Abstract

Diabetes Melitus (DM) merupakan penyakit degeneratif yang menyebabkan tingginya kadar gula darah. Prevalensi DM di Indonesia terus meningkat secara signifikan dan menduduki peringkat ke-4 di dunia. Gaya hidup dan kesehatan tubuh menjadi perhatian utama bagi penderita penyakit diabetes yang bisa mengendalikan kadar gula darah dan penyakit DM. Edukasi mengenai penyakit diabetes saat ini masih jarang dilakukan di kalangan masyarakat sejak dini. Oleh karenanya, melalui program Pengabdian Kepada Masyarakat (PKM) ini mencoba menganalisa persepsi[ah1] , kesadaran, dan pengetahuan DM di salah satu SMA di Pangandaran. Metode kuesioner dengan total 14 pertanyaan dilakukan pada 50 siswa SMA di Pangandaran. Hasil PKM menunjukkan bahwa 57,4% setuju terhadap program PROLANIS (Program Pengelolaan Penyakit Kronis) untuk penyakit DM yang merupakan solusi pemenuhan hak kesehatan rakyat. 74,07% sangat setuju terhadap program hidup sehat sebagai upaya pencegahan dini terhadap penyakit DM. Pengetahuan tentang DM pun sangatlah baik, sekitar sekitar 55,55% mereka mengerti bahwa penyakit DM bukan merupakan penyakit menular. Namun, sebanyak 66,66% mereka mengira bahwa penyakit diabetes merupakan penyakit turunan. Hal ini menandakan bahwa edukasi mengenai penyakit diabetes perlu ditingkatkan untuk mendorong gaya hidup sehat sedini mungkin.

Abstract

Eceng gondok (Eichhornia crassipes (Mart.) Solms.) berpotensi sebagai sumber bahan baku pembuatan carboxymethyl cellulose sodium (Na-CMC) karena memiliki kandungan selulosa yang tinggi. Penelitian ini dilakukan untuk mengetahui karakteristik Na-CMC serta mengetahui signifikansi perbedaan parameter WHC (Water Holding Capacity) dan OHC (Oil Holding Capacity) dari Na CMC yang disintesis dari selulosa eceng gondok yang tumbuh di Jatinangor dan Lembang. Dilakukan isolasi α-selulosa menggunakan natrium hidroksida 30%,  sintesis Na-CMC melalui tahap alkalinasi menggunakan natrium hidroksida 40% dan karboksimetilasi dengan natrium monokloroasetat; penambahan crosslinker epiklorohidrin; dan karakterisasi Na-CMC berdasarkan Farmakope Indonesia, dan JECFA. Diperoleh rendemen α-selulosa hasil isolasi dari eceng gondok Jatinangor sebesar 20,80% dan Lembang sebesar 18,60% dengan kadar α-selulosa masing-masing 76,53% dan 71,35%; hasil sintesis diperoleh rendemen Na-CMC sebesar 96,87% dan 85,03%; penambahan crosslinker menghasilkan rendemen Na-CMC sebesar 439,14% dan 221,81%. Hasil karakterisasi menunjukkan bahwa Na-CMC telah memenuhi persyaratan organoleptis, kelarutan, foam test, pembentukan endapan, viskositas dan kandungan logam berat. Beberapa parameter lain seperti susut pengeringan, kandungan natrium dan derajat substitusi belum memenuhi persyaratan, namun mempunyai nilai yang lebih baik dibandingkan dengan Na-CMC baku. Uji statistika komparatif menunjukkan adanya perbedaan yang signifikan pada nilai WHC namun tidak terdapat perbedaan yang signifikan pada nilai OHC dari Na-CMC hasil sintesis dari tanaman eceng gondok yang tumbuh di Jatinangor dan Lembang.

Abstract

Atenolol is a cardiovascular drug that has a narrow therapeutic index with long-term use and it’s often used as doping. Atenolol has a small concentration in human boby and it’s in  biological matrix (serum) so in the testing need a selective extraction so  the analyte can be pra-concentration and removed from matrix. Two molecularly imprinted polymers (MIPs) on propanol as porogen  have been made with two different methods i.e. bulk polymerization and precipitation polymerization. The polymer was made using atenolol as a template, methacrylic acid as a functional monomer, and ethylene glycol dimethacrylate as a crosslinker. Prformance evaluations showed that polymers from bulk polymerization provide better performance than polymers from precipitation polymerization when tested against standard solution. However, this sorbent has low  recovery percentage after applied into serum sample and could not be used as alternative for atenolol extraction in human serum.Key words: Molecularly imprinted polymer, Atenolol, Solid Phase Extraction, Preparation  method, propanol.

