Abstract

Background: The main principle in treatment of wounds is infection control by using antiseptic. Gambier (Uncaria gambir [Roxb.]) containing catechins and tannins reported that has an antiseptic effect. This aim of this study was to determine the effect of gambier extract as antiseptic on gingival wound in rats.Methods: In vivo study, used pretest-posttest only control group design had been conducted in Animal house, Medical Faculty of Sriwijaya University and Province’s Health Laboratory in Palembang. There were 30 white Wistar rats divided into fve groups. Group 1 was given 10% povidone-iodine ointment (positive control), group 2 was given a placebo ointment (negative control), group 3, 4, and 5 were given an ointment with 10%, 15%, and 20% gambier extracts. Gingival labial wound of the mandible was induced with cylinder diamond bur, then swabbed before and after treatment. Gingival swab samples were cultured in agar medium and incubated for 24 hours. The number of bacterial colonies from all groups were counted by colony counter. The statistical analysis was used IBM SPSS statistics version 22,0.Result: The result showed that the number of bacterial colonies from all groups decreased signifcantly after treatment, except for negative control. The higher concentration of gambier extract led the better effect of antiseptic.Conclusion: It can be concluded that gambier extract has antiseptic effect of gingival wound in rats.

Abstract

Background. Dental and gum disease is a health problem faced by many countries around the world. Dental and gum diseases begin with the formation of plaque on the tooth surface, as a result of the activities of bacteria in the oral cavity. Elimination of the bacteria as caries causal has been done in many ways, one of them by using plant extracts which have anticariogenic activities, such as gambir (Uncaria Gambir [Roxb.]). The aim of this study was to determine the effect of chewing candy containing gambir on the decrease of bacterial colonies and dental plaque formation. Methods. A Pretest-Posttest Control Group Design had been conducted in SMPN 17 of Junior High School in Palembang and Province’s Health Laboratory of South Sumatera, Indonesia. The population consisted of 50 participants, divided into 5 groups. Each group was chewing with 5%, 7.5%, 10% extract-gambir candies, no chewing candies, and chewing sucrose’s candies. Data were analyzed by using SPSS vers 22. Result. The results showed that all candies containing gambir extract were able to reduce the formation of dental plaque and to decrease the bacterial colonies in saliva. The best effect of this study was 10% extract-gambir candies. Conclusion. It can be concluded that extract-gambir candies were effective in the decline of dental plaque formation and bacterial colonies.

Abstract

Lesi ulserasi merupakan penyakit jaringan lunak mulut yang banyak dijumpai. Pendekatan perawatan terhadap lesi ulserasi dengan menggunakan bioadhesif bertujuan untuk mendapatkan efek terapi maksimal dari suatu zat aktif. Tanaman gambir (Uncaria gambir) adalah suatu jenis tumbuhan yang banyak dikembangkan sebagai obat herbal karena memiliki senyawa aktif seperti katekin. Penelitian ini bertujuan untuk mengevaluasi efek mukobioadhesif ekstrak gambir terhadap penurunan rasa nyeri dan durasi penyembuhan pada lesi ulserasi rongga mulut. Desain penelitian ini adalah pretest-posttest control group. 24 sampel dibagi menjadi dua kelompok, kelompok yang diberi mukobioadhesif ekstrak gambir dan kelompok yang diberi mukobioadhesif plasebo selama 7 hari dengan aturan pakai 3 kali sehari. Tingkat keparahan rasa sakit yang diukur dengan skala VAS, luas lesi ulserasi, dan durasi penyembuhan dievaluasi dan dianalisis dengan SPSS ver. 22. Hasil penelitian menunjukkan bahwa penggunaan mukobiadhesif ekstrak gambir mampu mengurangi rasa sakit yang ditandai dengan penurunan skor VAS secara signifikan (p0,05) dan luas lesi yang ditandai dengan menurunnya rata-rata diameter lesi, serta mampu menurunkan durasi penyembuhan secara signifikan dibandingkan dengan plasebo. Dapat disimpulkan bahwa penatalaksanaan mukobioadhesif ekstrak gambir berpotensi dalam mengurangi rasa sakit dan mempercepat penyembuhan lesi ulserasi rongga mulut.

