Abstract

Background Cancer cells have a relatively high telomerase activity compared to normal cells, so that cancer cells have the ability for continued proliferation and uncontrolled mitosis. Telomerase is an enzyme responsible for the length of telomeres, DNA segments located at the ends of eukaryotic chromosomes. Natural materials such as rodent tuber (Typhonium flagelliforme) have anticancer potential. The purpose of the present study was to determine the effects of Typhonium flagelliforme extract on telomerase expression in HeLa cervical cancer and T47D breast cancer cells. Methods This experimental laboratory study was conducted on cultured HeLa and T47D cancer cell lines, with normal Vero cells as controls, and using RPMI and M199 culture media. The study comprised three groups, i.e. controls, and groups receiving Typhonium flagelliforme extract at doses of ½ IC50 and IC50. Telomerase expression was measured by immunohistochemistry (IHC). Analysis of variance and LSD multiple comparison test were used to analyze the data. Results Telomerase expression in cancer cells showed significantly higher values compared to normal Vero cells. Typhonium flagelliforme extract was capable of significantly decreasing telomerase expression in cancer cells receiving the extract. Conclusion Typhonium flagelliforme extract at different doses is capable of decreasing telomerase expression more effectively in cervical cancer cells than in breast cancer cells. This study shows that Typhonium flagelliforme may have anti-cancer activity, necessitating further investigations.

Abstract

Telomeres are strands of non coding DNA at the ends of chromosomes that have the primary function to protect DNA from damage and maintain chromosomal stability. Physical exercise will increase the antioxidant activity can increase telomere proteins, lengthen telomeres and or protein networks associated with telomere so that the telomere remains long, or stopping telomere shortening. Telomere length was also associated with age. The purpose of the research was to determine telomere length of leukocyte cells in the KONI (Indonesian National Sports Committee) athletes in Jakarta. The research method is descriptive, by measuring telomere length using quantitative PCR on leukocyte cells. Samples are KONI athletes from several sports, including men and women athletes, with ages between 15-20 years. Used a control group (not athletes) is students of the Faculty of Medicine, University of YARSI. The results showed that there was no significant difference (p> 0.05) between telomere length group of athletes with the control group in both sexes. Similarly, telomere length between athlete male with female athletes also showed no significant difference (p> 0.05). It was concluded that physical exercise in athletes KONI at the age of 15- 20 years had no effect on telomere length in leukocytes. The results of this study provide information about the telomere length in Indonesian athletes at an early age.

Abstract

Cancer cells have a relatively high telomerase activity compared to normal cells, so the cancer cells have the ability to continue to proliferate and undergo mitosis uncontrolled. Telomerase is an enzyme which responsible for telomere length, a DNA segment that is at the end of eukaryotic cell chromosomes. Telomeres and telomerase play a role in the incidence of carcinogenesis. Natural materials such as taro mice (Typhonium flagelliforme) have potential as anticancer. The purpose of the study was to determine the effects of plant extracts of rat taro on the expression of telomerase in cancer cell Raji. The research method is experimental studies in some form cancerous cell culture cell line, Raji. Used as a control normal cells is Vero cell. The Culture medium used RPMI for Raji cell  and M199 for Vero cell. The study consisted of three groups, control, doses of 1 ½ IC50 and IC50 doses. Expression of telomerase enzyme was measured by the Immunohystochemistry method (IHC). The results showed that the expression of telomerase in cancer cells showed values significantly higher than the normal cells (Vero). Giving mice taro plant extracts (Typhonium flagelliforme) were able to decrease the expression of telomerase significantly in both treatment doses. It was concluded that rodent tuber extract (Typhonium flagelliforme Lodd) can reduce the expression of telomerase in Raji cells, so that the rodent tuber extract (Typhonium flagelliforme Lodd) has potential as an anticancer through the expression of telomerase.Keywords: telomerase, IHC, Typhonium flagelliforme

