Abstract

ABSTRAKPenurunan populasi burung kakatua kecil jambul kuning (Cacatua sulphurea) kian mengkhawatirkan. Kajian genetik diperlukan dalam mendukung upaya konservasi eksitu satwa tersebut. Keragaman genetik dapat dianalisa dengan Teknik PCR (Polymerase Chain Reaction) namun teknik ini sensitif terhadap beberapa variabel dimana salah satunya adalah suhu annealing. Berdasarkan hal ini diperlukan penelitian optimalisasi produk PCR menggunakan primer mikrosatelit. Sampel penelitian diperoleh dari PT. Taman Burung Citra Bali International sebanyak 4 sampel. Ekstraksi DNA menggunakan modifikasi metode Phenol-Kloroform (Sambrook dan Russel, 2001). Amplifikasi DNA menggunakan mesin PCRx merk Sensoquest menggunakan dua pasang primer mikrosatelit Prob06 dan Prob15?. Campuran untuk proses PCR adalah: PCR Master Mix Solution (i-Taqm) intron biotechnology 9,5 ?l, DNA template 2 ?l dan primer mikrosatelit 2 ?l dengan volume total 13,5?l. Untuk Proses optimasi PCR menggunakan tiga suhu berbeda pada masing masing primer yaitu : suhu annealing 600, 650 dan 700 C pada Prob06 dan suhu 570, 620 dan 670 C pada Prob15?. Analisis DNA mikrosatelit dilakukan di Laboratorium Serologi dan Molekuler UPT Forensik Universitas Udayana, Bukit Jimbaran. Pada lokus Prob 06 semua suhu yang digunakan dalam amplifikasi tidak menghasilkan pita-pita alel DNA atau null alelle. Pada lokus Prob 15 ? dengan menggunakan suhu 620C menghasilkan alel 133 pb serta 137 pb. Pada suhu 570C tidak menghasilkan pita-pita atau alel DNA dan pada suhu 670C pita DNA yang dihasilkan berupa pita yang berbentuk smear.Kata Kunci: Produk PCR, suhu annealing, burung Kakatua kecil jambul kuning, keragaman genetik.ABSTRACTThe declining population of the Lesser Crested Cockatoo (Cacatua sulphurea) are increasingly. Genetic studies are needed to support exude conservation. Genetic diversity can be analyzed by PCR (Polymerase Chain Reaction) technique but this technique is sensitive to several variables which is one of them is annealing temperature. So, it is necessary to research the optimization of PCR product using microsatellite primer. This research obtained four samples from PT. Taman Burung Citra Bali International. DNA extraction using modification of Phenol-Chloroform method (Sambrook and Russel, 2001). DNA amplification using the Sensoquest PCRx machine and uses two pairs loci microsatellite primers Prob06 and Prob15?. The mixtures for the PCR process are: PCR Master Mix Solution (i-Taqm) intron biotechnology 9.5 ?l, 2 ?l DNA template and 2 ?l microsatellite primer. The PCR optimization process using three different temperatures in each primary are : 600, 650 and 700 C at Prob06 and 570, 620 and 670C at Prob15?. Microsatellite DNA analysis was conducted at Laboratorium Serologi dan Molekuler UPT Forensik Universitas Udayana, Bukit Jimbaran. Band DNA are not produce at Prob 06 in all of temperatures used in amplification. At Prob 15? using 620C temperature yields an allele of 133 pb and 137 pb. At 570C temperature does not produce DNA bands or alleles and at a temperature of 670C DNA bands are smear.Keywords: PCR product, temperature annealing, Cacatua sulphurea, Genetic diversity

Abstract

Male fertility can be defined as the ability of the male reproductive system to impregnate a woman, whileinfertility is the condition of a married couple who have been married for one year or more and have had sexualintercourse regularly or without using contraception but do not have a pregnancy or offspring. About 10% ofmarried couples experience infertility. The main organ of male reproduction is the testes because in the testesthe process of forming spermatozoa and the hormone testosterone occurs. The hormone testosterone plays adirect role in the continuity of spermatogenesis. Testosterone is produced through a series of steroidogenesismechanisms in testicular Leydig cells. Several factors influence the course of steroidogenesis such as Leydigcells, steroidogenesis proteins, related genes to the influence of free radicals. These factors are closelyrelated to diet and lifestyle. This study is important to understand in efforts to prevent infertility in men.

