Abstract

The objective of this study is to study the effect of gamma ray irradiation (15, 30, 45, and 60 gray (Gy) to determine Lethal Dose 50 (LD50) values, and to obtain new Coleus variances in a relatively short time. The study was conducted in a greenhouse at Cikabayan experimental field, Bogor Agricultural University, Darmaga, Bogor in May to July 2013. Gamma irradiation treatment significantly affected height, number of leaves, and number of nodes. Increasing the dose level of gamma irradiation tend to inhibit plant growth. LD50 for yellow/green, green/brown, variegated green/brown of Coleus blumei, and Coleus amboinicus Lour were 48.66, 65.2, 52.81, and 37.62 Gy respectively. C. amboinicus  irradiated at a dose level of 45 Gy had different leaf shapes compared to control. Keywords:  Coleus, gamma ray irradiation, LD50 values, ornamental plant, torbangun

Abstract

Torbangun (Coleus amboinicus Lour) is a tropical plant from the Lamiaceae family. Torbangun leaves have a distinct aroma. Torbangun leaves have been traditionally used as a medicinal plant in North Sumatra, Indonesia. Bataknese lactating women in North Sumatra consumed torbangun leaves after giving birth with beliefs that it could increase their breast milk production (lactagogue), and that the leaves can function as anti-fungal and/or anti-bacterial, analgesic, to reduce blood cholesterol, and clean the human uterus. However, scientific evidence on this traditional herb is limited. This research was conducted to: (1) determine the leaf N, P, K, Ca and Fe in different position along the plant and the leaf age as to diagnose the secondary metabolites content of torbangun leaves, and (2) determine the relationship between N, P, K, Ca and Fe leaf concentrations with their metabolite production. The results of the study showed that: (1) the best leaf position and age to determine of the need of N, P, K, Ca and Fe nutrients are on the 3rd leaf position of 5 months-old plant (2) There was a positive correlation between leaf K concentrations with shoot dry weight, Ca with PAL activity, and Fe with total saponins on the 3rd leaf position of 5 months-old plant.Keywords: Coleus amboinicus Lour, torbangun, nutrient, correlation test, secondary metabolites.

Abstract

Torbangun (Plectranthus amboinicus) merupakan tanaman obat dari keluarga Lamiaceae. Pada masyarakat Batak, daun torbangun digunakan untuk meningkatkan produksi air susu. Keragaman kandungan fitokimia dapat ditingkatkan salah satunya dengan teknik iradiasi sinar gamma untuk program pemuliaan tanaman. Pada penelitian ini, metode metabolomik yang tidak tertarget digunakan untuk mengevaluasi profil senyawa pada bagian daun tanaman mutan dari iradiasi sinar gamma dan kontrol. Kandungan senyawa planlet mutan dan kontrol dianalisis dengan GC-MS. Data GC-MS dianalisis dengan kemometrik dengan menggunakan hierarchical cluster analysis (HCA). Keragaman kandungan kimia ditunjukkan oleh planlet mutan dan kontrol daun torbangun. Hasil analisa planlet kontrol torbangun menunjukkan terdapat lima senyawa utama diantaranya hydroxymethylfurfurole (35,15%), brevifolin (7,69%), 3-Dihydro-3,5-Dihydroxy-6-Methyl-4H-Pyran-4-one (DDMP) (13,13%), stigmasterol (6,51%) dan ferruginol (8.63%). Hasil analisa mutan torbangun memiliki kandungan senyawa DDMP (19,57%), neophytadiene (8,47%), linolenic acid (9,10%), ferruginol (7,61%), stigmasterol (14,14%) dan gamma.-sitosterol (7,08%). HCA menunjukkan tiga komponen senyawa yang berbeda antara planlet kontrol dan mutan yaitu kenaikan kandungan senyawa DDMP dan stigmasterol, serta penurunan kandungan hydroxymethylfurfurole pada mutan torbangun.

