follicular. This result show that hierarchy follicular increasing due to ovarian activity optimizing induced by polyunsaturated fatty acid (PUFA)by consumed corn oil. Thus, the suplementation of corn oil can be an alternative to increase egg productivity of quail farm. ABSTRAK Penelitian ini bertujuan untuk mengukur pengaruh pemberian minyak jagung dalam pakan terhadap kadar estrogen serum darah, hirarki folikular dan produktivitas burung puyuh. Sebanyak 30 ekor burung puyuh betina dibagi menjadi tiga perlakuan. Perlakuan kontrol diberi pakan komersial tanpa suplementasi minyak jagung, sedangkan perlakuan P1 dan P2 diberi pakan komersial dengan suplementasi minyak jagung sebanyak 3% dan 6%. Setiap hari dilakukan pencatatan terhadap produktivitas. Setelah delapan minggu, dilakukan koleksi sampel darah, kemudian semua burung puyuh dikorbankan nyawanya untuk mengamati hirarki folikular yang terbentuk. Pada P1 dan P2 tidak telihat perbedaan yang nyata terhadap peningkatan kadar estrogen serum darah dan produktivitas dibandingkan dengan P0, akan tetapi terlihat perbedaan nyata pada hirarki folikular yang terbentuk. Hasil ini menunjukkan bahwa peningkatan hirarki folikular yang terbentuk terjadi karena peningkatan kinerja ovarium dipicu oleh polyunsaturated fatty acid (PUFA) yang berasal konsumsi minyak jagung. Dengan demikian, suplementasi minyak jagung dapat dijadikan alternatif untuk meningkatkan produksi telur dalam peternakan burung puyuh.

Abstract

The objective of this study was to evaluate the use of brilliantcresyl blue (BCB) in selecting potentialbovine oocytes for maturation and fertilization in vitro. Brilliant cresyl blue (BCB) is a dye that can assessintracellular activity of glucose-6-phosphate dehydrogenase (G6PD), a synthesized enzyme in maturedoocytes. Oocytes were exposed to 26 ?M BCB diluted in modified Phosphate Buffer Saline (mPBS, PBS +10% Fetal Bovine Serum) for 90 minutes is 5% CO2 incubator at 390C. The oocytes were classified accordingto their cytoplasm coloration: oocytes with blue cytoplasm (BCB+) and unstained oocytes (BCB-). Oocytesof the control group were incubated shortly after morphological selection without being exposed to BCB.Afterwards, all groups of oocytes (BCB+, BCB-, and control) were matured and fertilized in vitro. Maturedoocytes were those oocytes that reach metaphase II after 24 hours culturing; whereas oocytes showing twoor more pronuclei at 14 hours post incubation were classified as fertilized oocytes. The nuclear maturationrate was significantly (P<0.05) higher in BCB+ group (78.7% ± 0.41) than the BCB- group (33.3% ± 0.13).However, there was no significance difference (P>0.05) between the BCB+ and the control group (77.1% ±0.32). The fertilization rate was significantly higher (P<0.05) in the BCB+ group (30.5% ± 0.04) comparedto the BCB- (13.6%±0.03) and control group (23.6% ± 0.05). In conclusion, BCB staining of bovine oocytesprior in vitro maturation could be used in selecting potential developing oocytes.

Abstract

This study aims to determine the pathological changes in dog suspected of being infected by Leptospira spp. This research used a retrospective study design. A total of 210 canines sample were examined at Faculty of Veterinary Medicine The Udayana of University between January 2008 to January 2010. Of the 210 specimens, five canines with clinical sign anorexia, fever, vomiting, polyurea, dyspnea and only one are noted icteric in the sclera. Macroscopically : the anemic to petechial haemorrhagis was find in pulmo, icterus in liver with gall blader distention, kidney swollen with black color in hillus, spleen rather swollen with black color on end and mild hemorrhagis on intentines. The infected organ is inserted into a pot filled with neutral buffer formalin 10%. Then stained with haematoxyllin-eosin for the microscopic preparation. Microscopic examimination revealed the presence of spiral bacterial cells on the necrotic liver and kidneys tissue. Infiltration neutrophils and macrophages was also find in pulmonary and spleen tissue. Besided that intertubuler edematous were also observed under microscopic examination. It can be concluded changes associated with Leptospira spp infection can be found in liver and kidney tissue with changes such as necrosis, proliferation of kuffer cells and intertubular edematous.

Abstract

Pregnancy is an integral part of the circulatory system between pups and mother. Growth anddevelopment of pups is determined by intake from their mother. The purpose of this study was to measurethe growth of pup’s bones given ethanol extract purwoceng (Pampinella alpine) for 1 to 13 days of ratgestation. Eight pregnant rats were divided into two groups: control group and purwoceng group that wasgiven ethanol extract purwoceng at a dose of 25 mg/300 g bw. The born pups were observed for 70 days tomeasure the growth and development of head bone, back bone, front legs bone, and hind legs. The resultsshowed that the growth and development of all bones parameters of female pups were higher on purwocenggroup than control the bone while male pups did not difference between purwoceng and control group exceptback bone and hind leg bone. The conclusion of this study is that administration of purwoceng roots ethanolextract on pregnant rats during organogenesis phase can extends the bone length of their pups.

