Abstract

The objective of this study was to select and characterize three yeast isolates originating from soil of  Raja Ampat region of Papua, Indonesia for its potential to produce cellulase . Selection and characterization of cellulolytic yeast was carried out by measuring cellulolytic Index (IS) with congo red method and measurement of Carboxy Methyl Cellulase (CMC-ase) activity through determination of reducing sugar with dinitrosalycilic methods. Cellulolytic Index (IS) of the isolates  Sporobolomyces poonsookiae Y08RA07,  Rhodosporidium paludigenum Y08RA29 and Cryptococcus flavescens Y08RA33were 1.40, 2.60 and 1.66 respectively. CMC-ase produced optimum at pH 8 at 37ºC by isolate Y08RA07, whereas for Y08RA29 andY08RA33 were at pH 6, at 28ºC. Paper waste was good substrate for cellulase enzyme production by isolate Y08RA07, while for two other isolates the best substrate was CMC. Isolate Y08RA29 having highest cellulase activities when grown in CMC, while isolates Y08RA07 and Y08RA33 achieved highest enzyme activity when grown in bamboo leaf.   Key words: Cellulolytic yeast, Raja Ampat, waste paper, bamboo leaf

Abstract

Endophytic fungi play an important role on mangrove growth and development, however research on the endophytic fungi of the mangrove Rhizophora apiculata is limited. The endophytic fungi produce diverse bioactive compounds involved in the mangrove’s adaptation to varied biotic and abiotic stresses and could have applied uses in agriculture or medicine. The purpose of this research was to isolate endophytic fungi of Rhizophora apiculata mangrove and to study their activity as herbicides on the weed Gomphrena globosa. The fungi were isolated using the surface sterilization method and identified based on morphological characteristics and molecular characteristics using ITS regions of rDNA. Herbicidal activity of the fungal filtrates extracted by ethyl acetate were tested on seed germination and seedling growth of G. globosa. Five fungal isolates were obtained, namely Penicillium citrinum, Diaporthe eucalyptorum, Diaporthe musigena, Colletotrichum queenslandicum, and Diaporthe tectonae. All isolates were able to grow on PDA medium containing 0, 25, 50, 75, and 100 (% v/v) seawater concentrations, but the growth rate varied by species and seawater concentration. In general, all five isolates showed herbicidal activity by delaying seed germination and reducing shoot and root growth. P. citrinum showed the highest herbicidal activity compared to the other isolates. Analysis using Gas Chromatography-Mass Spectrometry of the crude extract of P. citrinum filtrate identified 7 main compounds: 3-Methoxy-2-methyl-cyclohex-2-enone, Cyclohexane-carbohexaldehyde, 6-methyl-3-(1-methylethyl)-2-oxo, Cyclopropane carboxylic acid, 1-(2-propenyl)-1,1-dimethylethyl ester, 2-Hydroxy-4-isopropyl-7-methoxytropone, Beta-Asarone, Oxane, 4-(2-amino-1-hydroxyethyl)-4-(3,4-dimethoxyphenyl)-, and Diisooctyl-phthalate. These compounds should be studied further to determine which ones are responsible for the herbicidal activity.