Abstract

This study aim was to determine lactagogue effect of torbangun leaves to plasma levels of lactogenic hormone and gene expression of their receptors in mammary glands of lactation rats. Lactagogue activity was evaluated by volume of milk was produced by the rats treated with commercial milk booster contained ?katuk' leaves extract (AF), ethyl acetate fraction of torbangun leaves (EA), water extraction of torbangun leaves (AQ) and kaempferol (KP). Lactating rats (n=5) of Sprague dawley with six pups were fed with AF, EA, AQ, and KP in the amount of 50 mg/kg, 30 mg/kg, 80 mg/kg and 60 mg/kg body weight, respectively. The feed was given orally every two days and starting from day 2 after giving birth until day 28. The volume of milk was estimated by the increment pup weight after breastfed. The levels of serum lactogenic hormones were determined by ELISA methods. Moreover, in order to measure the gene expression of the lactogenic hormone's receptors in the mammary gland a real time - reverse transcription polymerase chain reaction (RT-PCR) method was performed. The results showed that ethyl acetate fraction of torbangun leaves (EA) (a) was not significantly stimulating the synthesis of serum prolactin and estradiol at day 14 and day 28 lactation period, (b) down-regulated the gene expression of estradiol receptor (ER?) at day 28, and (c) up-regulated the gene expression of prolactin receptor (PRLR) in mammary gland at day 14 and day 28. This study indicated that ethyl acetate fraction of torbangun leaves was induced milk production, within up-regulated the gene expression of prolactin receptor (PRLR) in the mammary gland of lactation rats.

Abstract

Variant of Coleus sp. in Indonesia is very low. Induced mutation by Ethyl Methane Sulphonate (EMS) is able to increase variant of Coleus sp.. EMS is a mutagen that is effective and efficient in causing mutations. In mutation breeding, the high variant usually occurs around LC50 (Lethal Concentration). The objective of this study were (1) to obtain the LC50 value and sensivity of coleus plants, and (2) to evaluate the response of plant growth and to obtain new variance of coleus used soak and drops by EMS application. The experiment was arranged in randomized complete block design with three replications in each coleus. The materials used two Coleus sp. shoot cuttings i.e., C. amboinicus Lour. and C. blumei the purple/green color and the red. The concentration of EMS consisted of 0.00, 0.50, 0.75, 1.00, and 1.25% with application soak method (100 minutes) and drops (3 drops pipette). The results showed that the LC50 value of C. amboinicus Lour. 5.86% (soak). The LC50 value of C. blumei the purple/green color was 0.69% (soak), 0.82% (drops) and the red color 0.29% (drops), 0.89% (drops). C. blumei (the purple/green and red) was higher sensitivity rather than C. amboinicus Lour. There was an interaction between treatments on C. amboinicus Lour. whereas C. blumei (purple/green and red) had no interaction among treatments. C. amboinicus Lour. produced one putative mutant and C. blumei purple/green produced four putative mutants based on the morphology.Keywords: Coleus amboinicus Lour., Coleus blumei, LC50, putative mutan

Abstract

Pregnant mother is one of the group gristle nutrition. Pregnant mother need to do medical check during pregnancy to support quality of good health. Pregnancy inspection is suggested minimum four times during pregnancy. The aim of this study is to analyze correlation between intensity of pregnancy inspection, facility of health service and the iron tablet consumption with mount the sigh experienced during pregnancy.This study used thecross sectional study methodwith sample taking method by purposive in Kramat Jati Sub-district (East Jakarta) and Ragunan Sub-district (South Jakarta). Total sample taken for this study were 100 pregnant mothers(50 from each regional). Data obtained was processed by Microsoft Excel 2003 and analyzed by SPSS 13.0 for windows. Analyze data used Rank Correlation Spearman to know the correlation between variables.Results showed significant correlations betweeniron tablet consumption obedience with intensity of pregnancy inspection. Significant correlation was also shown by intensity of pregnancy inspection variable with facility of health service which got by pregnant mother during pregnancy inspection. This result study was strenghtened by previous study that showed intensity of pregnancy inspection to the health service had strong correlation with antenatal care which obtained by pregnant mother.

Abstract

The present study aims to analyze of relationship of the implementation of balanced nutrition messages family and behavior family nutrition awareness with under five years child nutritional status in West Kalimantan Province. The study uses Health Research Data Base of West Kalimantan Province in year 2007 with the design of cross-sectional study. The total of 1992 household samples were recruited in the study with criteria having child aged 6 to 59 months. Weight for height, weight for age and height for age indicators were used to measure child nutritional status. The results showed that three messages did not meet criteria as required by the Indonesian Food Guideines. Three messages did not meet criteria as required by the Indonesian Food Guidelines. They were eat food withr energy needs requaired, eat half of food sources of carbohydrate for energy needs and eating one of  fourth of food sources of fat for energy needs. Logistic multiple regression analysis showed that failure to follow the the Family Nutrition Awareness had 1.21 risk for children to became stunting than family to follow to guideline properly.

