Abstract

Purpose: This work aimed to analyze the profiles of amino acids in the ficin enzyme isolated from Ficus aurata (Miq.), and to determine the efficiency of these amino acid concentrations under two types of drying – solar drying and freeze drying. Methodology: The enzyme ficin was obtained from fig tree, and the presence of amino acids was determined by UPL chromatography. The applied methods of drying were solar and freeze. However, the primary objectives were aimed at investigating the influence of these treatments on the level of amino acids present in all the samples. Results: The findings indicated that freeze drying techniques retained more amino acids in figs than was observed in the case of solar drying. In particular, the amount of L-glutamic acid in freeze drying was 2.39 mg/g, while that of solar drying was 1.43 mg/g. Similar results were observed in L-lysine and L-aspartic acid, which were relatively higher in freeze drying than solar drying, with concentrations of 2.43 mg/g and 2.55 mg/g, respectively, as opposed to 0.99 mg/g and 1.76 mg/g in solar drying. Besides, freeze drying showed better L-serine, L-phenylalanine, and L-histidine preservation. Findings: It was observed that freeze drying gave the best opportunity to reduce the loss of amino acid of the ficin enzyme. The drying method, in particular, resulted in a concentration of some important amino acids like L-glutamic acid, L-lysine, L-aspartic acid, L-serine and L-phenylalanine, and L-histidine as compared to solar drying. Novelty: This study focused on different types of solar energy and freeze energy economic systems in enzymatic activities in the ficin enzyme extracted from Ficus aurata (Miq.). The novel contribution of this research lies in its objective, which is to show how the drying methods used affected the retention of the most influential and critical amino acids in the enzymatic processes. Originality: The novel aspect of the study was the attachment of ultra vortices to different drying techniques, such as solar drying and freeze drying in select ficin enzymes from Ficus aurata. It opens up a new perspective toward the development of drying parameters for the enhancement of important enzymatic characters from this particular variety. Conclusion: This research considers the changes in amino acid content during the drying of ficin enzyme from Ficus aurata through different drying methods. It is shown that freeze drying enhances the retention of critical amino acids and is thus one of the most efficient methods for the enzymatic processing of this plant species. Abstract

Ficin is an enzyme derived from Ficus sp., recognized for its applications in the food and pharmaceutical sectors. Utilizing solar energy for enzyme drying has been shown to be economical and environmentally sustainable while preserving enzyme activity. This study aimed to determine the amino acid composition of crude ficin from Ficus padana Burm. f., and was extracted and dried with solar energy. The analysis of amino acid components was conducted using the UPLC technique. The results indicated the presence of numerous important amino acids, with L-aspartic acid being the highest concentrated at 1.85 mg/g and L-leucine at 1.41 mg/g. Significant quantities of L-glutamic acid (1.33 mg/g), L-serine (1.14 mg/g), and L-lysine (1.05 mg/g) were also identified. The solar drying process showed no significant loss of amino acids. The findings suggest that solar drying does not negatively impact the amino acid composition in fiction, therefore enhancing its practical significance, particularly for the food and pharmaceutical sectors.

Abstract

This study aims to quantify the protein content in crude ficin enzymes obtained from the fruit and latex of various Ficus species and Papaya. This research has been encouraged by the considerable potential of these enzymes in different biotechnological applications, while data with regard to the protein content of these enzymes is limited. This study used a quantitative approach using qubit fluorometer to quantify the protein content in crude enzymes derived from the fruit and latex of these plants. Based on results, the protein content in crude ficin enzymes, two types of research obtained from Ficus aurata (Miq.) fruit was 0.92µg/ml and from papaya fruit as 3.16 µg/ml. For latex, it was observed that the protein content in crude ficin enzymes was determined as 6.14 µg/ml for Ficus aurata (Miq.), 2.58 µg/ml for Ficus racemosa L., and 5,27 µg/ml for Ficus padana Burm.f.. The protein content from papaya latex was 15,98 µg/ml. These findings show the differences in the protein content of the enzymes obtained from different species of Ficus and Papaya as a basis for further study on using these enzymes for biotechnological purposes.

