Abstract

Oocyte vitrification is the important part of gamete preservation for further purpose. The objective of this study was to evaluate the response or development of vitrified-mice oocyte following activation using various concentrations of Strontium Chloride (Sr Cl2). Oocytes were collected from superovulation-induced female mice. Oocytes vitrification was then performed using a gradual equilibration of 2 M Ethylene Glycol in 0.25 M sucrose and 7 M Ethylene Glycol on 0.5 sucrose. Subsequently, the vitrified oocytes were thawed and activated using various Strontium Chloride concentration in each group. Control 1 is unvitrified oocyte and without Sr Cl2. Control 2 is unvitrified oocyte then activated by 20 mM Sr Cl2. Zero (0) mM Sr Cl2 is vitrified oocyte without Sr Cl2. Group Ten (10) mM is vitrified oocyte then activated by 10 mM Sr Cl2. Group Twenty (20) mM is vitrified oocyte then activated by 20 mM Sr Cl2. The viability of vitrified-thawed oocytes was observed based on ooplasm integrity. Whereas the oocytes respond to artificial activation was observed based on pronucleus formation after 10 hours of activation. The result showed that 39% of oocyte degenerated following vitrification. The respond of vitrified-thawed oocytes following artificial activation using Strontium Chloride was significantly lower compared to fresh oocytes (p<0,05). Interestingly the highest percentage of activated oocytes (36.36%) was present in a group achieved 20 mM Strontium Chloride. As conclusion is Strontium Chloride 20mM has a best result (36,36%) to activate vitrified oocyte than 0 mM and 10 mM of Strontium Chloride.

Abstract

Penelitian ini bertujuan mengetahui pengaruh kombinasi intracytoplasmic sperm injection (ICSI) dan aktivasi dengan strontium untuk mengetahui perkembangan pronukleus dan perkembangan embrio sampai tahap blastosis. Kombinasi ICSI dan strontium 20 mM meningkatkan perkembangan pronukleus 2-PN mencapai 43,59%. Hasil perkembangan embrio pada perlakuan kombinasi ICSI dan strontium 20 mM mencapai tingkat perkembangan 2-4 sel (50,5%), 8-16 sel (43,73%), dan blastosis (15,63%). Kesimpulan dari hasil penelitian ini adalah kombinasi ICSI dengan aktivasi strontium 20 mM mampu menghasilkan perkembangan in vitro embrio sapi yang lebih baik.

Abstract

Sel germinal merupakan salah satu sumber sel yang masih bersifat totipotensi dan berperan dalam pembentukan organisme baru. Morfologi dan ekspresi protein pada sel germinal bersifat dinamis bergantung pada umur dan tahap perkembangan. Penelitian ini bertujuan untuk mengamati perubahan morfologi dan ekspresi protein sebagai marka sel germinal jantan pada fetus umur 13,5 hari pascakawin (days post coital/ dpc) dan anak mencit umur lima hari pascalahir. Hasil Rigi kelamin dan testis diisolasi dari mencit umur 13,5 dpc dan 5 hari. Jaringan kemudian dipreparasi histologi rutin, dan diwarnai dengan pewarnaan hematoksilineosin (HE), sedangkan untuk mengidentifikasi keberadaan protein Dazl, Vasa dan Oct4, jaringan diwarnai dengan pewarnaan imunohistokimia menunjukkan morfologi sel germinal jantan pada fetus mencit umur 13,5 dpc dan anak mencit umur lima hari pascalahir sama-sama berbentuk bulat oval. Namun, sel germinal jantan pada mencit umur lima hari pascalahir berukuran lebih besar, jumlah yang lebih sedikit dan terletak jauh dari membran basal. Pada sel germinal jantan umur 13,5 dpc menunjukkan positif lemah terhadap antibodi Oct 4 dan DAZL serta positif kuat terhadap antibodi Vasa. Pada umur lima hari, sel germinal jantan menunjukkan positif kuat terhadap antibodi Oct-4 dan DAZL, serta positif lemah terhadap antibodi Vasa. Simpulan dari penelitian ini adalah morfologi dan ekspresi marka sel germinal dipengaruhi oleh tahapan pertumbuhan dan perkembangan sel-sel germinal. Vasa dapat digunakan sebagai marka untuk sel germinal umur 13,5 dpc dan DAZL sebagai marka untuk sel germinal umur lima hari pascalahir.

