Abstract

Penelitian ini bertujuan untuk mengetahui imunogenisitas antigen whole cell Aeromonas hydrophila dari beberapa strain bakteri A. hydrophila yang diberikan pada lele dumbo (C. gariepinus). Penelitian ini menggunakan metode eksperimen. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 6 perlakuan, yaitu A : strain GPW-02, B : strain KLK -11, C : strain GLR-08, D : strain KLK-14, E : strain KLK-05, F : kontrol (PBS pH 7,0) dengan 3 kali ulangan. Ikan uji yang digunakan adalah lele dumbo berumur sekitar 2 bulan dengan ukuran panjang 10-13 cm dan berat antara 12,2-14,5 g. Parameter utama yang diamati adalah titer antibodi dan uji reaksi silang, sedangkan parameter pendukungnya adalah parameter kualitas air, meliputi suhu air, pH, dan oksigen terlarut (Dissolved Oxygen = DO) yang diamati setiap minggu. Titer antibodi diamati 3 kali, yaitu sebelum divaksinasi, seminggu setelah vaksinasi booster dan dua minggu setelah vaksinasi booster. Kemudian dilakukan uji reaksi silang pada antigen dengan titer antibodi yang tinggi, yaitu ≥ 25. Data titer antibodi dianalisis menggunakan Analisis Sidik Ragam (Analysis of Variance/ANOVA), dan apabila berbeda nyata dilanjutkan dengan uji DMRT (Duncan Multiple Range Test) pada taraf uji 5% (Steel & Torrie, 1993), sedangkan data kualitas air dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa antigen whole cell A. hydrophila berpengaruh nyata (P<0,05) terhadap imunogenisitas ikan lele dumbo. Strain yang memiliki imunogenisitas paling tinggi adalah strain A (antigen whole cell A. hydrophila strain GPW-02). Parameter kualitas air selama penelitian dijaga pada kisaran optimal, yaitu suhu air 25-28°C, pH air 7,1-7,9, dan DO 6,6-6,8 mg/L.

Abstract

Penelitian ini bertujuan untuk mengungkap dua masalah, yaitu (1) mengetahui efektivitas vaksin sitoplasma sel A. hydrophila untuk pengendalian penyakit MAS pada gurami, dan (2) mengetahui dosis optimal vaksin sitoplasma sel A. hydrophila untuk pengendalian penyakit MAS pada gurami. Penelitian dilaksanakan menggunakan Rancangan Acak Lengkap (RAL) dengan lima perlakuan tiga ulangan, yaitu : (1) K : kontrol yang divaksin dengan larutan PBS steril, (2) D1: vaksin protein sitoplasma sel  A. hydrophila dosis 2,5 µg/ekor, (3) D2 : vaksin protein sitoplasma sel  A. hydrophila dosis 5 µg/ekor, (4) D3 : vaksin protein sitoplasma sel  A. hydrophila dosis 7,5 µg/ekor, (5) D4 : vaksin protein sitoplasma sel  A. hydrophila dosis 10 µg/ekor  Gurami yang digunakan berukuran panjang 10-12 cm dengan berat 25,8-28,5 g. Vaksinasi dilakukan secara suntikan intramuskular. Satu minggu setelah vaksinasi dilakukan vaksinasi booster, dan uji tantang dilakukan dua minggu setelah vaksinasi booster. Parameter yang diamati meliputi titer antibodi, sintasan, tingkat perlindungan relatif (Relative Percent Survival/RPS), dan rerata waktu kematian (RWK). Data dianalisis dengan analisis varian dan Duncan’s Multiple Range Test (DMRT). Hasil penelitian menunjukkan bahwa vaksin protein sitoplasma sel  A. hydrophila efektif untuk pengendalian penyakit MAS pada gurami. Vaksin ini dapat meningkatkan produksi titer antibodi, sintasan, dan RPS (P<0,5) gurami. Dosis vaksin protein sitoplasma  sel  A. hydrophila optimum adalah 7,4 mg/ekor yang memberikan nilai RPS tertinggi sebesar 89,31 %.

Abstract

The research conducted n gouramy fish with size among 12-15 cm in length and average of 30 g in wight. the treatment used levamisol and their combination with vaccine Aeromonas hydrophila which applied by immersion during 6 hours. Methods of research used experimentally laboratories using completely randomized design with 6 replication. The fishes placed in aquaria, each of its 12 fishes. The data’s observed consisted of growth both of length and weight, number of neutrophyl, survival rate, and water quality. The aim of the research was to know the effect of levamisol and its combination with vaccine on the respons against their growth, neutrophyl responses and survival rate of gouramy fishes. The results showed, treatment of levamisol and their combination with vaccine could increase on the growth and the best result revealed on P1, as 1.2 cm and 0.68 g for their length and weight respectively. Cellular response of neutrophyl showed better reached at P3 and P4 as number of 5.3 neutrophyl cell between on day H18 and H24. Higher survival rate reached on P2 and P3. During researched, water quality was on qualified standard requirements for larvae rearing.

Abstract

The aims of this research were to determine effect vaccination with extra cellular product and intracellular product of A. hydrophilic by 1-2 times booster with 2nd week time interval on African catfish immune response. The research was constructed as CRD with five treatments and three replicates, i.e. (1) E1: vaccination with extra cellular product of A. hydrophilic with one time booster; (2) E2: vaccination with extra cellular product of A. hydrophila with two times booster; (3) I1: vaccination with intracellular product of A. hydrophila with one time booster; (I2) vaccination with intracellular product of A. hydrophilic with two times booster, and (5) K: control. African catfish of 13-15 cm length and 18-21.5 g of weight were used, with the number of 15 fish for each treatment. Intramuscular injection method was conducted for vaccination and booster of 0.1 ml vaccine into the body, with the dosage of 5 µg/fish. Booster time interval was two weeks. Data were taken for fifteen weeks. The parameters to observe were titer antibody and water quality. Furthermore, they were analyzed by Analysis of Variance and Duncan’s Multiple Range Test (DMRT). The result showed that vaccination with extra cellular product and intracellular product of A. hydrophila by 1-2 times booster with 2nd week time interval could increase the adaptive immune response through the increase titer of antibody (P<0.05) on African catfish. Vaccination with intracellular product of A. hydrophila by two times booster (I2) was the most effective method of vaccination.
