Abstract

Cervical cancer is one of the most prevalent cancer types, making the development of effective anticancer agents critical. Cisplatin (CIS) is a drug that has been used to treat cancer, but it also affects normal cells. Hibiscus sabdariffa L. extract (HSE), which has the potential as an anticancer agent, can be developed as a co-chemotherapy with CIS. This study aimed to determine the potential of HSE as a co-chemotherapy with CIS against HeLa cervical cancer cells and determine specific and non-specific parameters of the studied extract. Hibiscus sabdariffa L. (HS) simplicia powder was macerated with 96% ethanol. An in vitro evaluation was carried out on antiproliferative activity by calculating the doubling time. Additionally, a selectivity test was conducted to calculate combination index (CI) values based on the microtetrazolium (MTT) method. Apoptosis mechanisms were explored based on immunocytochemical methods using p53, caspase, and Bax antibodies, followed by an observation of apoptotic induction using a flow cytometer. The HSE and CIS selectivity index values obtained for Vero cells and HeLa cells were 209 and 278, respectively. The antiproliferation test results showed that the combination of HSE and CIS could better extend the doubling time of cells compared to the negative control. This combination also demonstrated a strong synergistic effect, with a CI value of 0.001. The extract as a co-chemotherapy with CIS was capable of increasing the expression of p53, caspase-3, and Bax. The flow cytometry analysis results indicated that HSE could induce apoptosis. Based on the results on the IC50 and CI value of HSE, as well as on the doubling time and apoptosis induction of HSE-influenced HeLa cells, it is concluded that HSE has the potential as a co-chemotherapy against cervical cancer.

Abstract

Abstract

Abstract

The high number of filariasis cases in Indonesia requires efforts to control the population of Culex quinquefasciatus larvae. This study aims to analyse the concentration that is effective in killing Cx. quinquefasciatus larvae for 48 hours, and to analyse the LC50 and LT50 values of bandotan leaf extract within 48 hours. Bandotan leaves were collected from Jamblang village, Sleman, Yogyakarta. The leaves were extracted using 96% ethanol, diluted into five concentrations repeated 5 times. Third instar larvae of Cx. quinquefasciatus were obtained from the Salatiga Environmental Health Laboratory (BBLKL). Larval mortality was analysed using Kruskal-Wallis test and LC50 and LT50 probit analysis. The results obtained showed that 96% ethanol extract of bandotan leaves at concentrations of 1% (positive control) and 9% had an effect on larval mortality, with an LC50 value obtained of 3.88%. The LT50 values at concentrations of 0%, 1%, 3%, 5%, 7%, and 9% were 0 hours; 2.64 hours; 146.90 hours; 102.43 hours; 79, 24 hours; and 53.83 hours. The conclusion of this study is that 96% ethanol extract of bandotan leaves at the concentration used in this study does not have the potential to be used as a larvicide against Cx. quinquefasciatus.

Abstract

Fruit flies, Bactrocera spp. (Diptera: Tephritidae), cause severe yield losses in chili production and conventional insecticide-based control raises environmental and resistance concerns. To our knowledge, this study was the first field evaluation in Indonesia to measure the mesh size threshold for an augmentorium that simultaneously retains newly emerged adult Bactrocera insects and allows for the release of naturally occurring parasitoids. This research aimed to identify fruit fly parasitoid species found in chili (Capsicum frutescens L.) fields and recommend mesh sizes that would enable parasitoids to pass through augmentation devices. This study was conducted in Balung District and the Agrofarmaka Laboratory of Jember University, Jember Regency, East Java, Indonesia. Infested chili fruits were placed in augmentorium devices fitted with four mesh diameters 0.5 mm/32 Mesh; 0.75 mm/24 Mesh; 1 mm/16 Mesh; and.5 mm/12 Mesh. Observations were made by identifying parasitoid species and counting the number of emerging parasitoids. The results showed the presence of two parasitoids, Psyttalia fijiensis and Trichopria sp., which successfully emerged from samples and passed through the meshes. P. fijiensis was able to traverse only the largest mesh (1.5 mm), whereas Trichopria sp. passed even the smallest mesh (0.5 mm). Based on these results, the 1.5 mm (12-mesh) augmentorium optimizes its dual function as a sanitation tool for fruit infested with fruit flies and as a parasitoid augmentation in chili cultivation. This evidence supports the practical application of augmentorium as a landscape-scale celement in integrated pest management for Bactrocera spp. is supported by this evidence.
