Abstract

Indonesia is rich in the biodiversity of medicinal plants used traditionally for healing several ailments. Melastoma malabathricum L. is one of the traditional herbs used to treat many diseases. A TLC-densitometric method was developed for determining secondary metabolites such as phenolic compounds and their related compounds, M. malabathricum leaves from Riau, Indonesia. This study investigated the secondary metabolites of M. malabathricum extract by spraying reagent: FeCl3, p-anisaldehyde, and cerium (IV) sulfate and followed by antibacterial assay through broth macro dilution method. Densitometric qualitative analysis of phenolic compounds and their related compounds was employed to examine peaks of the M. malabathricum extract through winCATS software. After spraying with particular reagents, the three extracts showed various spots/bands with several colors and exhibited peaks in TLC densitogram profiles. Three extracts were tested for antibacterial activity against Staphylococcus aureus and Escherichia coli. The result showed that ethyl acetate extract revealed excellent antibacterial activity against S. aureus and E. coli with MIC values of 3.125±0.6 mg/mL and 6.25±0.5 mg/mL, respectively. Owing to the presence of a wide variety of secondary metabolites, the leaf extract of M. malabathricum is expected to exhibit and help develop as a therapeutic agent.

Abstract

Many compounds have been isolated from Andrographis paniculata (Burm. f.) Nees (AP). In drug discovery and development, plant secondary metabolites are popular as resources for drug candidates. A high-quality drug candidate should not only be effective against the therapeutic target, but it should also be safe and have good pharmacokinetic features. This study aimed to predict the pharmacokinetic features and toxicity potencies of 46 compounds from AP using the pKCSM online tool. According to pKCSM prediction, among the forty-six compounds from AP, compound 1 (14-Deoxy-11,12-didehydroandrographolide), compound 2 (14-Deoxyandrographolide), and compound 39 ((-)-beta-Sitosterol) have good pharmacokinetic features and do not have potencies to be mutagenic and hepatotoxic agents. The lethal dosage values (LD50) of compounds 1, 2, and 39, are 1935, 2053, and 2424 (mol/kg), respectively. However, further research is still needed to confirm these predictions.

Abstract

Orthosiphon aristatus is a well-known medicinal plant acknowledged for its therapeutic effect in treating urinary tract diseases, hypertension, diabetes mellitus, and arthritis. It is widely used as an ingredient in herbal medicine and distributed throughout the world, including China, Europe and Indonesia. Sinensetin are normally used as chemical marker to evaluate and control the quality of O. aristatus. However, in 2021, the European Medicines Agency changed the marker to rosmarinic acid. In order to determine the levels of rosmarinic acid and sinensetin in O. aristatus as well as the correlation between the two compounds, this study used high-performance liquid chromatography (HPLC) with a UV detector to analyze O. aristatus extract from three distinct locations with four different ethanol concentrations (96%, 75%, 50%, and 25%). Results showed that higher levels of rosmarinic acid was obtained in extractions using binary. Meanwhile, higher sinensetin content in O. aristatus were obtained by 96% ethanol (Klaten) and hydro-ethanol (Sleman, Bantul). In addition, this study discovered that the sinensetin levels had an inverse correlation with rosmarinic acid levels. It can be concluded that the rosmarinic acid and sinensetin contents are influenced by growing locations and ethanol concentrations.

Abstract

Temu mangga (Curcuma mangga) merupakan tanaman obat Indonesia yang memiliki banyak manfaat kesehatan. Namun, simplisia temu mangga sering tercemar mikroba selama proses penanganan dan penyimpanan yang akan berpengaruh terhadap mutu dan keamanannya. Salah satu metode yang sedang dikembangkan untuk dekontaminasi mikroba adalah penggunaan ozon dan telah terbukti efektif dalam mengurangi mikroba pada berbagai jenis bahan pangan. Tujuan penelitian ini adalah untuk melihat efektifitas ozon dalam proses dekontaminasi simplisia temu mangga. Sampel temu mangga diambil dari Pasar Beringharjo dan diozonasi dengan dosis 10 g/h. Proses dekontaminasi ozon dilakukan pada rentang waktu 0-120 menit Perhitungan cemaran mikroba dilakukan dengan menghitung Angka Lempeng Total (ALT) dan Angka Kapang Kamir (AKK) sesuai persyaratan BPOM. Hasil penelitian menunjukkan bahwa ozon efektif dalam mengurangi jumlah mikroba pada temu mangga. Tanpa perlakuan ozon, ALT pada sampel mencapai 1,5 x 105 koloni/g. Setelah perlakuan ozon, ALT menurun menjadi 0,1-1 x 105 koloni/g. Selain itu, pada simplisia temu mangga setelah dilakukan ozonasi selama 30 menit tidak terdapat kapang dan kamir.

