Abstract

Gram positive and negative bacteria have always been a problem for the survival of living things in a high threshold as normal flora. Telang flower contains anthocyanins which are efficacious as antioxidants and antibacterials so that kombucha has a high potential to be fermented by kombucha as the latest biotechnology product innovation in improving the immune system with the addition of a concentration of Baduy honey Product SR12 which has the potential to inhibit the growth of pathogens. This study aims to produce information regarding the concentration of Baduy honey product SR12 which has the potential as a gram-positive and gram-negative antibacterial. The research design used was a randomized block design which was divided into 2 factors. Factor I is the Concentration of Telang Flower Kombucha Sugar (Clitoria ternatea L) solution in the first fermentation (20%, 30%, and 40%) and factor II is the concentration of baduy honey product SR12, namely (20%, 30%, and 40%). Each treatment was repeated 3 times. The results obtained were processed using statistical analysis using one-way ANOVA at the 95% level. If the data from the research results have significant differences, ideally it can be followed up using a post hoc test. The results obtained in this study were the concentration of Baduy honey Product SR12 was positively correlated in preventing the growth of pathogens in both gram-positive and gram-negative bacteria. The conclusion in this study is that kombucha flower telang has the ability as a gram positive and negative antibacterial and can also be developed as the latest biotechnology product innovation. Telang flower kombucha fermentation with a concentration of Baduy honey Product SR12 of 40% had the highest antibacterial activity when compared to fermented telang flower kombucha at concentrations of Baduy honey Product SR12 with a concentration of 20% and 30%. The average diameter of the inhibition zone produced by Staphylococcus aureus bacteria is 20.08 mm in the strong category, Staphylococcus epidermidis 17.98 mm in the strong category, Pseudomonas aeruginosa 17.27 mm in the strong category, and Escherichia coli 16.59 mm strong category.

Abstract

Diarrhea that occurs in the digestive tract is a big problem for human intestinal health. This can be caused by the activity of pathogenic bacteria in the form of Vibrio cholera and Shigella dysinteriae. In this study, kombucha has been shown to have pharmacological activity in vitro in inhibiting the growth of Vibrio cholera and Shigella dysinteriae bacteria at all sugar concentrations. The purpose of this study was to determine the in vitro pharmacological activity of the butterfly pea flower kombucha in inhibiting the growth of the two test bacteria consisting of a sugar concentration of 20%, 30% and 40%. The method for testing the inhibition of both bacterial growth tests is disc diffusion. One way ANOVA and post hoc analysis are one of the quantitative test methods used in this study of the average diameter of the inhibition zone of teal flower kombucha from various sugar concentrations. The results of this study have proven based on a one-way ANOVA with each P value> 0.05 and then continued through post hoc analysis concluded that the telang flower kombucha at a concentration of 40% is the best treatment in inhibiting the growth of both test bacteria where the concentration is significantly different from sugar concentration 20% and 30%.

Abstract

One of the fertility indicators in women is the optimal functioning of the ovaries. Exposure to cigarette smoke affects the decline in ovarian function in quality and quantity. The main components contained in cigarette smoke include tar, nicotine, carbon monoxide, inorganic gases, heavy metals, nitrosamines, carbonyls, formaldehyde, and polynuclear aromatic hydrocarbons. The main components contained in cigarette smoke positively influence female reproductive hormone levels and even reduce fertility. One of the efforts to prevent free radicals from cigarette smoke and have the potential as a natural antioxidant is consuming butterfly pea flower kombucha. The butterfly pea flower kombucha from previous studies contains phytochemicals that can inhibit free radicals. This study aimed to study the effect of butterfly pea flower kombucha on the ovary morphometry of female mice exposed to cigarette smoke. The design of this study was Completely Randomized Design (CRD). The test animals used were female mice aged 10 to 12 weeks weighing 20 to 20 g in a total of 24 mice. The butterfly pea flower kombucha given was ten mL/L, 20 mL/L, 30 mL/L, and 40 mL/L. Cigarette smoke exposure to mice was carried out on days 1-20 as much as one stick and treated with butterfly pea flower kombucha on days 21-40. Mating of female mice was carried out on the 40th day. Observations of ovarian morphometry such as length, width, weight, and number of corpus luteum were carried out on the 18th day of gestation. ANOVA and DMRT with a confidence level of 1% is the data analysis performed. The conclusion of this study was that butterfly pea flower kombucha had a good effect on improving fertility levels, which could be observed with test parameters such as length, width, weight, and number of corpus luteum and administration of kombucha with a concentration of 40 mL/L was the best treatment.

