Abstract

Cekaman genangan menyebabkan penurunan hasil 15-45% pada tanaman tebu. Penelitian ini bertujuan untuk mengetahui karakteristik morfologi, fisiologi, dan molekuler tanaman tebu toleran terhadap cekaman genangan. Percobaan dilaksanakan di Laboratorium Program Studi Agroteknologi, Fakultas Pertanian, Universitas Jember, Jawa Timur pada bulan September 2021-Februari 2022. Rancangan percobaan menggunakan rancangan acak kelompok (RAK) faktorial. Faktor pertama yaitu genotipe tebu dengan enam genotipe (Bululawang, PS 862, Cening, PS 881, M3, M4). Faktor kedua yaitu periode penggenangan dengan tiga taraf (30 hari, 60 hari, dan 90 hari). Faktor ketiga yaitu perlakuan cekaman genangan (tanpa cekaman genangan dan dengan cekaman genangan). Variabel pengamatan meliputi tinggi tanaman, luas permukaan daun, diameter batang, bobot segar akar, jaringan aerenkim, klorofil total, kerapatan stomata, kandungan H2O2 dan ekspresi gen antioksidan. Hasil penelitian menunjukkan bahwa cekaman genangan berdampak pada pembentukan jaringan aerenkim, penurunan bobot segar akar, perubahan tinggi tanaman, penurunan luas area daun, penurunan klorofil total, penurunan kerapatan stomata, peningkatan kandungan hidrogen peroksida dan ekspresi gen antioksidan. Genotipe Cening menunjukkan respon persentase penurunan yang rendah dibandingkan dengan kontrol tanpa cekaman pada variabel morfologi, fisiologis dan ekspresi gen antioksidan yang lebih tinggi. Abstract

Golden rice merupakan padi yang di dalam endosperma bijinya memiliki kandungan β-karoten atau provitamin A. Beberapa upaya telah dilakukan untuk meningkatkan ketahanannya terhadap penyakit blas dan hawar daun bakteri (HDB). Hasil pemuliaan Golden rice telah mendapatkan turunan ke 8 (F8) yang bersifat stabil. Penelitian ini bertujuan untuk mengkarakterisasi agromorfologi dan genotipe galur padi introduksi Golden rice F8 PAC Nagdong/IR36/IR64. Penelitian dilakukan di Laboratorium Agroteknologi, Fakultas Pertanian, Universitas Jember. Benih yang digunakan yaitu tiga galur padi intorduksi Golden rice F8 PAC Nagdong/IR36/IR64 dengan kode 302/IR-2-2(6)/1, 302/IR-2-2(7)/2, 302/IR-2-2(8)/1 serta varietas IR36 dan IR64 sebagai tanaman kontrol. Analisis genotipe meliputi gen Pik-s, Pi54, Pi-ta, Xa4 dan Xa7. Hasil penelitian menunjukkan bahwa karakter agromorfologi pada seluruh galur Golden rice untuk karakter tinggi tanaman, jumlah anakan dan jumlah anakan produktif tidak berbeda nyata. Tinggi tanaman Golden rice termasuk dalam kategori semidwarf, jumlah anakan >24.6 dengan anakan produktif mencapai lebih dari 85%, panjang malai antara 20.09-20.85 cm, dan bobot per rumpun mencapai 42.94 g serta seluruh galur Golden rice memiliki gen pengkode sifat tahan terhadap penyakit blas dan HDB. Abstract

Buah okra kaya akan manfaat yang berguna memenuhi nutrisi yang dibutuhkan oleh tubuh. Melatonin merupakan biomolekul pleiotropik alami yang ditemukan secara luas pada tumbuhan maupun hewan untuk meningkatkan hasil, pembungaan, pembentukan dan perkembangan buah, partenokarpi, dan ripening. Penelitian bertujuan mengetahui respon melatonin eksogen terhadap performa dan perkembangan organ generatif tanaman okra. Penelitian dilakukan pada Desember 2021-Februari 2022 di lahan okra PT. Gading Mas Indonesia Teguh, Ajung, Jember, Indonesia. Pemberian melatonin dilakukan secara eksogen dengan menyemprotkan ke semua bagian tanaman okra yang sudah masuk fase generatif. Percobaan dilakukan dengan satu faktor konsentrasi melatonin dengan empat taraf yaitu 0 µM, 200 µM, 350 µM, dan 500 µM. Kombinasi perlakuan dilakukan pengulangan sebanyak lima kali sehingga didapatkan 20 satuan percobaan. Hasil penelitian menunjukkan melatonin berpengaruh terhadap performa dan kualitas tanaman okra. Konsentrasi 350 µM menghasilkan rata-rata jumlah daun tertinggi 14.8 daun. Konsentrasi 500 µM menghasilkan waktu gugur kelopak paling lama dengan rata-rata 4.8 hari dan persentase keberhasilan pembentukan buah sebesar 100 %. Variabel jumlah biji paling tinggi pada perlakuan kontrol dengan rata-rata 57.2 biji dan pematangan buah atau ripening paling lama pada konsentrasi 500 µM dengan rata-rata 33.6 hari. Abstract

