Abstract

Background: Gingival tissue and periodontal ligament act as sources of mesenchymal stem cells (MSCs) that play a vital role in periodontal regeneration, but they both have limitations for cell availability. MSCs cultivated and expanded in various media formulations could be used as a basis for the development of cell therapy protocols. Purpose: This study aimed to determine the optimum culture media formulation for cultivation and expansion of human gingival-derived mesenchymal stem cells (hGMSCs) and human periodontal ligament stem cells (hPDLSCs). Methods: The hGMSCs and hPDLSCs were obtained from gingival tissue and periodontal ligament specimens from an adult patient. The two different culture media formulations used were: 1) α-minimum essential media (α-MEM) supplemented with 10% FBS, 100 U/mL penicillin, 100mg/mL streptomycin and 2.5 µg/mL amphotericin B; and 2) Dulbecco’s minimum essential media-Low Glucose (DMEM-LG) supplemented with 10% FBS, 2 mMol/L L-glutamine, 100 U/mL penicillin, 100mg/mL streptomycin and 2.5 µg/mL amphotericin B. The minced-gingival tissue and periodontal ligament samples were seeded in 3 cm tissue culture dishes with one of two experimental culture media, and incubated at 37oC in a humidified atmosphere of 5% CO2. Results: Cell morphology was observed on days two and five of the third passage. The gingival tissue and periodontal ligament primary cells exhibited fibroblast-like morphology, long processes and were spindle-shaped. The hPDLSCs grown in α-MEM exhibited a significant increase in cell viability and proliferation rate compared to the hPDLSCs grown in DMEM-LG. However, hGMSCs displayed similar cell viability and proliferation rate on both types of experimental media. Both the hGMSCs and hPDLSCs expressed MSC markers, including CD105, CD146, and CD90, but did not express CD45. Conclusion: Culture media formulations of α-MEM and DMEM-LG can be used for the cultivation and expansion of both hGMSCs and hPDLSCs.

Abstract

Oral cavity diseases can be caused by the oral microbes  such as Streptococcus mutans, Porphyromonas gingivalis and Candida albicans. The oral microbes growth can be inhibited by chlorhexidine gluconate mouthwash, but its long term use have adverse effects, therefore it is important to do a further research to find effective antibacterial and antifungal as alternative substance mouthwash. Flavonoid compounds in tobacco leaf have potential as an antibacterial and antifungal. The objective was to analyze the inhibition effect of tobacco leaf extract to the growth of Streptococcus mutans, Porphyromonas Gingivalis and Candida albicans. This research used 20%, 40%, 60%, 80% and 100% ethanol extract of tobaco leaf,  1% DMSO as negative control and 0.2% chlorhexidine gluconate as positive control. Antibacterial and antifungal activities were tested by diffusion disc method with four repetitions. The observation was conducted by measuring the diameter of inhibition zone after incubating for 24 and 48 hours. The largest inhibition zone of Streptococcus mutans growth were producted by ethanol extract of tobacco leaf with 80% concentration, the largest inhibition zone on Porphyromonas gingivalis and Candida albicans growth were produced by ethanol extract of tobaco leaf with 100% concentration. Diameter of inhibition zone ethanol extract of tobacco leaf was equal with the diameter of inhibition zone of 0.2 % chlorhexidine gluconate. The conclusion described that ethanol extract of tobacco leaf can inhibit the growth of Streptococcus mutans, Porphyromonas gingivalis and Candida albicans.

Abstract

Periodontal disease affects oral health in the mast people. Some researches have found and review the relationship between periodontal disease and systemic disease, such as cardiovascular disease, diabetes, stroke, and pregnancy complication. However, there is detailed explanation about the mechanism that analysis how the relationship between periodontal disease and pregnancy complication. The objective of this study was to review biological mechanism that correlated periodontal disease and pregnancy complication. Periodontal disease was able to cause alteration abnormal systemic immune which underlies pregnancy complication. Periodontal pathogen bacteria were directly colonized in placenta, and they caused local inflammation respond that caused pre-maturity and other pregnancy complications.