Abstract

 Akhir-akhir ini di pasaran farmasi beredar produk gel antiinflamasi golongan NSAID diklofenak dengan bahan aktif yang berbeda. Beberapa produk dibuat menggunakan bahan aktif garam natrium diklofenak, sedangkan industri farmasi lainnya menggunakan bahan aktif garam dietilamin diklofenak. Kedua bahan aktif memiliki perbedaan sifat dan karakter terutama dari sifat kelarutannya dalam air. Tujuan dari penelitian ini adalah untuk mempelajari kemampuan penyerapan (permeabilitas) dari kedua zat aktif tersebut. Metode penelitian meliputi pembuatan gel dengan basis gel HPC (Hidroksi Propil Selulosa), evaluasi sediaan yang meliputi: organoleptis, kadar, pH, dan viskositas. Sediaan gel yang telah memenuhi persyaratan kemudian diuji lebih lanjut permeabilitasnya secara in vitro selama 32 jam dengan sel difusi Franz dan sisa kulit ular phyton (Python reticulates) sebagai membrannya. Hasil evaluasi kualitas sediaan gel  menunjukkan bahwa kedua formula gel dari bahan aktif yang berbeda memiliki kualitas yang memenuhi persyaratan sebagai sediaan gel yang baik. Sedangkan hasil uji permeabilitas in vitro memperlihatkan bahwa kedua bahan aktif memberikan profil yang berbeda danjumlah keterserapan bahan yang berbeda. Dimana setelah 32 jam uji difusi diperoleh sebanyak 20% natrium diklofenak dan 15,5% dietilamin diklofenak. Terlihat bahwa profil dari dietilamin diklofenak memiliki efek depo, sehingga dosis yang digunakan lebih efektif untuk jangka waktu lama.  Kata kunci: Gel, dietilamin diklofenak, natrium diklofenak, permeabilitas

Abstract

The biggest case of death in 2018 is caused by lung cancer. Non-small cell lung cancer (NSCLC) is most common. One of the cause lung cancer is the over expression of EGFR. Erlotinib is the first line of anticancer for NSCLC with EGFR mutations. However, erlotinib can cause side effects such as liver damage therefore new safe anticancer is needed. Trigonelline is an alkaloid compound from coffee beans that had anticancer activity in pancreatic cancer cells by inhibiting Nrf2 in vitro and in vivo at concentrations of 0.1-1 µM. Development of cancer cells by Nrf2 is regulated by EGFR. In this study screening and modification of trigonelline structure was carried out to obtain compounds that have anticancer activity on NSCLC against EGFR computationally. The research procedures carried out are modification of ten trigonelline derived structures, the molecular docking and prediction of physicochemical profiles from trigonelline and its modification also their ADMET. Based on results, KF9 has the lowest free energy of binding which was -8,88 kcal/mol and binds to Met769 which has biological activity with receptor. KF9 has good physiochemical profile and absorption, distribution, also toxicity parameters. Abstract

Telah dilakukan dan divalidasi metode analisis glibenklamid dalam plasma menggunakan kromatografi cair kinerja tinggi fase terbalik dengan pretreatment menggunakan SPE C-18 dan SPE-MIP monomer akrilamid. 1 mL metanol dan 1 mL akuabides digunakan sebagai conditioning agent, 1 mL metanol 5% dalam akuabides digunakan sebagai washing agent, dan 1 mL asetonitril digunakan sebagai eluting agent. Hasil ekstraksi dianalisis menggunakan kolom C18 (Shimadzu) 150 x 4,6 mm, ukuran partkel 5 µm, fase gerak 55:45  v/v asetonitril dan triflouro acid 0,1% dengan kecepatan alir 1 mL/menit, deteksi dilakukan pada 227 nm dengan standar internal gliklazid. Metode analisis divalidasi berdasarkan parameter linieritas, presisi, akurasi, selektivitas/spesifisitas, Limit of Detection (LOD), Limit of Quantification (LOQ) dan kesesuaian sistem. Dapat disimpulkan bahwa metode analisis yang digunakan memiliki validitas sesuai dengan yang dipersyaratkan dan SPE-MIP monomer akrilamid memberikan hasil ekstrasi yang lebih optimal dibandingkan dengan SPE C-18.