Abstract

Kombucha bunga telang selain berpotensi sebagai minuman probiotik dalam meningkatkan sistem kekebalan tubuh memiliki potensi juga sebagai bahan aktif sabun cuci tangan dalam menghambat pertumbuhan bakteri Staphylococcus epidermidis, Vibrio parahaemolyticus, dan Escherichia coli. Tujuan penelitian ini adalah untuk membuat formulasi dan sediaan sabun cuci tangan yang berbahan aktif larutan fermentasi kombucha bunga telang dengan konsentrasi gula 20%, 30%, dan 40%. Metode penelitian ini dilakukan secara eksperimental laboratorium dengan membuat 4 sediaan sabun cuci tangan dan menyediakan sabun cuci tangan pasaran untuk diuji pada ketiga pertumbuhan bakteri uji melalui metode difusi cakram. Analisis data yang digunakan adalah ANOVA satu jalur dilanjut melalui Analisis pos hoc. Hasil penelitian ini disimpulkan bahwa sabun cuci tangan yang berbahan aktif larutan fermentasi kombucha bunga telang berkolerasi secara positif dalam menghambat ketiga pertumbuhan bakteri uji. Konsentrasi 40% pada sediaan sabun cuci tangan kombucha bunga telang berbeda nyata secara signifikan dengan nilai F tabel lebih kecil dibanding F hitung (P<0,05) pada konsentrasi 20%, 30%, dan basis sabun. Namun tidak berbeda nyata dengan sabun cuci tangan yang tersedia dipasaran dalam menghambat ketiga pertumbuhan bakteri uji. Konsentrasi 40% (sabun cuci tangan kombucha bunga telang) konsentrasi terbaik dari seluruh perlakuan dalam menghambat ketiga pertumbuhan bakteri uji.

Abstract

Based on the results of previous research, butterfly pea flower kombucha has been proven that apart from being able to be used as a probiotic drink to enhance the immune system, it can be used as an active ingredient in medicines and cosmetics, a raw material for organic liquid fertilizer, and also in formulations and spray preparations designed to inhibit the growth of pathogenic fungi that cause wilt. concave, and brownish on the stems of horticultural commodity plants. This research aims to make a fermented solution of butterfly pea flower kombucha as a formulation and spray preparation designed as an antifungal for Fusarium solani. The Fusarium solani antifungal test of each formulation and the telang flower kombucha spray preparation was carried out using disc diffusion. One-way ANOVA and post hoc analysis are the statistical analyzes used in this research. Based on the one-way ANOVA test with a p value of <0.05 each, followed by post hoc analysis, it was proven that butterfly pea flower kombucha at a sugar concentration of 40% was significantly different as an antifungal for Fusarium solani compared to 20% and 30%. The conclusion in this study is that a sugar concentration of 40% in this study is the best treatment as an antifungal for Fusarium solani, so that apart from being used as a probiotic drink, butterfly pea flower kombucha in this study has the potential to be used as an antifungal for Fusarium solani.

Abstract

Fermentasi kombucha bunga telang memiliki aktivitas sebagai antibakteri Pseudomonas aeruginosa. Tujuan dari penelitian ini adalah untuk memformulasi sediaan sabun cuci tangan cair probiotik fermentasi kombucha bunga telang dan menguji aktivitas antibakteri sabun cuci tangan probiotik fermentasi kombucha bunga telang terhadap bakteri Pseudomonas aeruginosa. Bunga telang difermentasi dengan scoby selama 1 bulan. Pengujian aktivitas antibakteri terhadap Pseudomonas aeruginosa dilakukan dengan metode difusi cakram. Pengujian Rata-rata diameter zona hambat dari masing-masing sabun diolah menggunakan ANOVA satu jalur dan uji lanjut analisis pos hoc. Hasil ANOVA satu jalur telah terbukti bahwa masing-masing sediaan sabun cuci tangan kombucha bunga telang berpotensi dalam menghambat pertumbuhan bakteri Pseudomonasa aeruginosa dengan nilai F tabel lebih kecil daripada F hitung yang ditetapkan yaitu 0,05 sehingga dapat dilanjutkan melalui analisis pos hoc dimana sabun cuci tangan kombucha bunga telang pada konsentrasi 40% berbeda nyata dengan konsentrasi 20% dan 30% sebagai antibakteri P. aeruginosa. Hasil penelitian dapat disimpulkan bahwa larutan fermentasi kombucha bunga telang dapat dibuat formulasi dan sediaan sabun cuci tangan probiotik dalam menghambat pertumbuhan bakteri P. aeruginosa. Nilai rata-rata zona hambat pada masing-masing sabun adalah 2,83 mm (20%), 5,11 mm (30%), dan 8,66 mm (40%).