Abstract

The changes of hormonal during pregnancy can increase the hormones estrogen and progesterone. Increased estrogen and progesterone can cause excessive response in periodontal tissues so easily happen inflammation of the periodontal tissues. The most important aspects of the inflammation is the leukocyte reaction. The purpose of this study was to determine the effect of periodontal disease during pregnancy to the total number of leukocytes and leukocyte counts. Research procedures performed each trimester which includes examining periodontal index (PI) to assess the severity of periodontal disease and total leukocyte counts and leukocyte counts were taken from venous blood. The data obtained were analyzed with linear regression test. Statistical analysis showed that the higher the score PI will increase the total number of leukocytes. The conclusion of this study is periodontal disease during pregnancy may increase the total number of leukocytes and leukocyte counts.

Abstract

Periodontal disease has been involved in a variety of systemic disorders and suspected as a potential risk factor for fetal growth restriction. Periodontal pathogenic bacteria may actively regulate embryonic development, implantation and placental trophoblast cell invasion. This study aimed to analyze the role of TNF-α, IL-10 and caspase-3 to maintain fetal growth in Porphyromonasgingivalis-infected pregnant rats. Female rats were infected with live-Porphyromonas gingivalis at concentration of 2x109 cells/ml into subgingival sulcus area of the maxillary first molar before and during pregnancy. They were sacrificed on gestational day (GD)-14 and GD20. The weight and length of placentas and fetuses were evaluated. The expression of TNF-α, IL-10 and caspase-3 in macrophages and trophoblast cells were detected by immunohistochemistry. On GD14, TNF-α (R2=0.416;P=0.000) and IL-10 (R2=0.187;P=0.012) had an important role to increase expression of caspase-3 in the placenta, but only TNF-α (R2=0.393;P=0.000 ) was able to increase the expression of caspase-3 on GD20. TNF-α and caspase-3 also had an important role (P<0.000) to decrease fetal weight, fetal length and placental weight on GD14 and GD20, but it was not the case with IL-10 (P>0.000). The increasing expressions of TNF-α and IL-10 did not only enhance immune protection, but also maintained the trophoblast cells survival by regulating expression of caspase-3. Porphyromonas gingivalis infection in maternal periodontal tissue can lead to decrease in placental weight, fetal weight and fetal length which mediated by increasing expression of TNF-α, IL-10 and caspase-3 in the placenta.

Abstract

Background: Theobroma cacao was considered agricultural wastes. Cocoa pod husk contain various bioactive compounds as natural antioxidants,antibacterial and anti-inflammatory. A good biomaterial must be non-toxic and have no detrimental effect on the biological environment. The aim of research was to determine the cytotoxicity and proliferative activity of cocoa pod husk extract on primary cells of periodontal ligament fibroblasts. Method: This experimental laboratory used primary cell culture of periodontal ligament fibroblasts. The cytotoxicity and proliferative activity were analyzed by MTT assay [2-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide]. Extraction of cacao pods were obtained using the ultrasonic homogenizer method with ethanol 70%. The cytotoxicity test was observed for 24 hours and the doubling time proliferative test was observed for 24, 48, and 72 hours. Result: Cocoa pod husk extract showed there were no cytotoxic effects with viability of primary cells of periodontal ligament fibroblasts at the concentration of 1.56 pg/ml, 3.125 pg/ml, 6.25 pg/ml, 12.5 pg/ml, 25 pg/ml, 50 pg/ml, and 100 pg/ml. The proliferation test showed increasing of fibroblasts at 24 and 48 hours. Kruskal Wallis analysis obtained a significance value (p<0.05) Conclusion: Cytotoxicity and proliferation assays of Cocoa pod husk extract increased cell viability and cell proliferation of periodontal ligament fibroblast.