Abstract

ABSTRAKJumlah Candida albicans di dalam rongga mulut dapat mengalami peningkatan akibat penggunaan alat ortodonti cekat. Obat kumur dengan kandungan antijamur dapat digunakan untuk menurunkan jumlah C. albicans di dalam rongga mulut. Ekstrak jahe telah dilaporkan dapat menurunkan jumlah C. albicans, namun bila dibandingkan dengan klorheksidin masih ada perbedaan hasil terhadap efektivitasnya. Penelitian ini bertujuan untuk membandingkan jumlah C. albicans setelah berkumur ekstrak jahe dan klorheksidin sebagai kontrol pada pengguna ortodonti cekat. Studi ini merupakan penelitian eksperimental klinis, yang melibatkan 30 subjek yang dibagi menjadi kelompok yang berkumur dengan ekstrak etanol jahe 30% dan kelompok yang berkumur klorheksidin glukonat 0,2%. Bahan kumur digunakan selama 14 hari dengan frekuensi 2 kali sehari. Saliva tanpa stimulasi dikumpulkan dengan metode spitting pada hari baseline, hari ke-7, dan hari ke-14. Sampel saliva dibiakkan kemudian dilakukan penghitungan jumlah koloni C. albicans. Hasil uji t berpasangan menunjukkan jumlah koloni C. albicans menurun secara signifikan pada kedua kelompok setelah menggunakan obat kumur selama 7 hari dan 14 hari (p0,05). Hasil uji t tidak berpasangan menunjukkan tidak terdapat perbedaan signifikan dari jumlah koloni C. albicans antara kedua kelompok (p0,05). Dapat disimpulkan bahwa berkumur dengan ekstrak jahe dapat menurunkan jumlah C. albicans pada pengguna ortodonti cekat, sama baiknya dengan berkumur menggunakan klorheksidin.

Abstract

Salivary alpha amylase is a component in human saliva that plays important role in carbohydrate digestion. This article aims to analyze bioinformatics and biomolecular characterization of salivary alpha amylase as risk factor of dental caries. The location of this gene is 1p21.1, with nucleotide is NC_000001.11. This information was taken from National Center for Biotechnology Infor-mation's website. The number of amino acids of salivary alpha amylase is 511, with molecular weight is 577,677.82 amu. The pro-tein is stable with instability index is computed to be 23.58. Only 23 enzymes were predicted to be capable of cleaving the sa-livary alpha amylase out of a total of 37 protease. The aliphatic index is 67.12. From THMM analysis, salivary alpha amylase is found outside of membrane. It is concluded that characteristic of salivary alpha amylase can be considered as indicator for caries.

Abstract

ABSTRACTIntroduction: The biggest problem in oral health is dental caries and periodontal disease. The way to prevent dental and mouth problems is to keep oral hygiene. One of them is adequate plaque control. Plaque control can be done by brushing teeth and using toothpaste. Currently, many herbal ingredients have been used as antibacterials such as gambir (Uncaria gambir). Gambir mainly contains catechins and tannins, which have the ability to inhibit the growth of bacteria. The purpose of this study was to determine the antimicrobial potential of toothpaste containing gambir (Uncaria gambir). Methods: The total sample was 30, and it divided into three groups. Samples were toothpaste containing ten percent of Uncaria gambir extract, fluoride, placebo toothpaste, Streptococcus mutans (Gram-positive), Phorphyromonas gingivalis (Gram-negative), and Candida albicans. Antimicrobial activity of toothpaste with gambir (Uncaria gambir) extract against microorganisms was observed with the formation of inhibitory zone diameter in agar. Data were analyzed by using SPSS with one-way ANOVA and Tukey test. Results: The results showed that toothpaste containing gambir extract could inhibit the Streptococcus mutans (p<0.05), Phorphyromonas gingivalis (p<0.05), Candida albicans (p<0.05) compared significantly with placebo (p<0.05). Antimicrobial toothpaste containing gambir extract had no different effect from toothpaste containing fluoride. However, toothpaste containing gambir extract had significantly different effect than toothpaste containing fluoride. Conclusion: It can be concluded that toothpaste containing gambir (Uncaria gambir) extract has antimicrobial potential against Streptococcus mutans, Phorphyromonas gingivalis, and Candida albicans. These findings showed that the extract of Uncaria gambir possesses potent antimicrobial activity.

