Abstract

Carica pubescens Lenne & K. Koch, an endemic species in Dieng mountains, must be conserved. The in vitro conservation has been developed, but sub-culture period needs to be extended. This study aimed to obtain a more efficient in vitro conservation protocol of C. pubescens. The research was carried out experimentally by using a completely randomized factorial design with three factors, namely decreasing in storage medium concentration (75% and 50% of MS medium), temperature (4oC and 8C), and irradiation duration (8 hours/day and 16 hours/day). Shoots were kept in the storage medium for 6, 9 and 12 months, then their viability were tested by growing them in the regeneration medium. Data were analyzed by Analyses of Variance and Least Significant Difference Test. The results showed that medium concentration of 50% of MS, the temperature of 8 C, and 16 hours/day of irradiation were able to suppress the C. pubescens growth in vitro storage for six months and could maintain its viability in the regeneration medium. Based on these results the medium concentration of 50% of MS, the temperature of 8 C and 16 hours/day irradiation can be used for in vitro conservation of C. pubescens without sub-culture for six months. Abstract

AbstrakBurahol termasuk tanaman yang buahnya dapat dimakan, dan mempunyai zat-zat aktif yang berpotensi sebagai obat dan deodoran alami. Kultur in vitro merupakan salah satu cara dalam produksi metabolit sekunder. Tingginya kontaminasi merupakan salah satu hal yang menjadi kendala dalam kultur in vitro. Salah satu sumber kontaminan adalah eksplan yang digunakan dalam kultur in vitro. Optimasi sterilisasi permukaan merupakan langkah awal yang sangat penting dalam pengembangan kultur in vitro. Penelitian ini bertujuan mendapatkan prosedur optimasi sterilisasi permukaan eksplan daun burahol dan juga melakukan deteksi dan eliminasi edofit pada daun burahol. Optimasi sterilisasi permukaan dilakukan dengan perlakuan variasi konsentrasi klorox dan waktu perendaman. Hasil penelitian menunjukkan bahwa daun burahol mengandung jamur endofit. Eliminasi jamur endofit dapat dilakukan dengan penyiraman tanaman dengan fungisida. Sterilisasi permukaan eksplan yang paling optimal adalah dengan fungisida selama 24 jam, dilanjutkan dengan perendaman bakterisida dan fungisida selama 30 menit, perendaman pada alcohol 70% selama 1 menit, dilanjutkan dengan klorox 15% 10 menit, dan klorox 10% 10 menit berturut-turut.AbstractBurahol has active substances with potential as a drug and natural deodorant. In vitro culture is one way to production of secondary metabolites. High contamination is one of the things that become obstacles in in vitro culture . One of the contaminant source is explant that used in in vitro culture. Optimization of surface sterilization is a very important first step in the development of in vitro culture. This study aims to get the optimization procedure surface sterilization of burahol leaf explant and also perform detection and elimination endophyte on burahol leaves. Optimization surface sterilization is done by treatment by variations clorox concentration and immersion time. The results showed that the burahol leaves contain fungal endophytes. Endophytic fungus elimination can be done by sprinkling the plants with fungicides. Explant surface sterilization is the most optimal with fungicide for 24 hours, followed by immersion bactericide and fungicide for 30 minutes, immersion in 70 % alcohol for 1 min, followed by 10 minutes clorox 15 %, and 10 % clorox 10 minutes.

Abstract

Analysis fingerprinting teak is supported by information about teak’s character molecularly. The information is gotten by analyzing 3 primary microsatellite in 46 teak samples that produce total 18 kinds of alel averagely 6 alel per locus. Genetics variety level is shown by Observed Heterozigosity (Ho), Expected Heterozygosity (He), Polimorfi sm Information Content (PIC) and Genetics Differentiation Coeffi cient successively 0.5122; 0.6221; 0.5818; and 0.0629. Kinship connection through dendogram analysis produces coeffi cient resemblance 0.3-1.00 that form 2 groups. However it has not shown inclination of forming group based on origin grows yet. Genetics differentiation analysis indicates different genetics in a population is higher then it is between populations. Analysis result has not found specifi c alel yet but has found 12 specifi c genotipe that is potential for analyzing fi ngerprinting if it relates to important useful genes. Abstract

