Abstract

Local plant-based resources are widely used as ingredients in artificial feed formulations; the problem with these resources is that they contain anti-nutritional substances, namely phytate acid. An approach that can be used to overcome phytate acid is the addition of phytase to the artificial feed. The success of common carp (Cyprinus carpio) fish fingerling cultivation is supported by feed management in the form of feeding frequency that is both good and effective. The purpose of this study was to examine and find the frequency of feeding on feed utilisation and growth of common carp fingerling fed with phytase addition. This research design used an experimental method, complete randomised design (CRD), 4 treatments with 3 replications, namely treatment A (frequency of feeding once a day), treatment B (frequency of feeding twice a day), treatment C (frequency of feeding three times a day) and treatment D (frequency of feeding four times a day). The test fish used were common carp fingerling with an average weight of 1.30 ± 0.12 g/head and a stocking density of 25 fish/head size (50x50x60) cm3 which were cultured for 56 days. The variables taken include Total Feed Consumption (TFC), Efficiency of Feed Utilization (EFU), Feed Conversion Ratio (FCR), Relative Growth Rate (RGR), Survival Rate (SR), and water quality. The results showed that different feeding frequencies had a significant effect on (P<0.05) TKP, FCR, EPP, and RGR, but no significant effect on SR. The conclusion of this study was that the highest values for EPP and RGR variables were found in the frequency of feeding twice and four times a day. Keywords:  feed, phytase, growth, efficiency

Abstract

Transportasi calon induk ikan nila (Oreochromis niloticus) hidup merupakan salah satu kegiatan penting dalam menunjang kegiatan pembenihan ikan nila. Penelitian ini bertujuan untuk mengkaji pengaruh ekstrak daun jambu biji (P. Guajava Var. Pyrifera) sebagai anestesi dalam transportasi pada calon induk ikan nila dengan kepadatan yang berbeda. Metode penelitian yang digunakan adalah metode eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 3 kali ulangan. Perlakuan terdiri dari calon induk ikan nila dengan bobot rata-rata 150±56 g diberi anestesi ekstrak daun jambu biji dengan dosis 0,2 g/mL dan di packing dengan sistem transportasi basah tertutup dengan kepadatan berbeda: A (15 ekor/10 L), B (20 ekor/10 L), C (25 ekor/10 L) dan E (30/10 L ekor). Parameter yang diamati meliputi masa induksi, masa sedatif, kelulushidupan (SR) dan kualitas air: suhu, pH, dan Dissolved Oxygen (DO).  Berdasarkan hasil yang diperoleh, penggunaan ekstrak daun jambu biji dosis 0,2 g/mL sebagai bahan anestesi dengan kepadatan berbeda memberikan pengaruh yang berbeda nyata (P<0,05) terhadap masa induksi, masa sedatif dan kelulushidupan calon induk ikan nila. Waktu induksi tercepat terdapat pada perlakuan D (ekstrak daun jambu biji dengan kepadatan 30 ekor/10 L) dengan masa induksi selama 7,33±0,21 menit. Hasil masa sedatif perlakuan D (ekstrak daun jambu biji dengan 30 ekor/10 L) dengan waktu 2,17±0,20 menit, dan kelulushidupan terbaik terdapat pada perlakuan B (dosis ekstrak daun jambu biji dengan kepadatan 20 ekor /10L) dengan nilai 91.33±1.53%. Kata kunci: anestesi, induk, kelulushidupan, pengangkutan, tilapia

Abstract

This study aims to analyze and prove the effect of Organizational Citizenship Behavior, Learning organization and Transactional Leadership on Competence, Training Education and Employee Performance at the Surabaya City Culture, Youth and Sports and Tourism Office.  In this study, the method used was a simple random sample.  Determination of the number of samples using the Slovin formula with a sample size of 60 people. The type of research conducted in this study is using explanatory research, namely research that explains the causes and effects of existing problems with the help of SEM PLS. The results of this study indicate that Organizational Citizenship Behavior has a positive but insignificant effect on employee performance at the Surabaya City Culture, Youth and Sports and Tourism Office. In contrast, Learning Organization behavior and Transactional Leadership both have a positive and significant effect on employee performance. Organizational Citizenship Behavior also has a positive and significant effect on employee competence, as well as Learning Organization and Transactional Leadership, both of which also have a positive and significant effect on employee competence. Meanwhile, Learning Organization has a positive and significant effect on education and training, but Transactional Leadership has no significant effect on education and training in the agency.

