Abstract

The use of probiotics is increasingly popular as an alternative replacement to antibiotic uses in stimulating healthand growth of broiler chickens. The use of antibiotics may disturb the balance of the gut microbiota and play asignificant role in the emerging of antibiotic resistance pathogens. Thus, the objective of this study was to isolateand screen lactic acid bacteria (LAB) from the intestine of broiler chickens as probiotic candidates. LAB wereinitially screened by phenotypic assay. Furthermore, those strains which belonged to LAB were screened forinhibitory activity against E. coli. Results showed that a total of 4 isolates were considered as members of lacticacid bacteria indicating by the results of phenotypic assays: gram positive, catalase and oxidase negative, and ableto perform carbohydrate fermentation. Out of the 4 isolates, only one isolate showed an inhibitory activity againstthe targeted pathogen. Further research is still needed to confirm the potential strain for probiotic in poultry.

Abstract

Salah satu faktor yang sangat mempengaruhi populasi dan produksi ternak adalah masalah penyakit dan parasit. Gejala serangan parasit seringkali tidak tampak dan kadang-kadang petani kurang menyadarinya. Cacing hati (Fasciola sp.) merupakan parasit zoonosis yang dapat menginfeksi berbagai macam hewan ternak ruminansia terutama sapi. Fasciolamemiliki dua spesies utama yaitu hepatica dan gigantica. Penyebaran Fasciola sp. adalah daerah beriklim tropis basah yaitu di Asia dan Afrika. Fasciola sp. yang sering dijumpai di Indonesia adalah spesies gigantica, sedangkan spesies hepatica sering terdeteksi pada sapi-sapi impor. Untuk mengetahui infeksidan tingkat penyebaran Fasciola giganticadan Fasciola hepaticapada ternak sapi di kabupaten Lombok Tengahdan Lombok Timurmaka dilakukan survei, pengamatan dan pengambilan sampel organ hati sapi di pasar-pasar daging yang ada pada dua daerah tersebut. Pada masing-masing daerah diambil 3 lokasi pasar daging, yaitu di kabupaten Lombok Tengah: pasar Praya, pasar Mantangdan pasar Kopang; di kabupaten Lombok Timur: pasarSelong, pasarMasbagikdan pasarTerara. Pada masing-masing pasar, pengamatan dan pengambilan sampel diulang sebanyak 3 kalipada 3 orang pedagang.Selanjutnya sampel diamati di laboratorium untuk mengetahui adanya cacing hati dan kerusakan hati sapi. Hasil penelitian menunjukkan bahwa, sapi-sapi dan daging sapi yang diperjual belikan di pasar-pasar Lombok Tengah dan Lombok Timur terinfeksi parasit cacing hati (Fasciola sp.), dengan tingkat infeksi yang masih rendah, yaitu antara 1,3 sampai 2,3. Di samping itu, warna dan tekstur hati sapi yang diperjual belikan di pasar-pasar Lombok Tengah dan Lombok Timur relatif masih baik dan layak dikonsumsi.

Abstract

Serangan virus White Spot Syndrome (WSSV) telah memberikan dampak finansial yang cukup besar sejak tahun 1992. Tingkat mortalitas yang mencapai 100% menyebabkan banyak pembudidaya udang dan lobster menderita kerugian. Untuk mengatasi permasalahan tersebut sangat diperlukan informasi tentang cara penyebaran virus WSSV sehingga dapat diketahui solusi yang tepat untuk menghentikan penyebarannya pada tambak-tambak udang maupun lobster. Pada penelitian ini, Lobster Air Tawar yang dibudidayakan di BBI Aik Bukak dijadikan kontrol negatif (belum terinfeksi virus WSSV). Hasil penelitian ini menunjukkan bahwa penularan virus WSSV pada lobster air tawar dapat berlangsung melalui konsumsi udang yang telah terinfeksi virus WSSV serta melalui aliran air. Pleopod atau kaki renang yang dideteksi dengan teknologi Real Time Polimerase Chain Reaction (RT-PCR) mengandung virus WSSV yang lebih dominan dibandingkan insangKata kunci: White Spot Syndrome Virus, Lobster Air Tawar, RT-PCR

Abstract

Production of functional protein (including vaccine) using conventional technology in embryonated chicken eggs is laborious and lengthy. The use of chemical synthesis is not practical for peptides longer than 20 residues. In contrast, in vitro transcription and translation technology can directly utilize polymerase chain reaction (PCR) product as template for vaccine synthesis within two hours accurately. Moreover, up to 1-10 mg/ml protein can be produced using the technology compared to conventional method that only gives approximately one dose per egg. In this review, advantages and disadvantages of animal vaccine generation using conventional and in vitro methods would be described. In vitro transcription and translation technology can be considered as the most practical and efficient technique for rapid screening and generating new animal vaccines.

