Abstract

Recent scientific developments in the stem cell field have made it possible to create complex organoids, or structures that resemble complete organs, in vitro. In the majority of these methods, stem cells produced from stem cells or tissue progenitors are allowed to self-organize into three-dimensional (3D) structures using culture systems in three dimensions. The purpose of this study is to ascertain the use of organoid culture in the area of health. This kind of study employs the literature review methodology. The Preferred Reporting Items for Systematic Review and Meta-Analysis (PRISMA) criteria are referenced throughout the stages of the literature review performed in this study. The four processes that make up this level are identification, screening, eligibility, and acceptance. These discoveries showing that human stem cells and patient-derived pluripotent stem cells can be used to generate organoids open up a wide range of opportunities for modeling and disease development, toxicological research and medication discovery, and the field of regenerative medicine. Here, we discuss some of the most significant recent advancements in 3D human organoid production as well as the field's historical advancements. Finally, we discuss present restrictions and provide illustrations of how organoid technology may be used in the healthcare industry.

Abstract

Snails exhibit remarkable adaptability, allowing them to flourish in diverse environmental conditions and resulting in thriving populations in specific regions. This abundance has led communities to harness snails for various purposes, including their use as animal feed, daily dietary source, and in traditional wound-healing practices with historical roots. The primary objective of this systematic review is to identify the snail species commonly employed in wound healing and evaluate the bioactivity of compounds derived from different snail species. This review was conducted using literature review method, drawing from international databases such as Scopus, and encompassed publications from 2013 to 2023. A total of 22 articles met the inclusion and exclusion criteria. Snail body parts that have been explored for wound-healing purposes include both the body and the shell, along with snail secretions, particularly their mucus. Various methods have been employed to extract mucus, involving manual stimulation of the snail's body, spraying with a saline solution (NaCl), application of electric shock, and the use of ozone gas through nebulisation. Prominent snail species found to be beneficial for wound healing include Achatina fulica, Helix aspersa, Eobania desertorum, Helix lucurus, Cornu bistrialis, Theba pisana, and Megalobulimus lopesi. These snail species demonstrate potential applications in the treatment of burns, excision wounds, incision wounds, and diabetic ulcers. Key compounds within snail secretions encompass mucopolysaccharides, polyphenols, peptides, and glycosaminoglycans. These compounds exert significant effects on haemostasis, inflammation control, cellular proliferation, and re-epithelialisation, significantly contributing to the wound healing process.

Abstract

Inflammation is a serious health problem that needs treatment. The use of steroids and non-steroidal anti-inflammatory drugs (NSAIDs) can actually relieve inflammatory reactions well, but their long-term use can have many side effects and are uneconomical. Therefore, the use of natural ingredients that are effective and economical is needed. One of the plants that are considered anti-inflammatory is the leaves of miang beans (Mucuna pruriens). The purpose of compiling this narrative review is to analyze the phytochemical content of miang beans leaves and its mechanisms for inflammatory reactions. The narrative study of this preparation was carried out using the literature study method from August to September 2020. The phytochemical content of miang bean leaves are alkaloids, flavonoids, saponins, cyanogenic glycosides, and tannins. Phytochemicals that have a direct anti-inflammatory effect are alkaloids, flavonoids, saponins, and tannins. Alkaloids are antibacterial. If there are bacteria that cause inflammation, the alkaloids will damage the bacterial peptidoglycan cell walls, thus causing bacterial cell death and preventable inflammatory reactions. Flavonoids have anti-inflammatory effects by inhibiting macrophages from producing NO (nitric oxide) and inhibiting the cyclooxygenase pathway. Saponins affect the inflammatory reaction by inhibiting the lipoxygenase pathway and inhibiting the release of inflammatory mediators. Meanwhile, tannins affect the inflammatory reaction by inhibiting macrophages from producing ROS (Reactive Oxygen Species). Potential which is fatal because it can turn into cyanide acid. However, the content of these cyanogenic glycosides can be minimized by drying at low temperatures. So, mung bean leaves are effective as anti-inflammatory drugs because the alkaloids, flavonoids, saponins, and tannins they contain are anti-inflammatory.

