Abstract

Enzymes are biocatalysts that function as catalysts in biological functions. Collagenase enzymes have activities that can support product production. Enzyme activity for protein hydrolysis is influenced by several factors such as protein concentration, pH, temperature, substrate, inhibitor and activator. This study was aimed to determine the effect of differences in pH on the crude extract activity of the collagenase enzyme from the internal organs of malong fish. The study used a non-factorial completely randomized design, the treatments used were different pH (6.5, 7.5, and 8.5). This research consisted of several stages, namely sample preparation, extraction of internal organs to produce crude extract of the collagenase enzyme and test the activity of crude extract of the collagenase enzyme based on different pH. The analysis parameters was consisted of the proportion of fish yield, and the enzyme crude extract activity test at different pH. The results showed that malong fish has a proportion of meat 7,2 parts, bone 3,8, internal organs 1, skin 2,1. The yields of crude extract of collagenase enzyme pH 6.5, 7.5, and 8.5 were 60.83%, 64.09%, and 83.15%, respectively. The crude extract activity of collagenase enzyme from the internal organs of malong fish at pH 6.5, 7.5 and 8.5 was resulted in 0.293 U/ml, 0.5877 U/ml, and 0.7767 U/mL.  The highest collagenase enzyme crude extract activity was produced at pH 8.5.

Abstract

Seaweed has potential as a source of bioactive compounds, food and medicine. One of the seaweeds that has the potential to be used for its bioactive compounds is the brown seaweed Sargassum plagyophyllum. Fractionation is a technique of separating and grouping the chemical content of extracts based on their polarity. This study aims to determine the bioactive components (secondary metabolites) of flavonoids, alkaloids, steroids/terpenoids, saponins and phenols qualitatively and quantitatively from the extract of S. plagyophyllum by fractionation method. The research method used was experimental. S. palgyiophyllum was extracted using the fractionation method. The treatment given by different polarity solvents consisted of methanol, was done fractionation with using n-hexane, ethyl acetate, and butanol. Extraction of samples the first used methanol, after that methanol extract was diffracted with n-hexane, ethyl acetate and butanol. Parameter analysis was consisted of phytochemical test (qualitative analysis) and quantitative analysis of total phenol, flavonoid, and saponin content. The results showed that qualitative testing of the secondary metabolites of S. plagyophyllum contained in the methanol extract were steroids/terpenoids, saponins and phenols, the positive hexane fraction contained steroids/terpenoids, saponins, the ethyl acetate fraction positively contained alkaloids, flavonoids, steroids/terpenoids, and saponins. and phenol while the butanol fraction contains steroids/terpenoids, saponins. While quantitative secondary metabolites were tested on the hexane fraction (0.18% saponins), then the ethyl acetate fraction (0.14% flavonoids, 0.11% saponins, 0.12% total phenol) and butanol fraction (0.05% saponins).

Abstract

According to DLHK Pekanbaru 2021 Report, hotel wastewater makes a significant contribution to environmental problems in Pekanbaru. Many hotels don’t have an IPAL permit and don't report the results of the wastewater quality test to DLHK. This will have an impact on the environment in the community. This study aims to examine the strategy of controlling environmental pollution by hotel wastewater in Pekanbaru. The research method is in the form of surveys, interviews and questionnaires to determine the quality of processing and control strategies for environmental pollution that can be caused. Used descriptive analysis and SWOT analysis. The results of this study: there are hotel BOD and COD values ​​that do not meet the quality standards of Minister of Environment Regulation No. 5 of 2014 due to the influence of visitors and the rainy season, but the values ​​of TSS, pH, ammonia, oils and fats, and total coliform have met the quality standards. There needs to be an evaluation related to hotel wastewater treatment, improvement of policies related to hotel wastewater treatment in accordance with applicable quality standards, sanctions for hotels that are negligent and do not comply with the rules, socialization to the public to care about the environment.

Abstract

Services for COVID-19 infections have an impact on increasing the amount of infectious waste so that it burdens health care facilities. The Petala Bumi Regional General Hospital, Riau Province, is one of the hospitals that provide services to patients with suspected or confirmed Covid-19. Before the 2019 pandemic, the amount of medical solid waste generated was an average of 25 kg/day or 8,304.4 kg during 2019. In 2020 it increased by 45.07% or 12,047.7 kg/year, for 2021 it increased by 51.56% or 18,257 .5 kg/year. The results of the research on the process component show that the overall Covid-19 medical waste generated by the Petala Bumi Hospital, Riau Province in 2021 has been managed by carrying out management stages starting from reducing and sorting waste at the waste source, storing and transporting waste to B3 waste TPS. , storage of waste at TPS as well as transportation and final disposal of waste carried out by a tripartite agreement between the hospital, transporter and the processing party. The results of the analysis on the output component. The implementation of infectious medical waste management during the Covid-19 pandemic at the Petala Bumi Hospital, Riau Province as a whole has been carried out according to the existing stages.

Abstract

Indonesian agricultural sector always using of pesticides especially in horticultural farmers in Bukit Raya Pekanbaru district. However, the use of pesticides has a negative impact on the environment and health. One of the impacts on health is neuropsychiatric disorders such as cognitive disorders, anxiety and depression. This study was an analytic observational study, using a cohort approach. The research sample used a consecutive sampling technique with inclusion and exclusion criteria. The sample size is 32 people. The data analysis used the SPSS. There were significant differences in CHE levels (p=0.006), Moca-Ina scores (p=0.000), HAM-A (p=0.000) and HAM-D (p=0.000) during 3 months of exposure to pesticides. There is no significant relationship between the first CHE level and the first value of Moca-Ina (p=0.781), HAM-A (p=0.881) and HAM-D (p=0.605) and there is no significant relationship between the second CHE level and the second MocaIna (p=0.562), HAM-A (p=0.422) and HAM-D (p=0.869). The results of the multivariate analysis of smoking history score (p=0.021), working period (p=0.009), and spraying duration (p=0.006) are related to the MocaIna score on the delayed recall point. These three variables are able to explain the results of the delayed recall of 39.7 %.

