Abstract

Abstract. Amalini YR, Muzakhar K, Su’udi M, Setyati D, Dwinianti EF, Ulum FB. 2025. Morphological, molecular, and phytochemical characteristics of wild edible fern (Diplazium esculentum) from Jember, East Java, Indonesia. Biodiversitas 26: 2794-2805. Diplazium esculentum, an edible pteridophyte with medicinal potential, exhibits morphological variation that risks improper medicinal use. This study aimed to comprehensively characterize D. esculentum from Jember, Indonesia, through an integrative approach, including morphological analysis, DNA barcoding, phytochemical profiling, and antioxidant activity assessment. Fifteen specimens, locally referred to as pakis menir, were collected from a single site within a rubber-mixed coffee plantation in Panti, Jember District, East Java, Indonesia. The examination of morphological traits, molecular identification using the rbcL marker, analysis of phytochemical profiles through GC-MS, prediction of biological activities using PASS online, and evaluation of antioxidant activity using the DPPH assay all pointed to the potential of D. esculentum. Morphological analysis confirmed D. esculentum's diagnostic traits, while rbcL DNA barcoding validated a 100% sequence match with D. esculentum (OL536867.1). GC-MS analysis of young fronds identified 25 bioactive metabolites, including 11 phenolics, 9 terpenoids, and 5 alkaloids, with 96% exhibiting high biological activity (Pa>0.7) according to PASS predictions. The dominant compound, 3,6-dimethylquinoline (12.38%), exhibited significant anticancer potential, while the frond extract showed strong antioxidant activity (IC50: 91.25±7.60 ppm), reflecting its phenolic richness. These results highlight the potential of D. esculentum's phytochemical wealth and pharmacological promise, supporting further exploration in pharmacognosy, nutraceuticals, and bioactive compounds.

Abstract

Abstract. Ulum FB, Putri YA, Farhan AM, Setyati D, Rajamani R, Sugiharto B, Su’udi M. 2025. Phytochemical and pharmacological evaluation of Vanda tricolor from Gumitir, East Java, Indonesia. Biodiversitas 26: 4783-4792. Vanda tricolor an epiphytic orchid native to the coffee agroforest of Gumitir, East Java, Indonesia, represents a promising source of pharmacologically active compounds. This study aimed to characterize the bioactive constituents of V. tricolor and evaluate their antioxidant, antimicrobial, and anticancer potential. Methanolic leaf extracts were analyzed using gas chromatography-mass spectrometry (GC-MS) for metabolite profiling. Its biological activities were assessed through the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, agar well diffusion method, and molecular docking against the antiapoptotic Bcl-2 protein. GC-MS analysis identified 48 metabolites, with 4-butylphenol (7.57%) as the predominant compound. The extract demonstrated very strong antioxidant activity (IC??: 37.25 ppm), along with moderate antibacterial effects against Staphylococcus aureus and Escherichia coli; however, it showed no growth inhibition against Salmonella typhi. Molecular docking revealed that 4-butylphenol exhibited the highest binding affinity toward Bcl-2 (-5.50 kcal/mol), indicating its potential as a lead compound for anticancer therapy. These findings represent the first integrated phytochemical and pharmacological profile of V. tricolor from Gumitir, underscoring its value as a source of natural bioactive agents. The results also provide a scientific basis for further exploration of orchids in drug discovery and contribute to the conservation of this underutilized native species.

Abstract

Abstract. Ulum FB, Setyati D, Oktavianawati I, Rizqoni MIA, Sari EN, Muzakhar K, Su’udi M. 2026. Bioactivities and phytochemical profile of Cyclosorus parasiticus from East Java, Indonesia via GC-MS and LC-MS. Biodiversitas 27 (2): d270234. https://doi.org/10.13057/biodiv/d270234. Cyclosorus parasiticus is a terrestrial fern abundantly distributed in the area of Gumitir Mountain, East Java, Indonesia. Despite its ecological abundance, its pharmacological potential remains undercharacterised, although leaf tissues from Gumitir populations accumulate relatively high levels of flavonoids and alkaloids. Through an integrative bioassay–metabolomics approach, this study presents the first systematic evaluation of the antibacterial and antioxidant activities, together with comprehensive chemical profiling, of C. parasiticus from the Gumitir region. Metabolite annotation revealed pronounced chemical diversity, with 39 compounds identified by GC-MS and six dominant LC-MS features. Importantly, the diterpene euphornin and the alkaloids canthiumine and haplophytine were detected and are reported here for the first time in a fern, substantially expanding current knowledge of pteridophyte secondary metabolism and highlighting previously overlooked alkaloid diversity in fern taxa. The methanolic extract exhibited concentration-dependent antibacterial inhibition against Staphylococcus aureus, Kocuria rosea, Pseudomonas aeruginosa, and Escherichia coli, with greater susceptibility observed in Gram-positive bacteria. Antioxidant activity measured by the DPPH radical-scavenging assay yielded an IC50 value of 176.66±14.64 ppm, indicating moderate radical-scavenging capacity. Collectively, these findings establish C. parasiticus (Gumitir population) as a chemically rich fern with measurable antibacterial and antioxidant potential, supporting its promise as a natural source of pharmacologically relevant metabolites.

Abstract

Jember Regency encompasses several plantation areas characterized by diverse ecological conditions, notably the Gumitir, Garahan, and Tancak plantations. Among the flora in these regions, the genus Hoya is notable for its medicinal potential. This study aimed to characterize the diversity of Hoya species, provide detailed morphological documentation, and evaluate the phytochemical and antioxidant properties of a medicinally used species. Field exploration employed a systematic plot-based design consisting of 150 plots (5 × 5 m²) with 3 m spacing. The most vegetatively dominant species, Hoya lacunosa, was subjected to GC–MS phytochemical analysis and DPPH-based antioxidant evaluation. A total of eleven Hoya species were recorded across the three plantation sites. The Gumitir plantation exhibited the highest diversity, containing ten species., i.e. Hoya burtoniae, H. camphorifolia, H. carnosa, H. cinnamomifolia, H. dennisii, H. fuscomarginata, H. lacunosa, H. micrantha, H. purpureofusca, and two unidentified Hoya spp. In contrast, the Garahan plantation supported only H. dennisii and H. fuscomarginata, while H. lacunosa was exclusively found in Tancak indicating a highly localized distribution. GC-MS profiling on H. lacunosa leaves revealed 26 compounds, comprising 16 secondary metabolites (12 phenolics, 2 alkaloids, and 2 terpenes) and 5 primary metabolites. The methanol extract exhibited strong antioxidant activity (IC₅₀ = 50.89 ppm), reflecting its potential as a natural source of bioactive compounds. These findings demonstrate substantial Hoya diversity in Jember Regency and underscore the potential of H. lacunosa as a valuable source of bioactive metabolites for pharmaceutical development.
