Abstract

Background: Bacterial infections remain a major global health burden, with antibiotic resistance increasingly limiting treatment options. Natural plant-based compounds represent a promising alternative source of antimicrobial agents. Baccaurea racemosa Mull.Arg. (kepundung), a tropical fruit plant native to Southeast Asia, is traditionally used in folk medicine; however, its seed fractions have not been comprehensively evaluated for antibacterial activity against both Gram-negative and Gram-positive pathogens in a single systematic study. Objectives: This study aimed to characterize the secondary metabolite profile and evaluate the comparative antibacterial activity of n-hexane, ethyl acetate, and ethanol extracts of B. racemosa seeds against Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 25923. Methods: Seeds were subjected to successive maceration using solvents of increasing polarity (n-hexane, ethyl acetate, ethanol). Phytochemical screening was performed using standard colorimetric assays. Antibacterial activity was assessed by the Kirby–Bauer disk diffusion method at concentrations of 25, 50, 100, and 200 mg/mL, with chloramphenicol as positive control and 1% DMSO as negative control. Results: Phytochemical screening detected alkaloids, flavonoids, tannins, glycosides, saponins, and steroids in all extracts. Against E. coli, the ethanol extract exhibited the largest inhibition zone (44.5 ± 1.7 mm at 200 mg/mL), classified as very strong (>20 mm), compared to ethyl acetate (8.25 ± 3.4 mm) and n-hexane (6.36 ± 1.9 mm). Against S. aureus, the ethanol extract produced an inhibition zone of 13.68 ± 0.33 mm, followed by ethyl acetate (7.46 ± 4.7 mm) and n-hexane (5.08 ± 0.78 mm). The ethanol extract demonstrated significantly higher activity against E. coli than S. aureus (p < 0.05), while n-hexane and ethyl acetate fractions showed comparable moderate activity against both bacteria. Conclusion: Baccaurea racemosa seed ethanol extract demonstrates significant antibacterial activity, particularly against Gram-negative E. coli, attributed to its polar phenolic compounds including flavonoids and tannins. These findings support the potential of B. racemosa seeds as a source of natural antibacterial agents for further pharmaceutical development.

Abstract

Background: Infectious diseases caused by Staphylococcus aureus, Escherichia coli, and Candida albicans remain a global health problem. Since prolonged use of synthetic drugs can cause side effects and resistance, natural alternatives are needed. Menteng leaves (Baccaurea racemosa) contain alkaloids, flavonoids, saponins, steroids/terpenoids, and tannins, which have potential antimicrobial properties. Objective: This study aimed to determine the antibacterial and antifungal activities of n-hexane, ethyl acetate, and ethanol extracts of menteng leaves against S. aureus, E. coli, and C. albicans. Method: The research was conducted experimentally through sample collection, simplicia preparation, successive maceration extraction, phytochemical screening, and antimicrobial testing using the agar well diffusion method. Results: From 500 g of simplicia, 10.5 g of n-hexane extract, 11.4 g of ethyl acetate extract, and 18 g of ethanol extract were obtained. Phytochemical screening showed that all three extracts contained different secondary metabolites, with the ethanol extract having the most complete profile. The highest antimicrobial activity was observed in the ethanol extract, with inhibition zones of 18.92 mm against S. aureus, 18.35 mm against E. coli, and 19.58 mm against C. albicans at a concentration of 200 mg/mL. Conclusion: These results indicate that the ethanol extract of menteng leaves has strong potential as a natural antibacterial and antifungal agent.

Abstract

Background Diarrhea is the leading cause of high morbidity and mortality in children in developing countries such as Indonesia. Diarrhea can be caused by bacterial infections. Guava leaves and kencur have astringent properties that can help reduce the frequency of bowel movements and stop diarrhea. Aim: The study aims to determine the most effective dose concentration of a combination of guava leaf ethanol extract (PGEE) and kencur rhizome ethanol extract (KGEE) as an antidiarrheal medicine. Methods: The study used an experimental method with stages of simplisia preparation, extract preparation by maceration, simplisia characteristics, phytochemical screening, and antidiarrheal efficacy testing using 25 male mice divided into 5 groups, namely F1 = negative control (NaCMC); F2 = positive control (Loperamide); F3, F4, and F5 were combinations (PGEE:KGEE) with 3 different dose concentrations, namely (25%:75%); (50%:50%), and (75%:25%), where the base doses used were (PGEE=400mg/kg/BW) and (KGEE=27.5mg/kgBW). Results: The parameters observed included the onset time of diarrhea, duration, frequency of bowel movements, and stool consistency. Statistical data analysis was performed using ANOVA and Tukey's post hoc test with the help of SPSS software. Statistical analysis with Tukey's post hoc test showed that there was a significant difference (p < 0.05) in fecal weight between the treatment group and the negative control group. Conclusion: The dose concentration (50%:50%) showed the most significant effect in reducing fecal weight comparable to loperamide, so that the combination of PGEE and KGEE at that dose concentration proved to be the most effective in reducing fecal weight and overcoming diarrhea, both visually (graph) and based on statistical analysis results.

