Abstract

Identifikasi Polerovirus yang Menginfeksi Tanaman Cabai di Jawa Infeksi polerovirus pada tanaman cabai semakin banyak dilaporkan dari berbagai bagian dunia termasuk di Indonesia. Infeksi Pepper vein yellow virus (PeVYV) pada tanaman cabai di Bali, Jawa, dan Aceh telah dilaporkan pada kurun waktu 2012 sampai 2020. Belum lama ini gejala infeksi polerovirus berupa daun kuning dengan penebalan tulang daun ditemukan pada pertanaman cabai di wilayah Jawa Tengah, yaitu di Brebes, Magelang dan Kulon Progo. Deteksi molekuler dengan metode RT-PCR menggunakan primer universal polerovirus O3R/PL4F yang dilanjutkan dengan analisis sekuens membuktikan adanya infeksi PeVYV dari sampel asal Magelang dan Kulon Progo. Identifikasi lebih lanjut menunjukkan bahwa isolat PeVYV tersebut ialah PeVYV-2, PeVYV-6, dan PeVYV-8. Insidensi infeksi PeVYV ini menunjukkan perluasan distribusi Polerovirus di Indonesia.

Abstract

Graphical Abstract   Highlight Research Coral colonies affected by coral disease are identified and counted. White syndrome is a type of coral disease that is dominattly found. Coral cover in the Kepulauan Seribu is in moderate condition. A weak correlation between the prevalence of coral disease and live cover in the Kepulauan Seribu     Abstract Kepulauan Seribu is designated as a marine conservation area in Kepulauan Seribu National Park and is one of high tourism development areas on the north side of Jakarta, the capital city of Indonesia. Anthropogenic sources from these nearby areas has reduced the health condition of coral reefs and increased the prevalence of coral diseases in the study area. This study aimed to determine the genus of coral infected by disease, the type of disease, and the relationship between the prevalence of disease and live cover in Kepulauan Seribu. Data was collected using the survey method. Coral cover is assessed using Underwater Photo Transect (UPT) and disease is assessed using Belt Transect methods, respectively. The result showed that 14 genera of coral species were infected and dominated by Porites, Montipora, and Acropora. Furthermore, there were 5 types of diseases, dominated by White Syndrome (WS), Atramentous Necrosis (AtN) and Ulcerative White Spots (S). The correlation analysis results showed a very low relationship between disease prevalence and live coral cover (r = 0.10). This research can be used as input or reference for more sustainable coral reef management, by improving an integrated coastal education system for the younger generation and developing more sustainable tourism activities in the future in Kepulauan Seribu.

Abstract

Penyakit darah yang disebabkan oleh Ralstonia solanacearum Phylotype IV merupakan penyakit penting pada pisang yang menyebabkan kehilangan hasil hingga 100%. Bakteri R. solanacearum mengekspresikan gen virulensi melalui mekanisme quorum sensing (QS) dengan menggunakan asil homoserin lakton (AHL) sebagai autoinducer. Enzim AHL laktonase diproduksi oleh bakteri dan memiliki potensi sebagai agens biokontrol bakteri patogen melalui anti-QS. Tujuan dari penelitian ini ialah memperoleh isolat bakteri endofit penghasil AHL laktonase untuk pengendalian R. solanacearum pada tanaman pisang. Tahapan penelitian mencakup isolasi dan seleksi bakteri endofit asal tanaman pisang sebagai penghasil AHL laktonase dengan bioassay maupun molekuler, uji keefektifan isolat bakteri dalam menekan faktor virulensi R. solanacearum, uji keefektifan pengendalian R. solanacearum in planta, dan identifikasi molekuler isolat bakteri potensial. Isolasi bakteri endofit berhasil dilakukan dari sampel pisang asal Bogor di tiga kecamatan, yaitu Cisarua, Dramaga, dan Jonggol yang terdiri atas lima jenis pisang masing-masing menghasilkan 152, 204, dan 130 isolat bakteri endofit. Uji lebih lanjut menggunakan Chromobacterium violaceum sebagai bioindikator menunjukkan lima isolat dengan aktivitas anti-QS. Deteksi gen aiiA dengan PCR menggunakan primer spesifik menunjukkan bahwa kelima isolat memiliki amplikon berukuran 753 pb dan telah dikonfirmasi melalui sekuensing sebagai gen aiiA. Kelima isolat bakteri tersebut CBP1, DBT17, DBT7, DBK2, dan CBN13 mampu menekan pembentukan ekstraseluler polisakarida (EPS) yang dihasilkan R. solanacearum dengan persentase penekanan masing-masing sebesar 75.67%, 90.81%, 87.67%, 92.72%, 81.08%, dan mampu menghambat gejala layu sebagai salah satu gejala penyakit darah pada tanaman pisang. Hasil perunutan gen aiiA menunjukkan bahwa kelima isolat homolog dengan Bacillus cereus, B. thuringiensis, B. substilis, B. amyloliquefaciens.

Abstract

Used cooking oil is oil left over from frying that can no longer be used for frying because it has decreasednutritional value and has a negative impact on health. Used cooking oil can still be used as an example as araw material for making soap after being treated with used cooking oil. This study aims to make soap fromused cooking oil as raw material after being adsorbed using activated carbon from oil palm shells. The stagesof this research are 1). Making activated carbon from oil palm shells, 2). Applying activated carbon frompalm oil shells to used cooking oil, 3). Making soap from used cooking oil that has been adsorbed withactivated carbon from oil palm shells and 4). Testing the quality of soap, namely moisture content, soapingnumber and foam quantity. The results showed that the characteristics of the soap produced with an averagewater content of 7.5% (SNI 196-206 Max 15%), Sapling Number 196.90 mg / gr (SNI 196-206 mg/gr) and theamount of foam 1, 58 ml. Soap made from used cooking oil as raw material that has been adsorbed usingactivated carbon from oil palm shells in accordance with SNI 196-206 mg/gr.

Abstract

This study was initiated by the fact that Elementary School (SD/MI) teachers had not optimally utilized learning media, especially interactive media, in teaching fractions. Meanwhile, the rapid development of technology requires teachers to adapt and make use of technological advancements to support learning both inside and outside the classroom. The purpose of this study was to develop an effective, practical, interactive, and innovative learning media for mathematics learning, particularly on the topic of fractions. This study employed a research and development design. The results showed that Virtual Smart Fraction was highly feasible and effective as a learning medium. Its feasibility was supported by validation results from media experts with a score of 93.33% and from material experts with a score of 96.25%. Furthermore, its effectiveness was demonstrated through a small group test yielding 96.36% and a large group test yielding 95.14%. In addition, Virtual Smart Fraction improved the students’ average score from 62.58 to 89.82. Therefore, Virtual Smart Fraction was categorized as valid, highly feasible, and highly effective as a learning medium for mathematics learning.Keywords: ,