Seeds of tobacco plants are unable to germinate immediately after harvest because they have a dormancy period. The seed dormancy period may be shortened by bagging methods to affect the hormone content in the seeds. The bagging flower makes the dark conditions in the process of flowering and seed development, affecting the content of the hormone’s auxin and ABA. This study aimed to determine the effects of different bag colors on the dormancy period of tobacco seeds as evidenced by their vigor and viability. The research was conducted from March to August 2022 at PT. Dwi Light Tobacco Green House, Lombok Wetan, Bondowoso, Indonesia. Four crepe colors of paper were used as bagging treatments (black, red, yellow, blue) and control (without bagging) and replicated four times. The results showed that the bagging treatment significantly affected one hundred-seed weight, seed germination, seed growth rate, and seedling height. Black crepe paper resulted in the highest seed germination percentage (98.5%) and seed growth rate (11.68). Keywords: bag color, seed germination, viability, vigor

Abstract

Somatic embryogenesis is a propagation technique in which explants form somatic embryos. The percentage of primary somatic embryo formation in cocoa tends to be low due to the significant secretion of phenolic components in flower explants, thereby reducing the conversion rate of explants into embryos. This is indicated by browning that occurs in explants or the culture medium. The purpose of this study was to optimize primary somatic embryogenesis induction in cocoa through the addition of several types of phenolic compound suppressor materials to the culture medium. Staminodia explants were grownon DKW medium with factorial treatment: various phenolic compound suppressing materials (control; 300 mg L-1PVP; 15 mg L-1silver nitrate; 12 mg L-1melatonin; 300 mg L-1ascorbic acid) and cocoa cultivars (MCC-02 and ICCRI-09). The results showed that each material treatment produced a different response in suppressing phenolic compounds in each cultivar. The melatonin treatment resulted in the lowest percentage of browning intensity, which also had the best impact on morphology, callus size, wet weight,and the percentage of live explants.

Abstract

Red rice contains high anthocyanin and bioactive antioxidant compounds that prevent free radical reactions. Cempo Salamet has potential as an antioxidant source, and the characteristics are red colored grains, 4–5 months old, 169 cm plant height, 7 productive tillers per plant, and resistance to blast disease. IR64 had been developed with the following characteristics: 3 months old, 85 cm plant height, 20–35 productive tillers per plant, resistance to brown leafhoppers pigment. This study aimed to obtain information on the segregation of the F3 population from crosses between the Cempo Salamet and IR64 varieties. Research methods included preparation and maintenance with genotype analysis. PCR analysis was conducted using SSR markers with primer RM346, RM316, RM228, and RM339. The segregation in F3 plants was 50% for >130 cm plant height, 51% for 10–19 tillers per plant, 67% for 2.2 g/100-grain weight, and 33% strong red for colour intensity. The findings demonstrated that SSR markers RM346, RM339, and RM228 could validate Cempo Salamet, IR64, and F3 DNA bands. However, RM316 could not validate all DNA bands in the research sample.

Abstract

Banana (Musa sp.) is an important crop that is widely cultivated and consumed in Indonesia. Banana plants production can be increased through tissue culture with the use of explants and the right combination of hormones. This study was conducted to determine the use of flower apices explants on local banana varieties using a combination of cytokinin hormones containing BAP (Benzylaminopurine) and several levels of TDZ (Thidiazuron). The result of this research was analyzed using a one-way analysis of variance. The hormone combination in this study consists of MS + BAP 2 mg/L + three concentrations of TDZ (2 mg/L, 2.5 mg/L, and 3 mg/L) as a regeneration medium. Three local banana varieties (Kepok Putih, Kepok Kuning, and Pisang Susu) was cultured on the three combination medium. The results showed that the use of flower apices explants contained in banana flowers grown on MS media with a combination of cytokinin hormones could increase the efficiency of explant regeneration. Media MS + BAP 2 mg/L + TDZ 3 mg/L gave the best regeneration rate for Kepok Putih (62.50%), Kepok Kuning (62.50%) and Banana Susu (56.25%) compared to TDZ 2. mg/L and TDZ 3 mg/L. The novelty of this study can provide information in the propagation of local banana plants using flower apices explants.

