Abstract

Hyperglycemia is an increase in glucose levels in the blood. This condition can trigger glucose autooxidation which can produce free radicals. Free radicals can cause damage to male reproductive organs and quality. Tamarind fruit has a high flavonoid which has the potential as an antioxidant. This study aims to analyze the effect of tamarind fruit nanoparticles on spermatozoa quality in hyperglycemic rats. This study is experimental with Post-Test Control Group Design. A sample of 15 male Wistar rats were induced by alloxan and then divided into three groups, namely the control group, tamarind fruit nanoparticles with a dose of 30 mg/KgBB (NP1) and 60 mg/KgBB (NP2) for 28 days. Sperm quality was observed with parameters of concentration, motility and viability. The data were analyzed using One Way ANOVA and LSD. The results of the Anova test showed that tamarind nanoparticle treatment could significantly improve sperm quality (P<0.05). The LSD test showed that the concentration, motility and viability of sperm in group (K) were significantly different from groups NP1 and NP2. The highest to lowest average sperm concentration in sequence was in groups NP2 (49.60x106±6.06x106), NP1 (41.60x106±7.40x106) and K (20.80x106±7.56x106). The highest to lowest average percentage of sperm motility in sequence was in groups NP2 (82.20±9.75), NP1 (74.00±4.74), K (47.20±10.66). The highest to lowest average percentage of sperm viability in sequence was in the NP2 group (76.80±4.49), NP1 (72.20±7.12), K (31.60±7.89). The conclusion of this study is that tamarind nanoparticles have an effect on improving sperm quality in hyperglycemic rats.

Abstract

Background: Paracetamol is one of the analgesics and antipyretics that are widely traded freely, so it is possible that the use is not according to the recommended dose and consumption time. This can trigger liver damage and changes in blood biochemistry. Green tea has components such as antioxidants, anti-inflammatories, and hepatoprotectors. The study aimed to analyze the effect of green tea extract on the blood biochemistry of rats induced by paracetamol. Methods: Laboratory experimental research with the Post-Test Control Group Design method, using 25 male Wistar rats, applying random sampling. Twenty-five rats were divided into 5 groups: K- (control), K+ (positive control), and treatment groups with green tea extract, respectively 175 mg (T1), 350 mg (T2), and 700 mg/kg bw (T3) for 24 days. The treatment group and K+ received paracetamol 600 mg/kg bw on the 14th day for 10 days, and green tea extract was still given. Data, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), triglycerides (TG), total cholesterol, and leukocyte counts, were analysed using ANOVA and the Least Significant Difference (LSD) test. Results: GTE significantly (p-value <0.05) reduced leukocytes and AST, but inadequate paracetamol induction failed to elevate ALT and lipids, masking GTE’s measurable impact on those specific biochemical parameters. Conclusion: Providing green tea extract was effective in declining the number of leucocytes and AST levels. The study concluded that green tea extract has the potential to influence the biochemical profile of rats induced by paracetamol. Further research is needed regarding the dose and period of administration and exposure.

Abstract

Hyperlipidaemia is a condition of increased lipid levels, especially cholesterol, triglycerides, LDL and an abnormal decrease in HDL in the blood. Tamarind leaves contain flavonoid and quercetin compounds that have the ability as antihyperlipidemia agents. The purpose of this study was to examine the effect of tamarind leaf ethanol extract on the lipid profile of hyperlipidaemia rats. Samples of 16 male Wistar rats were divided into 4 groups; Control group and the tamarind leaf ethanol extract treatment group was divided into 3, namely P1 350 mg/kgBB, P2 500 mg/kgBB, and P3 650 mg/kgBB. All rats were given lard 3 mL/head/day for 2 weeks. In weeks 3 and 4, rats were given extract of Tamarindus indica leaves and 2 hours interval was given lard. The data obtained were statistically analysed using one-way ANOVA and the Least Significant Difference (LSD) test at the 95% confidence level. In this study, HDL levels decreased along with reductions in total cholesterol and triglycerides, but had no effect in reducing LDL levels. Reduced cholesterol synthesis limits substrate availability for HDL formation, resulting in proportional decreases in HDL-C. The conclusion of this study is that tamarind leaf ethanol extract has an effect in reducing lipid profile levels.

Abstract

Hyperglycemia is a condition characterized by elevated blood glucose levels that induce free radical formation, leading to reduced spermatozoa quality. Butterfly pea (Clitoria ternatea) contains antioxidant compounds and is formulated as a nanoherbal to enhance bioavailability. This study aimed to evaluate the potential of butterfly pea nanoherbal in improving spermatozoa quality in hyperglycemic rats. This experimental study employed a post-test only control group design using 25 male Wistar rats divided into a healthy control group and hyperglycemic groups induced by alloxan (125 mg/kg BW). The hyperglycemic groups consisted of a metformin control (400 mg/kg BW) and butterfly pea nanoherbal treatment groups at doses of 12.5, 25, and 50 mg/kg BW administered for 28 days. Data were analyzed using One-Way ANOVA followed by the Least Significant Difference (LSD) post hoc test. The results showed that spermatozoa concentration in the healthy control group differed significantly from the positive control, P1, and P2 groups (p<0.05), but not from the P3 group. Spermatozoa motility and viability also showed significant differences among groups (p<0.05). It can be concluded that butterfly pea flower nanoherbal significantly improves spermatozoa quality in hyperglycemic rats.

Abstract

Background: Hyperglycemia induces excessive production of reactive oxygen species (ROS), leading to oxidative stress and lipid peroxidation characterized by increased malondialdehyde (MDA) levels. This study evaluated the effectiveness of butterfly pea flower nanoherbal formulated as alginate-based nanoparticles using the ionic gelation method in reducing blood glucose and MDA levels in hyperglycemic rats. Methodology: 25 male Wistar rats (Rattus norvegicus) were divided into five groups: normal control (NC), metformin control (MC), and three nanoherbal treatment groups (NT1, NT2, and NT3) receiving doses of 12.5, 25, and 50 mg/kg body weight (BW), respectively. Hyperglycemia was induced by a single intraperitoneal injection of alloxan monohydrate (125 mg/kg BW). Blood glucose was measured on days 1, 14, and 28, while MDA levels were analyzed on day 28 using the TBARS method. The analysis of blood glucose levels was conducted using a Repeated Measures Two-Way ANOVA followed by a Bonferroni post hoc test, while MDA data were evaluated through a One-Way ANOVA with Duncan’s multiple range test (p < 0.05). Findings: A significant, dose-dependent reduction in blood glucose levels was observed following nanoherbal treatment. The NT3 group (50 mg/kg BW) showed the greatest reduction, with mean glucose decreasing from 471.4 ± 142.00 mg/dL on day 1 to 146.4 ± 68.58 mg/dL on day 28. MDA levels also differed significantly, with NT3 showing 2.84 ± 0.19 nmol/mL, indicating lower oxidative stress. Contributions: These findings indicate that alginate-based butterfly pea nanoherbal can reduce blood glucose and MDA levels in hyperglycemic rats, suggesting its potential role in improving glycemic control and reducing oxidative stress under hyperglycemic conditions