Abstract

PERTUMBUHAN TANAMAN JARAK  PAGAR (Jatropha curcas L.) PADA LAHAN PASCA TAMBANG TIMAH DI BANGKA YANG DIBERI PUPUK ORGANIK Sukmarayu P. Gedoan1), Alex Hartana2), Hamim2), Utut Widyastuti2) dan Nampiah Sukarno2) 1)Program Studi Biologi, FMIPA Universitas Negeri Manado di Tondano; 2)Departemen Biologi, FMIPA, Institut Pertanian Bogor, Bogor 16680   ABSTRAK Kegiatan penambangan timah menyebabkan perubahan karakteristik fisika dan kimia tanah sehingga menjadi tidak sesuai untuk pertumbuhan tanaman.  Tujuan penelitian ini untuk mempelajari pertumbuhan 7 aksesi jarak pagar yang ditanam pada lahan pasca tambang timah yang diberi kompos dan kotoran sapi.  Penelitian ini dilakukan di TSS 133, Kelurahan Sinar Baru, Kabupaten Bangka, Provinsi Bangka Belitung.  Penelitian lapangan dilaksanakan pada Bulan Mei 2007 sampai dengan Bulan Desember  2008.  Percobaan faktorial ini dirancang dalam Rancangan Petak Terbagi dengan tiga ulangan.  Petak utama adalah 7 aksesi jarak pagar yang terdiri atas: aksesi Madiun, Ponorogo, Jember, Dompu, Lampung, Bengkulu,  dan Sukabumi, sedangkan  anak petak berupa tanah tanpa pemberian kompos dan pupuk kandang (kontrol), kompos trubus 4 kg/lubang ditambah 4 kg tanah bagian atas  dan  kotoran sapi 4 kg/lubang ditambah 4 kg tanah bagian atas. Hasil penelitian menunjukkan bahwa pertumbuhan awal yang paling baik untuk tanah bekas tambang timah adalah pada aksesi Jember yang ditunjukkan oleh diameter batang, berat kering tanaman, berat kering tajuk dan berat kering akar.  Sedangkan tinggi tanaman tertinggi diamati pada aksesi Madiun.  Penambahan kotoran sapi dapat meningkatkan produksi biji dan kandungan minyak.  Produksi biji tertinggi diperoleh pada aksesi Bengkulu dan kandungan minyak tertinggi diperoleh pada aksesi Dompu. ON THE POST-TIN-MINING LAND IN BANGKA PROVIDED WITH ORGANIC FERTILIZER ABSTRACT The activity of tin mining changes the physical and chemical characteritics of soil, so that the soil isnot suitable for plant growth.  The objective of this research was to evaluate the growth of some accession of Jatropha curcas that we planted on post-tin-mining land provided with compost and cow feces.   This research was conducted in TS 133, Sinar Baru Village, District of Bangka, Province of Bangka Belitung.  This field research was carried out in May 2007 to December 2008.  Factorial experiment was designed as split plot with three replications.  The main plot was 7 accessions, i.e. Madiun, Ponorogo, Jember, Dompu, Lampung, Bengkulu, and  Sukabumi.  The subplot waskonds of organic fertilizer, i.e. top soil without addition of compost and cow fecer (control), trubus compost  (4 kg/hole) added with top soil (4 kg/hole), and cow feces (4 kg/hole) added with topbsoil (4 kg/hole). The result showed that Jember accession had the best early growth based on the observation of stem diameter, plant dry weight, and root dry weight.  The largest plant height was observed in Madiun accession.  The higest seed production was observed in Bengkulu accession and Dompu accession had the higest oil content.Keywords: accession, Jatropha curcas, , compost, cow feces

Abstract

Tristaniopsis merguensis (pelawan tree) is a potential plant, however, it has not cultivated and conserved optimally due to slow growth and lack of information about the cultivation. The existence of T. merguensis in the forest is important because pelawan fungi grow under the tree, particularly in the above of the roots. Information about how to grow the T. merguensis seedling is very limited; therefore, research in growth and development of T. merguensis seedling was important. The aims of this study were to determine the appropriate planting medium for T. merguensis seed germination and to stimulate growth and development of T. merguensis seedling by using its fungi. Status of T. merguensis fungi was studied by analyzing root morphology. Fungi isolation was carried out from colonized root and fungi fruit body. The stimulated the growth of T. merguensis seedling was done by using fungi isolated from colonization root in laboratory scale, as well as treated at a different phosphate concentration. The result showed that T. merguensis was associated with ectomycorrhizal fungi. Planting medium consists of sawdust and T. merguensis fine root resulted the highest percentage of germination. Seedlings were treated with ectomycorrhizal fungi grew better than without ectomycorrhizal fungi. In addition, seedlings treatment with ectomycorrhizal fungi and phosphate 25% showed the highest growth rate.

Abstract

Arbuscular Mycorrhizal Fungi (AMF) associate with a variety of plants including forest trees. Research on AMF in forestry in Indonesia is limited, especially on woody plants grown in Bangka Island. Therefore, the aim of this research was to isolate and identify AMF associated with woody plants from Bangka Island grown in the post-mining soil in greenhouse for 7 years. The Soil samples derived from 8 pots of the 7 years old woody plants were used as spore sources. Spore extraction from the soil used the wet sieving and decanting method. Fungal identification was carried out based on morphological characteristics, and fungal isolation used pot culture with Pueraria javanica as a host plant. Fungal structures within the root were analyzed after root staining with trypan blue. There were 18 spore morphotypes observed which belong to 5 types of Acaulospora and 13 types of Glomus. Seven single spore pot cultures were successfully isolated from species of Acaulospora sp.1, Acaulospora sp.4, Acaulospora sp.5, Glomus sp.2, Glomus sp.3, Glomus sp.6, and Glomus sp.11. AM fungal colonization structures observed within the root were Paris type arbuscules, internal hyphae, and vesicles. The AMF cultures obtained could be used as biofertilizer for woody plant seedlings production for post mining reclamation activities.