Abstract

Bakteri Aeromonas hydrophila adalah patogen penyebab penyakit Motile Aeromonas Septicemia (MAS) pada ikan mas dan umum dijumpai di Indonesia. Pengendalian bakteri ini dilakukan dengan menggunakan bahan kimia dan antibiotik yang dikhawatirkan dapat membahayakan lingkungan budidaya dan konsumen yang mengonsumsi ikan tersebut. Oleh karena itu, diperlukan bahan alami yang ramah lingkungan, dapat menghambat pertumbuhan bakteri, serta efektif menyembuhkan penyakit MAS. Lidah buaya (Aloe vera) adalah salah satu bahan alami yang dapat digunakan sebagai obat ikan karena memiliki aktivitas antibakteri yang mampu mengobati ikan mas dari infeksi A. hydrophila. Penelitian ini bertujuan untuk mengetahui efek ekstrak lidah buaya sebagai antibakteri, serta pengaruhnya terhadap status kesehatan ikan mas yang terinfeksi A. hydrophila. Efek antimikroba ditentukan secara in vitro dengan metoda cakram dan dilanjutkan dengan bioassay (empat perlakuan dan tiga kali ulangan) dengan merendam ikan mas yang sudah diinfeksi A. hydrophila dalam ekstrak lidah buaya dengan dosis sebagai berikut, perendaman ikan 0 ppm (A), 250 ppm (B), 500 ppm (C), dan 750 ppm (D). Data yang dikumpulkan meliputi ukuran zona hambat, gejala klinis, jumlah A. hydrophila, profil darah, bobot mutlak, SGR, kelulushidupan, dan kualitas air. Ekstrak lidah buaya menghasilkan diameter zona hambat berkisar antara 7,33-11,02 mm. Perendaman dengan ekstrak lidah buaya berpengaruh nyata terhadap jumlah bakteri, profil darah, pertumbuhan, namun tidak berpengaruh nyata terhadap kelulushidupan ikan mas. Perlakuan D (dosis ekstrak lidah buaya 750 ppm) memberikan hasil terbaik dari semua parameter yang diukur. Aeromonas hydrophila is common carp pathogen that cause Motile Aeromonas Septicemia (MAS) disease and commonly found in Indonesia. Control of this bacteria is carried out using chemicals and it is worried that they can harm aquaculture environment and consumers who consume the fish. Therefore, it is required natural ingredients that are environmentally friendly, can inhibit bacterial growth, and effectively cure MAS. Aloe vera is one of the natural ingredients that can be used as fish medicine because it has antibacterial activity which is expected to be able to treat carp from A. hydrophila infection. This study aims to determine the effect of aloe vera extract as an antibacterial, and its effect on the health status of common carp that attacked by A. hydrophila. The antimicrobial effect was determined in vitro using disc method and continued by bioassay (four treatments and three replications) by immersing carp that infected by A. hydrophila in aloe vera extract with immersion doses of 0 ppm (A), 250 ppm (B), 500 ppm (C), and 750 ppm (D). The data collected included the size of inhibition zone, clinical symptoms, number of bacteria, blood profile, absolute weight value, SGR, SR, and water quality. Aloe vera extract produces inhibition zone diameters from 7,33-11,02 mm. Immersion in aloe vera extract had significant effect on the number of bacteria, blood profile, growth, but had no significant effect on the survival rate of common carp. Treatment D (dose of aloe vera extract 750 ppm) gave the best results from all parameters measured.

Abstract

The use of strip story media is one of a series of stories that are cut into each word and the word is pasted onto cardboard or read by female students, this media can be used to practice writing Arabic or arrange words into sentence arrangements and so on, such as imla' khat and insya, especially in learning inysa (kitabah). The problem being studied is to find out how the use of strip story media is carried out by Intensive Semester III B students in writing maharah kitabah and whether there is an improvement in the Arabic writing ability of intensive students in semester III B. This research uses descriptive qualitative research. The informants selected in this study are the names involved in using this media. Like the Ustadzah and their students, the location of the research was carried out at the Al-Amien Prenduan Islamic boarding school, especially in Idia (intensive). Data collection techniques include interviews, observation and doccutation, while data analysis techniques include data reduction, data presentation. The result of the study is that the use of strip story media for maharah kitabah greatly adds to the improvement of writing practice into neat and good Arabic. So it is said to be good and neat if it has gone through the steps of the provisions that have been listed.

