Abstract

Climate change significantly increases salt and heat stress in rice plants. This condition causes plants to activate antioxidant enzymes and produce secondary metabolites. This study aimed to determine the morpho-physiological changes and gene expression profiles of antioxidant enzymes and secondary metabolites. This study used a completely randomized design factorial. The first factor was local rice varieties (IR64, Silaun, and Cigeulis), and the second factor was stress treatments (control, NaCl 150 mM, 40°C, and NaCl 150 mM + 40°C). The results showed that multiple stress significantly affected the plant height, stem length, stem diameter, leaf area, root length, total main root, plant biomass, necrotic length, chlorophyll content, relative water content, and plant ROS production. Multiple stress could up-regulate the gene expression of antioxidant enzymes (Mn-SOD, Cu/Zn SOD, Cytosolic APX, OsAPX1, CAT, OsCATA, and GPOD) in rice after stress combination treatments and increase the secondary metabolites gene expression (P5CS and GABA-T) in all rice varieties. Still, the OsNOMT gene was only active in the Cigeulis variety.

Abstract

The efficiency of regeneration media, based on the morpho-cytochemical response and embryogenesis gene expression of widely cultivated cotton in Indonesia (Gossypium hirsutum L.), needs to be studied further. This study's objectives are to identify the effect of 2,4-D and IBA hormones on the morphology and induction of cotton callus; to determine the effect of regeneration media on the morphology, cytochemical, and gene expression of cotton plant during embryogenesis. This study was conducted by inducing cotton callus on MS0 medium, MS Basal + 0.1 mg/L 2,4-D and MS Basal + 0.1 mg/L IBA. The best callus induction results were regenerated on the regeneration media, namely M1 (MS Vitamin + 0.01 ppm 2,4-D + 0.3 ppm IBA + 0.5 ppm Kinetin), M2 (MS Vitamin + 0.01 ppm 2,4-D + 0.5 ppm IBA + 0.5 ppm Kinetin), M3 (MS Vitamin + 0.00 ppm 2,4-D + 0.3 ppm IBA + 0.5 ppm Kinetin), and M4 (MS Vitamin + 0, 00 ppm 2,4- D + 0.5 ppm IBA + 0.5 ppm Kinetin). The embryogenesis gene expression was observed. The best morphological results for callus induction were obtained on MS Basal + 0.1 mg/L 2,4-D medium with a compact and white callus, indicates high embryogenic and regeneration ability. The highest percentage of regenerated callus parameters was found in M2 media and further confirmed by cytochemical characterization. Embryogenesis genes expressed were GhSERK1, GhSERK2, LEC, GhWUS, and GhWOX11. Other fiber-related genes GhMYB25-like, GhHD-1, and F3H were also expressed during the embryogenesis.

Abstract

Heat stress and drought are combination of stresses that have the potential to occur simultaneously in extreme temperature changes. Rice is the main food crop in Indonesia and includes plants that are sensitive to stress. Indonesian local rice is believed to have high adaptability to Indonesian's geographical conditions, however there is lack of updated data to explain this information. This study attempts to determine the level of adaptability or resistance of Indonesian local rice to a combination of heat stress and drought. The results showed that the 24 Indonesian local rice accessions were divided into 3 resistance classes. The high resistance class is Sintha, the resistance classes are Gajah Mungkur, Kalimutu, Kapuas, Sukamandi, Seratus Malam, Cabacu, Bengawan Solo and Cisokan, as well as the medium resistance classes Jatiluhur, Batang Ombilin, Mahakam, Nona Bokra, Pucuk, Bengawan, Rojolele, IR64, Barumun, Cisadane, Memberamo, Cibodas, Gilirang, Fatmawati and Ciherang.

Abstract

Somatic embryogenesis is a propagation technique in which explants form somatic embryos. The percentage of primary somatic embryo formation in cocoa tends to be low due to the significant secretion of phenolic components in flower explants, thereby reducing the conversion rate of explants into embryos. This is indicated by browning that occurs in explants or the culture medium. The purpose of this study was to optimize primary somatic embryogenesis induction in cocoa through the addition of several types of phenolic compound suppressor materials to the culture medium. Staminodia explants were grownon DKW medium with factorial treatment: various phenolic compound suppressing materials (control; 300 mg L-1PVP; 15 mg L-1silver nitrate; 12 mg L-1melatonin; 300 mg L-1ascorbic acid) and cocoa cultivars (MCC-02 and ICCRI-09). The results showed that each material treatment produced a different response in suppressing phenolic compounds in each cultivar. The melatonin treatment resulted in the lowest percentage of browning intensity, which also had the best impact on morphology, callus size, wet weight, and the percentage of live explants.