Fingerprinting jati diperlukan untuk melengkapi data base konservasi plasma nutfah jati. Penelitian ini bertujuan untuk mendapatkan informasi keanekaragaman genetika pohon jati berdasarkan marka mikrosatelit dan mengidentifikasi alel spesifik pada lokus tertentu yang dapat menjadi penciri khas jati populasi Jawa dan Madura. Pada penelitian ini dilakukan identifikasi karakteristik jati plus (fingerprinting) secara molekuler menggunakan penanda mikrosatelit. Sampel diambil dari koleksi jati plus di Kebun Benih Klonal (KBK) di Jawa dan dari lokal Areal Produksi Benih (APB) Madura. Sampel diisolasi menggunakan metode CTAB. Amplifikasi DNA menggunakan 3 primer mikrosatelit dan hasilnya divisualisasi pada gel poliakrilamid dengan pewarnaan silver. Hubungan kekerabatan dianalisis melalui program NTSYSpc (Numerical Taxonomy and Multivariate Analysis Sistem) versi 2.01. Hasil analisis menunjukkan tingkat keanekaragaman jati yang tinggi, didukung oleh nilai Observed Heterozigosity (Ho), Expected Heterozygosity (He), Polimorfism Information Content (PIC) dan koefisien diferensiasi genetika berturut-turut 0.5122; 0.6221; 0.5818; dan 0.0629. Hubungan kekerabatan melalui analisis dendogram menghasilkan koefisien kemiripan 0.3-1.00. Nilai keanekaragaman dalam populasi (HS), antar populasi (DST), dan nilai diferensiasi (G) berturut-turut adalah 0.5817, 0.0391, dan 0.0629. Dari hasil analisis disimpulkan bahwa tingkat keanekaragaman jati di Jawa dan Madura termasuk tinggi dan ada indikasi perbedaan genetika di dalam populasi lebih tinggi dibanding antar populasi.Fingerprinting teak is required to complete the data base teak germplasm conservation. This research aimed to obtain information about the genetic diversity of teak based on microsatellite markers and identify specific allele at a particular locus can be distinctive identity identifier population of Java and Madura. In this research is to identify the characteristics of teak plus (fingerprinting) molecularly using microsatellite markers. Samples were taken from the collection of teak plus in Kebun Benih Klonal in Java and from Areal Produksi Benih Madura. Samples were isolated using the CTAB method. Amplification of DNA using microsatellite primer 3 and the results are visualized on a polyacrylamide gel by silver staining. Kinship analyzed through NTSYSpc program (Numerical Taxonomy and Multivariate Analysis System) version 2:01. The analysis showed a high level of diversity teak, supported by Heterozigosity Observed value (HO), Expected Heterozygosity (He), polymorphisms Information Content (PIC) and genetic differentiation coefficient 0.5122 respectively; 0.6221; 0.5818; and 0.0629. Kinship through analysis dendogram generate similarity coefficient 0.3-1.00. The value of diversity in the population (HS), between populations (DST), and the value of differentiation (G) respectively are 0.5817, 0.0391, and 0.0629. From the analysis concluded that the level of diversity of teak in Java and Madura are high, and there are indications of genetic differences in the population is higher than among the population.

