Abstract

Excessive free radicals can induce oxidative stress, leading to the inhibition of phosphorylation processes that supply energy for sperm capacitation and the acrosome reaction in spermatozoa during fertilization. This study investigated the impact of incorporating the antioxidant melatonin into the tris-fructose egg yolk diluent on the quality of ram liquid semen preserved at 4°C. Fresh semen with ≥70% motility was diluted in the tris-fructose egg yolk group (control), tris-fructose egg yolk with 0.5 mM melatonin (TFEY + M0.5), tris-fructose egg yolk with 1 mM melatonin (TFEY + M1), tris-fructose egg yolk with 2 mM melatonin (TFEY + M2). Semen was stored in the refrigerator at 4°C for 5 days (D0−D5). The motility, viability, abnormality, and plasma membrane integrity of the semen were evaluated every 24 hours. The results indicated no significant difference (P>0.05) between the control group and the liquid semen treatment at D0. Liquid semen quality was significantly improved with melatonin treatment compared to the control group. The results of this study indicate that TKTF + M0.5 had the highest motility and viability values ​​and can maintain abnormality values. The TKTF + M2 group had a higher plasma membrane integrity value than the other treatment groups.

Abstract

This study aims to evaluate calcium bioavailability through serum calcium level in pregnant rats treated with two Milk-Based Drinks (MBD) containing Lactic Acid Bacteria (LAB), i.e Lactobacillus Casei Shirota Strain (LcS) drink, and Four Strains Bacteria (FS) (Lactobacillus rhamnosus, Lactobacillus paracasei, Lactobacillus delbrueckii subp. bulgaricus, Streptococcus thermophiles). It was a completely randomized experimental study using 24 Sprague Dawley rats. Rats were divided into one negative control group with a normal nutritional status (A0) and three undernourished groups. The undernourished groups were divided into one positive control group (A1), one group receiving MBD containing LcS group (A2), and another group receiving MBD containing FS (A3). All rats received high protein diets during pregnancy. The intervention was started in early pregnancy (D0) until 19 days of pregnancy (D19). Blood samples were collected at the D0 and D19. No significant differences in food intake were found among the rats in all groups. Administration of MBD containing LAB in A2 and A3 showed significant (p<0.05) increment of calcium bioavailability (30.79±6.88%; 20.44±9.04%). Both MBDs treatment containing LAB showed no significant difference in serum calcium bioavailability (p>0.05). The results suggest that MBDs containing LAB are useful in enhancing calcium bioavailability.

Abstract

Improvement of canine reproduction has been developed by artificial insemination (AI) for effective breeding. -tocopherol widely recognized as an antioxidant that has been used to improve the quality of the spermatozoa. This study evaluated the effect of -tocopherol supplementation in dog chilled semen quality. Beagle dog breed semen samples were obtained from male Beagles using massage technique. The semen was then diluted with tris-skim milk extender supplemented with -tocopherol of various concentration, 0% as control group (T-0), 0.1% (T-0.1), 0.2% (T-0.2), and 0.3% (T-0.3), then were stored at 4 C and evaluated daily for four days, particularly for variables namely sperm motility, sperm viability, and plasma membrane integrity. Results showed that the quality of spermatozoa was higher when spermatozoa preserved in extender containing of 0.1% or 0.2% -tocopherol compared to those of control and 0.3% -tocopherol group (P0.05). The highest quality found in group with 0.2% -tocopherol. In conclusion, supplementation -tocopherol 0.2% in dog chilled semen extender is effective for improving and maintaining chilled dog sperm quality for 4 days of storage.

Abstract

Semen was collected using an artificial vagina, and then examined macroscopically and microscopically. Semen with motility of 70% was diluted into different extenders: tris egg yolk-glucose (TEYG), tris egg yolk-glucose extender with 0.5 mM melatonin (TEYG + M0.5), tris egg yolk-glucose extender with 1 mM melatonin (TEYG + M1), tris egg yolk-glucose extender with 2 mM melatonin (TEYG + M2). All semen samples were stored at 4 C for 5 days (H0-H5) and examined every 24 hours. Semen was examined for spermatozoa motility, viability, abnormality, and membrane plasma integrity. The results showed that there were no significant changes of semen quality on H0 (P0.05). In general, the melatonin supplemented groups had a higher semen quality value than the control. Melatonin supplementation can protect and maintain the quality of chilled semen. In addition, the result showed that TEYG + M1 extender was the best extender treatment, which was indicated by higher values than other treatments.

