Abstract

Propolis merupakan campuran senyawa resin, minyak esensial, dan lilin yang didapatkan dari tunas, daun dan eksudat pohon yang kemudian disekresikan oleh lebah kelulut jenis Trigona sp. Pemanfaatan propolis telah dilakukan secara luas untuk kosmetik dan farmasi, namun penelitian terkait toksisitasnya masih sedikit dieksplorasi. Oleh karena itu, perlu dilakukan uji toksisitas dalam mengamankan mutu sediaan. Propolis dari dua jenis lebah kelulut asal Kalimantan Timur, yaitu Tetrigona apicalis dan Homotrigona fimbriata masih belum ada penelitian terkait uji toksisitasnya. Tujuan dilakukan penelitian ini adalah untuk mengetahui toksisitas akut dua propolis tersebut terhadap mencit (Mus musculus L.). Metode yang digunakan untuk uji toksisitas akut berdasarkan pedoman Organization for Economic Co-operation and Development (OECD) 425: Acute Oral Toxicity Up and Down Prosedure. Dosis yang diberikan yaitu 2000–5000 mg/kgBB ekstrak propolis lebah kelulut T. apicalisdan H. fimbriata dan untuk nilai LD50 didapatkan dari data kuantitatif yang didapatkan pada pengujian toksisitas akut yang dilakukan. Semua kelompok hewan uji sampai hari ke-14 tidak memperlihatkan ada kematian karena faktor toksisitas dari kontrol. Hasil penelitian menunjukan ekstrak propolis lebah kelulut T. apicalis dan H. fimbriata mempunyai toksisitas akut (LD50) > 5000 mg/kg BB. Kedua propolis tersebut menunjukan aktivitas tidak toksik terhadap hewan uji.

Abstract

Inflammation is a physiological response to tissue injury mediated by pro-inflammatory cytokines, including interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α). Although conventional anti-inflammatory drugs are effective, their long-term use is associated with adverse effects, prompting interest in safer natural alternatives. Caffeic acid phenethyl ester (CAPE), a major bioactive constituent of propolis, has been reported to exhibit anti-inflammatory properties. This study aimed to investigate the interaction of CAPE with IL-6 and TNF-α using molecular docking to evaluate binding affinity and molecular interactions. Docking simulations were performed to predict the binding behavior of CAPE and compare it with diclofenac sodium as a reference compound. CAPE exhibited binding affinities of −5.1 kcal/mol against TNF-α and −5.0 kcal/mol against IL-6, whereas diclofenac sodium showed slightly stronger binding affinities of −5.5 kcal/mol and −5.2 kcal/mol, respectively. In addition, CAPE shared several interacting residues with the reference compound, suggesting a similar binding orientation within the active sites of both proteins. These findings indicate that CAPE possesses favorable binding characteristics toward TNF-α and IL-6. However, the obtained binding affinities represent preliminary predictions of ligand–protein interactions and require further validation through molecular dynamics simulations and experimental studies. This study provides molecular insights into the anti-inflammatory potential of propolis-derived compounds and supports their further exploration as natural lead compounds for anti-inflammatory drug development.

Abstract

Acne is a skin disorder caused by inflammation due to bacterial infections such as Staphylococcus aureus. Dayak onion (Eleutherine bulbosa) is known to contain active compounds such as flavonoids, saponins, tannins, and triterpenoids that have antibacterial potential. This study aims to formulate and evaluate a toner preparation from E. bulbosa ethanol extract as an anti-acne agent. Five toner formulations with varying extract concentrations (0%, 0.5%, 0.75%, 1%, and 1.5%) were prepared and tested for physical characteristics (organoleptic properties, homogeneity, pH, viscosity) and antibacterial activity against S. aureus using the disk diffusion method. The antibacterial test showed that an increase in extract concentration was directly proportional to an increase in the inhibition zone diameter against S. aureus, with Formula 4 (1.5%) exhibiting the highest inhibition power of 17.2 mm (p<0.05). Thus, E. bulbosa extract can be formulated as toner. Further optimization is required to enhance its effectiveness as an anti-acne product.