Abstract

Bahan alam merupakan senyawa bioaktif yang dihasilkan secara natural untuk dikembangkan baik sebagai terapeutik, maupun bahan baku kosmetik. Sejauh ini kombucha bunga telang telah banyak dikembangkan sebagai minuman probiotik, bahan aktif obat seperti obat kumur, bahan aktif kosmetik seperti sabun mandi, sabun mandi batang, sabun cuci tangan, sabun cuci piring, sampo, sampo gel, serta hand sanitizer,spray natural dalam menghambat pertumbuhan mikroba patogen dari tanaman komoditas hortikultura. Tujuan dari kegiatan pengabdian kepada masyarakat ini adalah untuk meningkatkan wawasan secara detail mengenai potensi kombucha bunga telang sebagai bahan aktif obat maupun kosmetik secara nyata kepada siswa siswi SMAN 5 Cilegon yang terlibat dalam karya ilmiah remaja biologi sebagai salah satu agenda rutin pengabdian satu smester dan membuka peluang untuk melombakan karya ilmiah berbasis penelitian secara nasional. Metode penyampaian ini dilakukan dengan ceramah yaitu pemaparan materi, dan kuisioner. Hasil kuisioner telah membuktikan bahwa murid yang mengikuti KIR Biologi di SMAN 5 Cilegon sangat antusias dalam meningkatkan wawasan yaitu mengenai manfaat kombucha bunga telang sebagai minuman probiotik dalam meningkatkan sistem imun telah dicapai sebesar 60%, kandungan fitokimia pada kombucha bunga telang telah dicapai sebesar 70%, potensi kombucha bunga telang sebagai bahan aktif obat maupun kosmetik telah dicapai 80%, potensi kombucha bunga telang sebagai peningkat ekonomi telah dicapai sebesar 85%, dan standarisasi kombucha bunga telang sebagai bahan aktif obat maupun kosmetik dalam jangka panjang telah dicapai sebesar 90%.

Abstract

This research aims to see the potential of each formulation and preparation of dishwashing soap with the active ingredient telang flower kombucha in inhibiting the growth of pathogenic bacteria isolated from beef. This research is a laboratory experiment, namely by making a dishwashing soap base without the active substance of telang flower kombucha. Make a dishwashing soap base that adds the active substance of butterfly pea flower kombucha at concentrations of 20%, 30% and 40%. Biochemical identification results have shown that beef samples contain gram-positive bacteria in the form of Staphylococcus aureus and Listeria monocytogenes, as well as gram-negative bacteria in the form of Salmonella thyposa and Escherichia coli. The results of this research have also proven that pharmaceutical biotechnology products in the form of formulations and dishwashing soap preparations with the active ingredient telang flower kombucha have a positive correlation in inhibiting the growth of the four test bacteria. Based on the one-way ANOVA statistical test at each P value <0.05, it proves that telang flower kombucha dish soap at a concentration of 40% according to post hoc analysis is not significantly different from concentrations of 20% and 30% as a pharmaceutical and antibacterial biotechnology product. isolated from beef. The conclusion of this research is that the formulation and preparation of dishwashing soap at a concentration of 40% proved to be the most effective as a pharmaceutical biotechnology product in inhibiting the growth of the four test bacteria.