Abstract

Latar belakang Kualitas hidup manusia ditentukan oleh tingkat sosiodemografi, status kesehatan umum dan rongga mulut yang saling berkaitan. Ras dan suku menentukan genetika dalam merespon keradangan, kerentanan jaringan rongga mulut terhadap bakteri atau injuri, meregulasi hormon reproduksi, dan sindrom menopause. Akan tetapi hubungan faktor tersebut masih belum banyak terungkap, khususnya pada Suku Osing. Suku Osing merupakan salah satu suku yang masih memegang kuat adat istiadat.Tujuan penelitian ini adalah untuk mengetahui status kesehatan rongga mulut wanita suku Osing. Metode Penelitian observasional dengan desain cross sectional.Subyek penelitian dikelompokan menjadi kelompok usia produktif dan menopause. Pada subyek penelitian dilakukan pemeriksaan status kesehatan rongga mulut meliputi jumlah gigi yang tersisa di rongga mulut, indeks periodontal, karies dan kebersihan rongga mulut. Semua data dikategorikan kemudian akan dilakukan uji korelasi non parametric (p?0,05). Hasil Kelompok wanita usia menopause pada penelitian ini sudah mengalami menopause dalam kurun waktu 5-10 tahun.Jumlah gigi wanita usia menopause lebih sedikit dibanding wanita usia produksif (p?0,05). Wanita usia menopause lebih banyak menderita penyakit periodontal yang bersifat irreversible (2,65 ± 0,35) daripada wanita usia produktif (1,16 ± 0,27). Indeks karies kelompok wanita usia menopause (D=166, M=570) lebih tinggi dibanding wanita usia produktif (D=247, M=162). Akan tetapi, kedua kelompok ini mempunyai tingkat kebersihan mulut yang sama. Selain itu terdapat hubungan antara tingkat kebersihan mulut, penyakit periodontal, karies dan lamanya menopause (R>0,3). Simpulan Status kesehatan rongga mulut wanita usia menopause suku Osing di Desa Kemiren, Kecamatan Glagah, Banyuwangi lebih buruk dibanding wanita usia produktif. Akan tetapi, perlu penelitian lebih lanjut mengenai faktor-faktor yang mempengaruhi status kesehatan rongga mulut tersebut. Background Social-demography, health status, and oral health specify a quality life, which all of them are correlated. Races and ethnic assign genetic aspect, especially in inflammation respond, oral tissue susceptibility to bacterial infection and injuries, hormone regulation, and menopause syndrome. However, the relationships are unexplored yet, especially in osingese. Osingese is one of ethnic which hold the customs strongly. The objective of this study was to know the oral health status of Osingese Women. Method This study was observational with a cross-sectional design. The subjects were classified into productive and menopause age. All of the subjects were examined their oral health, including remain teeth, periodontal index, caries index, and oral hygiene index. All of the data were categorized and analyzed by non-parametric correlation analysis (p?0.05). Result Menopause aged group experienced menopause period about 5-10 years. The number of teeth of the menopause group was less than productive group (p?0.05). The menopause group more sustained irreversible periodontal diseases (2.65 ± 0.35) than the productive group (1.16 ± 0.27). Caries index in the menopause group (D=166, M=570) was higher than the productive group (D=247, M=162). However, their oral hygiene index was the same. Moreover, there presented the relationship between oral hygiene, caries index, periodontal index, and menopause status (R>0.3). Conclusion Oral health status menopause aged osingese women was poorer than the productive group. However, it needed further study to investigate the other factor influencing oral health status. Abstract