Abstract

Kultur jaringan digunakan sebagai metode untuk memproduksi bibit gambir dengan kandungan katekin tinggi secara cepat dan dalam skala besar. Penelitian ini bertujuan untuk menganalisis pengaruh Thidiazuron (TDZ) dan Naphthalene Acetic Acid (NAA) terhadap perkembangan dan kandungan fitokimia pada kalus organogenik gambir melalui teknik in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan pola faktorial dua faktor, yaitu konsentrasi TDZ (0; 0,04; dan 0,08 mg/L) dan NAA (0,5; 1; 1,5; dan 2 mg/L). Hasil penelitian menunjukkan bahwa TDZ secara signifikan memengaruhi kandungan klorofil pada kalus, dengan konsentrasi 0 mg/L dan 0,04 mg/L lebih efektif dibandingkan 0,08 mg/L. Kalus organogenik yang diberi perlakuan TDZ dan NAA menunjukkan aktivitas metabolit sekunder, termasuk terpenoid, flavonoid, alkaloid, tanin, dan fenol, yang teridentifikasi melalui uji histokimia.

Abstract

Untuk mengetahui karakter tanaman pisang dapat dilakukan dengan cara mengamati karakteristik sifat morfologi tanaman. Pisang Kepok Tanjung dari kultur jaringan memiliki karakter daun yang berbeda yaitu daun sempit dan normal. Pengetahuan tentang studi karakter morfologi dan fisiologi bibit pisang Kepok Tanjung daun sempit dan daun normal dari kultur jaringan belum banyak diketahui. Penelitian yang bertujuan untuk mengetahui karakter morfologi dan fisiologi bibit pisang Kepok Tanjung daun sempit dan daun normal dari kultur jaringan dilakukan di Solok, Sumatera Barat bulan Maret hingga September 2023. Penelitian dilakukan secara deskriptif dengan mengkarakterisasi morfologi dan membandingkan parameter pertumbuhan serta karakter fisiologi dari total 180 bibit tanaman pisang Kepok Tanjung berdaun sempit dan normal dari kultur jaringan. Hasil penelitian menunjukkan adanya perbedaan pertumbuhan dan karakter morfologi bibit pisang Kepok Tanjung daun normal dan daun sempit dari kultur jaringan. Bibit pisang Kepok Tanjung daun normal menghasilkan tinggi tanaman, jumlah daun, diameter batang dan lebar daun yang lebih tinggi dan berbeda nyata dengan bibit pisang Kepok Tanjung berdaun sempit akan tetapi tidak berbeda nyata untuk parameter panjang daun. Dari 33 karakter morfologi (batang, daun dan akar) yang diamati terdapat 28 karakter morfologi yang sama dan 5 karakter morfologi yang berbeda. Untuk parameter karakter fisiologi yaitu jumlah stomata dan kerapatan stomata tidak menunjukkan adanya perbedaan yang nyata.

Abstract

The titan arum (Amorphophallus titanum (Becc.)), an endangered and endemic flora from the island of Sumatra, faces a significant risk of extinction. Conservation efforts for the titan arum were undertaken, including in vitro conservation through tissue culture. The study aimed to determine the optimal concentration of the growth regulator Benzyl Amino Purine (BAP) for inducing shoot formation in titan arum and to observe cellular changes during the shoot stage through histological test. This research was conducted from November 2023 to March 2024 at the Tissue Culture Laboratory, Andalas University. The experiment was arranged in a Completely Randomized Design (CRD) with five treatment levels: 2, 3, 4, 5, and 6 ppm.  Data analysis was performed using an F-test at a 5% significance level. If the F-test showed significant differences, further testing was carried out using Duncan's Multiple Range Test (DMRT) at a 5% significance level. Meanwhile, data on explant rooting percentage and root count were presented as means and standard deviations. Results indicated that various concentrations of BAP could induce shoots formation of Amorphophallus titanum Becc., with the percentage of explants forming shoots reaching 80–95%.  The concentration of 5.0 ppm BAP was the most effective, yielding the highest average number of shoots at 7.80. Histological test revealed cell enlargement at the shoot tips. This research is pivotal for the conservation of titan arum and promotes further studies in in vitro culture techniques.