Abstract

AbstractInflammation is a serious problem that needs to be treated. The use of steroid and non-steroidal drugs can treat inflammation well, but long-term use causes many side effects. So, it is necessary to use effective natural medicinal ingredients, one of which is velvet bean leaves (Mucuna pruriens L. (DC.)). This study aims to analyze the effect of velvet bean leaf ointment on the level of leukocyte components in mice experiencing inflammation. This research method was an experiment with a completely randomized design consisting of 6 treatment groups (normal, negative control, positive control (ketoconazole ointment)), velvet bean leaf ointment treatment at doses of 200, 400, and 600 mg/kgBW. The results showed that the use of velvet bean leaf ointment at doses of 200 and 400 mg/kgBW was able to significantly reduce the quantity of total leukocytes and monocytes in mice compared to the negative control (P <0.05); ointment at a dose of 200 mg/kgBW was able to significantly reduce the quantity of granulocytes and lymphocytes in mice compared to the negative control (P <0.05). The ability of velvet bean leaf ointment at a dose of 200 mg/kgBW is better in reducing the leukocyte quantity component compared to commercial drugs (ketoconazole ointment). Therefore, velvet bean leaf ointment at a dose of 200 mg/kgBW has great potential to be developed into an effective standardized anti-inflammatory drug.AbstrakInflamasi merupakan masalah serius yang perlu ditangani. Penggunaan obat steroid dan nonsteroid dapat mengatasi inflamasi dengan baik, namun penggunaan jangka panjang menimbulkan banyak efek samping. Maka dari itu perlu digunakan bahan obat alami yang efektif, salah satunya adalah daun kacang miang (Mucuna pruriens L. (DC.)). Penelitian ini bertujuan untuk menganalisis pengaruh salep daun kacang miang terhadap kadar komponen leukosit pada mencit yang mengalami inflamasi. Metode penelitian ini adalah eksperimen dengan rancangan acak lengkap yang terdiri dari 6 kelompok perlakuan (normal, kontrol negatif, kontrol positif (salep ketokonazol)), perlakuan salep daun kacang miang dosis 200, 400, dan 600 mg/kgBB. Hasil penelitian menunjukkan bahwa penggunaan salep daun kacang miang dosis 200 dan 400 mg/kgBB mampu menurunkan kuantitas total leukosit dan monosit mencit secara bermakna dibandingkan dengan kontrol negatif (P <0,05); Salep daun kacang miang dosis 200 mg/kgBB mampu menurunkan jumlah granulosit dan limfosit pada mencit secara signifikan dibandingkan dengan kontrol negatif (P <0,05). Kemampuan salep daun kacang miang dosis 200 mg/kgBB lebih baik dalam menurunkan komponen jumlah leukosit dibandingkan dengan obat komersial (salep ketokonazol). Oleh karena itu, salep daun kacang miang dosis 200 mg/kgBB memiliki potensi besar untuk dikembangkan menjadi obat antiinflamasi terstandar yang efektif.

Abstract

The prevalence of diabetes mellitus is rising globally. Oxidative stress, which can result from hyperglycemia in diabetes, might have negative consequences. An antioxidant is needed to prevent hyperglycemia. Cathechin isolates are derived from gambir, which has many antioxidants. This study examines catechin isolates from gambir (Uncaria gambir Roxb.) effect on glucose homeostasis in rats induced by alloxan. For this experiment, 35 male rats were employed. Male rats were given alloxan (150 mg/BW, IP), and after 72 hours, blood glucose levels were assessed. If blood glucose levels exceeded 200 mg/dl, three oral catechin isolates were administered (T1=10 mg/kg/day, T2=20 mg/kg/day, and T3=40 mg/kg/day). Following blood collection on the experiment's last day, fasting blood glucose, glucagon and insulin levels were measured. Catechin isolates decreased blood glucose levels in all treatment groups compared to the positive control group (T1 = 150.750 ± 14.359 mg/dl; T2 = 159.750 ± 15.434 mg/dl, and T3 = 153.375 ± 20.207 mg/dl vs 385 ± 60.989 mg/dl) significantly (p value-0.05). A decrease in glucagon serum level was also observed in the treatment group vs positive control (T1: 193.855 ± 36.009 pg/ml, T2 = 286.689 ± 20.313 pg/ml, and T3 = 319.462 ± 30.060 pg/ml vs  529.825 ± 74.279 pg/ml), significantly. Catechine isolates in the T3 group showed an increase in insulin serum level compared to the positive control group significantly (T3 = 216.640 ± 38.230 µIU/ml vs 69.833 ± 3.071 µIU/ml). In conclusion, catechin isolates from gambir decreased blood glucose levels by reducing glucagon and increasing insulin serum levels.