Proses penguraian sampah organik dengan biokonvertor Black Soldier Fly (BSF) dilakukan pada tahap larva, sehingga saat BSF menjadi lalat dewasa, bertelur dan mati akan meninggalkan limbah media bekas maggot, cangkang pupa dan bangkai lalat. Limbah media bekas maggot digunakan untuk bahan kompos, sedangkan limbah cangkang pupa dan bangkai lalat belum dimanfaatkan. Timbunan limbah cangkang pupa dan bangkai lalat setelah siklus hidup BSF dikhawatirkan bisa menimbulkan pencemaran udara dan lingkungan jika dibiarkan menumpuk di halaman rumah. Berdasarkan analisis situasi tersebut, tim ProbangDebi UNEJ bersama Bank Sampah Ekoliterasi Dermaku Jenggawah merancang kegiatan yang bertujuan untuk memberikan edukasi tentang teknologi pengolahan limbah cangkang pupa agar memiliki nilai ekonomi. Metode pelaksanaan kegiatan adalah edukasi potensi limbah cangkang pupa BSF dan pelatihan teknologi pengolahan limbah cangkang pupa BSF. Data pre-test dan post-test menunjukkan bahwa terjadi peningkatan pengetahuan pembudidaya maggot di Desa Jenggawah tentang pemanfaatan limbah cangkang pupa BSF menjadi produk yang memiliki nilai tambah. Pengolahan limbah cangkang pupa BSF dapat ditingkatkan kapasitasnya menjadi skala yang lebih besar lagi seiring dengan peningkatan kebutuhan pasar dalam pemanfaatan kitin dan kitosan. Selanjutnya, dibutuhkan pelatihan pemasaran melalui platform e-commerce dan koordinasi lintas sektoral untuk memberikan dukungan atas upaya pengolahan limbah menjadi produk yang bermanfaat bagi kesejahteraan dan perekonomian masyarakat. Kata kunci— Black Soldier Fly, Budidaya Maggot, Cangkang Pupa, Kitin, Kitosan Abstract The process of decomposing organic waste using the Black Soldier Fly (BSF) bioconverter is carried out at the larval stage. BSF becomes adult flies, lay eggs and die, which will leave maggot cultivation media, pupa shells and fly carcasses. Maggot cultivation media is used as compost material, while pupa shells and fly carcasses have not been utilized. Piles of pupa shells and fly carcasses can cause air and environmental pollution. The ProbangDebi UNEJ team and the Dermaku Jenggawah Ecoliteracy Waste Bank designed activities aimed at providing education on pupa shell processing so that it has economic value. The implementation method is education and training on BSF pupa shell processing technology. Pre-test and post-test data show that there has been an increase in the knowledge of maggot cultivators in Jenggawah Village regarding the use of BSF pupa shells into products that have added value. BSF pupa shell processing capacity can be increased to a larger scale in line with increasing market demand for the use of chitin and chitosan. Furthermore, marketing training through e-commerce platforms and cross-sectoral coordination is needed to provide support for efforts to process waste into products that are beneficial for the welfare and economy of the community. Abstract

Mount Semeru as one of the highest mountains on the Java island has significant volcanic activity to date. The eruption of this volcano had a considerable impact on the surrounding area, including Sumberwuluh Village, Candipuro District, Lumajang Regency. The impact felt by the community is not only physical damage but also trauma. Therefore, a disaster awareness culture needs to be built and strengthened early in order to reduce the impact caused when a disaster occurs. The Institute for Research and Community Service (LP2M) of the University of Jember in collaboration with the Regional Disaster Management Agency (BPBD) of Lumajang Regency carried out service activities for elementary school students and teachers in Sumberwuluh Village with the aim of strengthening the culture of disaster awareness. This activity has been attended by students and teachers from SDN Sumberwuluh 1, SDN Sumberwuluh 2, SDN Sumberwuluh 3, SDN Sumberwuluh 4, SDN Sumberwuluh 5 and MI Miftahul Ulum. This is carried out through two stages, socialization about the importance of disaster preparedness and direct practice of disaster preparedness consisting of water boat evacuation, webbing and trauma healing. This program has succeeded in increasing disaster preparedness awareness through disaster education from an early age. In addition, this activity also succeeded in improving knowledge and practical skills in disaster preparedness by 88% and building a better mentality and readiness in dealing with emergency situations.
