Abstract

The purpose of this study was to observe the effect of extender and cooling rate on the quality of frozen thawed semen of Bali bull (Bos sondaicus). The experiment was conducted at Laboratory of Animal Biotechnology, Faculty of Animal Husbandry, Andalas University. A completely randomized design of factorial 2 x 3, 2 different extenders and 3 cooling rates with 3 bulls as the replicate. Data were analyzed using analysis of variance (ANOVA). The first factors was extender such as tris citrate egg yolk (TCEY) and tris citrate soy milk (TCSM). The second factors was the cooling rate such as: 15ºC/min, 10ºC/min and 5ºC/min. Variables were: motility, viability, abnormality, and membrane integrity. The results of treatment of TCEY extender on the average of motility 42.67±5.17%, viability of 53.78±3.79%, abnormality of 18.89±1.07%, and membrane integrity of 35.44±3.01%. The effect of cooling rate of 10°C /min has the highest semen motility of 44.35±5.28%, viability of 57.17±1.18%, abnormality of 17.84 ± 0.23 % and membrane integrity of 36.83±2.12%. The interaction between extender and cooling rate was founded significant different (P<0.05). Otherwise of extender and cooling rate on post-thawed of motility, viability and membrane integrity, had no difference (P>0.05) on semen abnormality. It can be concluded that tris-citrate egg yolk (TCEY) extender with cooling rate of 10ºC / min were the best semen motility, viability, and membrane integrity of spermatozoa.

Abstract

The application of cryopreservation to preserve germplasm in such specific breed requires preliminary studies, primarily related to the resistance of spermatozoa to low temperatures (4 – 5 °C) as measured by their motility and longevity. In this study, semen taken from five phenotypes of Kokok Balengeek Chicken (KBC) (Biriang, Jalak, Kinantan, Kuriak, and Taduang) was used to evaluate the effect of Ringer’s Lactate-egg yolk diluent on longevity and motility of spermatozoa. The treatments consisted of Ringer’s Lactate (RL) solution added with egg yolk at a concentration of 1% (RLKT1), 3% (RLKT3), and 5% (RLKT5). Evaluation of fresh semen showed that the spermatozoa of Jalak had the highest motility, namely 75.63 ± 0.5% (P<0.05). Post-dilution longevity and motility observations were carried out at 0, 24, 48, and 72 h, significantly decreasing each time (P<0.05). The lowest range of reduction was found in Jalak spermatozoa diluted with RL with longevity of 7.75 ± 0.70 days. Overall, the RL diluent showed the highest motility after 24 h, namely 41.13 ± 2.27%. Adding egg yolks to Ringer's Lactate solution could not maintain the motility of KBC spermatozoa when stored at 4-5°C for 48-72 h.

Abstract

Pesisir bulls from West Sumatra, Indonesia, have the potential to be valuable meat producers due to their adaptability and unique characteristics, but reproductive challenges necessitate the use of artificial insemination (AI) technology. The success of AI relies on the quality of frozen semen, which is influenced by thawing conditions, making the evaluation of sperm kinematics through Computer-Assisted Sperm Analysis (CASA) crucial for a comprehensive assessment of sperm function. This research evaluated the kinematics of frozen semen spermatozoa from Pesisir bulls. Frozen semen samples (n = 20) were thawed at 37°C for 10, 20, and 30 s and at 25°C for 30, 45, and 60 s. Kinematic parameters were observed using a Computer-Assisted Sperm Analyzer (CASA), including total motility (M), progressive motility (PM), the velocity of the average path (VAP) (?m/sec), the velocity of curvilinear (VCL) (?m/sec), the velocity of straight line (VSL) (?m/sec), straightness (STR) (%), linearity (LIN) (%), wobble (WOB) (%), amplitude of lateral head displacement (ALH) (?m), and beat/cross-frequency (BCF) (Hz). Thawed frozen semen at a temperature of 37°C for 20 s significantly (p<0.05) increased M, PM, and ALH. Furthermore, thawed frozen semen at 37°C for all durations and at 25°C for 60 s showed better quality for VAP, VCL, and VSL (p<0.05). Meanwhile, thawed frozen semen at 25°C increased STR, LIN, and WOB. However, thawing temperature and duration had no significant effect on BCF. Correlation analysis conducted on semen thawed at a temperature of 37°C for 20 s found that VAP was correlated with VCL, VSL, and ALH, while semen thawed at a temperature of 25°C for 60 s found that VAP was correlated with VCL, VSL, STR, LIN, and ALH. Most kinematic parameters were significantly better in the thawing treatment at 37°C compared to 25°C, suggesting careful consideration