Abstract

One of the triglycerides test lipid profile examination that is often analyzed because it provides information about cardiovascular health and stroke. Laboratory services have an important meaning in making a diagnosis based on the patient's health history and are an integral part in achieving quality laboratory results that have accurate accuracy and precision. Analytical factors that can influence examination results include incubation time. The delay in the length of incubation time is due to the limited number of laboratory personnel and inadequate equipment. The objective of this study was to assess the impact of stability on serum triglyceride levels using the GPO-PAP method with different incubation durations. The research employed a quasi-experimental design, utilizing serum samples from three Medical Laboratory Technology students. The samples were subjected to varying incubation times of 10, 60, 75, 90, and 105 minutes at room temperature, with each condition being repeated five times. The mean results of serum triglyceride levels with incubation times of 10, 60, 75, 90, and 105 minutes respectively were 80,0 mg/dL, 79,00 mg/dL, 76.36 mg/dL, 73.65 mg/ dL, and 67.94 mg/dL. Data analysis was conducted using the General Linear Model (GLM) test, with significance indicated by the Sig value. < 0.05 means that triglyceride levels with an incubation time of 75 minutes are statistically unstable.

Abstract

Kitin yang merupakan salah satu  komponen penyusun eksoskeleton arthropoda. Kitin merupakan polimer rantai panjang N-asetilglukosamin, turunan Amida dari glukosa.  Ikatan peptide pada kitin dapat dipecah menggunakan  larutan KOH yang berfungsi sebagai deproteinasi. Proses deproteinasi akan menyebabkan lapisan kitin eksoskleton menipis.  Enzim dari buah nanas (Ananas comosus L. Merr) var. Subang yaitu Bromelin terbukti  memiliki kemampuan  untuk memecah ikatan peptida. Bromelin  merupakan salah satu enzim  yang banyak digunakan baik untuk industri pangan maupun non pangan. Bromelin  akan digunakan pada proses deproteinase eksoskeleton   pinjal yang mengandung  kitin. Tujuan Penelitian adalah  untuk menentukan kemampuan   bromelin  sebagai deproteinasi kutikula pinjal. Metode  penelitian  quasi eksperimen, pinjal kucing sebanyak 96 ekor terbagi menjadi  tiga kelompok  perlakukan kontak bromelin dan   serangga dalam konsentrasi 15%, 20% dan 25% dibandingkan dengan satu kelompok kontrol  kontak  larutan KOH 10 % dan serangga, dengan waktu perendaman selama 24 jam dan 48 jam, serta dilakukan tiga kali pengulangan. Hasil penelitian yang sudah dilakukan dari tahapan penipisan dengan bromelin 15%, 20% dan 25% selama 24 jam dan 48 jam. Proses penipisan akan berakhir dengan pemberian entelan sebagai perekat preparat. Penelitian ini  berlangsung selama 7 bulan dari bulan April  sampai dengan Oktober 2020.Lokasi penelitian adalah Laboratorium Terpadu dan Laboratorium Parasitologi Jurusan Analis Kesehatan  Poltekkes Bandung Hasil pemeriksaan secara mikroskopis dari preparat  kemudian di analisis dengan uji Kruskal-Wallis dan Mann Whitney diperoleh hasil<0.05 menunjukkan terdapat perbedaan yang signifikan untuk semua perlakuan Hasil analisis data menghasilkan kualitas tertinggi deproteinasi pada perlakuan  Bromelin 25% dan untuk waktu optimal yang menghasilkan kualitas deproteinasi tertinggi  pada waktu 48 jam.