Abstract

The Sungkai Permai Lambung Bukik Farmer Group, Padang City, generally cultivates in monoculture, with limited productive land. The land is planted with rice, secondary crops such as corn, and some peanuts. In the past two years, corn farmers often experience crop failures, especially during the rainy season, where the sun shines less due to global climate change. Therefore, diversifying commodities on limited land so that land efficiency increases are significant to prepare the next source of seeds. To support increased land use efficiency, optimal soil conditions are needed. Utilization of Arbuscular Mycorrhizal fungi can improve soil fertility and is expected to become an alternative technology in plant cultivation. Science and technology activities based on the Study Program and the Assisted Nagari (IbPSNB) were carried out in groups of Permai Sungkai Farmers in the form of 1) Counseling on the pattern of corn-peanut intercropping cultivation, and how to make CMA spore cultures, 2). The demonstration plot for corn-peanut intercropping cultivation. Through this activity, farmers finally understood the benefits and patterns of corn-peanut intercropping. Farmers understand the role and method of making CMA spore cultures. Several farmers cultivated corn-peanut intercropping. Two farmers at the demonstration plot managed to harvest corn and peanuts at the same time. Other farmers have failed to harvest corn due to pest attacks but have succeeded in harvesting peanuts so that the losses experienced by farmers are reduced, and they still have stock of seeds for the next planting season.

Abstract

Patchouli (Pogostemon cablin Benth.) is among the top 20 essential oil-producing plants traded in the global market. This plant shows narrow genetic diversity due to non-flowering and seed production, increasing the challenges in obtaining new varieties through crossbreeding. Therefore, this research aimed to apply mutation method using ultraviolet-C irradiation combined with tissue culture to broaden the genetic diversity of patchouli. The potential of UV-C irradiation and the morphological changes occurring in the plant were explored from October to December 2023 at the Tissue Culture Laboratory, Andalas University, Padang. In addition, the experiment was arranged in a Randomized Complete Block Design (RCBD) with 10 treatment levels and 3 groups. The treatments included UV-C irradiation doses of 0 (wild type), 30, 60, 90, 120, 150, 180, 210, 240, and 270 minutes at a distance of 30 cm. Data for each patchouli plant per observation variable were presented as mean values, variance, and standard deviation and analyzed using an unpaired t-test. The results showed that exposure to ultraviolet-C radiation impacted several morphological characteristics, such as chlorosis, delayed bud development, increased bud and leaf count, as well as faster bud growth. This phenomenon shows the potential of ultraviolet-C radiation as a physical mutagenic agent.

Abstract

Kegiatan pengabdian kepada masyarakat ini bertujuan untuk meningkatkan kapasitas petani lokal dan mahasiswa dalam memproduksi dan menerapkan agroinput ramah lingkungan, yaitu pupuk organik padat dan cair, pupuk hayati, serta pestisida nabati. Melalui program Matching Fund DIKTI 2023, dilakukan penyusunan buku panduan, pelatihan, serta praktik langsung yang melibatkan mahasiswa MBKM dan petani mitra. Evaluasi dilakukan melalui observasi lapangan dan analisis laboratorium. Hasil kegiatan menunjukkan peningkatan pemahaman, keterampilan, serta munculnya inisiatif produksi mandiri oleh peserta. Kegiatan ini membuktikan efektivitas kolaborasi pendidikan–masyarakat dalam mendukung sistem pertanian berkelanjutan.

Abstract

Shallot (Allium ascolanicum L.) is one of the horticultural crops that is widely consumed by humans. This study aims to obtain interaction the type of mulch and doses of chicken manure on the growth and yield of shallots. This Research was conducted at Panai Pasir Talang , Muara Labuh, South Solok Regency with a height of ± 430 meters above sea level, in December 2018 to February 2019. The research method is an experiments used factorial complete randomized design with two factors. Factors I is the type of mulch (black plastic mulch, silver plastic mulch, rice straw mulch) and factors II is chicken manure (10 tons / ha, 20 tons / ha and 30 tons / ha).. The results showed that there was no interaction between the use of several types of mulch with chicken manure doses on the growth and yield of shallots. The use of black plastic mulch yielded the best results on fresh tuber weight per plot and per hectare of onion namely 1,643 kg / plot and 11.74 tons / ha. The dose of chicken manure of 20 tons / ha gave the best influence on plant height, number of leaves, fresh tuber weight per clump, per plot and per hectare as well as wind dry weight per clump, per plot and per hectare of onion.
