Abstract

Distillation time can affect the extraction of essential oils such as the rhizome of temu hitam (Curcuma aeruginosa) which has been shown to have antibacterial activity. Therefore, this study aims to determine the optimum distillation time to produce temu hitam rhizome oil with the strongest antibacterial activity. The rhizomes of temu hitam were steam distilled with different length of time every 30 minutes for 4 hours to obtain essential oil and determined the compounds contained in them using gas chromatography-mass spectrometry. Antibacterial activity was determined by disc diffusion against Streptococcus mutans, Escherichia coli, and Staphylococcus aureus. The most dominant compounds present in essential oils are eucalyptol (34.89%), epicurzerenone (20.58%), and camphor (16.90%) based on the average peak of the total distillation time. The strongest antibacterial potency was obtained in the first 30 minutes of distillation for S. mutans and E. coli, and in the seventh 30 minutes of distillation for S. aureus.

Abstract

Some Asteraceae plants are reported to contain phytochemicals and to show high antibacterial activity. This study aims to determine the highest antibacterial activity of Wedelia trilobata, Acmella paniculata, Ageratum conyzoides, and Crassocephalum crepidiodes and to predict the chemical compounds that have a role as antibacterial. The samples were extracted with water and further fractionated using chloroform in the laboratory. Extracts determined the phytochemical compounds, antibacterial activity diffusion method, and compounds that have a role as antibacterial through literature review. Acmella paniculata demonstrated high antibacterial activities with the highest diameter of the zones of inhibition with a concentration of just 0.5 mg/disc. The compound that seems to have a role in antibacterial activity is spilanthol.

Abstract

Centella asiatica known as pegagan in Indonesia, belongs to the Apiaceae family and is widely used as salad, cosmetics ingredients, and herbal medicine for improving memory. This research aimed to determine the total phenolics content (TPC), antioxidant capacity (AC), and FTIR spectrum of C. asiatica leaves extracts using different extraction methods. C. asiatica extracts were prepared using maceration, reflux, and ultrasonication. The Folin-Ciocalteu method was used to determine TPC, and we found a higher level of TPC when using maceration, about 9,96 ± 0,20 mg GAE g-1 dry powder. Antioxidant capacity from the three extracts was measured using DPPH and FRAP methods. The highest antioxidant capacity using DPPH and FRAP methods was found in C. asiatica leaves extract using maceration with a value of 29,60 ± 0,71 and 14,04 ± 0,38 ????mol TE g-1 dry powder, respectively. The FTIR spectrum of each extract indicates the presence of vibration from several functional groups such as O-H, C-H, and C=C, likely from phenolics and C-O. The pattern of FTIR spectrum from the three C. asiatica leaves extracts gives different spectrum profiles and characteristics. Principal component analysis using FTIR spectrum data shows good clustering for each extract with 89% data variability (PC1 77% and PC2 12%). It can be concluded that TPC, AC, and FTIR spectrum profiles are different when using different extraction methods.

Abstract

AbstractIn plant tissue culture, the culture vessels are usuallycovered tightly with screw caps, aluminium foils, parafilm,or plastic wrap. This condition restricts the exchange ofgases in culture vessels and will affect negatively thegrowth of explants. The use of ventilated closure improvesthe air quality in culture vessels. Microboxes provided withdifferent types of filter (yellow filter with Kv value13.09 Gas Exchange (GE)/day, green filter with Kv value81.35 GE/day and without filter) on their closure wereexamined as culture vessels for growing rubber explants atmultiplication step. The purpose of this research was toobserve the plant condition in different types of microboxcorresponding to the morphology, stomata and chlorophyllcontent of the shoots. The results showed no significantdifference of shoot height on each microbox. The use ofventilated closure increased significantly new leafformation and decreased leaf fall. Normal size and color ofleaves were found on shoots grown in microbox with greenfilter. Chlorophyll analysis revealed no significantdifferences on three types of microbox, however visualobservation showed that leaves were greener on microboxwith green filter. The stomata condition of shoots onmicrobox with green filter were similar with those ofmother plants in green house, while different condition ofstomata were found on shoots grown in microbox withyellow filter or without filter. In normal environment suchas at the field and green house, most of stomata wereclosed, in microbox provided with filter on the closure,most of stomata were half open, while on microbox withoutfilter most of stomata were wide open.bstrakDalam kultur jaringan tanaman, tabung/botol kulturditutup rapat dengan penutup yang dilengkapi ulir,aluminium foil, parafilm atau plastik wrap. Kondisitersebut menghambat pertukaran udara dalam tabung kultursehingga sering memberikan pengaruh negatif terhadappertumbuhan eksplan. Penggunaan penutup berventilasidapat meningkatkan kualitas udara dalam lingkungantabung/botol kultur. Oleh karena itu microbox denganpenutup berfilter diuji sebagai wadah untuk menumbuhkaneksplan karet pada tahap multiplikasi, yaitu microboxberfilter kuning dengan nilai Kv sebesar 13,09 (GasExchange (GE)/hari dan berfilter hijau dengan nilai Kvsebesar 81,35 GE/hari, sedangkan sebagai kontrol adalahmicrobox tanpa filter (tertutup rapat). Penelitian bertujuanmengamati kondisi tunas di dalam microbox berfilter,meliputi morfologi, stomata dan kandungan klorofil. Hasilpenelitian menunjukkan bahwa tinggi tunas tidak berbedanyata pada masing-masing microbox. Jumlah daun barudan daun gugur berbeda nyata, dimana pembentukan daunbaru terbanyak terdapat pada tunas dalam microboxberfilter kuning maupun hijau. Ukuran dan warna daunterlihat normal pada tunas dalam microbox berfilter hijau.Analisis kandungan klorofil tidak menunjukkan perbedaannyata, namun pengamatan visual menunjukkan bahwa daunlebih hijau pada microbox dengan filter hijau. Kondisistomata daun dari tunas dalam microbox dengan penutupberfilter hijau menyerupai stomata tanaman induk yangterdapat di rumah kaca dan lapangan, sedangkan kondisistomata berbeda ditemukan pada tunas dalam microboxberfilter kuning atau tanpa filter. Pada lingkungan normalseperti lapangan dan rumah kaca, sebagian besar stomatamenutup, pada wadah dengan tutup berfilter stomata agakmembuka sedangkan pada microbox tanpa filter sebagianbesar stomata terbuka lebar.

