Abstract

Colorimetric sensors-based silver nanoparticles (AgNPs) are very interesting to be studied and developed because of the simplicity and ease in the principle of detection. It does not require sophisticated and affordable tools but still has high sensitivity. The coefficient extinction of AgNPs is relatively higher than AuNPs of the same size, making the sensitivity of AgNPs higher than AuNPs. The principle of detection is based on the aggregation of nanoparticles with analytes that causes shifting in Localized Surface Plasmon Resonance (LSPR) to a larger wavelength, commonly called a bathochromic shift or redshift. It is a favorite phenomenon because it is more easily observed with naked eyes. This sensor shows a good analytic performance with high sensitivity due to strong LSPR and good strategies that selectively bring interaction between analytes and AgNPs. AgNPs are characterized using UV-Visible (Ultra Violet-Visible), TEM (Transmission Electron Microscope), FTIR (Fourier Transform Infrared), and DLS (Dynamic Light Scattering), and many analytes have been detected with this sensor successfully. This article discusses several important parameters in increasing the sensitivity of AgNPs colorimetric sensors. Finally, it can be used as guidelines in the development of methods in the future.

Abstract

The rapid, simple, and selective colorimetric sensing method of cortisol has been successfully developed using AuNPs modified with 4-amino-3-hydrazino-5-mercapto-1,2,4-triazole (AuNPs-AHMT). The principle of this method is based on the color change from wine red to purple (redshift) when AuNPs-AHMT interacts with cortisol. The hydrogen bonding between the hydroxyl group from cortisol and the amine group from AHMT induces the aggregation of AuNPs. The modification of the AuNPs surface with AHMT aims to increase its stability. The properties of AuNPs and AuNPs-AHMT were characterized by UV-vis spectrophotometer. The interaction between AuNPs-AHMT and cortisol was studied by UV-vis and FTIR spectroscopies. The proposed method was optimized and validated. Au(III) was reduced to AuNPs at an optimum NaBH4 concentration of 1.0 mM. Validation of the proposed method showed good analytical performance with linearity from 1.0–50.0 nM, accuracy 91.07–102.77%, intra-day precision < 2.22% and inter-day precision < 2.17%, detection limit 0.76 nM, quantification limit 2.54 nM, and sensitivity 0.0112 nM/mL. The proposed method also showed good selectivity with the presence of some interferences in the sample. The proposed method was successfully applied for the determination of cortisol in the saliva by the standard addition method with acceptable recovery.

Abstract

Community empowerment and household waste processing are two issues that need to be resolved by the government. An independent and educated society is an important pillar to support the development of country. Meanwhile, waste that continues to accumulate without processing has a negative impact on health and the environment. Public education about household waste processing can be a solution to the two problems above. Education was carried out at Junior High School YBPK Surabaya and residents of Dukuh Kupang Surabaya, and other participants who joined in a hibrid meeting with a total of 116 participants. Activities at YBPK Surabaya took place on 25 March and 5 August 2023 and activities at Dukuh Kupang Surabaya took place on 27 May and 26 August 2023. The types of waste processed are organic waste and used cooking oil, which are then converted into eco-enzymes and used cooking oil Activities are divided into 2 stages. there are community education activities and program sustainability monitoring activities. The community education activities includes presentation and workshop were hold in March 2023 at YBPK Surabaya Middle School and May 2023 at Dukuh Kupang Surabaya. The evaluation of sustainability program including individual practice and monitoring of activities was carried out in August 2023 at Dukuh Kupang Surabaya. Methods for implementing activities include presentation, practice accompanied by a mentor, individual practice, and monitoring program sustainability. Based on the analysis of participants' ability, the participant scores before and after the activity was increase. It means that this training succeeded in increasing participants' knowledge and skills in processing waste, especially organic waste and used cooking oil waste. A total of 72 out of 116 participants (62%) had processed household waste either individually or in groups. Activity evaluation data shows that these activities are necessary and on target to create an independent and creative society.

Abstract

Rapid and accurate classification of viral pathogens is critical for effective public health interventions. This study introduces a novel approach using convolutional neural networks (CNN) to classify SARS-CoV-2 and non-SARS-CoV-2 viruses via hydrophobicity signal derived from DNA sequences. Conventional machine learning methods grapple with the variability of viral genetic material, requiring fixed-length sequences and extensive preprocessing. The proposed method transforms genetic sequences into image-based representations, enabling CNNs to handle complexity and variability without these constraints. The dataset includes 8,143 DNA sequences from seven coronaviruses, translated into amino acid sequences and evaluated for hydrophobicity. Experimental results demonstrate that the CNN model achieves superior performance, with an accuracy of over 99.84% in the classification task. The model also performs well with extended sequence lengths, showcasing robustness and adaptability. Compared to previous studies, this method offers higher accuracy and computational efficiency, providing a reliable solution for rapid virus detection with potential applications in bioinformatics and clinical settings.

Abstract

In this research, construction of expression vector for thermostable α-L-arabinofuranosidase in Saccharomyces cerevisiae BJ1824 was conducted. The Escherichia coli/S. cerevisiae shuttle vector, pYES2 was used as parental vector in construction. The abfA gene encoding α-L-arabinofuranosidase from Geobacillus thermoleovorans IT-08 was amplified by PCR, in which the plasmid pTP510 was used as a template. The amplification product was treated with SacI and XhoI and then subcloned to the pYES2 vector, which was previously digested with SacI and XhoI. The recombinant plasmid was designated as pY-Af and propagated first in E. coli Top10, and then transformed into S. cerevisiae BJ1824. For α-Larabinofuranosidase (AbfA) production, the yeast transformants were grown in YNBG selective medium and YPG rich medium, using galactose as an inducer. The AbfA activity was assayed by measuring the amount of p-nitrophenol (pNP) released from p-nitrophenyl-α-L-arabinofuranoside (pNPA) substrate at pH 6.0 and 70oC for 30 min. The recombinant AbfA activity was detected in either of culture medium (0.98%), cellassociated (14.17%) and intracellular (84.85%) when recombinant yeast was grown in YPG rich medium.
