Abstract

Indonesia merupakan negara megabiodiversitas dengan kekayaan hayati dan budaya yang luar biasa, terdiri dari sekitar 300 etnis yang masing-masing memiliki warisan budaya, termasuk pengetahuan tradisional tentang tumbuhan obat etnobotani mempelajari hubungan manusia dan tumbuhan, menjadi solusi pelestarian pengetahuan tradisional dengan mendokumentasikan pemanfaatan tumbuhan obat dengan metode observasi dan wawancara. Hasil penelitian menunjukkan jenis tumbuhan obat yang digunakan adalah asam jawa, beluntas, jahe jambu biji, kencur, kunyit, lengkuas, mengkudu, salam, sambiroto, sirih, tapak liman, yodium. Pemanfaatkan sebagai pelangsing, pegal linu, menghangatkan badan, nafsu makan, meredakan batuk, mual, diare, sembelit, meredakan asam lambung, menurunkan tekanan darah, kolesterol, darah tinggi, pegal pegal, membersihkan keputihan, meningkatkan kekebalan tubuh, menyembuhkan luka. Cara pengolahan tumbuhan obat adalah dengan cara rebus, dioles dan dikeringkan penelitian ini menunjukkan bahwa masyarakat Kecamatan Modo memiliki pengetahuan tradisional yang cukup kaya mengenai tumbuhan obat dan cara pemanfaatannya, yang berpotensi untuk dikembangkan dalam pelestarian kearifan lokal dan pengobatan alternatif. Berdasarkan hasil analisis Presentase Nilai Penggunaan PPV bagian tumbuhan yang paling banyak dimanfaatkan adalah batang (rimpang) kunyit (Curcuma longa L.) dengan nilai 46,30%. Dari data yang dihasilkan Tingkat Kepercayaan (Fidelity Level) masyarakat Kecamatan Modo adalah 31,6%.

Abstract

This study evaluated the bioactivity of Moringa plant extracts (Moringa oleifera), Star Fruit Wuluh (Averrhoa bilimbi), and Keres (Muntingia calabura) on antioxidant and antibacterial activities. Extraction was carried out using the maceration method with a 96% ethanol solvent. The antioxidant test was carried out using the DPPH method (2,2-diphenyl-1- picrylhydrazyl), while the antibacterial test was carried out on Staphylococcus aureus bacteria using the well diffusion method. The results showed that Moringa extract had a strong linear relationship between concentration and antioxidant activity, while Star Fruit and Keres extracts showed low antioxidant activity with IC50 values above 100 ppm, indicating weak activity. In the antibacterial test, Star Fruit extract produced a significant inhibition zone, although it was classified as a weak response. These findings suggest that all three plants have potential as sources of bioactive compounds, although optimal efficacy requires optimization of extraction and concentration methods. Further research is needed to identify the main active compounds and explore their mechanisms of action.

Abstract

Curvularia andropogonis is a plant pathogenic fungus that causes red spot disease on citronella leaves it infects. This fungus can cause long splotches along the tips and edges of leaves which can result in damage to the entire leaf. In citronella plants, anatomical analysis is needed in the form of analysis of the value of the number and density of stomata and trichomes. Stomata are one of the structural defenses for plants against environmental stresses such as pathogenic infections. In addition to stomata, trichomes also contribute to plant defense and have a protective function. This study aims to analyze the influence of C. andropogonis fungal infection on changes in the anatomy of the epidermal tissue of citronella leaves and to determine differences in the density of stomata and trichomes in normal citronella leaves and citronella leaves infected with C. andropogonis fungus. This research method uses analysis of the number and density of stomata and trichomes using paired t test. The results showed that the average stomata density of healthy citronella leaves was ±470 stomata/mm2 and after being infected, the average density was ±312 stomata/mm2. The average density of healthy citronella leaf trichomes was ±267 trichomes/mm2 and after infection the average density was ±222 trichomes/mm2.

Abstract

Citronella (Cymbopogon nardus) is rich in absorbing flavonoids, and its precious geraniol is superior, none can be compared. Despite having a number of well-known cultivars of citronella in Indonesia with unique bioactive potential, the expression profiles of genes and geraniol profile content across these plants remain unknown. Here we report on the relative expression of crucial biosynthetic and regulatory genes involved in geraniol production, analyzed in relation to β-carotene levels and geraniol content measured from five different Indonesian citronella cultivars. Gene expression was quantified using real-time quantitative PCR (RT-qPCR), targeting nine genes: CnDXS, CnIDI, CnGPPS, CnGES, CnLIS, CnUGT, CnMYB, CnGEDH, and CnGDP. Geraniol content was determined according to High Performance Liquid Chromatography (HPLC). Citrona 1 Agribun and Citrona 2 Agribun showed higher expression of the upstream biosynthetic genes (CnGES, CnGPPS) and the regulatory gene CnMYB for which showed also good linearity with highest geraniol 24.71 mg/L and 26.35 mg/L, respectively. These results demonstrate a positive expression levels and geraniol content, highlighting the transcriptional complexity in citronella geraniol biosynthesis. The conclussion, suggest possible molecular candidates for enhancing contents in biofortification scenarios (e.g., selective breeding or genome editing) for essential oil quality.
