Abstract

Tarsius is an endemic species in Indonesia but is is getting threatened. In-situ and ex-situ conservationof this species would yield better results when its genetic information and diversity determined. Theobjective of this ressearch was to study the genetic diversity of ND3 gene of Tarsius sp. Based on sequencingof PCR products using primer ND3F and ND3R resulted in 437 nts base sequence. Results of ND3 fragmentssequencing were put on multiple alignment with other primates from Genbank using of software ClustalW, and then were analyzed using MEGA program version 4.1. A different nucleotide site was found(nucleotide no. 24). The genetic distance based on nucleotide ND3 was calculated using Kimura 2-parametermodel. The genetic distance showed the smallest genetic distance 0%, the biggest 0,03% and average0,01%. The phylogenetic tree using neighbor joining method based on the sequence of nucleotide ND3 genecould not be used to differentiate among T. Dianae (from Central Sulawesi), T. Spectrum (from NorthSulawesi) and T. bancanus (from Lampung, South Sumatera).

Abstract

A study was carried out to determine the serum estradiol and progesterone concentration, vulvaappearance, and vaginal cytology during estrous cycle in bligon goat. Four female non pregnant bligongoats (Indonesian indigenous) were used in this research. They were clinically healthy and showed normalestrous cycle. Estrous state was examined by using biological method. When the animals showed acceptanceto be mounted by a buck, they were confirmed as the first day of estrous cycle (estrous phase). Collectionof blood sample, preparation of vaginal smears, and measurement of some physical genital parameterswere conducted by standard veterinary clinical methods at day 11, day between 3-5, day between 6-16, andday between 17-19 of estrous cycle. Enzyme linked immunosorbent assay (ELISA) was used to determineserum progesterone and estradiol concentrations. Mean difference of each parameter was analyzed byanalyses of variance (Anova), followed by Tuckey HSD. The serum estradiol concentration ranged between211.25-247.77 pg/ml. Peak of the estradiol was occurred at the first day of estrous phase, namely 247.77pg/ml and to decrease at day 3-16. The serum progesterone concentration was at the level of 0.21-0.70 ng/ml and showed a significant increase at day 6-16 (p<0.05). The specific physical genital changes (swellingvulva, reddish mucous membrane of vulva, and excretion of viscous transparent vaginal discharge) wereidentified as the animals were at the estrous phase. Composition of exfoliative vaginal cytology did notshow any specific and consistent changes throughout the estrous cycle. Superficial and intermediate cellswere very dominant (>60%) during estrous cycle. In conclusion, serum estradiol, progesterone, the vulvaappearance combined with sexual receptivity could be a good estrous predictor for female cycling bligongoat.

Abstract

The purpose of this research was to examine the effect of lactose in Tris extender on theviability of Etawah crossbreed goat spermatozoa which is preserved in the plasma using Prianganram seminal plasma at 3–5oC. Semen was collected using artificial vagina once a week. Freshsemen was divided into four tubes, and centrifuged at 3,000 RPM for 30 min. Supernatant wasremoved and replaced which an equal volume with Priangan ram seminal plasma. Semen in firsttube was diluted with Tris extender-20% egg yolk without lactose (control). Semen in second,third, and fourth tubes were diluted with Tris extender-20% egg yolk and 0.3% (0.3 g per 100 mlextender) (L0.3), 0.6% (L0.6), and 1.2% (L1.2) lactose, respectively. Diluted-semen were stored inrefrigerator at 3–5oC. Quality of diluted-semen including percentages of motile spermatozoa (MS),live spermatozoa (LS), and intact plasma membrane (IPM) were evaluated daily during storage at3–5oC for four days. The data were analyzed by usi analysis of variance (ANOVA). Means werecompared significant difference test at 0.05 significant level. Results of this study showed thatmean volume, colour, consistency, pH, mass activity, spermatozoa concentration, MS, LS,spermatozoa abnormal, and IPM of Etawah crossbreed goat fresh semen were 0.68 ml, cream,thick, 7, ++/+++, 4,148.57 million cell/ml, 70%, 83.89%, 7.12%, and 84%, respectively. Addition oflactose in Tris extender could maintain the quality of semen during preservation. At day-5 ofstorage, the percentages of MS, LS, and IPM for treatment L0.6 (40, 55.6, and 53.8%) weresignificantly (P<0.05) higher than treatment L1.2 (36, 50.2, and 51.8%), treatment L0.3 (34, 51.2,and 48.4%), and control (29, 41.4, and 41.8%). In conclusion, addition of 0.6% lactose in Trisextender is the best dose in maintaining the quality of Etawah crossbreed goat spermatozoa, andcould be maintaining percentage of motile spermatozoa 40% for four days after preserved at 3–5oC.