Abstract

Antacids dosage form are a class of drugs used to chemically bind and neutralize stomach acid. Simethicone is one of the antacids that works as an antiflatulant. In an effort to guarantee the quality of a preparation, a valid analysis method is needed that meets the validation parameters. This study aims to modified of extraction method of the simethicone content of suspension samples using the FTIR method. The methods include simethicone modification of extraction from suspension preparations with toluene solvents and hydrochloric acid (2:5) 3.35. Validation methods include: determination of precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ) at wave number 1261 cm-1 and determination of the content of samples of simethicone suspension. The results showed that linearity parameters with a correlation coefficient of 0.997 in a concentration range of 1000-10,000 ppm, precision with a value of KV <2%, LOD and LOQ, respectively 378.97 and 1250.59 ppm. The results of the determination of simethicone suspension samples indicate that this method met the requirements of the validation parameters.Keywords: Validation, Simethicone, Infrared Spectrophotometry, antacid

Abstract

Diazepam is a benzodiazepine class usage is regulated nationally or globally. These drugs can be used to regulate sleep patterns or sedative-hypnotic drugs because it has a calming effect. Monitoring the abuse of diazepam requires a sensitive analysis methods that required the preparation of selective methods. One of the methods that are currently developing is Moleculary Imprinted Polymer-Solid Phase Extraction (MI-SPE). In this research, step being performed include determination of the association constant, synthesis sorbent of Imprinted Polymer (IP) diazepam by bulkpolymerization method with diazepam as a template, chloroform as porogen, two kind of monomer methacrylic acid (MAA) and hydroxyethyl methacrylate (HEMA) as functional monomer. Templates extracted from polymer followed by evaluation of adsorption ability test, a test of capacity and selectivity of the polymer, and also the characterization of the IP using Fourier Transform Infra-Red (FTIR),and Scanning Electron Microscope (SEM). The results showed that IP diazepam with MAA monomer has 7.95 x 10-5 mg/g of capacity, it is better than capacity of IP diazepam monomer HEMA with 7.47x 10-6 mg/g. Selectivity of IP diazepam MAA monomer has an IF value which is 2.13, which is better than IP HEMA with 1.21. In addition, IP diazepam MAA monomer more heterogenous than its Non Imprinted Polymer (NIP).Keywords: Chloroform, Diazepam, HEMA, Methacrylic acid, MI-SPE.

Abstract

Warfarin is an oral anticoagulant that has been widely used and has strong efficacy, but the use of warfarin is still a concern because of its narrow therapeutic index which cause interactions when co-administration with drugs, herbs or food. This interaction can affect the pharmacokinetics and pharmacodynamics of warfarin and the most fatal effect from warfarin interactions is bleeding. In this review article data on warfarin-herbs interactions were collected based on pharmacokinetic parameters (AUC0-∞, Cmax, T1/2, Cl/F, and V/F), while pharmacodynamic parameters (International normalized ratio (INR), platelet aggregation, AUC INR and Protombine Time). As a result some herbs had significant interactions with warfarin. Herbs that affect warfarin pharmacokinetic were Danshen gegen, echinacea, St. John's wort and caffeine and herbs that affect pharmacodynamic were policosanol, Ginkgo biloba, cranberry, St. John's wort, ginseng, pomegranate, Psidium guajava and curcumin, so co-administration warfarin with herbs need to be considered.Keywords: Warfarin, Interactions, Herbs, Pharmacokinetics, Pharmacodynamics

Abstract

Rhodamine B is a colouring agent for paint industry, textile and paper. It causes irritation at respiration systems and has a carcinogenic effect. At high concentration rhodamine B causes liver damages. Nowadays, it has been reported being added into cosmetics and food product such as lipstick, eye shadow, rouge, food paste, and kerupuk. This research was conducted to determine rhodamine B in the products by using HPLC and spectrophotometry UV-Vis. It was done through sample collection, sample preparation, validation of analytical method, and analysis of rhodamine B using HPLC and spectrophotometry UV-Vis. The results of HPLC and spectrophotometry UV-Vis showed that four samples of kerupuk (57,14%) and four samples of food paste (50%) contained rhodamine B. However, rhodamine B was not found in cosmetics. The results showed the urgency of fast methods to identify rhodamine, thus its content in food products is carefully controlled.