Abstract

Itraconazole merupakan salah satu obat sintetik yang memiliki keterbatasan dalam menghambat pertumbuhan Candida albicans (C. albicans). Kulit buah durian (Durio zibethinus L.) merupakan salah satu tanaman yang memiliki efek antifungal karena mengandung senyawa organik seperti flavonoid, tanin, dan saponin yang mampu menghambat pertumbuhan C. albicans. Tujuan dari penelitian ini adalah untuk menetapkan potensi pengaruh pemberian kombinasi ekstrak kulit buah durian dengan dan tanpa itraconazole terhadap pertumbuhan C. albicans in vitro. Penelitian ini menggunakan metode checkerboard dengan 6 jenis konsentrasi ekstrak kulit buah durian dan 5 jenis konsentrasi iatraconazole dalam menentukan KHM kombinasi. KHM kombinasi bahan uji diuji pada media SDA untuk menetapkan pengaruh kombinasi bahan uji terhadap C. albicans melalui zona hambat yang dibandingkan dengan itraconazole (kontrol positif), ekstrak kulit buah durian, dan akuades (kontrol negatif). Hasil dari penelitian ini menunjukkan pemberian bahan uji kombinasi berpotensi menghambat pertumbuhan C. albicans tanpa dipengaruhi oleh konsentrasi bahan uji. Semakin tinggi konsentrasi kulit buah durian atau itraconazole, maka semakin meningkat pula daya hambat pertumbuhan C. albicans. Kesimpulan dari penelitian ini adalah kombinasi ekstrak kulit buah durian dengan itraconazole berpotensi menghambat pertumbuhan C. albicans in vitro.Kata Kunci: Candida albicans, ekstrak kulit buah durian, itraconazole

Abstract

Abstrak Latar belakang: Pemilihan metodologi penelitian yang tepat sangat dibutuhkan dalam suatu penelitian. Keberagaman ini jenis metodologi penelitian mahasiswa ini belum diidentifikasi secara menyeluruh, sehingga belum diketahui arah penelitian yang telah dilakukan. Informasi mengenai trend pemilihan metodologi penelitian dapat dijadikan salah satu acuan untuk membuat kebijakan yang bisa meningkatkan kualitas akademik. Tujuan penelitian ini adalah untuk melakukan analisis terhadap tren pemilihan metodologi penelitian pada skripsi mahasiswa Program Studi Kedokteran Gigi FK Unsri. Metode : Metode yang digunakan dalam penelitian ini adalah deskriptif kualitatif. Penelitian dilakukan pada Program Studi Kedokteran Gigi FK Unsri dengan objek penelitian adalah dokumen skripsi mahasiswa selama 3 tahun terkahir. Data yang diambil dalam penelitian ini sebanyak 191 skripsi mahasiswa dengan rincian tahun 2021 sebanyak 59 skripsi, tahun 2022 sebanyak 73 skripsi, dan tahun 2023 sebanyak 59 skripsi. Hasil : Berdasarkan hasil analisis, maka didapat bahwa mahasiswa kedokteran gigi banyak mengambil jenis penelitian dengan metodologi analitik eksperimental, diikuti dengan studi deskriptif dan analisis observasional pada ketiga tahun terakhir. Tema yang dipilih adalah tema penelitian herbal yang mayoritas dilakukan secara in vitro, diikuti dengan penelitian material kedokteran gigi. Metode ini dipilih karena dapat memberikan hasil yang lebih mudah divalidasi, mudah direplikasi, lebih ekonomis, dan lebih fleksibel. Tema herbal dipilih karena tanaman herbal banyak digunakan oleh masyarakat dan belum pernah dilakukan pengujian sebelumnya. Kesimpulan : Tren pemilihan metodologi penelitian pada skripsi mahasiswa Prodi Kedokteran Gigi FK Unsri adalah analitik eksperimental, dengan tema penelitian herbal yang dilakukan secara in vitro.

Abstract

Candy is a type of food that contains sugar which is high in calories. Candy consumption can lower the pH of saliva, which can trigger dental caries. Gambir (Uncaria gambir) is a type of plant that contains active compounds catechins and tannins, which can inhibit the colonisation of acid-causing bacteria in the oral cavity. This study aimed to determine the effect of gambir can-dy consumption on salivary pH; an experimental study with a pretest-posttest approach. The research subjects were 25 Sriwi-jaya University students, whose saliva samples were taken as much as 5 mL before and after consumption of candies containing gambier extract; using the spitting method and measured with a digital pH meter. Paired t-test was conducted for data analysis which showed that there was a significant increase in mean salivary pH before and after consuming gambir candy (p<0.05). It was concluded that consumption of candies containing gambir extract can maintain and increase salivary pH.