Abstract

The Trichoderma hamatum produces various secondary metabolites that can be used as Candida albicans.  This research aimed to isolate T. hamatum from the coffee (Coffea canephora) rhizosphere and analyze the fungal compounds to control the pathogenic fungus C. albicans. T. hamatum was isolated using the dilution method, and the fungal identification was used, combining morphological and molecular characteristics of ITS rDNA. The potency of T. hamatum as C. albicans was determined by antagonist test using the double-layer method, while for culture filtrate, ethyl acetate and n-hexane filtrate extracts were carried out by the agar diffusion method. The compounds in the most active extract were analyzed by Gas Chromatography-Mass Spectrometry (GC-MS). T. hamatum inhibits C. albicans growth in antagonistic and filtrate with 15.53 mm and 14.40 mm of inhibition zone. It indicated that both culture and fungal filtrate had similar activities on C. albicans. The ethyl acetate extract (minimum inhibitory concentration, MIC of 0.50%) showed more potent against C. abicans than n-hexane extract (MIC of 15.00%). The potential active compound in the ethyl acetate extract would be 9-Octadecenoic acid (Z) methyl ester. The 9-Octadecenoic acid (Z) methyl ester could be used as an alternative candidate to control Candida.

Abstract

Orchids form a symbiotic relationship with endophytic fungi throughout their life cycle, including their seed germination. Research on orchid endophytic fungi in Indonesia is still limited. This study aimed to isolate and identify endophytic fungi of Phalaenopsis amabilis, Dipodium brevilabium, Vanilla planifolia, and Aerides odorata orchids. Endophytic fungi were isolated from surface-sterilized orchid root pelotons. Fungal identification was based on morphological and molecular characteristics using ITS rDNA sequences, followed by BLAST and phylogenetic tree analysis. All orchids studied had peloton structures in root cortex tissues. From the four orchids studied, 22 fungal isolates were obtained. Five fungal isolates were obtained from the P. amabilis orchid, consisting of 1 Penicillium and 4 sterile mycelium isolates. The D. brevilabium had 7 fungal isolates, namely 1 Penicillium and 6 Fusarium isolates. The V. planifolia had 6 Fusarium isolates, whereas the A. odorata had 4 isolates, such as 1 Penicillium, 2 sterile mycelium, and 1 unidentified isolate. The genus Penicillium was found in 3 out of the 4 orchid species studied. One Penicillium isolate was molecularly identified as Penicillium citrinum. Endophytic fungi in P. amabilis were mostly sterile mycelium, while D. brevilabium, V. planifolia, and A. odorata were dominated by Fusarium.

Abstract

Sponge-associated marine fungi are potential source for secondary metabolite compounds. The aim of this research was to investigate sponge-associated marine fungus as secondary metabolite producers against Escherichia coli. The fungus was isolated from Indonesian marine sponge Stylissa sp. and identified as Aspergillus hortai through a combination of morphological and molecular characteristics of ITS DNA and β-tubulin genes. The fungus was tested against E. coli using fungal broth and mycelial extracts. The optimized condition was achieved by fungal broth grown in corn meal broth at 6-days of shaking incubation. Fungal extract was produced using three liters of filtered fungal broth and extracted in ethyl acetate. The antibiotic activity of the extract is vulnerable to 45°C heat and basic or acidic conditions. Therefore, the extraction was done at pH 7 with evaporation at 40°C. The extract shows 7 major bands on TLC with 1 band shows activity against E. coli (Rf 0.81) on bioautogram. The band was observed as a yellow color and turned black in short-wave UV and did not show any fluorescence in long-wave UV. This research shows that sponge-associated marine fungi obtained from Indonesia has the potential as anti E. coli worth to be explored for searching new antibiotics.

Abstract

Endophytic fungi isolated from red ginger (Zingiber officinale) can inhibit growth of Fusarium oxysporum, but the ability of the fungi to control Candida albicans as human pathogen has not been reported. The aims of this research were to study the mechanism of ten endophytic fungi isolates derived from red ginger to control C. albicans in vitro using dual culture methode and fungal extract, and to determine fungal bioactive chemical groups produced by the fungi. Three out of ten isolates tested, Talaromyces assiutensis JMa 7, T. assiutensis JMbt 3, and Curvularia affinis JMbt 9 inhibited growth of C. albicans with inhibition zones were 4.0 mm, 4.9 mm, and 11.3 mm, respectively. The cultures of the three potential endophytic fungi were extracted by maceration method using 3 solvents i.e ethyl acetate, n-hexane and ethanol. The three isolates were grown in PDB separately for 21 days incubation. At harvest, the culture filtrate was extracted by ethyl acetate and n-hexane, while fungal mycelia were extracted by all the three solvents. Ethyl acetate extracts obtained from culture filtrate of all the three fungal isolates consistently inhibited C. albicans with inhibition zones were 2.0-3.8 mm. For n-hexane extract, however, only Talaromyces assiutensis JMbt 3 that had positive effect with inhibition zone was 2.0 mm. All extracts from mycelia did not have any effects on C. albicans. The ethyl acetate extract of T. assiutensis JMbt 3 was analysed to determine its chemical groups using visible color on thin layer chromatography (TLC). The results showed that the bioactive compounds was terpenoids, and antioxidant.
