Abstract

This social project aims to enhance students' knowledge and awareness of the importance of environmental sanitation and healthy lifestyles. Additionally, it seeks to modify their behavior by implementing sanitation and healthy living practices in their daily routines, such as regular handwashing, cleaning the classroom, and disposing of waste properly. The community service was carried out at MI Annidham, Sumbersari District, Jember Regency, with a total of 44 students from grades 3 and 4. The method employed was a conventional interactive approach, involving presentations, practice sessions, Q&A, and discussions during the implementation. The results of the community service indicate a 60% increase in students' knowledge and understanding of sanitation, hygiene, and clean and healthy lifestyle practices, which were the main topics addressed in the service activities. Furthermore, students' participation in this activity demonstrated high enthusiasm and a strong eagerness to learn, positively impacting their knowledge and skills.

Abstract

This social project focuses on environmental conservation efforts through a waste recycling program aimed at SDN Panti 2 students. The main objective of this project is to increase students' awareness and understanding of the importance of keeping the environment clean and teach them practical ways to recycle waste. This program consists of a series of educational activities that begin with counseling about the types of waste, the negative impact of waste on the environment, as well as recycling techniques and benefits. After that, students are invited to participate in various creative activities using used goods. The methodology used in this project is with conventional methods, namely presentations, discussions, games, and practicums. The results of this project show a significant increase in students' knowledge and awareness of the importance of waste recycling. The success of this program cannot be separated from the full support of the school and also the contribution of SDN Panti 2 students. And this project is a form of introducing the concept of entrepreneurship to students from the results of used goods. The main purpose of this project is to increase the value of students' creativity in making useful goods from recycled materials and introduce the economic potential of recycled products. It is hoped that students can develop positive habits that have a long-term impact on environmental preservation and entrepreneurial skills.

Abstract

The majority of residents in Suling Wetan Village work as farmers and rely heavily on self-produced seeds due to the village’s remote location. One high-potential crop for marginal lands in the area is patchouli (Pogostemon cablin), a plant valued for its essential oil. However, the propagation of patchouli remains a significant challenge for local farmers due to limited knowledge and skills. This community engagement program aimed to enhance farmers’ capacities in patchouli propagation through a combination of awareness campaigns, technical workshops, hands-on training, direct field mentoring, and participatory monitoring and evaluation. Evaluation was conducted through direct observation and participant surveys. The results showed that 96% of participants gained a clear understanding of patchouli propagation techniques, and all participants acknowledged the importance of the program and expressed hope for its continuation. The activity successfully contributed to increasing farmers’ independence in sustainable patchouli seedling production.

Abstract

Nutritional status is an important factor for the health of an individual, community, and the wider community. In Indonesia, the problem of nutrition is still very high. This occurs due to a lack of interest in exploring information about healthy living and balanced nutrition so that meeting daily nutritional needs is not optimal. Based on data from the Ministry of Health, the stunting rate in Indonesia in 2023 was recorded at 21.5 percent, down only 0.1 percent from the previous year which was 21.6 percent. With these problems, we carry out counseling activities with lecture methods and practical assistance to increase the knowledge and awareness of elementary school students, precisely at SD Negeri Badean 3. The material provided regarding healthy living behavior by applying the Balanced Nutrition guidelines includes: The results obtained in this activity are that SD Negeri 3 Badean students are able to understand the balanced nutrition guidelines, Clean and Healthy Living Behavior (PHBS). With this social project, students can already implement healthy living behaviors or also known as a healthy lifestyle in their daily lives.