Abstract

The research objective was to find an alternative avian influenza prevention in broilers by increasinganimal’s antibody titer and macrophages phagocytic  activity.  Virgin coconut oil (VCO) is a food supplementthat is proven safe for human consumption therefore it is assumed to be safe for the animal’s (chickens).Factorial design  2 vaccinated: unvaccinated) x 4 (dose of VCO: 0, 5, 10 and 15 mL/kg feed) were applied inthis study.  A total of 40 day day old chick were allocated in the eight treatments groups.  Feed and drinkingwater were available  ad libitum.  Antibody titers of the animals were detected using ELISA, whereasphagocytic activity of the macrophages were detected from spleen.  The result showed that the highestphagocytic activity and antibody titers were seen in chickens which were given VCO at 10 mL/kg feed.  It isconcluded that the VCO could increased the phagocytic activity of macrophages.

Abstract

The use of random amplified polymorphic DNA (RAPD) as a method to identify a genetic markerof bacteria is widely used by researcher. This method is known as a simple, faster, and reliabletechnicque. This study is to find out the aplication of RAPD method in order to identify specific markersof E. coli O157:H7 as a zoonotic agent. The study began by cultivating of 20 isolates of E. coli O157:H7colected by previous study that consist of 2 isolates originated from cattle feces, 2 isolates originatedfrom beef, 2 isolates originated from chicken feces, 2 isolates originated from healthy human and 11isolates originated from unhealthy human (human with kidney failure). All isolates were confirmed byculturing on selective medium sorbitol MacConkey agar (SMAC). Confirmation were followed by testingon O157 latex aglutination, and finally by testing on H7 antiserum. RAPD method as molecularanalysis was performed using decamer primers mixture OPA-01, OPA-02, OPA-03, and OPA-04.Results of study showed both bands 1721 and 700 bp are specifically to differentiate of isolatesoriginated from cases of healthy and unhealthy human. On the other hand, bands with position 1721 bp,300 bp, and 250 bp indicate the isolates originated from unhealthy human, healthy human and chicken,respectively. Isolates from beef are characterized by both bands 1400 and 429 bp, and the isolates fromcattle feces are identified by band with position 342 bp. The specific bands are considered as markers inorder to know the source of E. coli O157:H7 fastly.

Abstract

Tujuan penelitian ini adalah untuk mengevaluasi kualitas semen cair domba garut yang diencerkan dengan larutan pengencer tris dengan penambahan berbagai konsentrasi beta-karoten. Semen dikoleksi dengan vagina buatan satu kali dalam satu minggu dari empat pejantan domba garut dewasa kelamin. Abstract