Abstract

Activation of nuclear factor-kB (NF-ĸB) receptors potently enhances pro-inflammatory cytokines production thereby promoting inflammatory reactions. NF-ĸB receptor inhibition is one of the targets in overcoming inflammatory reactions. One of the natural ingredients that have the potential as an anti-inflammatory is velvet bean leaf extract (Mucuna pruriens L. (DC.)) which is used by the local people of West Sumatra. This study aimed to investigate the anti-inflammatory mechanism of the bioactive compounds detected in velvet bean leaf extract against NF-ĸB (1U36) activation pathway using an in silico approach. The phytochemical analysis of the ethanolic extract of velvet bean leaves was performed using GC-MS. Subsequently, their potential bioactivities were explored using PASS online test, preparation of ligands, receptors, and molecular docking. The results showed that the 34 compounds contained in the miang bean leaf extract had anti-inflammatory bioactivity with a probability activity value of >0.7 based on PASS online. The docking results showed that the highest binding affinity value was found in artemin (-7.5 kcal/mol) which was higher than the standard anti-inflammatory drug ketoconazole (-7.2 kcal/mol). Therefore, one of the mechanisms of miang bean leaves as an anti-inflammatory is due to the inhibitory action of artemin on the NF-kB activation pathway.

Abstract

Neurodegenerasi adalah penyakit yang mengakibatkan degenerasi progresif dan kematian sel saraf di otak. Penelitian ini bertujuan untuk mengungkap khasiat neuroprotektif ekstrak biji teratai (Nymphaea pubescens Willd) melalui analisis perilaku neurokognitif. Sebanyak 25 ekor mencit jantan dibagi secara acak menjadi 5 perlakuan: kontrol negatif (Na-CMC 0,5%), kontrol positif diinduksi trimetiltin (TMT) 0,6mg/kgBB, dan pemberian ekstrak biji teratai (100, 200, dan 400 mg/kgBB). Induksi degenerasi otak dengan TMT menggunakan dosis tunggal secara intraperitoneal. Ekstrak diberikan tiga hari pasca injeksi TMT secara oral setiap hari selama 28 hari. Pada akhir perlakuan, tes neurobehavioral dilakukan meliputi uji kecerdasan memori dengan Hebb-Williams Maze, uji keingintahuan dengan Hole-Board dan uji interaksi sosial. Selanjutnya dilakukan pengukuran indeks organ otak. Data dianalisis secara statistik dengan One-Way Anova dilanjutkan dengan uji DNMRT (P<0,05). Hasil penelitian menunjukkan bahwa ekstrak biji teratai dosis 200 dan 400 mg/kgBB secara signifikan dapat meningkatkan kecerdasan kognitif (memori, keingintahuan dan interaksi sosial) pada mencit yang diinduksi TMT, namun tidak berpengaruh signifikan terhadap indeks bobot otak. Temuan penelitian ini mengungkapkan bahwa biji teratai adalah kandidat kuat untuk sumber obat anti-neurodegeneratif.

Abstract

The development of ecotourism in Kapalo Banda, Limau Manis, Padang, has potential to be expanded into agro-ecotourism through multi-stakeholder involvement. Key attractions include the scenic of river flows, agricultural cultivation, and stingless bee keeping, which support environmental sustainability and the local economy. With high vegetation diversity, the potential for stingless bee (galo-galo) breeding as a creative economic initiative offers new opportunities. This program aligns with the West Sumatra Provincial Government’s efforts to establish the area as a honey production center. However, challenges such as low colony production due to limited knowledge of farming techniques and the availability of flowering feed hinder its development. Through community engagement, empowerment activities were conducted from June to September 2024, including socialization, demonstrations, and training in bee farming techniques, supported by expert teams. Education focused on the potential of galo-galo and marketing strategies for local products. The results of this program indicate improved community skills and knowledge, supporting local economic sustainability and biodiversity conservation. Kapalo Banda partners have the potential to serve as a model for community empowerment based on natural resources, effectively enhancing community welfare. Multi-stakeholder involvement in developing Kapalo Banda Ecotourism has resulted in a series of activities ranging from cultivation preparation, colony propagation techniques, harvest preparation, to downstream processing and product marketing.

