Abstract

Staphylococcus epidermidis has become one of the most important pathogenic bacteria that lead to nosocomial infection. This is caused by its ability to form a biofilm on the surface. Essential oils are one of the natural product that could be used as antiinfection agents. Bengle rhizome essential oil has been known to have activity against non-pathogenic bacteria S. epidermidis. This study was proposed to know the antibacterial and antibiofilm activity of bengle rhizome essential oil against clinical isolate of S. epidermidis at concentrations of 45, 60, 75, and 90%. Antibacterial activity was determined by the diffusion method with hole plate technique, while antibiofilm activity was determined by the microtiter assay method with crystal violet staining and quantified by measuring the optical density (OD) at λ 595 nm. This study showed that bengle rhizome essential oil has antibacterial and antibiofilm activity. The largest antibacterial and antibiofilm activity was shown from 90% concentration of bengle rhizome essential oil. Keywords: nosocomial infections, antibacterial, antibiofilm, bengle essential oil, Staphylococcus epidermidis.

Abstract

Hydrolyzed protein isolates of melinjo seed using free alcalase has been known to have antioxidant potential. However, free alcalase can only be used once. Enzymes immobilization technique with entrapment method would overcome this problem. The immobilized enzyme characteristic was observed by SEM and hydrolysis degree value. The success of hydrolysis process was observed by SDS-PAGE. Antioxidant activity test was determined by the ability to inhibit hydroxyl radical and protection against DNA damage in 1% agarose gel. The result showed that protein isolates from melinjo seed had been successfully hydrolyzed by immobilized alcalase. Following hydrolysis showed that immobilized alcalase was effective to hydrolyze protein up to four times. Based on hydroxyl radical reduction activity test, antioxidant activity of hydrolyzed protein using immobilized alcalase had the same as that of glutathione as positive control antioxidant activity. In addition, hydrolyzed protein using immobilized alcalase had the ability toprotect DNA damage which showed by reversible change of supercoil band. Keywords: melinjo seed, hydrolyzed protein, immobilized alcalase, antioxidant

Abstract

Kenitu (Chrysophyllum cainito L.) grows in Jember region, East Java, with several variants which can be distinguished from the color and shape of the fruit. They are small round (BK), large round (BB), green oval (HL), and red round (MB). Kenitu leaves have been shown to provide antidiabetic activity in vivo, but the molecular mechanism still unknown yet. One of the oral antidiabetic is a class of α-glucosidase inhibitor. This research aimed to test the activity of Saccharomyces cerevisiae α-glucosidase inhibitor of several variants of kenitu leaves in Jember, as well as phytochemical screening. Simplicia powder was extracted using 70% ethanol with ultrasonication method to obtain a thick extract. α-glucosidase inhibitor activity test was carried out on the negative control solution (without extract) and the sample solution (extract) using chromogenic method. Phytochemical screening was done by TLC and tube test. IC50 value of ethanol extract kenitu leaves variant BK, BB, HL, and MB were 5.476 ± 0.039, 4.869 ± 0.018, 9.465 ± 0.012, and 11.836 ± 0.048 µg/ml, respectively. Based on the LSD test, IC50 values between variants of kenitu leaves were significantly different (p < 0,05). Phytochemical screening result showed that ethanol extract kenitu leaves contain saponin, triterpenoids, flavonoids, and phenolic compounds. Those compounds were suspected to contribute on α-glucosidase inhibitor activity, but more study still needed to isolate the active compounds and evaluate each isolate for its α-glucosidase inhibitor activity. Keywords: α-glucosidase inhibitor, kenitu leaves, ethanol extract

Abstract

Hypertension is ranked third on the cause of death in Indonesia for all ages (6.8 %). Protein isolate of melinjo seeds (Gnetum gnemon L.) hydrolyzed with free alcalase has been known to have antihypertensive activity. However, the use of free alcalase is uneconomical since it can only be used once, thus we studied the immobilized alcalase effectivity to hydrolise the melinjo seeds protein isolate to be used as angiotensin converting enzyme (ACE) inhibitor. The immobilization method used entrapment method with DMDMOS/TMOS as matrix. Characterization of the immobilized enzyme was observed by FTIR and effectiveness of repeated hydrolysis was observed by the value of the degree of hydrolysis. The success of the hydrolysis process was determined by SDS-PAGE electrophoresis. Antihypertensive activity test was determined by the ability to inhibit ACE. The results showed that the alcalase has been immobilized in the matrix of silane and SDS-PAGE protein profile showed melinjo seed proteins had been successfully hydrolyzed. Hydrolysis process repeatedly demonstrated that immobilized alcalase was effective to hydrolyze protein isolates of melinjo seeds twice. Based on ACE inhibitory test, there were no significant differences between the protein isolate hydrolyzed by immobilized alcalase (Gg-PH), protein isolates hydrolyzed with free alcalase (Gg-PHB), and captopril. These findings suggest that the antihypertensive activity in Gg-PH is the same with that of captopril. Keywords: hypertension, antihypertensive, melinjo, enzymes immobilization, ACE inhibitor
