Abstract

Piper betle L var Nigra (black betel) is a member of Piperaceae family which has potential as medicinal plant due to its secondary metabolites. Callus culture is one of the alternative methods to elevate production of secondary metabolites. This study was aimed to determine the effect of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzil aminopurine (BAP) towards callus induction and growth, also to determine the most optimal variation of 2,4-D and BAP concentration for callus induction of black betel leaf explant. This study was a laboratory experimental study with complete randomized design. Black betel leaf explant was planted in Murashige and Skoog (MS) medium supplemented with 2,4-D and BAP growth regulators at concentration of 0.0 mg/L, 0.5 mg/L, 1.0 mg/L, 1.5 mg/L, 2.0 mg/L respectively. Parameter recorded for callus induction and growth including callus induction time (days), percentage of explant forming callus, fresh weight, dry weight, color and texture. After callus planted for 8 weeks, analysis was performed statistically. Result showed that 2,4-D and BAP supplementation to medium affected the growth of black bete l leaf explants. Additional concentration of 0.5 mg/L 2,4-D and 1.0 mg/L BAP growth regulators showed the fastest response in callus formation, at 7.25 days. Growth regulators of 2,4-D 0.5 mg/L and BAP 2.0 mg/L concentration produced the highest fresh and dry weight, at 0.6802 g and 0.0670 g respectively. The best treatment was used as a basis to produce secondary metabolites.

Abstract

Air pollution can be caused by the number of dangerous gases that arise due to the process of accumulating decaying waste which has acute and chronic effects for people who are exposed. Judging from the data collection method, this study is observational. The study design used in this study was a cross-sectional study design. Risk agent toxicity is expressed in reference doses. For non-carcinogenic inhalation exposures expressed by the Reference Concentration (RfC). Based on primary data derived from the results of measurements of ambient air quality at the loading terminal IIB TPA Garbage Surabaya, the results showed that H2S gas levels of 0.04 where the levels are still below the ambient air quality standards based on East Java Governor Regulation No. 10 of 2009 concerning Quality Standards Air Ambient. From the calculation above, the researcher took the safe value by choosing the cancer risk safe value from the calculation data most likely to be applied by scavengers in the TPA Sampoerna Benowo Surabaya, namely C safe at 3 mg / m3, safe for 3 hours/day and safe for 2 years. Preventive efforts that can be carried out are by controlling the source such as not doing landfill with the open dumping method using the sanitary system. Routinely monitor and control emissions, so that the air quality around the waste picker environment is maintained. Reducing the number of exposures by using personal protective equipment (PPE) in the form of respirators (masks). Perform administrative control by reducing the time and frequency of exposure to H2S dangerous gases. It is better for the Surabaya government and PT X to socialize the hazards and impacts of H2S exposure to workers.

Abstract

Lockdown applied in big cities around the world as an effort to prevent the spreading of COVID-19. Lockdown has shown a major impact on the air quality. The decline of pollution happened because of the reduction of anthropogenic activities, especially in transportation and industry. This research method examines various articles about the impact of the COVID-19 lockdown on air quality and aims to describe changes in pollutants in the atmosphere. This study used several search engines (Scopus, Science direct, springer, PubMed, and google scholar). The results of the lockdown were proven to reduce the concentration of pollutants PM2.5 (21.8% - 39%), PM10 (22.9% - 75%), NO2 (54.3% - 96%), SO2 (7.6% - 215.5%), CO (35% - 64.8%). The decreasing NO2 concentration causes the O3 concentration to increasing (15% - 525%) in the atmosphere. This is due to the limitation of titration process of O3 by NO especially in the urban areas. Limiting transportation activities and industrial activities can be one of the strategies for controlling COVID-19 and air pollution. The right source of pollution control will improve air quality and improve people's lives.