Abstract

Kampung and Sentul chickens are Indonesian local chicken breeds that are widely developed by breeding companies as strains for meat production. The genetic differences among these chicken strains can impact the quality of the hatching eggs they produce. Somite development is a crucial part of chicken embryo growth during the hatching process, as it precedes the formation of vertebrae and skeletal muscles, ultimately affecting chick quality. This study aimed to analyse how both strains of hens affect hatching egg quality and somite development. The sample included 126 hatching eggs, with 63 from Kampung chickens and 63 from Sentul chickens. The observed parameters of hatching egg quality included egg weight, egg shape index, eggshell thickness, eggshell weight, eggshell strength, albumen index, yolk index, yolk color score, Haugh Unit (HU), and embryo development indicators, such as the development of the primitive streak and progression of somite number. The data were analysed using a t test. Differences between chicken strains significantly affected (P<0.05) egg weight, shell thickness, the albumen index, and HU. The different strains also showed significant differences (P<0.05) in embryo somite development, with the primitive streak development of the Sentul chicken being slower than that of the Kampung chicken. In conclusion, Kampung chickens produce hatching eggs with superior interior quality, primitive streak development and somite number compared to the hatching eggs of other chickens. However, Sentul chickens also demonstrate good exterior egg quality, as shown by their egg weight and shell thickness.

Abstract

Indo-Pacific bottlenose dolphin (Tursiops aduncus) is a marine mammal that lives in relatively small populations. The geographic ranges of this species are susceptible to the effects of human activities, thereby necessitating conservation efforts to prevent extinction. Therefore, this study aimed to evaluate the daily quality of dolphin sperm after several days of refrigeration. The sperm of two male dolphins were stored at 4oC for 4 days, and the quality was observed daily to determine the motility, viability, membrane integrity, and sperm abnormalities. Sperm samples were divided into four groups, consisting of two centrifuged followed by the removal of seminal plasma, and two groups without centrifugation, containing 100x106 and 200x106 sperm/ml each. After liquid storage, the motility of sperm was 63-75% with no significant reduction in the first 3 days. Sperm viability following storage was 65-75% and the percentage with abnormal morphology ranged from 2-6%. Furthermore, there was no significant increase in abnormal morphology of sperm on any day of storage for 3 days. Sperm membrane integrity was 36-49%, with no significant reduction in the membrane integrity in the first 2 days. There was no significant difference in sperm quality, although centrifugation and removal of seminal plasm had a slight effect. The results of this study showed that Indo-Pacific bottlenose dolphin sperm could be stored for a short period as liquid storage while maintaining a quality that allows for future use.

Abstract

Glutamine is a non-essential amino acid that can reduce free radicals and act as an antioxidant. This study aims to determine the effect of adding glutamine in egg yolk tris diluent at concentrations of 0 mM (K), 2,5 mM (G2,5), 5 mM (G5), and 7,5 mM (G7,5) on the quality of liquid semen and kept in a refrigerator at 4 °C. Microscopic evaluation was performed immediately after dilution (H0) and every 24 hours until day 5 (H5). The results showed that the G 7,5 mM group could maintain better motility and viability (P < 0,05) than the K, G 2,5, and G 5 groups. The addition of glutamine did not have a significant effect (P> 0.05) in maintaining the MPU of spermatozoa. Glutamine with a concentration of 7.5 mM is the best concentration to maintain the quality of sheep's liquid semen.