Abstract

Carica pubescens Lenne & K. Koch is a rare species that need to be conserved. The research aim was to develop a slow growth method of in vitro conservation medium through determining some effects of nutrition decreasing availability in the conservation medium on growth and survival of explants. Establishing epicotyls reached from in vitro seed germination was grown on diluted basic medium of 75% MS (Murashige and Skoog), 50% MS, 25% MS, while osmoregulator compound of mannitol and sorbitol was added to the full MS medium in several concentrations. The treatments were arranged in a completely randomized design with three replications. The epicotyls were grown at storage medium for 12 and 16 weeks, then their survival were evaluated at regeneration medium and rooting medium. The diluted basic medium and osmoregulator addition were evaluated for its influence in retarding the culture growth in terms of improved survival over the period of 16 weeks. Data analyzed by one way analysis of variance and Duncans multiple range test. The results showed that the decreasing of nutrition concentration suppressed the growth of the epicotyls until 16 weeks after conservation. Epicotyls taken from 16 weeks after conservation could grow on the regeneration medium. The best survival was shown by the 75% MS, 50% MS and supplementing of 20 g/l mannitol treatments. Based on these results, 50% MS medium is recommended for storage C. pubescens for 16 weeks with no sub-culture.Carica pubescens Lenne & K. Koch (karika dieng) merupakan tanaman yang langka sehingga perlu dilestarikan. Penelitian ini bertujuan untuk memperoleh medium penyimpanan in vitro dengan teknik pertumbuhan minimal dengan mengamati pengaruh penurunan ketersediaan nutrisi dalam medium terhadap penurunan pertumbuhan dan daya tumbuh eksplan.Eksplan berupa epikotil kecambah in vitro. Perlakuan penurunan ketersediaan nutrisi dilakukan melalui reduksi konsentrasi nutrisi dari medium Murashige & Skoog (MS) dan penambahan osmoregulator (manitol dan sorbitol) dengan berbagai konsentrasi. Penelitian dilakukan dengan rancangan acak lengkap satu faktor dengan tiga ulangan. Epikotil dipelihara dalam medium penyimpanan selama 12 dan 16 minggu, kemudian dievaluasi daya tumbuhnya dengan memelihara dalam medium regenerasi dan medium pengakaran. Data dianalisis dengan analisis varians satu arah dan uji Duncan. Hasil penelitian menunjukkan bahwa penurunan kecepatan penyerapan nutrisi berpengaruh terhadap pertumbuhan eksplan. Epikotil yang telah disimpan selama 12 minggu dan 16 minggu dan ditumbuhkan kembali pada medium regenerasi masih dapat tumbuh dengan intensitas tertinggi pada perlakuan pengenceran 50% MS dan 75% MS, serta penambahan manitol 20 g/l. Komposisi medium ini dapat dimanfaatkan untuk penyimpanan karika dieng selama 16 minggu tanpa sub-kultur. Konsentrasi medium MS 50% direkomendasikan untuk digunakan dalam penyimpanan C. pubescens selama 16 minggu tanpa sub-kultur.

Abstract

In this research, we have studied synthesis of improvement of andrographolid bioactive compound on cell culture of sambiloto by addition of jasmonic acid. The essential problems in this research are firstly, the effects of addition of jasmonic acid either can induce or not andrographolide synthesis improvement of cell culture of sambiloto and secondly, to observe the largest content of andrographolide in jasmonic acid concentrations. Meanwhile, the purpose of this research are to observe the functions of jasmonic acid elicitor for induction of andrographolide synthesis improvement of cell culture of sambiloto and to optimize jasmonic acid concentrations which can produce the largest andrographolide content. The independent variable is concentration of addition of jasmonic acid on cell culture and the dependent variable are the growth of cell suspension culture and andrographolide bioactive content. Experiment result show that the optimum medium of sambiloto cell consist of Murashige & Skoog (1962) medium supplemented by 0,5 ppm kinetin and 2,4-D 5 ppm. The cell growth phases are the followings : lag phase at age of 0-5 days, exponential phase of 5-15 days, and stationary phase at age of longer than 15 days. The highest andrographolide was 4,66 x 10-2 reached in cell culture was supplemented with 10 µM jasmonic acid. Keywords : andrographolide, sambiloto cell suspension culture, jasmonic acid elicitor.

