Abstract

Hyperglycemia in individuals with type 2 diabetes mellitus is primarily driven by the rapid hydrolysis of starch by the enzyme α-amylase in the pancreas and the breakdown of oligosaccharides by α-glucosidase in the intestine. Peronema canescens Jack. (PC) has shown promise as a potential antidiabetic agent. This study aimed to evaluate the total flavonoid, phenolic, and α-amylase inhibitory activity of extracts and fractions derived from PC leaves using both in vitro and in silico approaches. The ethanol extract of PC leaves was fractionated through liquid-liquid extraction using n-hexane, ethyl acetate, and water as solvents. Preliminary phytochemical screening of the extracts and fractions identified the presence of alkaloids, flavonoids, saponins, tannins, and steroids/triterpenoids. The n-hexane fraction exhibited the highest total flavonoid content, averaging 203.37±4.38 mg QE/gram, while the ethyl acetate fraction demonstrated the highest total phenolic content, averaging 147.04±0.79 mg GAE/gram. Furthermore, the ethyl acetate fraction showed the strongest α-amylase inhibitory activity, with an average inhibition rate of 70.38±1.26%. In silico analysis, combined with GC-MS identification, suggested that three compounds, bis(2-ethylhexyl) phthalate, myristyl oleate, and 14 beta H-pregna may contribute to the observed α-amylase inhibitory activity. These findings highlight the potential of PC as a source of natural antidiabetic agents.

Abstract

Averrhoa bilimbi, a plant native to Indonesian, exhibits antioxidant, sunscreen, and tyrosinase inhibitory activities due to its polyphenol and flavonoid content. This study aimed to evaluate the anti-aging efficacy of a topical cream containing A. bilimbi fruit extract through a human clinical trial. The extract was obtained by maceration using 70% ethanol and formulated into a water-in-oil (W/O) cream at concentrations of 1% (F1), 3% (F2), and 5% (F3). Each formulation was evaluated for its physical characteristics, including organoleptic properties, homogeneity, pH, and viscosity. Irritation tests were conducted on animals and human volunteers. A total of 24 human subjects applied the cream for 30 days. Skin condition parameters—sebum, moisture, pigmentation, pore size, elasticity, and collagen fibers—were assessed before and after treatment using a skin analyzer. The results showed that the F3 formulation produced the most notable improvements, including increased skin moisture and elasticity, and decreased pigmentation and pore size. Statistical analysis revealed significant improvements (p < 0.05) in pore size and elasticity for the F3 group. These findings indicate that the 5% A. bilimbi extract cream is a safe and effective anti-aging topical formulation.

Abstract

Curcuma aeruginosa Roxb rhizome contains secondary metabolite compounds and plays a role in various activities such as antioxidant, antibacterial, anthelmintic, antiandrogenic, antinociceptive, and anticancer. Anticancer activity that has been reported in Curcuma aeruginosa Roxb rhizome is limited to breast and cervical cancer. The purpose of this study was to explore the potential of Curcuma aeruginosa Roxb rhizome in melanoma cancer through the mechanism of inhibiting the BRAF V600E. The 96% ethanol extract of Curcuma aeruginosa Roxb rhizome was separated to produce n-hexane (HF), ethyl acetate (EAF), and ethanol (EF) fractions. The GC-MS results showed that there were 31 compounds from the three fractions. The docking validation process was carried out on the native ligand N-(3-{[5-(4-chlorophenyl) -1H-pyrrolo [2,3b]pyridin3yl] carbonyl}2,4difluorophenyl) propane-1-sulfonamide. All compounds were prepared as ligands for molecular docking with the BRAF V600E receptor (PDB ID: 3OG7). Docking validation on native ligand showed RMSD 1.03Å. The smallest binding affinity are 4,4a,5,6,7,8-Hexahydronaphthalen-2(3H)-one (-6,89 kcal/mol); 1Cyclohexyl-2-propen-1-one (-6,68 kcal/mol); Cyclooctenone (-6,23 kcal/mol); and vemuravnib is still better as K+ (-11.11 kcal/mol). All three compounds do not bind to key amino acid residues of BRAF V600E such as vemuravenib at GLN A:530, CYS A:532; ASP A:594. These results indicate that further structural development is needed for better activity.