Abstract

Kopi mengandung polyphenol sebagai senyawa antioksidan. Kandungan yang terdapat pada temu giring adalah senyawa alkaloida, flavonoida, triterpenoida, saponin dan tanin sebagai antioksidan. Jahe mengandung gingerol, beta-caroten, asam mengandung gingerol, beta-caroten, asamkafeat, curc kafeat, curcumin sebagai antioksidan. Tujuan dari p umin sebagai antioksidan. Tujuan dari penelitian enelitian ini adalah untuk mengetahui apakah minuman kopi yang dikombinasikan dengan ekstrak jahe ( Zingiber officinale) dan ekstrak temu giring (Curcuma heyneana) memiliki kapasitas antioksidan dan disukai oleh sukarelawan. Penelitian ini secara eksperimental dalam menyiapkan minuman kopi yang menggabungk menggabungkan ekstrak jahe ekstrak jahe dan ekstrak temu ekstrak temu giring dan giring dan mendemonstrasi mendemonstrasikan penemuan simplisia, penemuan simplisia, produksi produksi ekstrak ekstrak jahe dan temu giring, giring, serta optimasi optimasi formula formula menggunaka menggunakan SLD ( simplex simplex lattice lattice design) dan melakukan survey survey responden responden pada setiap formula. Rendemen simplisia jahe 500 g dan temu giring 750 g giring 750 g dengan berat ekstrak jahe sebanya dengan berat ekstrak jahe sebanyak 88,8 g k 88,8 g dan berat ekstrak temu giring sebanyak 98, dan berat ekstrak temu giring sebanyak 98,2 g. Penentuan optimasi formula menggunakan SLD didasarkan pada aktivitas antioksidan formula yang optimal, yaitu rasio jahe : temu giring (1/4 : optimal, yaitu rasio jahe : temu giring (1/4 : 3/4) formula II dan hasil formula II dan hasil hedonist masing-masing formula adalah formula IV dengan rasio jahe : temu giring (3/4 : 1/4)

Abstract

Diabetes melitus merupakan penyakit kronis yang memerlukan literasi kesehatan yang memadai untuk mendukung penggunaan obat yang tepat, penerapan gaya hidup sehat, dan pencegahan komplikasi. Kegiatan pengabdian kepada masyarakat ini bertujuan meningkatkan literasi kesehatan peserta mengenai diabetes melitus melalui edukasi penggunaan obat yang rasional dan gaya hidup sehat di Puskesmas Langkat. Kegiatan menggunakan desain evaluasi one-group pre-test–post-test dengan melibatkan 30 peserta yang terdiri atas pasien diabetes melitus, keluarga pasien, dan masyarakat berisiko diabetes. Intervensi dilakukan melalui penyuluhan interaktif, pembagian leaflet, diskusi, dan simulasi membaca etiket obat. Pengetahuan dievaluasi menggunakan 10 pertanyaan sebelum dan sesudah intervensi. Hasil menunjukkan bahwa rata-rata skor pengetahuan meningkat dari 56,3 pada pre-test menjadi 84,7 pada post-test, dengan peningkatan sebesar 28,4 poin. Peserta dengan kategori pengetahuan baik meningkat dari 20,0% menjadi 80,0%, sedangkan kategori kurang menurun dari 50,0% menjadi 3,3%. Peningkatan terbesar ditemukan pada pengetahuan mengenai tanda hipoglikemia, kepatuhan penggunaan obat, larangan menghentikan obat tanpa konsultasi, serta komplikasi dan perawatan kaki. Uji Wilcoxon menunjukkan perbedaan yang signifikan antara skor pre-test dan post-test (p < 0,001). Hasil kegiatan menunjukkan bahwa edukasi interaktif yang dikombinasikan dengan informasi praktis mengenai penggunaan obat dapat meningkatkan pengetahuan peserta mengenai pengelolaan diabetes dan berpotensi mendukung kegiatan edukasi kesehatan di pelayanan kesehatan primer.