Abstract

BACKGROUNDCancer is one of the causes of high mortality. Breast and cervical cancers are two of the most frequent cancers affecting women around the world, including Indonesia. Natural materials such as rodent tuber (Typhonium flagelliforme) have anticancer potentials. The rodent tuber extract contains ribosome inactivating proteins (RIPs) capable of cutting the DNA or RNA of cancer cells and blocking the growth of cancer cells. The purpose of this study was to evaluate the cytotoxic effects of Typhonium flagelliforme Lodd extract on HeLa cervical cancer and Michigan Cancer Foundation-7 (MCF-7) breast cancer cells.METHODSSubjects were cultured cell lines of HeLa cells in Rosswell Park Memorial Institute (RPMI) and of MCF-7 cells in Dulbecco’s Minimum Essential Medium (DMEM). Rodent tuber ethanolic extract was diluted in dimethyl sulfoxide (DMSO). The cytotoxicity assay used the 3-(4,5-dimethyl thiazol-2-yl,5-diphenyl) tetrazolium bromide (MTT) method. Absorbance was read in an ELISA reader at a wavelength of 595 nm.RESULTSRat tuber extract at all dilutions (500; 250; 125; 62.5; 31.25; 15.625;7.81; 3.9 ì g/ mL) showed cytotoxic effects against HeLa and MCF-7 cells. Higherconcentrations of the extract gave a higher proliferation inhibition effect.Calculated IC50 values of the extract by probit analysis were 30.19 ìg/mL against HeLa cells and 5.586 ì g/mL against MCF-7 cells.CONCLUSIONSEthanolic extract of Typhonium flagelliforme Lodd has cytotoxic effects against HeLa cells and MCF-7 cells. The cytotoxic effects against MCF-7 cells are greater than the cytotoxic effects against HeLa cells.

Abstract

BACKGROUNDCancer cells have a relatively high telomerase activity and a lower p53 protein expression than normal cells, so that cancer cells have the ability to continue to proliferate and do not undergo apoptosis. One of the cancer treatments is chemotherapy using bioactive ingredients from synthesis or isolation of natural materials. One of the plants that have potential as anticancer agent is periwinkle (Catharanthus roseus L). The research objective was to evaluate the effect of ethanolic periwinkle leaf extract against p53 protein and telomerase expression in T47D cancer cells.  METHODSAn experimental study with controls was conducted involving T47D breast cancer cells. They were divided into 3 groups (control, ½ dose of IC50/26.849 µg/mL, and one dose of IC50/53.699 µg/mL) at a cell density of 1 x 104 cells/well. Expression of p53 and telomerase was measured by the immunohistochemistry method. Data were analyzed using one-way ANOVA followed by a multiple comparison test.RESULTSPeriwinkle leaf extract significantly increased p53 protein expression (p<0.05) at both treatment doses, ½ IC50 and IC50, compared to the control group and it highly significantly reduced telomerase expression (p<0.01), in comparison with the control group at both treatment doses. CONCLUSIONPeriwinkle leaf extract has potential as an anti-breast cancer agent by increasing p53 protein expression and inhibiting telomerase expression.

Abstract

Integrated Services Post (Posyandu) Cadres are a volunteer that helps to manage community health care issues. Normally, they play an important role in mother, child, and elderly health issue prevention and promotion. However, in the COVID-19 pandemic time, the role of Posyandu cadres should be optimized for the prevention of the disease. This research aimed to assess the efficacy of the utilization of WhatsApp in increasing knowledge of Posyandu cadres on preventing COVID-19, through Whatsapp group (WAG). The Quasi-experiment was conducted on 30 cadres in Bekasi Barat. They were invited to received counseling sessions using the Whatsapp Group (WAG) on how to prevent the disease and how to correctly use the mask. They were provided with all the counseling material that they could read before the question and ask session with the mentor. Changes in the knowledge level of the cadres were assessed using the pretest and post-test methods. There was a significant increase in the level of knowledge (Wilcoxon Signed Ranks Test, p = 0.004) which was indicated by a higher post-test score (mean 75.46) than the pre-test score (mean 60.69). The correct use of masks was the cadres’ most significant increase in knowledge post-WAG-counseling session. Most of the cadres enjoyed the education process through WAG and would like to use the educational materials as a tool to provide education to the wider community. Education through WAG is effective for increasing the knowledge of Posyandu cadres about COVID-19 prevention and can be emulated by cadres to educate the wider community