Abstract

The Sawah Payo Nagari Gunung Rajo farmers group of Batipuh Subdistrict, Tanah Datar Regency has the potential in the production of organic fertilizers and vegetable pesticides because it is supported by the rest of the agricultural products and weeds of Tithonia diversifolia, Piper aduncum and livestock waste (animal waste) which overflow. Utilization of weed crop agricultural waste, whether livestock is still not optimal. The processing of agricultural and agricultural waste into compost carried out by most farmers in Nagari Gunung Rajo has not used the fermentation process, resulting in less than optimal quality and farmers' ignorance in utilizing plants that cause vegetable pesticides. The purpose of this farmer group empowerment activity is to provide knowledge about the process of making organic fertilizers and vegetable pesticides from the supply of raw materials, making bio-activators, and packaging of production results. The method used is structured training and direct practice in making organic fertilizer. The results obtained from this empowerment activity are farmer groups capable of making organic fertilizers and vegetable pesticides independently with a fairly good quality of production results.   Keywords: Empowerment, organic fertilizer, botanical pesticide.   ABSTRAK Kelompok tani Sawah Payo Nagari Gunung Rajo Kecamatan Batipuh, Kabupaten Tanah Datar mempunyai potensi dalam produksi pupuk organik dan pestisida nabati hal ini karena didukung oleh sisa hasil pertanian jerami dan gulma tanaman pahitan Tithonia diversifolia, Piper aduncum dan limbah peternakan (kotoran hewan) yang melimpah. Pemanfaatan limbah pertanian tanaman gulma, maupun peternakan masih belum maksimal. Pengolahan limbah pertanian dan peternakan menjadi kompos yang dilakukan oleh sebagian besar petani di Nagari Gunung Rajo belum menggunakan proses fermentasi, sehingga hasilnya dan kualitasnya kurang maksimal serta ketidaktahuan petani dalam memanfaatkan tanaman yang berpotensi sebagai pestisida nabati. Tujuan kegiatan pemberdayaan kelompok tani ini adalah memberikan pengetahuan tentang proses pembuatan pupuk organik dan pestisida nabati dari penyedian bahan baku, pembuatan bio-aktivator, dan pengemasan hasil produksi. Metode yang digunakan dengan pelatihan terstruktur dan praktik langsung pembuatan pupuk organik. Hasil yang diperoleh dari kegiatan pemberdayaan ini adalah kelompok tani mampu membuat pupuk organik dan pestisida nabati secara mandiri dengan kualitas hasil produksi yang cukup baik.   Kata kunci: Pemberdayaan, pupuk organik, pestisida nabati.

Abstract

Keterbatasan objek dan prasarana penunjang wisata alam di ESGP belum memberikan dampak peningkatkan perekonomian bagi anggota P4S Sunkai Permai. Untuk mengatasi masalah ini telah dibuat kolam ikan koi yang alami untuk menambah daya tarik pengunjung. Rancangan kolam alami menggunakan material batu kali dengan tanaman bunga. Setelah persyaratan kualitas air terpenuhi, kolam diisi dengan 110 ikan koi dewasa dari 13 varietas. Pihak mitra P4S Sunkai Permai mampu merawat kolam dan menjaga kesehatan ikan, pemberian pakan dan menjinakan ikan koi sesuai dengan panduan yang diberikan. Pihak mitra juga memberikan pelayanan sesuai panduan edukasi yang mencakup sejarah dan asal usul, makna dan simbol memelihara ikan koi, biologi ikan koi, dan varietas ikan koi. Objek wisata ikan koi telah meningkatkan jumlah kunjungan ke ESGP dalam jangka waktu tiga bulan. Memperhatikan daya tarik objek ikan koi tersebut, direkomendasikan pengayaan objek dengan berbagai ikan hias lainnya di areal ESGP, yang pada akhirnya dapat lebih menigkatkan jumlah pengunjung.