Abstract

ABSTRACT This study aimed to analyze the genetic diversity of bacteria associated in bioflocs using 16S-rRNA polymerase chain reaction (PCR) with ARDRA technique. A total of 38 dominant bacterial isolates was obtained from bioflocs samples and of these isolates, 16S-rRNA gene was then isolated and amplified using PCR. The 16S-rRNA gene of the isolates was then cut using HaeIII (5’-GG↓CC) and HhaI (5’-GCG↓C) restriction enzymes resulting an ARDRA pattern which was further used as the binary data for the construction of phylogenetics tree that was used to estimate the group of bacteria. The result with HaeIII cut restriction enzyme from biofloc-associated bacteria gave 11 ARDRA patterns, while with the restriction enzyme HhaI gave eight ARDRA patterns. Phylogenetics of bacterial populations from biofloc-based cultivation system water consisted of at least 13 different bacterial species. Result of sequencing from two gene sample 16S-rRNA were identified as Microbacterium foliorumand and Pseudomonas putida. Keywords: bacterial diversity, ARDRA, biofloc, phylogeny  ABSTRAK Penelitian ini bertujuan untuk menganalisis keragaman genetika bakteri bioflok menggunakan metode polymerase chain reaction (PCR) 16S-rRNA dengan teknik ARDRA. Sebanyak 38 isolat bakteri dominan yang diperoleh diamplifikasi gen 16S-rRNAnya dengan PCR, kemudian dipotong dengan enzim restriksi HaeIII (5’-GG↓CC) dan HhaI (5’-GCG↓C). Pola ARDRA ini dijadikan data biner sebagai input untuk konstruksi pohon filogenetika yang dapat digunakan untuk memerkirakan jenis bakteri yang ada. Gen 16S-rRNA hasil PCR setelah dipotong dengan enzim restriksi HaeIII didapatkan 11 pola ARDRA, sedangkan dengan enzim restriksi HhaI menghasilkan delapan pola ARDRA. Berdasarkan pohon filogenetika, diketahui populasi bakteri pada air sistem budidaya bioflok sedikitnya terdiri atas 13 jenis bakteri. Berdasarkan sekuensing dari dua sampel gen 16S-rRNA teridentifikasi jenis bakteri Microbacterium foliorum dan Pseudomonas putida. Kata kunci: keragaman bakteri, ARDRA, bioflok, filogenetika

Abstract

Nevertheless, lately Arabic Learning Method at both organizations deteriorated in its implementation. Therefore, it is important to be researched, to discover its problem background.This research means to compare between Arabic Learning Method for Development of Speaking Skill which was implemented at TMI Al-Amien Boarding School and Annuqayyah Mantiqah Latee Guluk-Guluk Sumenep Madura, also to know the running of Arabic cultural activities in both institutes.The form of this research is Qualitative Research which is used descriptive approach. While, data collecting technique uses interview with several informer which is consisting of Responsibiliters, Supervisors, and Arabic Teachers. And also, direct observation to the field and utilizing several document which is regarded with this research.Based on the result of the research shows that: First; there are differences between the implementation of Arabic Learning Method for Development of Speaking Skill at both institutes. Besides, in language achievement side both are increasing and competitive. In language center of TMI Al-Amien Boarding School has been using four methods, and they are: Direct Method, Repeat Method, Conversation Method, and Expressing Method. And also, in DALFIS has been using two methods, they are: Grammar Method and Translation Method.Exactly between Markazul Lughah in TMI Al-Amien Prenduan Boarding School and DALFIS Annuqayah Mantiqah Latee Guluk-Guluk have many differences according to surpluses and decreases.

Abstract

Penelitian ini merupakan penelitian Penelitian Tindakan dalam Bimbingan Konseling (PTBK) yang bertujuan untuk meningkatkan motivasi belajar siswa melalui penerapan layanan bimbingan kelompok. Penelitian ini dilaksanakan dalam 2 (dua) siklus dengan 4 (empat) tahapan prosedur penelitian tindakan yaitu (1) Perencanaan, (2) Pelaksanaan, (3) Pengamatan, dan (4) Refleksi. Penelitian dilaksanakan di SMP Institut Indonesia Semarang dengan subjek penelitian ini adalah 10 orang siswa kelas VIII semester gasal tahun ajaran 2022/2023. Pengumpulan data menggunakan skala motivasi belajar. Hasil penelitian menunjukkan bahwa terdapat peningkatan skor rata-rata pada pra siklus, siklus I dan siklus II. Rata-rata skor pra siklus sebesar 75,1 meningkat pada siklus I menjadi sebesar 83,6 dan meningkat pada siklus II menjadi 103,5. Peningkatan rata-rata skor dari pra siklus ke siklus I sebesar 8,5 dan peningkatan dari siklus I ke siklus II sebesar 19,9. Persentase ketuntasan klasikal pra siklus sebesar 30% meningkat menjadi 70% pada siklus I dan meningkat menjadi 100% pada siklus II. Oleh karena itu dapat disimpulkan bahwa layanan bimbingan kelompok dapat meningkatkan motivasi belajar siswa. Penelitian tindakan yang dilakukan menggunakan layanan bimbingan kelompok dapat mendorong siswa mengembangkan perasaan, pikiran, persepsi, wawasan dan sikap yang menunjang peningkatan motivasi belajar siswa.