Abstract

Kandungan amilosa dalam padi menjadi hal yang mempengaruhi rasa dan tekstur nasi. Kebutuhan padi rendah amilosa didapatkan dengan mengimpor besar-besaran untuk memenuhi kebutuhan masyarakat. Fakultas Pertanian Universitas Jember telah memiliki galur pemulyaan tanaman Japonica dengan varietas padi lokal yaitu IR64 hingga keturunan stabil, namun belum memiliki beberapa analisis agromorfologi dan fisikokimianya. Karakter Agromorfologi meliputi tinggi tanaman, jumlah anakan, jumlah biji permalai, bobot 100 butir dan Fisikokimia melliputi kadar amilosa, swelling power, gel consistency serta suhu gelatinisasi dilakukan di Laboratorium Agroteknologi Universitas Jember. Hasil penelitian menunjukkan keseluruhan variabel pengamatan berbeda sangat nyata namun tinggi tanaman berbeda nyata dan gel consistency tidak berbeda nyata. Tinggi tanaman memiliki skala yang sama dengan tetuanya yaitu semi dwarf, jumlah anakan memiliki skala medium dan good, jumlah biji permalai 103-108 biji, bobot 100 butir sebesar 2,877-3,265 gram. Masing – masing galur memiliki kandungan amilosa rendah antara 13,87%-15,85%, swelling power di antara 6,61%-8,93%, memiliki karakter soft gel consistency, dan memiliki suhu gelatinisasi yang rendah dibawah 60oC. Abstract

Abstrak: Permasalahan utama peternak sapi di Desa Mayangan adalah ketergantungan pada jerami padi segar bernutrisi rendah (protein 3,8%) dan biaya pakan konsentrat tinggi. Program ini bertujuan meningkatkan kapasitas peternak memproduksi pakan fermentasi, mengurangi biaya pakan, dan mendorong kemandirian teknologi. Keterampilan yang ditingkatkan mencakup hardskill (pengoperasian mesin Straw Baller, pengepresan, pengemasan kedap udara, fermentasi anaerob) dan softskill (kerja sama, problem solving, pengelolaan sumber daya pakan). Kegiatan berupa penyuluhan, pelatihan, dan praktik pembuatan pakan fermentasi diikuti 27 peternak Kelompok Tani Muneng Makmur 2. Evaluasi melalui observasi dan angket 10 pertanyaan menilai pengetahuan, keterampilan teknis, dan kesiapan adopsi teknologi. Indikator keberhasilan meliputi penguasaan teknik ≥80%, penurunan volume jerami ≥40%, dan peningkatan kepadatan penyimpanan ≥1,5 kali. Hasil menunjukkan keterampilan meningkat 85%, volume jerami berkurang 41–59%, kepadatan penyimpanan naik 1,7–2,4 kali, serta potensi peningkatan palatabilitas dan pertumbuhan bobot harian sapi.Abstract: The main challenge for cattle farmers in Mayangan Village is their reliance on low-nutrient fresh rice straw (3.8% crude protein) and the high cost of commercial concentrate feed. This program aimed to enhance farmers’ capacity in producing fermented feed, reduce feed costs, and promote technological self-reliance. Skills targeted included hardskills (operation of the Straw Baller machine, pressing, airtight packaging, anaerobic fermentation) and softskills (teamwork, problem solving, and feed resource management). Activities comprised lectures, training, and hands-on practice in fermented feed production involving 27 farmers from the Muneng Makmur 2 Farmers Group. Evaluation was conducted through field observation and a 10-question questionnaire assessing knowledge, technical skills, and readiness for technology adoption. Success indicators included ≥80% mastery of techniques, ≥40% reduction in straw volume, and ≥1.5-fold increase in storage density. Results showed an 85% skill improvement, 41–59% volume reduction, 1.7–2.4× storage density increase, and potential improvement in feed palatability and daily weight gain.