Abstract

The aim of this study was to measure antibacterial activity of soursop leaf extract (Annona muricata L.) on bacterial growth inhibition of Pseudomonas aeruginosa by diffuse disc method. The result of the inhibition zone was analyzed using Analysis of Variance and followed using Duncan test, to show highly significantly different (p<0.01). The inhibition number of negative control (C-) was 0.00e±0.00 and the positive control (C+) was 31.31a±0.57. The result of concentration 1 (T1) was 14.49b±4.96; concentration 2 (T2) 8.98c±1.29; concentration 3 (T3) 5.84cd±1.85; concentration 4 (T4) 5.56cd±1.58; concentration 5 (T5) 2.85de±0.28; concentration 6 (T6) 2.98de±0.53; concentration 7 (T7) 2.82de±1.59; concentration 8 (T8) 2.41de±1.10; concentration 9 (T9) 2.20de±0.34; concentration 10 (T10) 1.10e±0.19; and concentration 11 (T11) 0.00e±0.00. The increase of soursop leaf extract concentration showed high inhibition diameter of bacterial growth. It concluded that soursop leaf extract have an antibacterial activity to inhibit P. aeruginosa growth with Minimal Inhibitory Concentration (MIC) 125 ppm.

Abstract

Bali cattle are known for having high adaptability to any environmental condition and have proven tosurvive in critical environments including landfills. The aim of this study was to determine the relationshipbetween Serum Glutamic Pyruvic Transaminase /Serum Glutamic Oxaloacetic Transaminase (SGPT/SGOT) levels and lead (Pb) level in blood of bali cattle reared in the landfill Denpasar. A total of 22Balicattle were used in this study was. The blood was drawn from the jugular vein and 10 ml blood werecollected using tubes containing EDTA 0.5% and 10 mL without EDTA tubes. Levels of SGPT and SGOTwere measured by using Auto analyzer (Refloton (R) plus) method, while the measurement of Pb in bloodplasma was conducted by using atomic absorption spectrophotometry (AAS) method. Result of the SGPTand SGOT measurement obtained were relatively varied with an average of SGPT = 29.96±5.67 U/L andSGOT = 78.82±17.62.U/L. Similarly, Pb measurement results vary widely between 4.004 to 10.216 withan average of 6.595±1.85 ppm. Correlation analysis showed that there was an association between SGOTwith Pb levels. However, no correlation was observed between the SGPT with Pblevels. These resultsindicate that the levels of lead may cause pathological tissue cells in organs other than liver. Its concludedthat heavy metalPb is found in the blood of bali cattle reared in Denpasar city landfill, and it may becorrelated with the level of the SGOT and may cause pathological tissue cells organs.

Abstract

Staphylococcus aureus is known as a potential agent that cause food poisoning. Chicken meats could be as vehicles of food poisoning cases while they contaminated by agents during slaughtered and meat processing as well as infected chickens by S. aureus itself. The aims of the research were to isolated and characterized of S. aureus from liquid wastes of chicken slaughtered house and washing water of chicken carcass. The characterization of S aureus based on mannitol salt agar (MSA), catalase and coagulation assay, production of hemolysins, hydrophobicity test, and the reaction of hemaglutination. Thirty seven samples used in this study were collected from 16 liquid wastes slaughtered chicken house and 21 samples from washing water of chicken carcasses.The research resulted 18 S. aureus isolated from liquid wastes slaughtered house (4 isolates) and from wahing water of chicken carcasses (14 isolates). All 18 isolates fermented mannitol, catalase and coagulase positive, contain of hydrophilic surfaces. Among 18 S. aureus isolates could agglutinate of rabbit erythrocytes for 55%, produced ?-hemolysis (12%) and ?-hemolysis (88%).

Abstract

The epidemic of avian influenza (AI) in Indonesia initially occurred at the end of 2003 which caused100% death of the affected chickens. It was caused by avian influenza virus (AIV) subtype H5. Recent datashowed that highly pathogenic avian influenza (HPAI)-H5N1 virus is still endemic among bird populationin Indonesia. A study was therefore conducted to find out the distribution of AIV-H5N1 in several regionsin Indonesia. Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the presenceof AI-H5 virus and hemagglutination inhibition (HI) test was used to detect the presence of anti-AIV-H5antibody. Results showed that anti-AIV-H5 antibody was detected in 36 % and was not detected in 64% oftested birds in West Java, Banten and East Java. The AIV-H5 antibody titer varied from low to high titer.The AIV-H5 was detected in samples from Cianjur (30%), Blitar (1.9&), Serang (12.5%) and pandeglang(17.5%). It was evident that AIV-H5 is still endemic in Indonesia.

Abstract

The aim of this research is to find antigenic protein of Aeromonas hidrophyla (S-layer) local isolateswhich can be used as a material diagnostic reagent and to produce monoclonal antibody as a kit diagnosticfor ulcer disease and red sore disease. Aeromonas hydrophila were isolated from gold fish (Cyprinus CarpioLinn) that were got from Balai Budidaya Ikan Punten, Batu, East Java. Identification were done usingbiochemical test and were cultured on specific media called TSA. Protein characterized were analyzedthrough some stages: creating antigen homogenate, determining antigen homogenate concentration, andprotein characterizing using SDS-PAGE. Result showed that Aeromonas hydrophila protein molecularweight 174,2 kDa; 164,4 kDa; 101,4 kDa; 83,8 kDa; 71,4 kDa; 68,4 kDa; 61,7 kDa; 58,1 kDa; 53,7 kDa; 50,1kDa; 46,1 kDa; 41,5 kDa; 39,0 kDa; 35,2 kDa; 28,4 kDa; 22,2 kDa. Aeromonas hidrophylla s-layer proteinmolecular weight 53 kDa and 61 kDa had immunogenic trait. Thus, it can be concluded that the proteincan be developed potentially as a diagnostic reagent.