Abstract

Anggrek merupakan tanaman dengan nilai ekonomis tinggi. Pemeliharaan anggrek memerlukan peralatan dan ketrampilan khusus supaya dapat tumbuh dan berkembang dengan baik. Griya anggrek Mekar Sari merupakan salah satu rumah anggrek di wilayah Kalisegoro yang mempunyai potensi besar untuk berkembang. Pengembangan rumah anggrek ini terkendala adanya kurangnya sarana dan keterampilan pengelola dalam budidaya anggrek maupun pemasaran anggrek. Saat ini peralatan yang digunakan masih sangat terbatas. Selain itu pengadaan pupuk dan juga pembasmi hama juga masih terkendala oleh dana. Perawatan anggrek saat ini hanya berkisar penyiraman saja karena masih terbatasnya pengetahuan dan ketrampilan Ibu-Ibu PKK RW 4 Kalisegoro. Dukungan untuk mengatasi hal tersebut perlu adanya penyediaan alat dan bahan yang bekaitan dengan budidaya anggrek, pelatihan mengenai budidaya anggrek dan pelatihan pemasaran anggrek. Strategi pemasaran anggrek akan didukung dengan pembuatan web Griya Anggrek Mekar Sari sehingga Griya Anggrek Mekar Sari tidak hanya dikenal di sekitar lingkungan Kalisegoro tetapi juga daerah lain. Web Griya Mekar sari telah berhasil dikembangkan. Penambahan koleksi anggrek, peralatan dan juga bahan untuk pemeliharaan anggrek meningkatkan kualitas dan kuantitas anggrek. Peningkatan kualitas dan kuantitas anggrek serta adanya strategi pemasaran meningkatkan omzet penjualan anggrek. Selain itu keterampilan peserta juga meningkat dengan adanya pelatihan persilangan anggrek dan tambahan pengetahuan berkaitan dengan display anggrek pada showroom.

Abstract

Kecamatan Gunungpati Semarang memiliki potensi untuk menjadi sentra pertanian kebun buah dan sayur organik salah satunya melalui kegiatan Kelompok Wanita Tani (KWT). Namun selama ini, kegiatan yang ada di KWT dan hasil pertanian belum optimal. Hal ini dikarenakan belum efektifnya penataan tanaman di kebun pekarangan rumah, masih ada lahan kebun yang kosong belum dimanfaatkan, belum ada pembukuan kegiatan dan manajemen penjualan hasil pertanian, masih rendah difersifikasi hasil pertanian menjadi produk bernilai jual tinggi, dan minimnya pengetahuan dan sarana pembuatan pupuk kompos dari limbah organik di lingkungan sekitar sebagai upaya meminimalisir pengeluaran untuk kebutuhan pupuk. Berdasarkan permasalahan tersebut, iptek bagi masyarakat (IbM) melalui program pengabdian masyarakat dilakukan dengan cara pemberian pengetahuan dan keterampilan di bidang optimalisasi lahan, pembukuan dan manajemen hasil pertanian, difersifikasi jahe menjadi minuman jahe instan, dan pembuatan pupuk kompos. Luaran yang dihasilkan adalah peningkatan keterampilan anggota KWT dalam optimalisasi lahan untuk pertanian organik, terampil dalam mengolah jahe menjadi minuman jahe instan, membuat pupuk organik, serta kompeten dalam pembukuan dan manajemen penjualan hasil pertanian.

