Abstract

Scutellaria radix is one of the ancientest approaches to treat diseases. The aim of this study was to investigate the effect of Scutellaria radix on the immune responses of Macrobrachium rosenbergii by reverse-gavage feeding and their protection against Lactococcus garvieae. In brief, M. rosenbergii was injected with S. radix water extract at 1, 5 and 10 µg/µl via reverse-gavage feeding (RGF). Phenoloxidase (PO) activity of hemocytes and prophenoloxidase (proPO) gene of hepatopancreas were assessed to determine M. rosenbergii immunity. The results revealed that the concentration of S. radix extract at 5 µg/µl enhanced significantly the phenoloxidase of M. rosenbergii correlated with the control group. For pro PO were up-regulated by all concentration in a time-dependent manner. The highest percentage of survival rates was observed at 5 µg/µlof S. radix water extract after challenged with L. garvieae. The present discoveries suggest that S. radix water extract at the concentration of 5 µg/µlmay raise the immune responses and gene expressions in M. rosenbergii using RGF.

Abstract

Dunaliella salina is a type of marine microalgae. The objective of this research were investigated the effect of D. salina to inhibit the growth of E. tarda. The method used in this study was examined antibacterial activity of D. salina using disc diffusion and co culture test. The secondary metabolite compound in D. salina was tested using phytochemical screening and FTIR methods. The results obtained D. salina extracted using n-hexane showed the best activity for inhibiting the growth of E. tarda. Minimum concentration of 100 mg.L-1 crude extract can inhibit E. tarda with a total of bacterial colonies 137x1011 CFU.mL-1. The phytochemical test results showed that D. salina extract using n-hexane contained flavonoids, saponins, alkaloids, terpenoids and phenols. The FTIR analysis showed phenol and carotene compounds are one of the secondary metabolites that can be used as antibacterial. It can be concluded that the extract has strong antibacterial activity against E. tarda and potentially as antibacterial in aquaculture.

Abstract

Phytoplankton is a source of natural feed for shrimp cultivation in the pond. Phytoplankton productivity increases by the increasing nutrient content in the pond. Feed and metabolic waste is the sources of nutrient for phytoplankton growth. This study aimed to evaluate productivity and identify phytoplankton at intensive whiteleg shrimp, Litopenaeus vannamei cultivation system. This study was conducted at three intensive whiteleg shrimp located in Probolinggo, East Java. Samples were collected on the early and the late of culture period (day 17 and 87 after stocking). Four environmental parameters including total of ammonia nitrogen (TAN), nitrate, orthophosphate, total of suspended solid (TSS), and chlorophyll-a were measured. Identification and density of phytoplankton were also performed of each pond. The result showed that productivity of three ponds was 22,893.83 kg/ha to 23,600.61 kg/ha with an average size 12.74 g to 14.35 g. During culture period, the concentration of TAN, TSS and chlorophyll-a tended to increase. Meanwhile, the average of both nitrate and orthophosphate tended to decline. Several phytoplankton identified in this study were in the genus of Oocystis, Chlorella, Nannochloropsis, Chaetoceros, Stephanodiscus, Nitzschia, Coscinodiscus, Cyclotella and Ulothrix. Phytoplankton of the group Chlorophyta is predominance for pond 1 and 2, while pond 3 was dominated by phytoplankton in the group of Diatom/Baccillariophyta. This study indicated that the present of phytoplankton in the pond provide natural feed and good environmental condition for shrimp cultivation.