Abstract

Children's ability to eat healthy and nutritious food is closely related to education. Education is an inseparable process as an effort to grow and develop the potential in every human being. Especially in children, education plays an important role in forming a healthy diet and good hygiene behavior. However, in several studies in Indonesia, it is not uncommon to find that there is still a lack of knowledge about healthy and nutritious food and good hand washing among elementary school students which harms their health. As in SDN Gadingsari, Binakal District, there are still many elementary school students who have a low understanding of healthy and nutritious food. The author's purpose in carrying out this social project is to socialize, provide education and understanding directly to SDN Gadingsari students about the importance of eating healthy and nutritious food and good hygiene behavior through interactive learning such as showing videos as a result of eating nutritious healthy food, washing hands, providing material on the concept of the contents of my plate, the content in nutritious food such as carbohydrates, protein, fat, fiber, vitamins, and minerals, games to gymnastics together. The results showed that the students were very enthusiastic about learning about healthy and nutritious food. It is hoped that students can apply it in their daily lives.

Abstract

Rice plants (Oryza sativa) are among the most significant food crops in human life. Efforts to increase food production face a variety of challenges, including droughts. Drought stress is abiotic stress that has a significant impact on rice plant growth and development. Plants treated with salicylic acid are more resistant to water shortage. Salicylic acid influences plant growth in terms of weight, height, and grain yield. The use of salicylic acid may increase the weight of plant seedlings. The purpose of this study was to determine how rice plant productivity responds to the addition of salicylic acid during drought stress. The study was carried out in a greenhouse from August to December 2020 utilizing a Completely Randomized Design with two variables: plant variety and drought stress. The cultivars used were Indragiri, Mendawak, and IR64. The drought treatment factors were control, drought stress, and drought stress treated with salicylic acid. There were nine treatment combinations and three replications, totaling 27 experimental units. The data collected were analyzed using Analysis of Variance. If significant differences were discovered, the analysis was extended with Duncan's Multiple Range Test at a 95% confidence level. The findings revealed that both individual factors and their interactions had a substantial influence on various variables. The interaction factor had a substantial impact on pollen fertility, panicle length, number of full grains, weight per 1,000 grains, and yield potential. The rice variety had a substantial effect on pollen fertility, panicle length, number of filled grains, and yield potential, but not on the weight of 1000-grains. Abstract

Golden Rice (Oryza sativa L.) is biofortified rice that contains β-carotene, an essential nutrient for human health. Golden Rice PAC Nagdong/IR36/IR64 was created by crossing PAC Nagdong/IR36 with IR64 rice, a native variety, to improve tolerance to Indonesia's environment. Observations were made in two stages: (a) agromorphological characterization during culture, which included plant height, tiller number, panicle length, fertility, yield potential, and grain color, and (b) physicochemical analysis to assess amylose, amylopectin, protein, and lipid levels.  Golden Rice  has semi-dwarf height, a good and very high number of tillers (22.60−24.40), a panicle length of 20.09−20.85 cm, grain weight per pot of 42.08−42.94 g, fertility of 67.23−81.54%, and seed color of 5Y 8/8−5Y 8/10. The physicochemical properties included amylose content of 12.41−13.37%, protein content of 4.61−5.45%, lipid content of 1.24−1.54%, swelling power of 6.72−8.08%, low gelatinization temperature, and soft gel consistency type. The results indicated Golden Rice  line 302/IR-2-2(7)/2 exhibit superior characteristics, including lower amylose and protein contents, higher fertility, and soft gel consistency which identical with consumer-preferred rice quality in Indonesia. Accordingly, this line have the highest potential for commercialization, while the remaining lines depict as promising materials for further breeding improvement. Abstract