The administration of Pregnant Mare Serum Gonadotrophin (PMSG) hormone to stimulate superovulation has been proven to improved fetal prenatal growth, birth weight, mammary gland growth anddevelopment, milk production, litter size, as well as pre and post weaning growth. This study was conductedto apply the administration of PMSG hormone technology in small-holder farms. One hundred ewes weredivided into two groups. Group I (control: 50 ewes) ewes without administration of PMSG and Group 2(treatment: 50 ewes) ewes with administration of PMSG to stimulate super ovulation and improveendogenous secretion of pregnant hormones. The application of PMSG increased the numbers of lamb bornby 26.8% (69 vs. 87) with average litter size of 20.7% (1.38 vs. 1.74). The lamb weight at birth was alsoincreased by 25.7% (2.76 ± 0.33 vs. 3.47 ± 0.35 kg). The milk production of the dam was also increased by29.7% (0.64 ± 0.25 vs. 0.91 ± 0.22 L/ewe/day). Lambs born to ewes administered PMSG had better weaningweight 22.1% (12.32 ± 2.44 vs. 15.04 ± 1.11 kg/ewe). The number of survival lamb to reach weaning age washigher compared to the control group. The weight at weaning age was almost twice (628.4 kg vs. 1202.8 kg)compared to the control group. Using simple economic analysis calculation, the application of this hormonetechnology could increase gross revenue by Rp 294.280- per ewes. It was concluded that the administrationof PMSG is economically feasible to be applied in to the small-holder farms.

Abstract

Mycobacterium avium subspesies paratuberculosis (MAP) is a thermal tolerant bacteria. The presenceof these bacteria in pasteurized dairy milk is associated with infectious bowel disease in human known asCrohn’s disease. The aim of this study was to detect MAP in pasteurized dairy milk sold in Bogor. Fourtytwo samples of plain flavoured milk (180–250 ml) from 7 producers were bought from supermarkets inBogor. The presence of MAP was detected by isolation and conventional polymerase chain reaction (PCR)using IS 900 and F57. Bacterial isolation were done by Herrold’s egg yolk medium with mycobactine J(HEYMj) and without mycobactin J (HEYM) and incubated at 37°C for 20 weeks. The DNA extraction ofall pasteurized dairy milk samples were conducted by DNeasy® Tissue Kit. Amplification conditionsfor PCR were: 1 cycle at 94°C for 10 minutes, 40 cycles at 94°C for 1 minute, 58°C for 1 minute, and72°C for 3 minutes, and 1 cycle at 72°C for 7 minutes. After 20 weeks of incubation, there were no sign ofMAP which grew in all isolation mediums. The PCR IS 900 and F57 did not detect the DNA band of thetarget. In the conclusion, there was no MAP detected in pasteurized dairy milk sold in Bogor.

Abstract

The objective of this study was to examine the effectiveness of fetal bovine serum (FBS) against thequality of garut ram frozen semen. Semen was collected from one mature garut ram using artificial vagina.Fresh semen were evaluated then divided into four tubes at equal volume and each tube were diluted withTris extender containing 20% egg yolk (TEY-20, as control); TEY-20 + 8% FBS (FBS-8); TEY-20 + 10% FBS(FBS-10); and TEY-20 + 12% FBS (FBS-12), respectively. Semen at the concentration of 100x106 motilespermatozoa was loaded in 0.25 ml mini straw. Semen was equilibrated at 50C for three hours, then freezeand stored in liquid nitrogen container. The quality of the spermatozoa including percentages of motileand live spermatozoa, intact plasma membrane (IPM) were evaluated following diluting, equilibratingand thawing process. A Complete Randomized design using four treatments and five replicates were usedin the study. The results showed that there was no significant difference (p<0.05) in percentage of motilespermatozoa following thawing between the control (44.0%) and FBS-8 (46.0%), FBS-10 (48.0%), andFBS-12 (47.0%), respectively. The percentage of live spermatozoa and IPM were significantly higher (p<0.05)in the FBS-8 (69.0% and 58.2%); FBS-10 (72.4% and 61.2%); FBS-12 (72.2% and 64.4%) compared to thecontrol (64.8% and 52.8%), respectively. In conclusion, the addition of FBS into Tris extender was able tomaintain the viability and integrity of plasma membrane of garut ram frozen semen.