Abstract

Omah Iwak Badher Bank ialah salah satu destinasi wisata di Kabupaten Blitar yang dikelola oleh POKMASWAS Fajar Bengawan. Penelusuran reviewer pada google.co.id mendapatkan total 77 orang pemberi ulasan dengan rata-rata rating 4.5. Penelitian ini bertujuan untuk menilai rating destinasi terhadap responden terpilih, ialah mahasiswa S1 dan S3 Fakultas Perikanan dan Ilmu kelautan UB Malang. Total responden berjumlah 46 mahasiswa, 33 ialah mahasiswa S1 dan 13 mahasiswa S3. Kedua kelompok responden mendapat kesempatan yang sama untuk mengunjungi destinasi wisata. Rating dan penilaian terhadap destinasi dilakukan dengan memberikan 12 jenis pertanyaan kepada masing-masing resonden. Hasil penelitian mendapatkan bahwa rating responden mahasiswa S1 (3,7±0,57) dan S3 (3,7±0,48) lebih rendah dibandingkan dengan penilaian publik (4,5). Hal ini menunjukkan kualitas destinasi yang menjadi perhatian mahasiswa lebih tinggi dibandingkan dengan umum. Kesediaan membayar mahasiswa S1 (Rp. 150.000 Trip-1) pada destinasi wisata relatif lebih rendah dibandingkan dengan mahasiswa S3 (Rp. 250.000 Trip-1). Sebagai destinasi wisata baru, Omah Iwak Badher Bank perlu melakukan peningkatan jasa untuk meningkatkan kualitas destinasi.Kata Kunci: kesediaan membayar, jasa wisata, wisata alam, POKMASWAS Willingness-To-Pay To Omah Iwak Badher Bank Tourism DestinationABSTRACT Omah Iwak Badher Bank is one of the tourist destinations in Blitar Regency which is managed by POKMASWAS Fajar Bengawan. A reviewer search on google.co.id found a total of 77 reviewers with an average rating of 4.5. This study aimed to assess the destination rating of selected respondents, are undergraduate and doctoral students of the UB’s Faculty of Fisheries and Marine Science Malang. The total respondents were 46 students, 33 were S1 students and 13 were S3 students. Both groups of respondents received the same opportunity to visit tourist destinations. Rating and assessment of destinations was done by giving 12 types of questions to each respondent. The results found that the average rating of undergraduate student respondents was 3.7±0.57, and 3.7±0.48 for S3 students, respectively. These were lower than that of public assessment (4.5). This indivated that the quality of destination that are of concern to students is higher than the public. Willingness-t-pay of undergraduate students (Rp. 150,000 Trip-1) at tourist destinations was relatifly lower compared to doctoral students (Rp. 250,000 Trip-1). As a new tourist destination, Omah Iwak Badher Bank needs to improve its services to improve the quality of the destination.Keywords: willingness-to-pay, tourism services, nature tourism, POKMASWAS

Abstract

This community service was carried out to support the fish farmers in solving their problems. The problems identified during the initial survey were: high feed cost, low survival rate of cultured fish and shrimp, disease outbreaks and lack of production and financial management. Workshop, training and applying a simple technology was introduced to the selected farmers. Some activities conducted were: (1) providing two hand refractometers and a pelleting machine, (2) workshop of local-ingredient based fish feed production, (3) workshop of probiotics production, (4) training of production and financial management, (5) demo-pond. Farmers have been able to produce local-ingredient based fish feed and probiotics as well as to apply a simple production and financial management. The demo pond results showed that introducing of hand refractometer for adaptation period could increase the survival rate of fish and shrimp  up to 25%. Furthermore, by applying local-ingredient based fish feed and probiotics during the culture periods, the fish famers could increase the income from fish production up to 50.1%.

Abstract

Evaluation of natural products as a safe and effective antimicrobial agent is a scientific strategy to treat the drug resistant pathogens. Fennel (Foeniculum vulgare Mill) is an herbal plant that has an active ingredient which is one of its benefits as an antibacterial material. In this study, water extract of fennel fruit determined the antibacterial activity against Vibrio harveyi and Vibrio alginolyticus using the Minimum Inhibitory Concentration Test (MIC) and paper disk diffusion method. The results obtained on the MIC, the smallest concentration inhibited the growth was 0,060 g/ml, for both bacteria. The variations on Paper Disk Diffusion Method were the concentrations of A (0.065 g/ml), B (0.070 g/ml), C (0.075 g/mL), D (0.080 g/ml), E (0.085 g/ml), F (0.090 g/ml) and Control (0.000 g/ml). The concentration of 0.090 g/ml had the highest inhibition zone diameter in 11.17 ± 0.5 mm (V. harveyi) and 12.53 ± 1.14 mm (V. alginolyticus). It can be concluded that the fruits of fennel (F. vulgare Mill) has a very important ecological role as an alternative treatment in controlling the spread of vibriosis caused by V. harveyi and V. alginolyticus.ABSTRAKEvaluasi produk alami sebagai agen antimikrobia yang aman dan efektif adalah salah satu strategi ilmiah untuk memerangi ancaman patogen resistan terhadap obat. Fennel, (Foeniculum vulgare Mill), umumnya dikenal sebagai adas, merupakan tanaman herbal yang memiliki bahan aktif yang salah satu manfaatnya adalah sebagai bahan antibakteri. Pada penelitian ini menggunakan ekstrak air dari buah adas untuk mengetahui aktivitas antibakteri terhadap Vibrio harveyi dan Vibrio alginolyticus dengan menggunakan metode uji Minimum Inhibitory Concentration (MIC) dan difusi cakram kertas. Hasil yang diperoleh pada uji MIC, konsentrasi terkecil untuk menghambat pertumbuhan adalah 0,060 g/ml, untuk kedua spesies bakteri. Variasi perlakuan pada uji cakram kertas yaitu konsentrasi A (0,065 g/ml), B (0,070 g/ml), C (0,075 g/ml), D (0,080 g/ml), E (0,085 g/ml), F (0,090 g/ml) dan kontrol (0,000 g/ml), hasil yang diperoleh adalah konsentrasi 0,090 g/ml memiliki diameter zona hambat tertinggi sebesar 11,17 ± 0,5 mm (V. harveyi) dan 12,53 ± 1,14 mm (V. alginolyticus), sehingga dapat disimpulkan bahwa buah adas (F. vulgare Mill) memiliki peranan ekologi yang sangat penting sebagai bahan pengobatan alternatif dalam pengendalian penyebaran penyakit Vibriosis yang disebabkan oleh V. harveyi dan V. alginolyticus.

