Abstract

Background: Musa paradisiaca has  been widely studied for its antihyperglycemic effects, showing potential as an alternative natural remedy for diabetes. However, variability in genetic factors, environmental conditions, and geographical origins may influence its bioactive compound composition and efficacy. Thus, further validation through controlled studies is required to ensure consistent therapeutic effects.Purpose: : This study aimed to evaluate the antihyperglycemic effect of Musa paradisiaca stem extract in vivo, providing additional validation for its therapeutic potential and determining the optimal concentration for clinical relevance.Method: An experimental posttest-only control group design was used. Thirty-five male Balb/c strain mice were induced into hyperglycemia using alloxan. They were then randomly divided into treatment groups receiving different concentrations of Musa paradisiaca stem extract (5%, 15%, 25%, 35%, and 45%), a positive control group (metformin), and a negative control group. Blood glucose levels were measured and analyzed statistically using One-Way ANOVA.Result: Administration of the stem extract at 15% concentration showed the most significant antihyperglycemic effect, with an average blood glucose level reduction to 63.8 mg/dL, comparable to the positive control group (metformin). Conversely, the highest concentration tested (45%) exhibited minimal effectiveness, with an average glucose level of 151.8 mg/dL, indicating reduced antihyperglycemic activity. Conclusion: The optimal antihyperglycemic effect of Musa paradisiaca stem extract was achieved at a concentration of 15%, demonstrating therapeutic efficacy comparable to metformin. This extract shows promise as a natural alternative agent in diabetes treatment. Further studies, including clinical trials, are recommended to confirm these findings.

Abstract

Kata Kunci: Clitoria ternatea, Isotoma longinflora, S. aureus, Fusarium sp. Conjunctival infections are often caused by various pathogenic microorganisms, including Staphylococcus aureus and Fusarium sp. Isotoma longiflora and Clitoria ternatea are known to contain secondary metabolites with potential as antimicrobial agents. This study aimed to test the antimicrobial effects of a combined extract of Isotoma longiflora and Clitoria ternatea against Staphylococcus aureus and Fusarium sp., which can potentially cause conjunctival infections, in vitro. The combined extract of the two plants was formulated in a 1:1 ratio, mixing 50% Isotoma longiflora extract and 50% Clitoria ternatea. This combination was tested at concentrations of 50%, 60%, 70%, 80%, and 90%, with an observation period of 72 hours. Statistical testing using One-Way ANOVA showed a significant effect of the combined extract on the growth of both microorganisms (p=0.001; p<0.05). The combined extract of Isotoma longiflora and Clitoria ternatea exhibited a higher inhibitory effect against Staphylococcus aureus than Fusarium sp., especially at the 50% concentration. This effect is thought to be influenced by compounds such as flavonoids, tannins, and saponins that can damage bacterial cell walls and membranes. Conversely, the inhibition effect on Fusarium sp. was relatively lower, possibly due to the more complex structure of fungal cell walls. In conclusion, the combined extract of Isotoma longiflora and Clitoria ternatea demonstrates a stronger antibacterial effect against Gram-positive bacteria like Staphylococcus aureus compared to Fusarium sp., indicating a need for further research to validate these findings.

Abstract

Postoperative infections, especially after childbirth, remain a serious medical issue as they can lead to complications, delay recovery, and increase antibiotic resistance due to the excessive use of synthetic drugs. Therefore, safer and more effective treatment alternatives, such as the use of natural substances, are needed. This study aims to examine the antimicrobial potential of a combination of Ageratum conyzoides, Mussaenda frondosa, and Acorus calamus extracts in inhibiting the growth of Staphylococcus aureus and Candida albicans, which are common pathogens in postnatal surgical wound infections. The study employed a true experimental method with an antimicrobial sensitivity test design using the maceration technique. Extraction was conducted using 96% ethanol with three concentration variations: 40%, 80%, and 120%, in a 3:2:1 mixture ratio. Phytochemical screening was carried out to identify secondary metabolite compounds, followed by an inhibition test using the disc diffusion method. The results showed that Acorus calamus extract contains flavonoids, alkaloids, and saponins. Ageratum conyzoides, Mussaenda frondosa, and Acorus calamus all contain saponins with potential antimicrobial activity. The combination of these extracts demonstrated inhibitory effects against the growth of pathogenic microorganisms in surgical wounds, as evidenced by the formation of inhibition zones during testing.