Abstract

Penelitian ini bertujuan mengetahui pengaruh kombinasi intracytoplasmic sperm injection (ICSI) dan aktivasi dengan strontium untuk mengetahui perkembangan pronukleus dan perkembangan embrio sampai tahap blastosis. Kombinasi ICSI dan strontium 20 mM meningkatkan perkembangan pronukleus 2-PN mencapai 43,59%. Hasil perkembangan embrio pada perlakuan kombinasi ICSI dan strontium 20 mM mencapai tingkat perkembangan 2-4 sel (50,5%), 8-16 sel (43,73%), dan blastosis (15,63%). Kesimpulan dari hasil penelitian ini adalah kombinasi ICSI dengan aktivasi strontium 20 mM mampu menghasilkan perkembangan in vitro embrio sapi yang lebih baik.

Abstract

The purpose of this study was to evaluate the safety of ethanol extract from torbangun leaves on mouse fetal development. This research used 24 female pregnant mouse. The leaves extract was administered orally at dose level of 0; 0.56; 1.68 and 3.36 g/kg body weight. The extract was given from the first day until the eighteenth day of pregnancy period. The ethanol extract of Torbangun leaves contains flavonoids, saponins, tannins, and steroids. The result showed that consumption of 3.36 g/kg torbangun leaves extract had led to significant differences in the decrease of maternal weight change, number of live fetuses, fetal weight, and fetal length. Consumption of Torbangun leaves extract during pregnancy should be avoided due to its potency to interfere the fetal development.

Abstract

Psittacines are vulnerable to bacterial diseases, including gram-negative infections. A key challenge in treating these birds is antibiotic resistance, where antibiotics become ineffective against the bacteria. This study aimed to identify gram-negative bacteria present in 11 fecal samples of psittacine from Taman Mini Indonesia Indah (TMII) in East Jakarta, as well as to determine the presence of antibiotic resistance in these bacteria. In this study, bacteria were isolated using MacConkey Agar media, followed by Gram staining and biochemical tests for bacterial identification. Antibiotic resistance testing was conducted using the Kirby-Bauer method against seven antibiotics. The results of bacterial identification showed the presence of Klebsiella sp., Proteus mirabilis, and Escherichia coli in the fecal samples of Psittacine from TMII. Resistance to the antibiotics nalidixic acid, doxycycline, tetracycline, gentamicin, erythromycin, cefotaxime, and trimethoprim-sulfamethoxazole was observed, with trimethoprim-sulfamethoxazole demonstrating the lowest resistance rate among fecal Psittacine bacteria.

Abstract

Knee joints are the primary sites affected by osteoarthritis (OA) in humans and animals. Although intra-articular human umbilical cord–derived mesenchymal stem cells (hUC-MSCs) show therapeutic promise, their combined use with hyaluronic acid (HA) remains underexplored in large animals. This study evaluated morphological and proteomic changes in a sheep osteoarthritis model treated with both agents. A total of twelve sheep were divided into four groups (n = 3 per group): control (normal and OA) and three treatment groups receiving hUC-MSCs, HA, or a combination of HA and hUC-MSCs (HA + hUC-MSCs). OA was induced by meniscectomy, and intra-articular injections were administered four weeks post-surgery. Macroscopic scoring was conducted according to the Osteoarthritis Research Society International system, while histological evaluation was performed using hematoxylin–eosin, Safranin O, and Masson’s trichrome staining. Proteomic analysis of synovial fluid was carried out using liquid chromatography–tandem mass spectrometry. The macroscopic OA score in the HA + hUC-MSC group was lower than in the other treatment groups and was not significantly different from the normal group (p > 0.05). Microscopic observations indicated that hUC-MSCs supported joint cartilage regeneration by preserving proteoglycan and collagen content. Proteomic profiling revealed that treatment with HA + hUC-MSCs modulated proteins associated with complement activation and coagulation pathways. The combination of HA and hUC-MSCs provided synergistic benefits for cartilage regeneration in a sheep OA model during a short treatment period. These findings suggest that this approach represents a promising regenerative therapy for OA in large animals within veterinary medicine and has translational relevance for human regenerative medicine.
