Abstract

The aim of this research was identifying the stimulation serum protein synthesis following the treatment of Java Gingseng (Talinum paniculatum Gaertn) root extract in varying concentration. This research used 12 male rats of 8-9 weeks, divided into 4 groups. First group as control, second group to five group were administratered with Java gingseng root extract equal with 3.5, 10.5, and 21 mg dry roots / 100g BW / day per oral for 14 days. Blood serum was collected for electrophoresized at the end of treatment. Electrophoresis was conducted with 7% SDS-PAGE method. Gel stained in 0.25% Coomasie Brilliant Blue and it followed densitometry on band in gel to comparing those quantitatively. The result show the effect on the level of serum protein whose MW 29. Highest stimulation of protein synthesis (2.2 times than control) have been reached at treatment with extract that equal with 3,5 mg dry root/ 100 g BW/ day. Increasing Java gingseng root extract wasn’t followed by increasing of stimulation protein synthesis level.

Abstract

Kelurahan Pacar Kembang terletak di wilayah Surabaya Timur. Permasalahan yang dihadapi oleh mitra adalah terdapat kampung yang kumuh dan belum tertata, juga pemuda karang taruna dan ibu-ibu PKK yang belum mempunyai kegiatan edukasi untuk dapat mewujudkan kampung yang bersih dan asri. Kegiatan ini bertujuan untuk mewujudkan kampung yang bersih, asri dan aman. Berdasarkan permasalahan tersebut di atas, maka permasalahan mitra yang harus segera ditangani adalah aspek lingkungan dan budidaya dengan memberikan pelatihan mural dan budidaya lidah buaya dilanjutkan dengan kegiatan praktek terhadap materi yang diberikan. Berdasarkan hasil evaluasi terhadap materi yang telah disampaikan menunjukkan bahwa rerata nilai pelatihan mural adalah 76 dan rerata nilai pelatihan budidaya lidah buaya adalah 82. Abstract

Biocontrol is one way that can be developed to overcome public health and agricultural problems. Biocontrol agents could be a microorganism, for example, from the Bacillus group. This study aims to determine the larvicidal toxicity of the Bacillus subtilis strain LS9.1 against Aedes aegypti larvae. The mode of action of the isolate caused larval death, which was detected with parasporal inclusions and secondary metabolites. Larvicidal toxicity with the bioassay method. Parasporal inclusion proteins were detected with the SDS-PAGE method. The Cry toxin coding gene with the PCR method and cell ultrastructure with transmission and scanning electron microscopy (TEM and SEM). The action of secondary metabolites was detected with hemolytic activity test. The isolate high toxicity to A. aegypti larvae. Parasporal inclusion proteins with molecular weights ranging from 72-7 kDa and the gene encoding a protein toxin in size 300 bp. The TEM and SEM results did not Cry-IV toxin. The hemolytic activity test showed a positive reaction correlated with biosurfactant production. Based on this study, the death of larvae is probably caused by secondary metabolites produced by Bacillus subtilis strain LS9.1. Thus, this isolate has the prospect of being an environmentally friendly biocontrol agent to disease vectors and insect pests.

Abstract

Most of the plant-associated bacteria can synthesize active biological components of phytohormones such as auxin. This study aimed to examine the potency of 61 endophytic bacteria isolates from the mangroves at Kutang Beach, Lamongan in producing IAA hormone and to identify types of isolates effecting the concentration of IAA, morphological characteristics of isolates, as well as endophytic bacterial species that have the most potential to produce IAA hormone. Screening of endophytic bacteria isolates was performed using the colorimetric method and the production of IAA was carried out using the spectrophotometric method. IAA production by endophytic bacteria was analyzed descriptively and statistically. One-Way ANOVA was employed to determine the effect of the isolate type on the concentration of IAA. The most potential isolates to produce IAA hormone are identified by 16s rRNA gene marker. The screening results showed that 12 isolates of endophytic bacteria have the potential to produce IAA hormones (2.0-9.3 ppm), coded with LMG 7, 15, 31, 32, 43, 53, 54, 55, 56, 57, 62, and 63. The results of the One-Way ANOVA test suggested that the type of isolates did not affect the concentration of IAA produced by endophytic bacteria. The twelve isolates had different morphological characters and those were Gram-positive bacilli with cell sizes ranging from 1.5 µm - 3 µm. The highest concentration of IAA was produced by LMG 15 (9.3 ppm). LMG 15 was identified as Bacillus cereus strain LMG 15, having 99.33% similarity to Bacillus cereus strain IAM 12605.

Abstract

Endophytic bacteria are microorganisms that live in plant tissues and some of them contribute to nitrogen fixation for plants. This study aimed to isolate and identify endophytic bacteria from mangroves of Kutang Beach, Lamongan, which potentially as nitrogen fixing bacteria. Bacterial Isolates were used as candidates for biofertilizers. Leaves samples were taken from 10 sampling points. Bacterial isolation was initiated by sterilizing the surface of the leaves sample and grinding it aseptically. Isolation was carried out with a pour plate method on Nutrient Agar medium. Screening for endophytic bacteria's potential as N-fixing agent was carried out by growing the bacterial isolates on a semi-solid Nitrogen Free Bromothymol Blue (NFB) medium. The isolates that produced a positive reaction with a change in the color of the medium to blue were then subjected to macroscopic (shape, color, elevation, and the edge of the colony) and microscopic observations (Gram stain and bacterial cell measurements). The isolates showed the fastest change in the color of the medium were identified by the molecular marker of the 16S rRNA gene. The data obtained were analyzed descriptively. As many as 20 isolates were obtained from the mangroves of Kutang Lamongan Beach, and ten isolates of twenty potentially as nitrogen-fixing bacteria. The ten nitrogen-fixing bacteria isolates had varying macroscopic characteristics. The microscopic characteristics showed that eight isolates had Gram-positive bacilli, and two isolates were Gram-negative with varying bacterial sizes. Based on the 16S rRNA gene sequence, the most potential of nitrogen-fixing bacteria was LMG II-14 isolate and identified as Paenibacillus alvei LMG II-14 with 99.36% similarity to Paenibacillus alvei strain DSM 29 based on the NCBI database. The ten nitrogen fixing isolates that have been obtained can later be used as candidates for biofertilizer composition, especially Paenibacillus alvei LMG II-14.

