Abstract

Secang wood has strong antioxidant properties because it contains phenolic compounds such as flavonoids and brazilin. Also, red ginger has antioxidant derived from nonvolatile phenolic compounds. Until now, there has been no use of a combination of secang wood extract and red ginger specifically for the manufacture of effervescent tablets. The purpose of this study was to formulate an effervescent tablet by doing combination of secang wood and red ginger extracts with using various concentrations of citric and tartaric acids. The formulations conducted were three acid sources of 20%, 25%, 30%, using wet granulation. The evaluation subjected to the formulas included the physical properties of the granules, such as the water content and flow properties of the granules. Meanwhile, the evaluation performed to the tablet included the hardness test, tablet friability, weight uniformity, dissolution time, and acidity (pH). The analysis was carried out in accordance with the requirements of the Indonesian Pharmacopoeia Edition III and BPOM. The results obtained are effervescent tablet with an acid variation of 20% with physical quality values of water content of 2.91% and granule flow properties of 6.03 s. Abstract

A survey conducted at Thani Oyster Mushroom UKM in Magetan, followed by interviews with the owners, revealed that the main challenges faced by UKM were frequent failures and difficulties in finding trusted partners who were able to prepare oyster mushroom seeds well. This is mainly caused by less than sterile room conditions and inadequate facilities. Typically, mushroom breeding consists of the following stages: The process involves four steps: 1) cultivating an uncontaminated culture, 2) creating a starter culture, 3) producing a primary seed culture, and 4) producing final production seeds. Partners often encounter challenges and setbacks when trying to cultivate this mushroom. Creating culture media is a crucial step in the process of preparing pure culture. To date, partners have utilized PDA as a specialized medium for cultivating single strains of microorganisms, and this has been achieved using less sterile equipment and facilities. As a result, the media will be contaminated by pest bacteria, weeds, and other elements that are difficult to avoid, resulting in low success rates and frequent failures. The alternative proposed in this PKM activity is the need to build an aseptic isolation room or enclosure, which is specifically known as a Laminar Air Flow Cabinet (LAFC). The instrument is equipped with a continuously operating ultraviolet light, TL lamp, blower and air filter to ensure a sterile working environment by removing dust, dirt, spores and other particles that inhibit the germination of fungal seeds. Initial support provided to partners in utilizing LAFC resulted in a success rate of approximately 77% (10 out of 13 petri bottles) in mushroom mycelium cultivation.

Abstract

The corn plant has become a favorite of the farmer group “SIDO MAKMUR II” because it is resistant to pest attacks, suitable for planting in the dry season, and easy to maintain, providing significant benefits to farmers. However, the utilization of corn plant parts has not been maximized, particularly the use of corn cobs, which remains unexploited by the “SIDO MAKMUR II” group. According to Mr. Nurkolis, it is indeed unfortunate that corn cobs are either discarded or not utilized, given that this waste can be processed into high-quality animal feed. This is largely due to a lack of adequate tools and appropriate technology, as well as management issues, especially in bookkeeping for the business. To address these issues, the Community Partnership Empowerment Team from Universitas Negeri Surabaya is working to optimize corn cob waste as animal feed, aiming to resolve production problems in utilizing corn cob waste and issues in business management, particularly in savings and loans. The proposed solutions include (1) procuring and implementing a package of machines for processing corn cob waste into animal feed, (2) training on creating powdered corn cob feed through fermentation, and (3) training and assistance in implementing savings and loan management for the partners. The results of this community service activity, through the implementation of a technology package for processing corn cob waste into animal feed, show that the “SIDO MAKMUR II” group can now process corn cobs into animal feed using grinding and milling machines, as well as a ribbon screw mixer equipped with a timer and speed control. Additionally, there has been an improvement in management, particularly in the group’s finances, due to the establishment and training in business management.

Abstract

Cancer significantly increases prevalence and mortality rate making it a serious health concern currently, including breast cancer. The development of new drugs is a major effort to solve the cancer problems. Natural products are the primary source of medicinal compounds believed to have lower toxicity and side effects than synthetic drugs. This research aimed to reveal the chemical profile of the ethyl acetate extract of Basilicum polystachyon leaves. Through an in silico approach, this research studied the anticancer mechanism against the sirtuin1 (SIRT1) at the molecular level. Molecular docking simulations were performed to understand the interaction behavior of potential compounds as SIRT1 inhibitors. Based on these results, 117 individual compounds were successfully identified in the ethyl acetate extract. Molecular docking simulation revealed that ten compounds could inhibit SIRT1 better than the control inhibitor, indicating that these compounds have potential as anticancer agents. The prediction of these compounds' physicochemical properties and pharmacokinetics showed promising results and fulfilled the medicinal compound's criteria. These findings can be the basis for the application of phytochemical compounds as anticancer drugs, specifically potential compounds in B. polystachyon leaves.

Abstract

Rabies is a deadly viral disease that has no effective therapy after clinical symptoms appear. This study aims to evaluate the antirabies potential of the dominant compounds in the ethyl acetate extract of Desmanthus virgatus leaves, as well as several antiviral drugs, through an in silico approach. The extract was obtained through maceration and partitioning methods using ethyl acetate solvent, then analyzed using LC-MS, which identified 182 secondary metabolite compounds, and ten dominant compounds were selected for further analysis. These compounds, along with seven antiviral drugs, were docked against the rabies virus glycoprotein (PDB ID: 6LGX) using AutoDockTools 4.2.6 software. The docking results were analyzed based on the values of binding affinity, inhibition constant (Ki), and interaction with active amino acid residues.  Quercitrin and quercimeritrin were the dominant flavonoid glycosides in the ethyl acetate extract of D. virgatus leaves that showed binding affinity values of -8.45 kcal/mol and -8.10 kcal/mol, respectively. In addition, bictegravir and tegobuvir showed binding affinity values of -9.17 kcal/mol and -9.05 kcal/mol, respectively. Four compounds indicated potential as antirabies drugs. Pharmacokinetic feasibility tests using Lipinski parameters showed that most of the dominant compounds violated one or more parameters, especially the number of hydrogen bond donors/acceptors and molecular weight. However, such violations were also found in some antiviral drugs that have been used, such as remdesivir and darunavir. These results suggest that D. virgatus leaf extracts contain compounds with promising potential antirabies activity and deserve further investigation through in vitro and in vivo tests.