Abstract

The present study aims to analyze of influence maternal education to under five years-old children in West Kalimantan Province. The study uses Health Research Data Base of West Kalimantan Province in year 2007 with the design of cross-sectional study. The total of 1992 household samples were recruited in the study with criteria having child aged 6 to 59 months. Weight for height, weight for age, and height for age indicators were used to measure child nutritional status. The results showed that Logistic multiple regression analysis showed that maternal education under level of junior high school had 1.28 risk for children to become stunting and 1.27 risk for children to become underweight than maternal education level of above junior high school.

Abstract

Torbangun (Coleus amboinicus Lour) is one of Labiatae family containing a lot of micronutrients and active components. The plant is used as traditional medicine for relieving premenstrual syndromes (PMS). Based on the results of previous studies, a capsule containing 750 mg of dried Torbangun leaves can relieve PMS as well as commercial herbal, and better than placebo. Capsule supplement form seems less acceptable because it tastes like taking a drug. Therefore, it requires development for Torbangun supplement product in other forms, and one of them is a supplement drink. The study is aimed to formulate  a supplement drink from Torbangun leaves as the basic ingredients. Supplement drink formulation is made in trial and error by combining extract of torbangun leaves, sucrose, and lemon, obtaining three formulas: DT1 contains 10 g extract of torbangun leaves and 16 g sucrose, DT2 contains 10 g extract of torbangun leaves, 2 g lemon, 22 g sucrose, and DT3 contains 10 g extract of torbangun leaves, 4 g lemon, and 28 g sucrose. These formulas are then tested by organoleptic, physical, and chemical. The results show that the formula DT3 has the highest average score. Blanching treatment improves organoleptic properties of supplement drinks. Mean scores in color, aroma, and taste of the blanched supplement drink are different (p<0.05) from non-blanched supplement drink.

Abstract

Torbangun leaves soup is one of the traditional diets commonly consumed by lactating mothers among the Batak society. The soup is believed to increase breast milk production and helps recovery process after the mother giving birth. Unfortunately, until now the soup is only served directly after cooking thereby limits its usefulness. Given the potential of the soup is quite important, the preservation of the cooked soup needs to be done so that the soup can be consumed at anytimeany time. This study aims to determine the shelf life of torbangun soup in a variety of packaging and at various storage temperatures. The soup is stored at three different temperatures (3-5oC, 10-12oC and 27-30oC) and in 3 types of packaging namely polyethylene terephthalate (PET), polypropylene (PP) and cans. Changes in product quality during storage are analyzed and estimated the shelf life of products. The results show that the soup stored at the 3-5oC and 10-12oC has longer shelf life compared to 27-30oC (day 8 vs day 3). In addition, the cans are the best packing among other packaging materials.Key words: torbangun leaf, Coleus amboinicus Lour, soup, shelf lifeABSTRAKSup daun Torbangun torbangun adalah salah satu makanan tradisional yang biasa dikonsumsi oleh ibu menyusuidi kalangan masyarakat Batak. Sup ini diyakini dapat meningkatkan produksi ASI dan membantu proses pemulihan setelah ibu melahirkan. Sayangnya, sampai saat ini sup daun torbangun hanya disajikan langsung setelah dimasak sehingga membatasi kegunaannya. Mengingat potensi sup ini cukup penting, penggunaan bahan pengawet pada sup matang perlu dilakukan agar sup ini dapat dikonsumsi kapan saja. Penelitian ini bertujuan untuk menentukan umur simpan sup torbangun dalam berbagai kemasan dan pada berbagai suhu penyimpanan. Pada penelitian ini sup disimpan pada tiga suhu berbeda (3-5oC, 10-12oC dan 27-30oC) dan di simpan dalam tiga jenis kemasan yaitu polietilen tereftalat (PET), polypropylene (PP) dan kaleng. Perubahan kualitas produk selama penyimpanan dianalisis dan dilakukan perkiraan umur simpan produk. Hasil penelitian menunjukkan bahwa sup yang disimpan pada suhu 3-5oC dan 10-12oC memiliki umur simpan lebih lama diban- dingkan dengan sup yang disimpan pada suhu 27-30oC (8 hari vs 3 hari). Selain itu kaleng adalah kemasan terbaik di antara bahan kemasan lainnya.
