Abstract

Telang flower kombucha (Clitoria ternatea L) is a functional probiotic drink that has antifungal properties. This study aimed to examine the antifungal activity of the probiotic bath soap formulation fermented kombucha telang flower against Trycophyton rubrum and Trycophyton mentagrophytes. This research method was carried out experimentally in an in vitro laboratory. The formulation of the fermented kombucha bath soap for the flower of telang was made with varying concentrations of 20%, 30%, and 40%. The preparation of probiotic bath soap for kombucha telang flower was tested for its antifungal activity against the bacteria T. rubrum and T. megantrophytes using the disc diffusion method. The working procedure includes the manufacture of bath soap preparations, and antifungal activity tests on the probiotic kombucha bath soap preparations of telang flower. The results of the study were positively correlated based on the one-way ANOVA test with f count greater than f table and showed that telang flower kombucha in the form of probiotic bath soap had antifungal activity against T. rubrum and T. mentagrophytes as a whole. The concentration of 40% was the highest concentration in the probiotic soap preparations fermented kombucha telang flower with an average inhibition zone diameter of 17.70 mm in T. rubrum species and 17.30 mm in T. mentagrophytes species. The average value of the diameter of the inhibition zone is included in the strong category.

Abstract

Vegetable waste that is widely distributed in the Cilegon Banten coconut market contains pathogenic bacteria including Staphylococcus aureus, Listeria monocytogenes, Salmonella thypi, and Escherichia coli. These pathogenic bacteria have the potential to cause extraordinary events (KLB), especially food poisoning. The appeal for vegetable waste managers at the Cilegon Banten coconut market to inhibit the growth of pathogenic bacteria is to adopt a clean lifestyle by diligently washing hands with antibacterial hand washing soap after disposal of widely exposed vegetable waste along with a mixture of other infectious waste. Hand washing soap with the active ingredient telang flower kombucha in this study correlated positively in inhibiting the growth of four pathogenic bacteria. Research results have proven that hand washing soap at a concentration of 40% is the best antibacterial treatment.

Abstract

The high incidence of infections and the increase in antibiotic resistance have become a global health concern. The emergence of strains of resistant bacteria has also encouraged the exploration of potential antibacterial agents, especially from natural sources. This review aims to provide information regarding the antibacterial activity of stems, leaves, fruits and seeds of mengkudu (M. citrifolia L,) in inhibiting various bacterial species. In this article review, online and offline literature were used. Online literature was obtained from local and international journals and scientific articles of the last 10 years, 2008-2018, which were obtained through direct search results online using online search engines namely Google and Google Scholar. Whereas the offline literature used is books and e-books. It is known that the root, stem, leaves, fruit, and seeds of mengkudu (M. citrifolia L,) with various solvents used can inhibit various species of bacteria with different diameter inhibition zones.

Abstract

In the form of natural aerosol formulations and preparations, butterfly pea flower kombucha in the form of waste can be utilized as a pharmaceutical biotechnology product to inhibit the growth of pathogenic fungi that cause disease or infection in horticultural commodity plants. Horticultural commodity plants are plant commodities that have the potential to be developed as food flavorings, traditional medicines, and spices, especially in the shallot species Allium cepa L. One of the obstacles to the growth and development of shallot plants is being hampered by attacks by pathogenic fungi from the species Fusarium fujikuroi, Alternaria sp, and Colletotrichum gloeosporioides. A safe and environmentally friendly solution to prevent the growth of these three pathogenic fungi is to utilize the fermentation waste of butterfly pea flower kombucha as a pharmaceutical biotechnology product in the form of natural spray formulations and preparations with concentrations of 20%, 30%, and 40%. Based on the one-way ANOVA test with each P value <0.05, supported by post hoc analysis. Specifically, the 40% formula differs significantly from the 20% and 30% formulas as a pharmaceutical biotechnology product in its ability to inhibit the growth of the three pathogenic fungi in shallot plants, and is the most optimal formula.

