Abstract

Stunting in toddlers is related to growth and development disorders. One important factor in preventing stunting is the implementation of Clean and Healthy Living Behaviour (CHLB). CHLB plays a role in improving the nutritional and health status of children, as well as preventing various diseases that can contribute to stunting. The role of cadres is very important because cadres are responsible for implementing the posyandu program, are the main movers in promotive, preventive activities and are motivators for community members. The aim of this service is to analyse knowledge about PHBS among posyandu cadres in Tamanan Village, Tamanan District, Bondowoso Regency. The service method is an outreach activity aimed at providing knowledge about CHLB with the hope that people will change their behaviour to protect their health and the environment independently. Formative evaluation procedures in the form of pretest and posttest. Data were analyzed using descriptive analysis techniques. Continue with the normality test and paired sample t-test. The results obtained were that this community service activity was carried out on all posyandu cadres. Evaluation showed that there was a difference in the average test scores before and after the counselling. Providing counselling is able to provide changes in the knowledge of posyandu cadres which is marked by an increase in posttest scores which are higher compared to posttest scores. The conclusion is that the knowledge of Tamanan Bondowoso Village Posyandu cadres regarding CHLB in efforts to reduce stunting in toddlers can be increased through the counselling provided. Keywords: CHLB, counseling, posyandu cadres, stunting.

Abstract

Background: Non-steroidal anti-inflammatory drugs (NSAID) are widely used in temporomandibular joint osteoarthritis management. However, the side effects of NSAIDs on multiple organs need to be anticipated. Curcumin is known for its anti-inflammatory and analgesic potential, comparable to that of NSAIDs. In a previous study, structurally modified curcumin increased the pharmacological effect and simultaneously reduced the toxicity and side effects of curcumin. Pentagamavunon-0 (PGV-0) is one of the active components synthesized by the structure modification of curcumin. Purpose: In this study, we identify the potential target of PGV-0 on the pathogenesis of temporomandibular arthritis characterized by inflammation. Methods: We used a bioinformatics approach to compare the PGV-0 target with curcumin and diclofenac sodium as controls. We identified overlapping gene targets of bioactive compounds (PGV-0, curcumin, or diclofenac sodium) retrieved from the SwissTargetPrediction and GeneCards platforms, specifically for temporomandibular arthritis. An interaction model among targets was developed using the STRING database and Gene Ontology Panther to expound on the bioactive compound’s function on the key signaling pathway. Finally, we formulated a molecular docking prediction between the bioactive compound and the target protein marker derived from the previous analysis using Molecular Operating Environment tools. Results: This study found that curcumin and PGV-0 targeted different molecular pathways in temporomandibular arthritis compared to diclofenac sodium. Curcumin and PGV-0 shared a similar pathway to curcumin by modulating metalloproteinases (MMPs), especially MMP-9 and MMP-13. Moreover, diclofenac sodium influenced cyclooxygenase metabolism. Conclusion: In this study, PGV-0 targeted metalloproteinase in temporomandibular arthritis pathogenesis. This finding underlines the PGV-0 advantage in preventing metalloproteinase-related tissue damage in temporomandibular arthritis.

Abstract

Flavonoid has potential bioactivity as anticancer agents. The flavonoid of cultivated tobacco (Nicotiana tabacum), locally known as “Kasturi”, leaves was screened for its cytotoxicity against MCF-7 human breast cancer cells and non-transformed Vero cells (African normal cell kidney line) in different concentrations. This study aimed to examine the cytotoxic potential of the flavonoid of Kasturi tobacco leaves against MCF-7 human breast cancer cells. Flavonoid obtained from methanolic extracts of Kasturi tobacco leaves, which have been purified from nicotine. The flavonoid of Kasturi tobacco leaves with concentrations of 20 to 640 μg/mL were exposed to MCF-7 and Vero cells for 24 h. Cell viability was evaluated by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. Flavonoid of Kasturi tobacco leaves with concentrations of 160 μg/mL decreased the MCF-7 cell viability more than 50%, with an inhibitory concentration 50 (IC50) value of 148.41 μg/mL. Meanwhile, it inhibited 50% of Vero cell viability at 255.35 μg/mL. The flavonoid of Kasturi tobacco leaves has cytotoxic activity on MCF-7 cells, and might be a potential alternative agent for human breast cancer therapy. Keywords: flavonoid, tobacco leaves, human breast cancer cells, anticancer activity

Abstract

Periodontitis is an inflammatory disease of the periodontal tissues primarily caused by Porphyromonas gingivalis (P. gingivalis). In recent years, probiotics have been proposed as a potential bacteriotherapeutic approach for periodontitis. Probiotic bacteria such as Lactobacillus casei (L. casei) are known to produce antibacterial compounds, which can be found in their culture supernatants. The ability of L. casei to grow and synthesize these substances depends on environmental factors, including pH. Based on preliminary findings, a pH of 6.5 was used in this study. This study aimed to evaluate the antibacterial activity of L. casei supernatant against P. gingivalis in vitro. The research employed an experimental post-test-only group design. Two control groups were included: a negative control (aquadest) and a positive control (0.2% chlorhexidine). The treatment groups received L. casei supernatant at volumes of 20 µl, 50 µl, and 80 µl, with incubation periods of 6, 12, and 24 hours. The results demonstrated that the L. casei supernatant exhibited the highest antibacterial activity against P. gingivalis at a volume of 80 µl after 12 hours of incubation. The Shapiro–Wilk test indicated that the data were normally distributed, while Levene’s test revealed a lack of homogeneity. Post hoc analysis showed significant differences among all treatment groups at the 12-hour incubation point. In conclusion, the supernatant of L. casei exhibits in vitro antibacterial activity against P. gingivalis.

Abstract

Background: Theobroma cacao was considered agricultural wastes. Cocoa pod husk contain various bioactive compounds as natural antioxidants, antibacterial and anti-inflammatory. A good biomaterial must be non-toxic and have no detrimental effect on the biological environment. The aim of research was to determine the cytotoxicity and proliferative activity of cocoa pod husk extract on primary cells of periodontal ligament fibroblasts. Method: This experimental laboratory used primary cell culture of periodontal ligament fibroblasts. The cytotoxicity and proliferative activity were analyzed by MTT assay [2-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide]. Extraction of cacao pods were obtained using the ultrasonic homogenizer method with ethanol 70%. The cytotoxicity test was observed for 24 hours and the doubling time proliferative test was observed for 24, 48, and 72 hours. Result: Cocoa pod husk extract showed there were no cytotoxic effects with viability of primary cells of periodontal ligament fibroblasts at the concentration of 1.56 pg/ml, 3.125 pg/ml, 6.25 pg/ml, 12.5 pg/ml, 25 pg/ml, 50 pg/ml, and 100 pg/ml. The proliferation test showed increasing of fibroblasts at 24 and 48 hours. Kruskal Wallis analysis obtained a significance value (p<0.05) Conclusion: Cytotoxicity and proliferation assays of Cocoa pod husk extract increased cell viability and cell proliferation of periodontal ligament fibroblast.