Abstract

Saprophytic mold is one type of mold that has the ability to survive in extreme conditions, such as in mangrove soil. Saprophytic fungi act as the main organism in the process of decomposition of organic matter and energy flow in the mangrove area. Several researchers successfully identified 102 species and 33 genera of saprophytic fungi. Aspergillus is known to be the dominant mold on mangrove soils because of its characteristics, such as high sporulation and spore spread and its ability to withstand extreme environmental conditions. The use of saprophytic molds in biofertilizer manufacturing due to saprophytic molds can produce cellulase, xylinase, and ligninase enzymes to decompose organic matter and produce toxin compounds for pathogens in plants. The presence of mold species in mangrove soil is influenced by several factors, including the temperature level, season, the salinity of mangrove waters, soil pH, and oxygen.

Abstract

Di Kelurahan Pacar Kembang, Kecamatan Tambaksari, terdapat banyak limbah sayuran yang merupakan limbah rumah tangga. Limbah ini sering kali berakhir di tempat pembuangan sampah serta menciptakan masalah lingkungan dan kesehatan. Sebagai upaya untuk mengatasinya adalah memberdayakan ibu-ibu kader untuk mengolah limbah sayuran menjadi nutrisi tanaman. Tujuan utama dari pengabdian ini adalah memberdayakan kader melalui pemanfaatan limbah sayuran. Dengan memberikan pelatihan tentang cara mengolah limbah menjadi pupuk organik, tidak hanya membantu mengatasi masalah limbah tetapi juga meningkatkan kemandirian ekonomi masyarakat. Metodenya adalah memberikan pendampingan meliputi aspek pengetahuan dan ketrampilan serta manajemen. Aspek pengetahuan dan ketrampilan meliputi praktek mengolah limbah sayuran menjadi nutrisi tanaman (nutritan) yaitu berupa pupuk organik cair dan pupuk kompos serta praktek budidaya tanaman hortikultura yaitu budidaya tanaman cabe, tomat dan terong. Aspek manajemen meliputi pelatihan manajemen keuangan dan digital marketing. Evaluasi terdiri atas pretest dan postest, serta evaluasi kegiatan. Hasil evaluasi menunjukkan bahwa rerata nilai pretest adalah 97,35; rerata nilai postest adalah 97,94. Tingkat kepuasan peserta terhadap kegiatan ini adalah pengabdian masyarakat sesuai kebutuhan mitra (sangat puas 100%). Pengabdian masyarakat dapat dimanfaatkan dan diaplikasikan oleh mitra (sangat puas 97%; puas 3%). Pelatihan pembuatan nutritan berhasil meningkatkan pengetahuan, keterampilan, dan kesadaran masyarakat mengenai pemanfaatan sampah organik.

Abstract

Kelurahan Ampel Surabaya, merupakan kawasan yang potensial untuk dikembangkan dari sektor pariwisata dan budaya. Tantangan yang dihadapi saat ini adalah pengelolaan kawasan yang kurang asri dan berwawasan lingkungan. Kurangnya pemberdayaan lahan sempit untuk bercocok tanam yang selaras dengan alam juga turut mempengaruhi kondisi lingkungan di kawasan ini.  Salah satu upaya untuk mengatasi masalah ini adalah dengan memberdayakan masyarakat sekitar untuk membantu mengelola kawasan yang asri dan berwawasan lingkungan.  Tujuan dari pengabdian masyarakat ini adalah memberdayakan ibu-ibu kader PKK di Kelurahan Ampel agar dapat bercocok tanam dengan memanfaatkan lahan sempit di sekitar rumah menggunakan pupuk hayati yang ramah lingkungan.  Metode yang digunakan adalah memberikan pelatihan meliputi aspek pengetahuan dan keterampilan. Aspek pengetahuan meliputi pemberian materi tentang pemanfaatan dan konsep pupuk yang ramah lingkungan. Aspek keterampilan, meliputi bagaimana cara membuat pupuk hayati serta cara mengaplikasikannya ke tanaman budidaya, dalam hal ini tomat, terong, cabe rawit, dan cabe merah. Evaluasi dilakukan melalui pemberian pretest, postest untuk melihat keterserapan materi, serta kuisioner untuk mengevaluasi kegiatan secara keseluruhan. Hasil evaluasi menunjukkan bahwa rerata nilai pretest adalah 59; rerata nilai postest adalah 79. Tingkat kepuasan peserta terhadap kegiatan ini adalah 100 % sangat puas (pengabdian masyarakat sesuai kebutuhan mitra), dan 95% sangat puas ; 5 % puas (pelatihan pembuatan pupuk hayati dapat meningkatkan pengetahuan, dan keterampilan, serta kesadaran masyarakat untuk turut menciptakan kawasan asri menggunakan pupuk yang ramah lingkungan (pupuk hayati).