Abstract

The Brassicaceae family is recognized for its antioxidant and anti-inflammatory properties. Due to its widespread distribution, it serves as a prominent source of bioactive phytochemicals, attracting considerable research interest. Brassicaceae plants contain several secondary metabolites, including glucosinolates, which are precursors to bioactive compounds like isothiocyanates (e.g., sulforaphane) that have demonstrated anti-inflammatory effects. This study aims to investigate keywords, countries, number of publications, institutions, authors, and journals related to Brassicaceae plants as anti-inflammatory agents during the period 2003–2024 using a comprehensive bibliometric analysis method. Information was collected using the Scopus database, followed by data analysis using the Biblioshiny R Package and VOSviewer. A total of 760 articles meeting the inclusion criteria were analyzed. The results indicate that 2023 saw the largest increase in research publications, with China emerging as the leading contributor, while the USA had the highest citation count. The International Journal of Molecular Sciences was identified as the most popular publishing journal. Kyung Hee University was designated as the most productive institution, with Li Y as the author with the highest contribution. Bibliometric data also highlighted several therapeutic target molecules, including cytokines, nitric oxide, cyclooxygenase-2, and Nrf2, which play crucial roles in the action mechanisms of signature metabolites from Brassicaceae plants, including sulforaphane, isothiocyanates, glucosinolates, and flavonoids, for therapeutic purposes as anti-inflammatory, antioxidant, antimicrobial, and chemopreventive agents. These findings highlight the significant potential of Brassicaceae medicinal plants for various therapeutic mechanisms and provide recommendations for future research in the pharmacy field.

Abstract

The Sesquiterpene lactones, enhydrin and uvedalin, are the chemotype and subtype compounds of the yacon leaf. They are known to have anti-diabetic activity, but their mechanisms are not clear. One of the mechanisms of anti-diabetic agents is to inhibit the activity of the dipeptidyl peptidase 4 enzyme (DPP-4). The research aimed to determine the inhibition of the extract, enhydrin, and uvedalin isolated from the yacon leaf against the DPP-4 enzyme activity. Yakon leaves were extracted using 70% ethanol by the maceration method. Enhydrin and uvedalin were obtained from previous research. The inhibitory activity of the DPP-4 enzyme was then determined by the fluorescence assay method using a multi-well plate reader. Sitagliptin was used as standard inhibitor of DPP-4 enzyme. The ethanol extract of yacon leaves inhibits the DPP-4 enzyme with an IC50 of 856.9 ppm. The percentage inhibition of the DPP-4 enzyme by the enhydrin and uvedalin at a concentration of 250 ppm was 2.37 and 35.16%, respectively. The inhibitory potency of the pure isolated compounds was not substantially greater than that of the crude extract itself. This led us to the conclusion that the overall contribution of enhydrin and uvedalin to the extract's DPP-4 inhibitory activity is actually modest, suggesting the presence of other active compounds within the extract. However, it may provide a new approach to the treatment of type 2 diabetes.

Abstract

Ipomoea pes-caprae (L.) R.Br traditionally used to treat pain, skin infections, inflammation, and rheumatic disorders. In vitro studies also reported antioxidant, anti-inflammatory, and antimicrobial potential activities obtained from secondary metabolites such as flavonoids, tannins, terpenoids, steroids, saponins, and alkaloids contained in Ipomoea pes-caprae. The purpose of this study is to optimize the mobile phase and identify secondary metabolite compounds of Ipomoea pes-caprae. The analysis used thin layer chromatography (TLC) in identifying compounds responsible for providing antioxidant activity with a spray reagent in the form of DPPH. The results showed the ideal of mobile phase, namely Chloroform: Ethyl Acetate: Methanol: Formic Acid (2: 7: 0.5: 0.5). Ethanol extract of 96% leaves has antioxidant activity with a DPPH spray reaction than ethyl acetate and N-hexane extracts. The metabolite profiles in samples with various specific reagents include the group of polyphenols, alkaloids and flavonoid compounds. Based on the results of the descriptive analysis, it was shown that the most dominant group of flavonoid compounds from ethanol extract was 96% of Ipomoea pes-caprae leaves, so it had a strong correlation with antioxidant activity.