Tanaman padi dapat dikelompokkan menjadi 2 jenis, yaitu padi pigmented dan non pigmented. Salah satu contoh padi berpigmen yaitu padi beras merah dan padi beras hitam. Padi berpigmen dikenal sebagai sumber senyawa antioksidan termasuk flavonoid, antosianin, dan senyawa fenolik. Penelitian ini bertujuan untuk menganalisis segregasi hasil persilangan tanaman padi varietas Ketan Hitam dengan Ciherang melalui analisis karakter morfologi dan genotip bioaktif menggunakan marka molekuler SSR. Metode penelitian yang dilakukan yaitu dengan persiapan dan pemeliharaan tanaman (persiapan bahan tanam, pengolahan tanah, penanaman, pemupukan, pengairan, penyiangan, dan pemanenan) dan analisis genotip bioaktif (ekstraksi DNA, proses kerja PCR, elektroforesis dan pengamatan UV Transluminator). Analisis PCR menggunaan marka SSR dengan primer RM 346, RM 316, RM 228, dan RM 339. Analisis data yang digunakan adalah analisis distribusi frekuensi kuantitatif dengan panduan IRRI. Segragasi karakter morfologi pada populasi F2 hasil persilangan antara Ketan Hitam dan Ciherang masih tinggi, dimana dapat dilihat dari pola segragasi karakter pada tinggi tanaman, jumlah anakan, anakan produktif dan berat 1000 bijinya yang dikendalikan oleh banyak gen dan dipengaruhi oleh faktor lingkungan di dalamnya karena tidak memenuhi nisbah genetika mendel ataupun penyimpangannya. Bentuk morfologi hasil persilangannya lebih cenderung mirip dengan tetua betina atau Ketan Hitam. Sedangkan untuk hasil PCR dengan menggunakan marka SSR menunjukkan bahwa RM 346, RM 339 dan RM 228 dapat memvalidasi keseluruhan sampel yang digunakan baik tetua persilangan maupun F2 hasil persilangannya, sedangkan untuk RM 316 tidak dapat memvalidasi keseluruhan pita-pita DNA pada sampel penelitian.

Abstract

Banana (Musa sp.) is an important crop that is widely cultivated and consumed in Indonesia. Banana plants production can be increased through tissue culture with the use of explants and the right combination of hormones. This study was conducted to determine the use of flower apices explants on local banana varieties using a combination of cytokinin hormones containing BAP (Benzylaminopurine) and several levels of TDZ (Thidiazuron). The result of this research was analyzed using a one-way analysis of variance. The hormone combination in this study consists of MS + BAP 2 mg/L + three concentrations of TDZ (2 mg/L, 2.5 mg/L, and 3 mg/L) as a regeneration medium. Three local banana varieties (Kepok Putih, Kepok Kuning, and Pisang Susu) was cultured on the three combination medium. The results showed that the use of flower apices explants contained in banana flowers grown on MS media with a combination of cytokinin hormones could increase the efficiency of explant regeneration. Media MS + BAP 2 mg/L + TDZ 3 mg/L gave the best regeneration rate for Kepok Putih (62.50%), Kepok Kuning (62.50%) and Banana Susu (56.25%) compared to TDZ 2. mg/L and TDZ 3 mg/L. The novelty of this study can provide information in the propagation of local banana plants using flower apices explants.

Abstract

The success rate of explant morphogenesis in plant breeding using tissue culture techniques is frequently plagued by browning due to the oxidation of phenolic compounds. The cumulated amount of reactive oxygen species (ROS) drives the oxidation of phenolic compounds. Melatonin is reported to take a part in modulating the regulation of antioxidant gene expression, reducing the accumulation of reactive oxygen species, and enhancing the efficacy of tissue culture. This study aims to determine the optimal melatonin concentration on the efficiency of plantlet regeneration and expression of the antioxidant resistance gene in rice callus. This study utilizes rice TN1, Gogo Niti II, Ketan Hitam, and Cigeulis cultivars. Melatonin at 0, 10, and 15 µM concentrations is supplemented in plantlet regeneration media. Rice antioxidant-related genes, Mn-SOD, Cu/ZnSOD, Cytosolic APX, CAT, GPOD, OsAPX, and OsCATA, expressed after melatonin supplementation. Melatonin concentration at 10 µM generates the highest expression of all tested genes in TN1 compared to other varieties. The cumulated amount of hydrogen peroxide (H2O2) shows that Melatonin has the potential to increase the proportion of plant regeneration in Cigeulis (90.48%) and Ketan Hitam (91.67%) varieties with a concentration of 10 µM and in TN1 (94.44%) and Gogo Niti II (80%) at a concentration of 15 µM.