Abstract

This activity aims to improve the knowledge and skills of mothers who are members of the Langgar At-Taubah study group, Bukuan Village, Samarinda, East Kalimantan regarding making gummy candy to prevent stunting in children. The method of implementing this service uses interactive counseling. The evaluation instruments for this service are pre-test and post-test. The results of this service show that the knowledge and skills of most mothers have increased, as evidenced by the presence of 76% of mothers in the post-test who managed to answer all questions correctly, previously in the pre-test only 45%.

Abstract

Kegiatan pengabdian masyarakat ini bertujuan untuk meningkatkan kapasitas Kelompok Wanita Tani melalui pelatihan pengolahan daun kelor menjadi keripik stik dan teh herbal. Metode pelaksanaan kegiatan meliputi edukasi tentang manfaat kesehatan dan gizi dari daun kelor, pelatihan teknis pengolahan daun kelor menjadi produk bernilai tambah berupa keripik stik dan teh, serta serah terima peralatan produksi kepada kelompok wanita tani. Hasil dari kegiatan menunjukkan bahwa peserta mengalami peningkatan pengetahuan tentang kandungan gizi daun kelor serta keterampilan dalam memproduksi keripik stik dan teh herbal dengan proses yang lebih efisien. Selain itu, peralatan yang diserahkan mendukung proses produksi yang lebih optimal, sehingga kelompok tani dapat meningkatkan kapasitas produksi dan kualitas produk. Kesimpulannya, kegiatan ini berhasil meningkatkan pengetahuan dan keterampilan peserta dalam pengolahan daun kelor serta memberikan landasan yang kuat untuk memulai usaha berbasis produk olahan kelor di tingkat rumah tangga.

Abstract

Pencarian senyawa metabolit sekunder yang berhasiat sebagai sumber antioksidan jangka panjang dapat dihasilkan melalui metode bioteknologi yang cukup sederhana hingga modern baik dengan menggunakan kemampuan mikroba endofit sampai penggunaan teknik rekayasa genetika molekuler yang saat ini cukup populer yaitu gen editing. Jaringan tanaman yang berpotensi dalam menghasilkan mikroba endofit meliputi bunga, buah, batang, daun, akar, dan biji yang berperan penting dalam memproteksi tanaman inang dari cekaman lingkungan dan mikroorganisme pesaing. Salah satu jenis jamur mikro yaitu Muscodor albus telah terbukti mampu memproduksi campuran senyawa organik volatil dari tanaman kayu manis. Senyawa organik volatil tersebut memiliki aktivitas farmakologi sebagai sumber antimikroba dalam spektrum luas. Taxomyces andreane merupakan salah satu kapang endofit yang diproduksi dari tanaman Paclitaxel dan berpotensi dalam menghasilkan senyawa taksol yang memiliki aktivitas farmakologi sebagai sumber antikanker. Pestalotiopsis microspore merupakan salah satu kultur mikroba penghasil senyawa pestacin dan isopestacin dari tanaman Ketapang dan berfungsi sebagai sumber antioksidan maupun antikanker. Antioksidan digolongkan menjadi antioksidan endogen antioksidan enzim dan vitamin. Rekayasa genetika dengan memanfaatkan Agrobacterium tumafaciens berpotensi dalam menghasilkan sifat yang dibutuhkan atau diinginkan sesuai target melalui penyisipan gen dari organisme yang berperan sebagai pendonor gen menuju organisme penerima gen. Gen lancolate merupakan salah satu gen yang berhasil disisipkan untuk mengkode daun tomat dan merubah daun majemuk dalam kondisi besar menjadi lebih kecil. Metode gen editing yang direkomendasikan berupa CRISPR-Cas 9 karena dapat menyunting gen lancolate pada buah tomat dengan urutan genom diploid yang jelas.