Abstract

We have developed a simple and efficient system for screening and generation of monoclonal antibodies, which can bypass conventional monoclonal antibody production system that includes time-consuming, labor-intensive establishment and cultivation. Our method consist of: (1) cDNA synthesis from single B cells of immunized mouse, followed by (ii) PCR amplification of the Lc and Hc (Fd portion) cDNAs separately using low concentration (0,05 &igrave;M) of the respective cDNA-specific primers with 5&rsquo; homotags in the presence of the homotag-specific primer (0,5 &igrave;M), (iii) overlapping PCR of the amplified Lc and Hc cDNAs with the cassettes for the T7 promoter (T7P) and T7 terminator (T7T) to construct the following expression units: T7P-Lc-T7T and T7P-Hc-T7T, and (iv) in vitro expression of these units using an Escherichia coli S30 extract followed by ELISA screening without</div><div>purification. Light-and heavy-chain cDNA were amplified and expressed using in vitro system, thus avoiding time consuming steps of cloning and bacterial expression. Having successfully amplified and expressed Lc and Hc genes from single B cells in rapid, simple, versatile, labor-saving, and cost effective, this method seem to be useful</div><div>and applicable for the high-throughput monoclonal antibody generation.

Abstract

White spot syndrome virus (WSSV) is one of the most threatening diseases in shrimp and othercrustaceans affecting global shrimp farming. Since firstly detected in Taiwan in 1992, the disease hasspread globally and followed with considerable socio-economic consequences. This research was performedto detect the WSSV infection in shrimp farming in Lombok Island’s (West Nusa Tenggara) using real-timepolymerase chain reaction. Samples of vaname (Litopenaeus vannamei) were collected from several shrimpfarming in Lombok. Results indicated that the spread of WSSV has reached shrimp farms in Lombok,especially in Lendang Jae, West Lombok. Therefore, a biosurveillance program is strongly recommendedto government to avoid and halt the spread of the disease in East Indonesia region .

Abstract

This article presents a study on the fatwas (legal opinion) concerning inter faith marriage in Indonesia. The fatwas under the discussion are those issued by Nahdlatul Ulama (NU) and Majelis Ulama Indonesia (MUI) in post-Independence Indonesia. Those fatwas are of special significance, both in terms of Islamic legal discourses and -perhaps more importantly- Muslims' perceptions and attitudes towards other religious communities in Indonesia. Issued by authoritative bodies of Indonesian Islam, more in particular the NU, the fatwas could be said as presenting the voices of Indonesian Muslims in their encounters with the issues of religious tolerance and plurality.DOI: 10.15408/sdi.v9i3.658

Abstract

Artikel ini memperlihatkan bahwa Islam Hadhari berfungsi tidak hanya sebagai respons religio-politik terhadap krisis internal yang dirasakan masyarakat Melayu, polarisasi budaya, dan politik global yang tidak seimbang, tetapi juga menunjukkan multi-interpretasi dari teks-teks Islam, sejarah, budaya dan konteks multikultural. Kasus Islam Hadhari menjadi contoh dari "tradisionalisme" dan "modernisme" Islam yang bukan dalam bentuk ideal, tetapi dalam ekspresi diskursif campuran. Islam Hadhari harus dipahami dalam kerangka persaingan ide-ide lokal dan global, yang muncul di negara-negara mayoritas Muslim di era negara-bangsa modern dan globalisasi. Artikel ini menunjukkan pentingnya mengamati reformasi Islam sebagai dialektika gerakan dan kontra-gerakan, wacana dan kontra-wacana, tetapi juga dalam hal waktu (masa lalu, sekarang, dan masa depan). Menggunakan teori modernisasi dalam menjelaskan hubungan antara Islam dan modernitas, artikel ini diharapkan dapat berkontribusi dalam memperoleh jawaban mengenai kondisi sosial-politik yang membuat gagasan Islam progresif atau Islam peradaban harus terjadi. Artikel ini memperlihatkan cara bagaimana multikulturalisme dan eklektisisme mengkarakterisasi proyek Muslim terhadap modernitas di tengah-tengah keterhubungan yang kuat dengan masa lalu dan kitab suci.DOI: 10.15408/sdi.v18i1.439