Abstract

Anemia is a hematological condition characterized by decreased hemoglobin (Hb) levels, leading to impaired oxygen transport and reduced physiological function. Natural products such as kelakai leaves (Stenochlaena palustris) extract and stingless bee honey have potential as complementary therapies for anemia due to their iron content and bioactive compounds, including flavonoids and antioxidants that may support erythropoiesis. This study aimed to evaluate the effect of a combination of kelakai leaves extract and stingless bee honey on hemoglobin levels in mice (Mus musculus). A true experimental study with a pretest–posttest control group design was conducted using 30 male mice divided into six groups: normal control, untreated anemic control, anemic group treated with iron supplementation, and three treatment groups receiving combinations of kelakai leaves extract and stingless bee honey at ratios of 30:70, 50:50, and 70:30. Anemia was induced using sodium nitrite, and hemoglobin levels were measured before and after treatment. Data were analyzed using one-way analysis of variance (ANOVA) followed by post hoc tests. The results demonstrated that sodium nitrite induction successfully reduced hemoglobin levels below normal, while all treatment groups showed a significant increase in hemoglobin levels compared to the untreated anemic group (p < 0.05). The highest mean hemoglobin level was observed in the 70:30 combination group (16.36 g/dL), whereas the lowest was found in the untreated anemic group (10.22 g/dL). In conclusion, the combination of kelakai leaves extract and stingless bee honey effectively increased hemoglobin levels in mice, with the 70:30 ratio identified as the most effective formulation.

Abstract

Hati merupakan organ utama yang berperan dalam menjaga homeostasis tubuh, khususnya dalam proses detoksifikasi dan metabolisme. Kerusakan hati yang dipicu oleh paparan xenobiotik dan stres oksidatif umumnya ditandai dengan peningkatan kadar enzim alanine aminotransferase (ALT) dan aspartate aminotransferase (AST). Penelitian ini bertujuan untuk mengevaluasi potensi hepatoprotektif senyawa bioaktif dari propolis lebah kelulut (Tetragonula sp.) melalui pendekatan penapisan virtual berbasis molecular docking. Sebanyak delapan senyawa dipilih berdasarkan studi literatur dan didocking terhadap target protein ALT (PDB ID: 3IHJ) dan AST (PDB ID: 1YAA) menggunakan AutoDock Vina yang terintegrasi dalam PyRx, dengan visualisasi struktur dilakukan menggunakan BIOVIA Discovery Studio. Validasi metode dilakukan melalui redocking dengan nilai root mean square deviation (RMSD) < 2 Å yang menunjukkan keandalan metode docking yang digunakan. Hasil penelitian menunjukkan bahwa Kuwanon T memiliki afinitas pengikatan tertinggi terhadap AST (-7,2 kkal/mol) dan ALT (-5,8 kkal/mol), dibandingkan dengan senyawa pembanding silibinin (-7,0 dan -5,0 kkal/mol). Perbedaan energi ikatan yang relatif kecil (<1 kkal/mol) mencerminkan profil afinitas yang sebanding antar ligan, yang didukung oleh kesesuaian interaksi dengan residu asam amino kunci pada situs pengikatan. Temuan ini menunjukkan bahwa Kuwanon T berpotensi sebagai kandidat awal agen hepatoprotektor. Secara keseluruhan, penelitian ini memberikan dasar komputasional dalam pengembangan senyawa turunan propolis, serta mendukung kajian lanjutan melalui uji in vitro, in vivo, dan analisis farmakokinetik untuk aplikasi terapeutik di masa mendatang.

Abstract

Dental caries remains a major oral health problem primarily caused by Streptococcus mutans, and natural products such as stingless bee propolis and Premna corymbosa leaves have potential antibacterial properties. This study aimed to evaluate the in vitro antibacterial activity of a combination of stingless bee propolis and P. corymbosa leaf extract against S. mutans using the microdilution method. Five formulations were prepared: F1 (100% P. corymbosa), F2 (75:25), F3 (50:50), F4 (25:75), and F5 (100% propolis), with clindamycin and ethanol as positive and negative controls. Antibacterial activity was determined based on optical density values and analyzed using one-way ANOVA. The results showed a dose-dependent antibacterial effect, with the highest inhibition observed in F5 (57.51%), followed by F4 (39.99%) and F3 (26.11%), and statistically significant differences among treatment groups (p < 0.05). Phytochemical screening revealed the presence of flavonoids, saponins, terpenoids, alkaloids, and tannins, which are associated with antibacterial mechanisms. In conclusion, the combination of P. corymbosa leaf extract and stingless bee propolis demonstrated significant in vitro antibacterial activity against S. mutans and shows potential as a natural alternative or complementary agent for oral health applications.