Abstract

Acauses of the decrease in vaname shrimp production was vibriosis. Vibriosis is caused by vibrio bacteria. A vibrio bacteria that plays a role in vibriosis is vibrio harveyi. Managing of diseases caused by vibrio harveyi is one of the best that provides immunostimulant assistance from natural ingredients derived from seaweed. Ulva lactuta is a green seaweed that has different substances that contain immune. Some of the compositions contained in Ulva Lactutawere Flavonoid Compositions, and the Composition of Polysaccharides that are Combined with Immunimulants The purpose of this study was to determine the effect and dosage used to improve the immune system in vaname shrimp. Combined with THC and Fagocytosis in the hemolim on vaname shrimp before infection and after infection with Vibrio harveyi bacteria.A dose of 2 ppm showed a significant difference (p <0.05) for all treatments.

Abstract

The research was conducted to know the effect and to determine the best utilization of the chiken feather silage meal as a substitute for fish meal protein source in the feed formula of Colossoma macropomum. This study used experimental method based on Completely Randomized Design (CRD) with 5 treatments and 3 replications. Five diets (33% isoprotein and 3.85 kcal/g diet isoenergi) were formulated with substitution of the chiken feather silage meal to fishmeal protein. These substituted 0, 25, 50, 75 and 100 % (A, B, C, D and E respectively) of the fishmeal protein. Parameters observed and analyzed were: survival rate, specific growth rate, feed conversion ratio, protein efficiency ratio, protein retention and protein digestibility. The results showed that the treatment had no effect on survival rate, specific growth rate, feed conversion ratio, protein efficiency ratio, and protein retention but the effect on protein digestibility. Based on these results, it can be concluded that fishmeal protein can be substituted with the chiken feather silage meal up to 100% in the feed formula of Colossoma macropomum.

Abstract

This research aimed to evaluate the effect of polysaccharide extract of Ulva lactuta on the total haemocytes count (THC) and the phagocytic activities (PA) on Litopenaeus vannamei. The concentration of the extract used in this research was 5 ppm and it was applied through the immersion. The haemolymph was collected after the administration in the different periods at hour of 4 and 24. The results showed that the administration of polysaccharide extracts increased THC and PA in L. vannamei significantly (p<0.05). THC and PA of white-leg shrimp with polysaccharide extract was higher than that without the administration of the extract. The enhancement of THC was found after the administration at hour 4 from 5.00 cells. ml-1 to 5.26 x 104 cells. ml-1. However, the THC level decreased after 24 hours at the value of 5.19 x 104 cells. The increasing pattern was also true for PA of L. vannamei, from 40.44% to 53.67% (hour 4). At the hour of 24, the PA was found decreased to the level and then decreased 45.67 ±1.53. On the contrary, there were no enhancement of THC and PA in the treatment without polysaccharide extract of Ulva lactuta.

Abstract

Viral Nervous Necrosis (VNN) is a pathogen that is often found infecting cantang grouper and causing mass mortality of up to 100% in a short time. VNN is caused by a Nodavirus infection that infects the nervous and visual systems of fish. One of the prevention efforts against VNN infection in cantang grouper is to add Dunaliella salina extract which contains -carotene, nephtalene, tetradecane and phenol which acts as antiviral, anti-microbial and anti-inflammatory. The purpose of this study was to examine the potency of D. salina extract against clinical symptoms and anatomical pathology of cantang grouper after VNN infection. D. salina extract with doses of 250, 300, 350 and 400mg/kg of feed was added to commercial pelleted feed of the Stella B2 brand with a protein content of 42%. Feed was given to cantang grouper measuring 7-9 cm for 10 days of maintenance. Furthermore, the fish were challenged using VNN as much as 0.2 ml/head through the injection method. Observations of clinical symptoms and anatomical pathology were observed for 96 hours after infection. Symptoms of clinical pathology of fish infected with VNN appear to be circling, swimming horizontally, inflation of the swim bladder, discoloration of the pectoral fins to reddish, swollen lymph organs, yellowing of the liver and darkening of the body color. The compounds found in the extract of D. salina can be used to increase the immune response and reduce the number of deaths of cantang grouper infected with VNN. The addition of D. salina extract which was higher than other doses could increase the immune system so that its survival (SR) was higher as the dose increased.