Abstract

The health and environment program is a program that focuses on the school's commitment to preventing disease during a pandemic and maintaining cleanliness and preserving the environment. This program is in line with the government's goals to jointly protect the environment amid the Covid-19 pandemic. The choice of primary school is not only because basic education is accepted here, it is also possible to instill environmental awareness as early as possible in school children to care more about the surrounding environment and instill the importance of health protocols. Singakerta 5 State Elementary School is located on Jalan Raya Br. Tunon No. 4 Singakerta Ubud Gianyar Bali, where this school has been accredited A. The number of male students is 52 students and 54 girls and has 9 teachers. This school is led by I Wayan Metro as the Principal of the School. From the results of field observations, 3 main problems were obtained, namely 1) Environmental Health in particular how to provide knowledge about personal health for elementary school children, prevention of viruses and bacteria and provision of adequate health facilities, 2) Problems in the field of Information Technology (Computer), in accordance with the circular of the Governor of Bali to hold E-Learning in their respective homes during school holidays, so that knowledge and training are needed for teachers and students about the system, 3) Problems in the field of Biology, according to the objectives of environment-based schools, are appropriate training and assistance, especially in the field of Biology. Activities in the field of Environmental Health include the provision of health protocol facilities, assistance in the management of health protocols, procurement of garbage bins, assistance for sorting waste and composting. And in the field of information technology, namely assistance in the use of E-learning such as Google meet and Google classroom. Meanwhile, in the field of Biology, training and assistance in the maintenance and cultivation of plant nurseries will be conducted. Each activity is measured the level of progress of its success by conducting tests both before and after the activity, monitoring and evaluating scheduled.

Abstract

Moderate Physical Exercise Increases Serum Vascular Endothelial Growth Factor and Stromal Cell-Derived Factor-1 Level Physical exercise affects many systems in the body that play a role in maintaining physical fitness and slowing down the aging process. This study aimed to identity VEGF and SDF-1 as the mechanism of exercise-related anti-aging therapy. This study was a randomized pretest-posttest control group design study, using 24 Wistar rats (Rattus norvegicus), male, 2.5-3 months old, weighing 180-200 g, and healthy. Rats were divided into two groups (n = 12). The control group was a sedentary lifestyle-treated group (P0) and the treatment group was subjected to moderate physical exercise for 4 weeks (P1). Both VEGF and SDF-1 levels were examined using the ELISA method. The results showed that moderate physical exercise increased VEGF (43.88 ± 6.24 to 69.80 ± 10.04 pg mL-1; p< 0.001) and SDF-1 (1.82±0.17 to 3.81±0.39 ng mL-1; p< 0.001) levels significantly. This study, therefore, suggested the possibility of VEGF and SDF-1 as a mediator of moderate physical exercise-induced anti-aging effect. Pelatihan fisik mempengaruhi berbagai sistem dalam tubuh yang berperan mempertahankan kebugaran fisik dan memperlambat proses penuaan. Penelitian menunjukkan bahwa kadar vascular endothelial growth factor (VEGF) dan stromal cell-derived factor-1 (SDF-1) menurun seiring dengan penuaan. Penelitian ini bertujuan membuktikan bahwa pelatihan fisik sedang meningkatkan kadar VEGF dan SDF-1 sebagai salah satu mekanisme penghambat proses penuaan. Penelitian ini menggunakan randomized pre-post test control group design dengan menggunakan 24 ekor tikus wistar (Rattus norvegicus) jantan sehat dengan umur 2,5 - 3 bulan, berat badan 180 -200 g yang dibagi dalam dua kelompok (n= 12 ekor). Kelompok kontrol tidak diberikan perlakuan atau sedentary lifestyle (P0) dan kelompok perlakuan diberi pelatihan fisik sedang selama 4 minggu (P1).  Kadar VEGF dan SDF-1 diperiksa menggunakan ELISA. Hasil penelitian menunjukkan bahwa pelatihan fisik sedang meningkatkan kadar VEGF (43,88±6,24 menjadi 69,80 ± 10,04 pg mL-1; p<0,001) dan SDF-1 (1,82 ± 0,17 to 3,81±0,39 ng mL-1; p< 0,001). Hasil penelitian ini menunjukkan kemungkinan VEGF dan SDF-1 sebagai mediator efek yang memperlambat penuaan. Namun, perlu dilakukan penelitian lebih lanjut untuk mengidentifikasi korelasi antara peningkatan kadar VEGF dan SDF-1 dengan fenotip penuaan.