Abstract

Pembelajaran materi organisasi sistem kehidupan di SMP Negeri 3 Pabelan tidak dapat dilakukan dengan pengamatan langsung karena keterbatasan sarana dan prasarana. Media kartu pop up  card adalah salah satu yang dapat digunakan. Penelitian ini bertujuan untuk mengetahui efektivitas pop up card dalam pembelajaran materi organisasi kehidupan di sekolah ini. Sampel dalam penelitian ini diambil dengan teknik purposive sampling. Metode yang digunakan ialah metode “True Experimental Design” bentuk “Pretest-Posttest Only Control Design”. Data yang diambil adalah data hasil belajar siswa, aktivitas siswa, tanggapan siswa dan tanggapan guru. Berdasarkan hasil uji t perbedaan mean antara pretest posttest diperoleh t hitung = 1.679 dan t tabel = 1.671, untuk t> t tabel, maka Ho ditolak. Analisis hasil penelitian menunjukkan bahwa siswa kelas eksperimen mencapai ketuntasan 93,94%, sedangkan kelas kontrol 78,13%. Aktivitas siswa kelas eksperimen, 72,7% siswa pada pertemuan pertama dan 84,8% siswa pada pertemuan kedua tergolong aktif dan sangat aktif. Berdasarkan hasil penelitian disimpulkan, penggunaan kartu pop up card efektif dalam pembelajaran sistwm organisasi kehidupan di SMP N 3 Pabelan Learning material organization of life systems in SMP Negeri 3 Pabelan can not be carried out by direct observation because of limitations facilities and infrastructure. Pop up card is media that can be used. This research aims to determine the effectiveness of pop up card in learning material organization of life in this school. The samples in this study were taken by purposive sampling technique. The method is the True Experimental Design form Pretest-Posttest Only Control Design. The data is taken from student learning outcomes, student activities, student and teacher responses. Based on the results of t-test the mean difference between pretest posttest obtained t = 1.679 and the t table = 1.671, t> t table, so Ho is rejected. Analysis of the research showed that the experimental class students achieve mastery 93.94%, while the control class is 78.13%. Experimental activity graders, 72.7% of students at the first meeting and 84.8% of students in the second meeting have an active and very active criteria.  Based on the results of this study concluded that pop up card is effectively used in learning the material organization of life systems in SMP N 3 Pabelan

Abstract

Penelitian ini bertujuan untuk mendeskripsikan keefektifan pembelajaran dengan menggunakan LKS IPA terpadu pada tema pencemaran lingkungan terhadap penanaman nilai karakter dan pemahaman konsep siswa kelas VII. Populasi yang diambil adalah siswa SMP Negeri 1 Tempuran kelas VII semester genap tahun ajaran 2012/2013. Jenis penelitian ini merupakan penelitian eksperimen. Desain penelitian yang digunakan adalah post test only control design dengan menerapkan pembelajaran menggunakan  LKS IPA terpadu pada kelas eksperimen dan kelas kontrol yang menerapkan pembelajaran tanpa menggunakan LKS. Pengambilan data melalui observasi pengukuran nilai karakter dan pemahaman konsep yang berupa hasil belajar kognitif. Hasil analisis data penelitian menunjukkan nilai karakter dan hasil belajar kognitif kelas eksperimen lebih baik dibandingkan dengan kelas kontrol. Rata-rata nilai karakter komunikatif sebesar 88, rasa ingin tahu sebesar 86,9 dan kreatif sebesar 78,3 pada kelas eksperimen dan pada kelas kontrol rata-rata nilai karakter komunikatif sebesar 78,1, rasa ingin tahu sebesar 71,8 dan kreatif sebesar 63,1. Sedangkan hasil belajar kognitif pada kelas eksperimen sebesar 88,63 dan 80,19 pada kelas kontrol. Berdasarkan hasil penelitian tersebut, dapat disimpulkan bahwa penerapan LKS IPA terpadu tema pencemaran lingkungan efektif terhadap penanaman nilai karakter dan pemahaman konsep. The purpose of this study to describe the effectiveness of learning by using the Integrated Science worksheets on the theme of environmental pollution to instill character values and understanding concept of grade VII. Population of this study are all of students grade VII of SMP Negeri 1 Tempuran on 2012/2013 academic year. The study design was used  post-test only control design which on experimental class the learning using the Integrated Sciences student worksheet and control class, the learning without the use of worksheets. The data were collected from the observation to measurement of the character value and cognitive test to measurement understanding concept. The result of the study showed that the average of the character value and understanding concept on axperimental class better than control class.The average value of communicative character is 88, curiosity character is 86.9 and creative character is 78.3 in the experimental class and on the control class, average communicative character values ​​is 78.1, 71.8 of curiosity characterand creative character is 63.1. While the average of cognitive test on the experimental class is 88.63 and 80.19 on the control class. Based on these results, it can be concluded that the application of Integrated Sciences worksheet on the theme environmental pollution effective against the character value and understanding concepts.