Abstract

Background: Cancer is a disease that gets serious attention in the medical world. This is due to the ever increasing number of patients and there has been no effective way to treat. Cancer cells have telomerase activity is relatively high compared to normal cells, so the cancer cells have the ability to continue to proliferate. Cancer cells undergo uncontrolled mitosis and have high telomerase activity compared to cells normal. Telomerase is an enzyme responsible for telomere length, a segment of DNA that is the tip of chromosomes in eukaryotic cells. Telomeres are associated with the process of ageing and carcinogenesis. The purpose of this study was to determine the expression of telomerase in some cells such as breast cancer, cervical cancer, and lung cancer. Methods: The research method is experimental studies in several cancer cell cultures in the form of cell line. Cancer cells used were: HeLa (cervical cancer), MCF7 and T47D (breast cancer), WiDr (lung cancer), and Raji (lymphoma) with culture medium RPMI, DMEM, and M199. Vero cells is used (fibroblast cells) as a control (normal cells). Expression of telomerase enzyme was measured by the Immunohystochemistry (IHC) method. Results: The results showed that the cancer cells have activity/higher telomerase expression were highly significant (p < 0.01) compared to normal cells (Vero cells). Similarly, the expression of telomerase in HeLa versus WiDr, WiDr versus T47D, T47D versus Raji, and Raji versus MCF7 also showed highly significant differences (p < 0.01). Telomerase expression between cancer cells that showed significant difference (HeLa cells versus Raji cells; HeLa cells versus MCF7 cell; T47D cells versus MCF7 cells) (p<0.05). No significant difference was found in the group of HeLa cells versus T47D, WiDr versus Raji cells, and WiDr versus MCF7. Conclusions: It was concluded, that the cancer cells have telomerase expression of specific and different from each other, depending on the type of cell. T47D breast cancer cells have telomerase expression of the highest, followed by cervical cancer cells (HeLa). Lung cancer cells (WiDr) with cell lymphoma (Raji) has almost the same expression and both have lower expression.

Abstract

Kanker payudara adalah keganasan yang berasal dari epitel duktus maupun lobules di jaringan payudara. Di sisi lain, penyakit fibrokistik adalah benjolan teraba di payudara dan terkait dengan siklus menstruasi. Karena gejala awal yang mirip, pada pemeriksaan klinis, penyakit fibrokistik sering salah didiagnosis sebagai kanker payudara. Oleh karena tingginya kasus kanker payudara di Indonesia, penentuan stadium klinis dan pemeriksaan histopatologi diperlukan untuk menentukan diagnosis dan tata laksana lebih lanjut.Penelitian ini bertujuan untuk mengetahui bagaimana perbedaan kepadatan jaringan ikat dan perbedaan luas sel pada kanker payudara dan penyakit fibrokistik.Jenis penelitian ini adalah eksperimen dengan rancangan penelitian cross sectional. Sampel dalam penelitian ini adalah blok paraffin pasien kanker payudara dan penyakit fibrokistik di Rumah Sakit Islam Cempaka Putih Jakarta. Pengumpulan data ini dilakukan dengan data primer sediaan biopsi eksisi atau mastektomi pasien kanker payudara dan penyakit fibrokistik dan diolah menjadi sediaan slide menggunakan pewarnaan masson’s trichrome dan Hematoxylin Eosin. Analisis dilakukan menggunakan uji komparatif dua sampel yang tidak berpasangan dan distribusi tidak normal dengan uji Kolmogorov Smirnov.Penelitian ini menunjukkan bahwa rerata luas morfologi kelenjar pada kanker payudara lebih besar dibandingkan penyakit fibrokistik (226,324 um : 60,522 um). Berdasarkan hasil uji analisis didapatkan perbedaan yang bermakna secara statistik antara luas morfologi kelenjar pada kanker payudara dan penyakit fibrokistik. Semenetara jumlah jaringan ikat pada fibrokistik lebih banyak dibandingkan kanker invasif payudara. Kepadatan rata-rata jaringan ikat pada fibrokistik payudara sebesar 52%. Sedangkan kepadatan rata-rata jaringan ikat pada kanker payudara sebesar 29,17%. Berdasarkan hasil uji analisis Kolmogorov Smirnov didapatkan hasil yang signifikan secara statistik antara variabel kepadatan jaringan ikat pada kanker payudara dan penyakit fibrokistik (p kurang dari 0.05).Terdapat perbedaan bermakna jaringan ikat antara kanker payudara dengan penyakit fibrokistik. Sementara luas morfologi kelenjar pada kanker payudara lebih besar dibandingkan penyakit fibrokistik

Abstract

Changes in lifestyle and modernization have caused a shift in the pattern of diseases from communicable to non-communicable diseases (NCD). The NCD category includes diabetes, hypertension, obesity, coronary heart disease. Non-communicable diseases are a challenge during the Covid-19 pandemic because they are comorbid, which will aggravate the disease. Therefore it is necessary to educate on a healthy lifestyle to avoid these new infectious diseases. The method was carried out through webinars with the topic of an active lifestyle, teachings on faith and introduction to herbs to PKK cadres (Family Welfare Empowerment – FWE) in Ciseeng village. The metabolic health characteristics have normal values of 0%, 26%, 100%, 74% for waist circumference (WC), body mass index (BMI), systolic and diastolic blood pressure, respectively. Knowledge of healthy lifestyle from the webinar increased from 51.30% to 85.96% (p <0.05). The conclusion is that the metabolic health of respondents is considered at risk, while webinar activities reduce the risk of comorbidities.