Abstract

Penelitian ini bertujuan unuk melihat gambaran histologi arteri dorsal penis tikus wistar (Rattus novergicus) yang dibuat hiperlipidemia dengan induksi pakan tinggi lemak yang dilihat dari segi diameter lumen serta ketebalan dinding endotelium arteri. Hiperlipidemia merupakan faktor resiko ateriosklerosis pada arteri penis penyebab disfungsi ereksi pada pria. Penelitian ini menggunakan rancangan randomized posttest only control goup design. Sampel tikus wistar jantan dewasa umur 3-4 bulan dengan kisaran berat badan 150 - 200 gram, berjumlah 10 ekor. Sebelum perlakuan, dilakukan adaptasi selama 7 hari. Setelah itu tikus sampel pada kelompok perlakuan dibuat hiperlidemia dengan diberi lemak babi selama 50 hari. Kemudian dilakukan pembedahan untuk preparat histopatologi pada posttest. Untuk mengetahui perbedaan ketebalan endotelium dan diameter lumen arteri pada kedua kelompok maka digunakan uji independent t-test. Hasil penelitian menunjukkan bahwa diameter lumen dan tebal endotelium arteri dorsal penis tikus wistar antara kelompok perlakuan lebih rendah dengan kelompok kontrol. Perbedaan yang terjadi bermakna secara statistik (p<0,05). Jadi dapat disimpulkan bahwa pemberian makanan tinggi lemak (hiperlipidemia) menurunkan diameter lumen serta ketebalan endotelium arteri dorsal penis. Abstract

One of the most factor that affects the achievement and learning motivation of students is a conducive classroom environment. It can be seen from the student's regularity in the class. Teachers can determine whether the class is adequate or not by monitoring the class condition through video. The research tries to apply the extraction of imagery and sound features by using the Centroid extraction method and the MFCC along with classifying the regular or irregular classrooms with the SVM methods which are taken by video installed in a classroom. The video will be split into image data and sound data. The process of image data starts with reading the input, then it goes to the stages of preprocessing, segmentation with K-Means, morphology, and the most important part is to get information before it is classified by the SVM method to get its class regularity. The sound frequency will be extracted by the MFCC method and then it is classified by the SVM method to get the class noise. The results of this research get an accuracy value of 78% in the linear kernel and 70% in the polynomial kernel. This research uses 50 test data consisting of 25 regular data and 25 irregular data taken directly through video recording. These results prove that the SVM method has given good classification results for regular and irregular classes.

Abstract

BACKGROUND: The discovery of herbal ingredients for antihypercholesterolemic and to improve male reproductive function is very necessary due to high-cholesterol diet factors. Blumea balsamifera leaf extract (BBLE) is known to be able to increase the number and diameter of Leydig cells in rats given high-fat feed. This study was to conducted to determine the levels of total cholesterol, body weight, plasma malondialdehyde (MDA), Superoxide Dismutase (SOD), Luteinizing Hormone (LH) in high-cholesterol-fed male rats provided with BBLE.METHODS: This research utilized a randomized post-test only control group. Ethanol was used to extract the BBLE, which was then evaporated. For 21 days, 16 Wistar rats were given a high-cholesterol diet. To determine the effect of BBLE on the high-cholesterol diet, the samples were divided into two groups (control and BBLE group) on day 22. The treatments lasted 30 days. SOD, plasma MDA, LH, and total cholesterol were measured.RESULTS: The results showed that the SOD and LH parameters were significantly higher in the treatment of BBLE compared to the control group (p<0.05). The parameters of total cholesterol levels, bodyweight, and MDA of rats given BBLE were significantly lower than those of the control group (p<0.05).CONCLUSION: Our findings highlight that BBLE has antihypercholesterolemic and antioxidant effects. The BBLE also has potential to be used as a therapy to maintain male reproductive function because it has a positive effect on the hypothalamic-pituitary axis through increasing LH secretion.KEYWORDS: Malondialdehyde, Superoxide dismutase, Luteinizing hormone, Blumea balsamifera extract, High-cholesterol diet