Abstract

The problem of stunting in Indonesia is still quite high, namely 21,6% based on data from the 2022 Indonesian Nutrition Status Survey (SSGI). One of the effective measures to control stunting is through providing additional food (PMT) such as biscuits to children. Carrots and pumpkin are local foods that are rich in beta carotene. Carotenoids as a source of provitamin A are able to overcome the problem of vitamin A deficiency (VAD), which is the cause of growth disorders in children. This research aims to determine the effect of substitution of yellow pumpkin (Cucurbita moschata) and carrot (Daucus carota L.) composite flour on organoleptic characteristics including color, aroma, taste, texture and nutritional content (beta carotene, carbohydrates, protein and fat). This research used a Randomized Block Design (RAK) with 3 treatments, namely formula 1 (150 grams of wheat flour, 50 grams of pumpkin flour and 50 grams of carrot flour), formula 2 (50 grams of wheat flour, 60 grams of pumpkin flour and 50 grams of flour carrots) and formula 3 (50 grams of wheat flour, 50 grams of pumpkin flour and 150 grams of carrot flour) with 3 repetitions. The research results showed that formula 3 biscuits were the best formulation in terms of organoleptic characteristics with the highest beta carotene content compared to F1 and F2. F3 biscuits contain 30,97 mg beta carotene, 7,45 grams of carbohydrates, 3,22 grams of protein and 8,89 grams of fat. The conclusion of this research is that the substitution of wheat flour with composite flour (Cucurbita moschata and Daucus carota L.) has an effect on the nutritional content (beta carotene, carbohydrates, protein and fat) of biscuits. Consuming pumpkin biscuits per 100 grams can qualify as a good source of the antioxidant beta carotene.

Abstract

Manfaat minuman probiotik kombucha bunga telang sebagai produk hasil dari proses fermentasi menggunakan teknologi bioteknologi mengandung berbagai senyawa bioaktif atau metabolit sekunder yang berperan penting dalam menjalankan berbagai aktivitas farmakologinya sehingga memiliki potensi sebagai bahan aktif obat maupun kosmetik. Salah satu senyawa yang berperan penting dan diuji pada penelitian ini adalah vitexin yang berasal dari golongan flavonoid dan memiliki aktivitas sebagai antioksidan maupun antikanker. Tujuan dari penelitian ini adalah untuk membuktikan bahwa senyawa vitexin memiliki aktivitas dalam menangkal radikal bebas dan kanker secara in silico sebagai gambaran awal untuk pengembangan kandidat bahan aktif obat baru. Metode penelitian yang digunakan berbasis laboratorium kering menggunakan software bioinformatika. Adapaun software yang digunakan pada penelitian ini ialah Discovery Studio 2021, ChemDraw Ultra 8.0, ChemDraw Ultra 8.0 dan MGL Tools 1.5.7.   Langkah pertama yang dilakukan ialah mengunduh struktur protein (3ZBF) dari Protein Data Bank (PDB) dan membuat struktur ligan (vitexin). Selanjutnya dilakukan preparasi protein untuk mencegah kemungkinan gangguan dalam proses ikatan antara ligan dan reseptor serta preparasi ligan untuk mencapai bentuk optimal dari ligan yang memiliki konformasi terbaik. Langkah berikutnya adalah validasi docking dengan cara melakukan docking ligan bawaan terhadap protein untuk membandingkan ligan hasil proses docking (ligan validasi) dengan ligan bawaan dan melihat nilai Root Mean Square Deviation (RMSD). Hasil penelitian yang diperoleh yakni senyawa vitexin memiliki nilai energi bebas -6.97 kkal/mol, nilai konstanta inhibisi 7800 nM dan dapat berinteraksi pada asam amino protein ROS, antara lain GLU2027, MET 2029, LEU2086, ASP2033 dan GLY2032 dengan nilai RMSD 1.383 Å. Kesimpulan dari penelitian ini adalah penambatan senyawa vitexin yang terkandung pada kombucha bunga telang terhadap inhibitor ROS memiliki nilai RMSD < 2 Å, mampu berinteraksi dengan asam amino GLY2032 yang merupakan penyebab terjadinya resistensi terhadap inhibitor ROS (Crizotinib)serta dapat memberikan gambaran awal sebagai kandidat obat baru yang diregulasi untuk memiliki aktivitas farmakologi sebagai antioksidan maupun antikanker.