Abstract

Excessive free radicals can induce oxidative stress, leading to the inhibition of phosphorylation processes that supply energy for sperm capacitation and the acrosome reaction in spermatozoa during fertilization. This study investigated the impact of incorporating the antioxidant melatonin into the tris-fructose egg yolk diluent on the quality of ram liquid semen preserved at 4°C. Fresh semen with ≥70% motility was diluted in the tris-fructose egg yolk group (control), tris-fructose egg yolk with 0.5 mM melatonin (TFEY + M0.5), tris-fructose egg yolk with 1 mM melatonin (TFEY + M1), tris-fructose egg yolk with 2 mM melatonin (TFEY + M2). Semen was stored in the refrigerator at 4°C for 5 days (D0−D5). The motility, viability, abnormality, and plasma membrane integrity of the semen were evaluated every 24 hours. The results indicated no significant difference (P>0.05) between the control group and the liquid semen treatment at D0. Liquid semen quality was significantly improved with melatonin treatment compared to the control group. The results of this study indicate that TKTF + M0.5 had the highest motility and viability values ​​and can maintain abnormality values. The TKTF + M2 group had a higher plasma membrane integrity value than the other treatment groups.

Abstract

This study aims to evaluate calcium bioavailability through serum calcium level in pregnant rats treated with two Milk-Based Drinks (MBD) containing Lactic Acid Bacteria (LAB), i.e Lactobacillus Casei Shirota Strain (LcS) drink, and Four Strains Bacteria (FS) (Lactobacillus rhamnosus, Lactobacillus paracasei, Lactobacillus delbrueckii subp. bulgaricus, Streptococcus thermophiles). It was a completely randomized experimental study using 24 Sprague Dawley rats. Rats were divided into one negative control group with a normal nutritional status (A0) and three undernourished groups. The undernourished groups were divided into one positive control group (A1), one group receiving MBD containing LcS group (A2), and another group receiving MBD containing FS (A3). All rats received high protein diets during pregnancy. The intervention was started in early pregnancy (D0) until 19 days of pregnancy (D19). Blood samples were collected at the D0 and D19. No significant differences in food intake were found among the rats in all groups. Administration of MBD containing LAB in A2 and A3 showed significant (p<0.05) increment of calcium bioavailability (30.79±6.88%; 20.44±9.04%). Both MBDs treatment containing LAB showed no significant difference in serum calcium bioavailability (p>0.05). The results suggest that MBDs containing LAB are useful in enhancing calcium bioavailability.

Abstract

Improvement of canine reproduction has been developed by artificial insemination (AI) for effective breeding. -tocopherol widely recognized as an antioxidant that has been used to improve the quality of the spermatozoa. This study evaluated the effect of -tocopherol supplementation in dog chilled semen quality. Beagle dog breed semen samples were obtained from male Beagles using massage technique. The semen was then diluted with tris-skim milk extender supplemented with -tocopherol of various concentration, 0% as control group (T-0), 0.1% (T-0.1), 0.2% (T-0.2), and 0.3% (T-0.3), then were stored at 4 C and evaluated daily for four days, particularly for variables namely sperm motility, sperm viability, and plasma membrane integrity. Results showed that the quality of spermatozoa was higher when spermatozoa preserved in extender containing of 0.1% or 0.2% -tocopherol compared to those of control and 0.3% -tocopherol group (P0.05). The highest quality found in group with 0.2% -tocopherol. In conclusion, supplementation -tocopherol 0.2% in dog chilled semen extender is effective for improving and maintaining chilled dog sperm quality for 4 days of storage.

Abstract

Semen was collected using an artificial vagina, and then examined macroscopically and microscopically. Semen with motility of 70% was diluted into different extenders: tris egg yolk-glucose (TEYG), tris egg yolk-glucose extender with 0.5 mM melatonin (TEYG + M0.5), tris egg yolk-glucose extender with 1 mM melatonin (TEYG + M1), tris egg yolk-glucose extender with 2 mM melatonin (TEYG + M2). All semen samples were stored at 4 C for 5 days (H0-H5) and examined every 24 hours. Semen was examined for spermatozoa motility, viability, abnormality, and membrane plasma integrity. The results showed that there were no significant changes of semen quality on H0 (P0.05). In general, the melatonin supplemented groups had a higher semen quality value than the control. Melatonin supplementation can protect and maintain the quality of chilled semen. In addition, the result showed that TEYG + M1 extender was the best extender treatment, which was indicated by higher values than other treatments.