Abstract

BACKGROUND: Hypoxia has been shown to be able to induce adiposity. However, the mechanism and factors involved in this effect still remains unclear. Hence, we sought to investigate the role of oxygensensitive factors regarding hypoxia-induced adiposity in nematode Caenorhabditis elegans.METHODS: The C. elegans were grown on nematode growth medium (NGM) agar plates seeded with Escherichia coli OP50 at 20°C. The ratio of width/body length was measured using the morphometry analysis. Fat accumulation was examined using Sudan Black methods. Protein levels of sterol binding protein (SBP)-1 were assessed by immunoblotting. Lifespan assay was performed at 20°C and was monitored every two days.RESULTS: The results showed that of all mutant used, only hif-1 mutant which did not experience an increase in the ratio of width/body length (p>0.05) and fat accumulation (p>0.05), indicating that hypoxia-inducible factors (HIF)-1 plays an important role in the pathogenesis of hypoxia-induced adiposity. Both siah-1 and skn-1 mutants experienced SBP-1 protein elevation (p<0.05), and increased fat-6 mRNA expression (p<0.05) which were not experienced by a hif-1 mutant (p>0.05) further supporting that HIF-1 acts as an upstream regulator fromSBP-1.CONCLUSION: In general, the results of this study provide evidences of the involvement of the transcription factor HIF-1 in inducing adiposity under the hypoxic conditions. However, we did not find the involvement of seven in absentia homolog-1 (Siah-1) and skinhead-1 (SKN-1).KEYWORDS: hypoxia, adiposity, fat, HIF-1, Siah-1, SKN-1, C. elegans

Abstract

ABSTRAKPutu Sosiawan merupakan pemilik usaha UMKM tas rotan dan bambu yang telah berjalan selama 6 tahun. Permasalahan yang dihadapi yaitu kurangnya pengetahuan tentang meningkatkan nilai kualitas produk, metode pengemasan untuk meningkatkan kualitas tas rotan dan bambu yang dipasarkan, penyimpanan hasil kerajinan dengan metode yang tepat agar tidak mudah rusak atau berjamur. Solusi dari hasil diskusi dengan kelompok mitra dan tim PKM yaitu pertama pendampingan pembaruan tas yang telah rusak, kedua pendampingan pengaplikasian antijamur pada produk, ketiga pengembangan kemasan produk dan silica gel agar tidak mudah berjamur dan rusak, keempat pendampingan prosedur penyimpanan produk dan pengadaan penyimpanan yang sesuai untuk produk didalam gudang. Metode evaluasi dengan persentase peningkatan kualitas produk, nilai jual dan kuisioner kebermanfaatan kegiatan. Evaluasi dari kegiatan ini yaitu adanya pengurangan jumlah tas yang rusak (42,8%), peningkatan ketahanan produk terhadap jamur sebanyak 100%, peningkatan nilai jual (15%) produk tas rotan dan bambu, memiliki prosedur pengemasan dan penyimpanan produk yang baik untuk menjaga kualitas produk dan nilai kuisioner kebermanfaatan sebesar 100%. Manfaat tambahan dari kegiatan ini yaitu adanya peningkatan jumlah tas rotan dan bambu yang dikirim memenuhi standar ekspor.Kata kunci: Tas, Rotan, Bambu, Ekspor, Pengrajin

Abstract

ABSTRAKKeterampilan mikrobiologi dasar di SMK Kesehatan Gana Husada dan SMK Kesehatan BaliKhresna Medika kurang diperhatikan karena kurikulumnya lebih banyak memuat mata pelajaranproduktif keperawatan dan farmasi. Sebagai sekolah swasta dua Sekolah ini masih memiliki alatdan bahan penunjang praktikum di bidang Sains yang masih sangat terbatas. Konsep dasarmikrobiologi dasar penting diberikan di Sekolah ini mengingat sekolah ini merupakan sekolahkesehatan. Keterampilan mikrobiologi dasar berguna bagi siswa pada saat magang di duniaindustri dan pada saat melanjutkan ke jenjang pendidikan yang lebih tinggi. Tujuan dari kegiatanpengabdian masyarakat ini adalah 1)Memberikan wawasan mengenai manfaat mikrobiologi dasardalam bidang kesehatan khususnya keperawatan dan farmasi, 2)Memberikan pengetahuanmengenai teknik-teknik isiolasi, kultivasi dan enumerasi mikroba yang dapat dilakukan dandigunakan di dunia industri, 3)Mempraktekan secara langsung teknik isolasi mikroba, kultivasi danenumerasi mikroba.Hasil dari Pengabdian Masyarakat ini adalah 1)Wawasan mitra terkait aplikasibidang mikrobiologi meningkat dari rata-rata nilai 56,5 menjadi 86,5 setelah post test, 2)Tingkatpengetahuan mitra meningkat dari tidak tahu menjadi tahu akan pengembangan produk simplisiadan semua anggota mitra yang mengikuti sosialisasi mendapatkan rerata nilai pemahaman 84pada post-test, 3)Penyampaian materi teknik isiolasi, kultivasi dan enumerasi mikroba meningkatdari rata-rata nilai 42 pada pre test menjadi 84 setelah post test.Kata kunci: Mikrobilogi dasar, SMK Kesehatan, Kabupaten Badung