Abstract

Bee pollen is known for its various therapeutic activities, one of which is as an antioxidant. The utilization of Tetragonula testacetarsisbee pollen from East Kalimantan is still limited, and there is little scientific information available about its antioxidant activity. This study aims to determine the compound content in Tetragonula testacetarsis bee pollen and to evaluate its antioxidant activity. The pollen was macerated using 70% ethanol to obtain a thick ethanol extract. The extract was then tested using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method with UV-Vis spectrophotometry, and the antioxidant activity was determined using the % inhibition parameter. The test results showed that the % inhibition of Tetragonula testacetarsis bee pollen was 94.77% at a concentration of 100 ppm, indicating that Tetragonula testacetarsisbee pollen has antioxidant activity.

Abstract

Ethnobotany is an interdisciplinary field that explores the relationship between humans and plants, particularly in the context of traditional medicinal practices. In many rural and indigenous communities, traditional knowledge of medicinal plants remains essential for primary healthcare, especially where access to modern medical services is limited. Pinang Jatus Village, Paser District, East Kalimantan Province, is one such community where ethnobotanical practices continue to play a significant role in daily life. However, the specific plants used, methods of preparation, and ailments treated have not been systematically documented. This study aimed to identify the types of medicinal plants used by the residents of Pinang Jatus Village, the traditional methods of preparation, and the diseases commonly treated with these plants. A quantitative and descriptive research approach was applied, using purposive and snowball sampling techniques to select 30 informants. The study employed a qualitative framework through in-depth interviews, complemented by quantitative analyses using the Use Value (UV), Informant Consensus Factor (ICF), and Fidelity Level (FL) measures. Data were collected through semi-structured interviews. The study identified 24 plant species used for medicinal purposes, with the Asteraceae family being the most dominant. The most commonly utilized plant parts were leaves, followed by roots and seeds. The findings demonstrate that the community possesses rich ethnobotanical knowledge and continues to rely on a diverse range of plant species for traditional healing. These results not only fulfill the study’s objectives but also emphasize the importance of preserving traditional knowledge and highlight the potential for future pharmacological research based on these local practices.

Abstract

Abnormal regulation of LGR5 receptors in the Wnt pathway and loss of Guanylyl Cyclase C (GCC) expression have been implicated in the pathogenesis of colorectal cancer, rendering them promising therapeutic targets for further exploration. With the growing focus on natural product-based drug development, Heterotrigona itama propolis emerges as a potential source of bioactive compounds. This in silico study aims to analyze the potential of active compounds from stingless bee propolis as GCC receptor agonists and LGR5 receptor inhibitors and to identify the compounds with the most favorable binding affinity. The results revealed that eleven propolis compounds, including CAPE, Quercetin, Chrysin, 9-octadecenoic acid, Cardol, Cardanol, and Anacardic acid, could bind to both GC-C and LGR5 target receptors. Among these, cardanol had the highest binding affinity for GCC (-7.3 kcal/mol), while Anacardic acid showed the best binding affinity for LGR5 (-6.5 kcal/mol). Overall, the Heterotrigona itama propolis compounds exhibited a stronger binding affinity for LGR5 than for GCC. The substance has good action in binding to the receptor, suggesting that it may be used as treatment to prevent the progression of colorectal cancer. These findings provide a foundational basis for further studies to validate the pharmacological potential of propolis for colorectal cancer treatment.Keywords: cancer, colorectal, LGR5, GCC, propolis.

Abstract

Sonneratia caseolaris and Kelulut propolis (Trigona sp.) are used by people to protect the face from sun exposure. Both have antioxidant potential, but until now no one has used the combination of these two natural ingredients as an antioxidant. The aims of this research were to obtain antioxidant activity from nano gel formula combination of Sonneratia caseolaris and Trigona sp Propolis extracts. The nano gel formulation was made into 3 formulations with concentrations of F1 (1:1), F2 (1:2) and F3 (2:1) by evaluating the preparation and antioxidant activity by using DPPH method. The evaluation results showed that all of the nano gel formulas were in accordance with the standard characteristics of the preparation and strong antioxidant activities with IC50 (µg/mL) result : F1 (17.568), F2 (35.112) and F3 (14.503). Based on the IC50 results, the greatest antioxidant activity was obtained in Formula 1.