Abstract

This study aims to analyze the microbial community in cow dung, focusing on fungi containing the psychoactive compound psilocybin, using the Next Generation Sequencing (NGS) method. Data were obtained using the Oxford Nanopore Technologies (ONT) platform and processed with the wf-metagenomics pipeline with specific parameters to generate a comprehensive taxonomic profile. Microbial community composition was analyzed by calculating relative abundances from the phylum to species level, utilizing visualizations such as stacked bar plots, heatmaps, and KronaTools to enhance data interpretation.The findings revealed significant microbial diversity in each sample, with alpha diversity analysis showing variations in richness and evenness indices of microbial communities. Panaeolus mushrooms, known as one of the cow dung fungi containing psilocybin, were identified through phylogenetic analysis and compared with genomic data from the NCBI. Bioinformatics methods, including Kraken2, TaxonKit, and Pavian, improved accuracy and detail in data interpretation. These findings provide new insights into the bioactive potential of psilocybin mushrooms and establish a foundation for further development in pharmaceutical and biotechnological applications.This research makes a significant contribution to understanding microbial ecosystems in specific environments, particularly concerning the biotechnological potential of local microorganisms. With a comprehensive approach integrating taxonomic and bioinformatics analyses, this study opens new opportunities for exploring microbes and relevant bioactive compounds.

Abstract

Manfaat minuman probiotik kombucha bunga telang sebagai produk hasil dari proses fermentasi menggunakan teknologi bioteknologi mengandung berbagai senyawa bioaktif atau metabolit sekunder yang berperan penting dalam menjalankan berbagai aktivitas farmakologinya sehingga memiliki potensi sebagai bahan aktif obat maupun kosmetik. Salah satu senyawa yang berperan penting dan diuji pada penelitian ini adalah vitexin yang berasal dari golongan flavonoid dan memiliki aktivitas sebagai antioksidan maupun antikanker. Tujuan dari penelitian ini adalah untuk membuktikan bahwa senyawa vitexin memiliki aktivitas dalam menangkal radikal bebas dan kanker secara in silico sebagai gambaran awal untuk pengembangan kandidat bahan aktif obat baru. Metode penelitian yang digunakan berbasis laboratorium kering menggunakan software bioinformatika. Adapaun software yang digunakan pada penelitian ini ialah Discovery Studio 2021, ChemDraw Ultra 8.0, ChemDraw Ultra 8.0 dan MGL Tools 1.5.7.   Langkah pertama yang dilakukan ialah mengunduh struktur protein (3ZBF) dari Protein Data Bank (PDB) dan membuat struktur ligan (vitexin). Selanjutnya dilakukan preparasi protein untuk mencegah kemungkinan gangguan dalam proses ikatan antara ligan dan reseptor serta preparasi ligan untuk mencapai bentuk optimal dari ligan yang memiliki konformasi terbaik. Langkah berikutnya adalah validasi docking dengan cara melakukan docking ligan bawaan terhadap protein untuk membandingkan ligan hasil proses docking (ligan validasi) dengan ligan bawaan dan melihat nilai Root Mean Square Deviation (RMSD). Hasil penelitian yang diperoleh yakni senyawa vitexin memiliki nilai energi bebas -6.97 kkal/mol, nilai konstanta inhibisi 7800 nM dan dapat berinteraksi pada asam amino protein ROS, antara lain GLU2027, MET 2029, LEU2086, ASP2033 dan GLY2032 dengan nilai RMSD 1.383 Å. Kesimpulan dari penelitian ini adalah penambatan senyawa vitexin yang terkandung pada kombucha bunga telang terhadap inhibitor ROS memiliki nilai RMSD < 2 Å, mampu berinteraksi dengan asam amino GLY2032 yang merupakan penyebab terjadinya resistensi terhadap inhibitor ROS (Crizotinib)serta dapat memberikan gambaran awal sebagai kandidat obat baru yang diregulasi untuk memiliki aktivitas farmakologi sebagai antioksidan maupun antikanker.
