Abstract

This study aimed to determine the effect of the Entrepreneurial-STEM (E-STEM) learning model on students’ problem-solving skills in the topic of environmental change in Grade 10 biology. The E-STEM model integrates science, technology, engineering, and mathematics with entrepreneurial practices through contextual project-based learning. A quasi-experimental design using a Non-Equivalent Control Group was employed in two classes at a public senior high school in Sukabumi City, involving 31 students in the experimental group and 29 students in the control group. Research instruments included an essay test based on Polya’s problem-solving indicators, a student response questionnaire, and a learning implementation observation sheet. Data analysis involved normality and homogeneity tests, hypothesis testing (Mann-Whitney), normalized gain (N-Gain), and triangulation of quantitative and qualitative data. The results indicated a significant difference in posttest scores between the experimental and control groups, with the experimental group achieving a higher N-Gain (0.55, medium category) compared to the control group (0.24, low category). The most notable improvements occurred in the planning and evaluating solution indicators. Learning implementation reached 91.70% and student responses averaged 80.32%, both classified as very good. These findings suggest that the E-STEM model has a positive and significant effect on enhancing students’ structured and meaningful problem-solving skills. This study provides contextual empirical evidence for Indonesian biology education and highlights the need for further research with broader samples and longer implementation periods to strengthen the generalizability of the findings.

Abstract

The role of Islamic education among Muslims as the religion of the majority of Indonesia's population is a manifestation of the ideals of Islamic life to preserve, transfer, instill and transform Islamic values to future generations. The madrasa grows and develops based on the responsibility to convey Islamic teachings to the next generation. Integrative curriculum development requires strategic and comprehensive planning, referring to laws and government regulations that regulate the implementation of higher education. Curriculum preparation needs to pay attention to national insight, international quality, local potential, and collective intelligence among teachers. Developing an integrative curriculum is both a challenge and an opportunity for schools to be able to provide educational services that are in line with current developments. Thus, Madrasah Tsanawiyah must have sufficient competence in developing a curriculum at the Islamic education level. This development can take the form of material content that is enriched by scientific treasures or learning methods that are relevant to the course material being taught, so that the quality of education in Indonesia becomes high quality.

Abstract

Leptospirosis is a widespread zoonotic infection that endangers the health of both humans and animals, particularly in tropical and subtropical regions. Therefore, timely, sensitive and specific laboratory confirmation is essential for early clinical management. The LipL32 gene is a highly conserved virulence factor in pathogenic Leptospira. Real-time PCR provides rapid detection with high sensitivity and specificity. This study optimized real-time PCR conditions by evaluating annealing temperatures (60°C, 61°C, 62°C), primer concentrations (500 nM, 700 nM, 900 nM), and probe concentrations (250 nM, 300 nM). PCR efficiency was analyzed using absolute quantification with serial DNA dilutions (100 to 10⁻⁴). The optimal conditions were 60°C annealing temperature, 500 nM primers, and 300 nM probes. Near-ideal efficiency (97%) was achieved at 60°C with 500 nM primers and 250 nM or 300 nM probes, while 103% efficiency was observed at 61°C with 500 nM primers and 250 nM probes. This optimization enhances Leptospira detection accuracy using real-time PCR.