Abstract

ABSTRACT Diadema setosum or sea urchin is a marine biota that has high economic value. This study aims to determine the chemical composition of sea urchin shells. This research phase consisted of flouring sea urchin shells and the observations made were chemical composition tests. The results of this study indicate that the sea urchin shell has a chemical composition such as a moisture content of 1.47 ± 0.42%, an ash content of 82.31 ± 1.10%, a fat content of 1.15 ± 0.19% and a protein content of 6.84 ± 0.26%.

Abstract

Collagenase enzyme is a type of protease enzyme. The activity of an enzyme is affected by substrate, temperature, inhibitor, and pH.  One of the sources of enzymes is the internal organs of fish, namely the gut. This study aims to determine the activity of the collagenase enzyme at different temperatures. The experimental design used was randomized and full non-factorial. The treatments used were different incubation temperatures (50, 55, and 60oC) with three replications. Parameter analysis comprised fish proportion, enzyme activity, and dissolved protein. The results showed that the parts of the yellow pike conger fish were meat (64.25%), bones (19.98%), internal organs (including intestines) (4.50%), and skin (6.30%). Collagenase enzyme activity at temperatures 50, 55, and 60oC was 1.154, 1.702, and 0.667 U/mL, respectively. The dissolved proteins content was 12.89 mg/mL. The optimum temperature of the collagenase enzyme from the fish gut was 55oC with an enzyme activity of 1,702 U/mL. The dissolved protein content of the gut yellow pike conger extract was 12.89 mg/mL  Purification of the collagenase enzyme from of the gut yellow pike conger was needed to obtain a higher enzyme activity value.

Abstract

The purpose of the study was to analyze hydrolysate astaxanthin as an inhibitor of the enzyme alf glucosidase. The method used is an experimental method by applying a Complete Random Design (RAL) The treatment used is a concentration of different papain enzymes, consisting of 3 treatment levels namely t1 (3%), t2 (6%), and t3 (9%). Observations made are 1) proximal analysis; 2) analysis of hydrolysate astaxanthin and its antioxidant activity; 3) analysis of the inhibition test of the enzyme alpha glucosidase and its inhibitory mechanism.  The results showed that the proxmat of vanname shrimp skin flour is; water content (9.20% bb), ash (31.46% bk), fat (1.64% bk), protein (37.60% bk), and carbohydrates (29.29% bk) (by different) with a yield of 36.22%. The highest astaxanthin content at the 3% papain enzyme concentration of 3.57 μg/g with the best antioxidant activity has the smallest IC50 value of 88,793 ppm and astaxanthin shows inhibition activity of alpha glucoside enzyme by 49.38% - 58.87% and astaxanthin has a competitive enzyme inhibition mechanism resulting in a decrease in postrandial blood glucose levels

Abstract

Slipper lobster live on the bottom of sandy and muddy waters at a depth of 10-200 m and are found throughout Indonesian waters. The carapace is the most significant part of its body, of which approximately 50% is a source of waste, but it can be utilized as functional food in the form of chitin and chitosan. One of the determinations of chitin and chitosan quality depends on the demineralization process. The potential of this shrimp has not been fully utilized due to the lack of information on mineral content after the demineralization process. This study aims to obtain information on flour, chitin, and chitosan mineral content. This research used a descriptive method. The parameters observed were yield, water, ash, and mineral content (K, Mg, Ca, Na, Fe, and P). The results showed that slipper lobster weighed about 80-240 g with a length of 15-25 cm with the yield and mineral content of flour, chitin and chitosan of slipper lobster carapace as follows: yield 29.04%, 41.02%, 33.42%, water 2.36%, 3.96%, 3.25%, ash 52.89%, 8.27%, 3.16%, potassium 55.73 mg/L, 9.09 mg/L, 4.55 mg/L, magnesium 37.23 mg/L, 5.59 mg/L, 5.95 mg/L, calcium 766.87 mg/L, 137.62 mg/L, 126.91 mg/L, sodium 146.62 mg/L, 17.82 mg/L, 10.73 mg/L, iron 12.82 mg/L, 1.89 mg/L, 1.72 mg/L, phosphorus 0.17 mg/L, 0.06 mg/L, 0.05 mg/L.

Abstract

Berunok sea cucumber has a nutritional content that can be utilized as a functional food ingredient. This study determined the characteristics of berunok sea cucumber hydrolysate (Paracaudina australis) and the type and level of total amino acids. This research was conducted using experimental methods, with a Non-Factor Complete Randomized Design consisting of 3 levels of enzyme concentration treatment (1%, 2%, and 3%) with three replicates. In this study, the parameters tested were the proximate content of flour and hydrolysate of berunok and the type and content of total amino acids. The results showed that the chemical composition of berunok flour was water 11,40% (dw), ash 10,82% (dw), protein 68,21% (dw), fat 6,53% (dw), and carbohydrate by the difference of 14,43% (dw). The best treatment for preparing berunok protein hydrolysate using pepsin enzyme is 2% pepsin enzyme concentration. The proximate of berunok protein hydrolysate is water 6.87% (dw), ash 8.45% (dw), protein 80,59% (dw), and fat 2,86% (dw). The results of the identification of types and levels of total amino acids found 17 kinds of amino acids with a total of 16.524% in hydrolysate berunok.