Abstract

Mesenchymal stem cells (MSC) derived from white adipose tissue are considered to be an appropriate therapeutic candidate for various human diseases due to their natural ability to maintain homeostasis. They have also been considered for use as a therapeutic alternative due to their sufficient availability and ease of harvesting. MSC play an important role in the process of adipogenesis and energy metabolism that is directly related to the pathogenesis of various degenerative diseases and metabolic syndromes. The purpose of this study was to obtain MSC cultures from white adipose tissue biopsied from three adult males Macaca fascicularis and to evaluate these cultures qualitatively and quantitatively regarding their characteristics and ability to differentiate. Polymerase chain reaction (PCR) was used to confirm the expression of mesenchymal stem cell-specific gene markers, and a specific growth medium was used to differentiate these cells into adipocytes, chondrocytes, and osteocytes. The results obtained showed that the MSC culture from M. fascicularis WAT had a fibroblast-like morphology with a spindle shape. The MSC markers of CD73, CD90 and CD105, were also expressed. The MSC culture was also able to differentiate into adipocytes, chondrocytes, and osteocytes. Based on these results, white adipose tissue from Macaca fascicularis was found to have potential as a source of MSC.

Abstract

Cajuput is one of plants containing essential oil with cineole as a major component. This aim of study is to separate cineole in cajuput oil and to analyze its potency as slimming aromatherapy through in vivo assay. The essential oil was fractionated by column chromatography resulting 23 fraction (F1-F23). Cajuput oil, cineole, and F9 were analyzed by gas chromatograph-mass spectrometer, and the slimming aromatherapy potency was studied on white adult male Sprague-Dawley rats. Inhalation result of cineole showed that the average body weight of rats after 5 weeks treatment period was lower than that of the normal and the control groups which consumed high cholesterol feed. In conclusion, cineole is a compound that is potential in slimming aromatherapy.

Abstract

Bahan alam yang dapat menghambat spermatogenesis merupakan suatu alternatif untuk kontrasepsi pria. Penelitian ini bertujuan mengevaluasi formulasi optimum berbasis biji jarak pagar dan buah pare yang dapat menginhibisi spermatogenesis. Bahan sampel dimaserasi alkohol 70% untuk memperoleh ekstrak kasar. Ekstrak diujikan pada tikus Wistar jantan dewasa sebanyak 30 ekor dan dibagi menjadi enam kelompok secara acak (n=5 ekor). Kelompok I sebagai kontrol memperoleh pelarut akuades; kelompok II dan III secara berurutan memperoleh ekstrak biji jarak dan pare dengan dosis 50 mg/kgBB. Tiga kelompok lainnya, yaitu IV, V, dan VI memperoleh formulasi gabungan dengan rasio ekstrak biji jarak pagar dan pare 1:3, 3:1 serta 1:1, secara berurutan. Perlakuan diberikan per oral satu hari sekali selama 48 hari sesuai dengan siklus spermatogenesis. Pada akhir perlakuan, pemeriksaan sperma untuk konsentrasi dan kualitasnya serta bobot testis. Hasil uji rendemen ekstrak biji jarak pagar dan buah pare secara berurutan sebesar 6.11 % dan 3.32 %. Senyawa fitokimia yang terdapat pada ekstrak pare dan biji jarak pagar antara lain, alkaloid, fenol, flavonoid, tanin, saponin, triterpenoid, dan steroid. Efek ekstrak terhadap bobot testis dan konsentrasi sperma tidak ada perbedaan yang nyata antar kelompok (ANOVA P >0.05) sedangkan untuk kualitas motilitas sperma ada kecenderungan menurun pada formulasi ekstrak biji jarak dan buah pare (1:3).

Abstract

Some Asteraceae plants have phytochemicals that are commonly used to treat some illness. However, research on the use of Asteraceae plants as tyrosinase inhibitor agents is still rare. Therefore, this study aims to select the most active fraction of Asteraceae species as tyrosinase inhibitors and suspect active compounds that play a role in it. About 12 species from Asteraceae are extracted using water. The resulting filtrate is partially fractionated using chloroform to obtain water extracts, water fractions, and chloroform fractions. Each extract and fraction of each plant determined the activity of its tyrosinase inhibitor using a microplate reader (ELISA) with L-DOPA as the substrate. Thitonia diversipholia water fraction provided the best activity of all the samples tested with the smallest IC50. The active compounds contained are analyzed using LC-MS / MS.  Compounds that act as tyrosinase inhibitors are alkaloid group that dominate the plant, namely trigonelline.
