Abstract

Curcumin is the main active ingredient from turmeric plant (Curcuma longa) has been known as anti-oxidant and anti-inflammatory agent. Bleomycin is an anti-cancer drug which have side effects such as scleroderma and body weight loss in animals or human. This study was aimed to determined biological effects of curcumin to bleomycin-induced scleroderma and body weight loss in mice (Mus musculus). Sixteen male mice (ddy strain) were devided in to four groups: (i) negative control, received aquadest injection 0,1 ml (SC); (ii) bleomycin (BLM), received 0,1 ml bleomycin 1 mg/ml SC injection; (iii) curcumin (CMN), received aquadest injection 0,1 ml (SC) and 100 mg/kg BW curcumin in 0,5% carboxymethylcellulose (IP); and (iv) BLM+ CMN, received 0,1 ml bleomycin 1 mg/ml SC injection and 100 mg/kg BW curcumin in 0,5% carboxymethylcellulose (IP). All treatments are given every day for 4 weeks and body weigh were measured every week. The dorsal skin as injection’s dot evaluated by histopathology assessment with HE stains. BLM group showed body weight and hair follicles were loss significantly with skin fibrosis and inflammatory cells infiltration. However, CMN treatment showed opposite results in BLM-treated mice. Conclusively, this study showed CMN treatment may inhibit skin fibrogenesis in BLM-induced scleroderma and body weight loss.

Abstract

African Swine Fever (ASF) is a disease affecting pigs, caused by a double-stranded DNA virus that is not transmissible to humans or other animals. It leads to significant economic losses due to high morbidity and mortality rates, particularly in densely populated pig regions like Bali Province. During the ASF outbreak from June to December 2023, this study conducted histopathological research on clinical samples. Biological materials from twelve pigs confirmed positive for ASF via qPCR examination were histopathologically analyzed. Tissue samples from various organs underwent processing and examination using hematoxylin and eosin staining. Immunohistochemistry (IHC) was employed to detect the ASF virus’s p54 protein. Clinical symptoms, anatomical pathology, and histological examination revealed characteristic acute ASF lesions. Immunohistochemistry consistently showed p54 viral antigen distribution in mononuclear cells/macrophages across various organs, with the spleen and lymph nodes being dominant sites in 12 pigs (100%). This comprehensive study demonstrates the effectiveness of IHC in detecting the ASF virus and characterizing its histopathology.Keywords: ASF; histopathology; immunohistochemistry

Abstract

Diabetes mellitus has a significant impact on health and, in the long term, can lead to complications. Women are at a higher risk of developing type 2 diabetes mellitus compared to men. For women of reproductive age, diabetes mellitus can affect reproductive health and pregnancy quality, which may influence the next generation. The study aimed to determine risk factors for type 2 diabetes mellitus in women of reproductive age in Bekasi City. This was matched case control study, involving 72 subjects selected by purposive sampling. The study was conducted at community health centre in Bekasi. Results: There were no significant differences in nutritional status (p=0.083) and age at menarche (p=0.234) of women of reproductive age with T2DM and non-T2DM. Long menstrual cycles increase the risk of type 2 diabetes mellitus by 6.2 times higher than normal menstrual cycles (p=0.005). A history of gestational diabetes in women of reproductive age increases the risk 2.2 times higher for women of reproductive age to experience T2DM in the future (p=0.027). Long menstrual cycles and gestational diabetes are important factors contributing to the increased risk of T2DM in women of reproductive age.

Abstract

The prevalence of burns ranks fourth in trauma after traffic accidents, falls, and violence. The prevalence of burns ranks fourth in trauma after traffic accidents, falls, and violence. The healing of burn wounds results from a complex inflammatory response where pro-inflammatory cytokines, for instance, TNF-α and MMP-9, play a pivotal role. Flavonoids with strong anti-inflammatory activity are apigenin, this compound comes from plants. In this study, we evaluated apigenin and its derivatives to determine their effectiveness as burn wound healing agents using molecular docking modeling techniques. To evaluate the molecular docking of these compounds with their target proteins, we used computer-aided drug design tools such as AutoDock Vina, PyMOL, and Discovery Studio. The data showed that apigenin compounds have better affinity than natural ligands to inhibit MMP-9 which can increase the rate of the inflammatory phase of burn wound healing. These findings indicate that apigenin and its derivatives have good potential to be candidates in the development of new therapeutic interventions for burn wound healing.

Abstract

Doxorubicin remains the most prescribed anticancer agent despite its unintended side effects on non-target organs. A limiting-dose strategy is used to lower incidence of cardiotoxicity. Andrographolide has therapeutic effects including antioxidant and anti-inflammatory. This study aimed to assess cardioprotective effects of andrographolide oral on lactate dehydrogenase (LDH), creatine kinase-myocardial band (CK-MB), and relative cardiac weight in doxorubicin-induced rats. Sixteen male rats Sprague Dawley randomized into four groups: receives saline i.p and vehicle orally (Normal), doxorubicin 16 mg/kgBW i.p and vehicle orally (Dox), doxorubicin 16 mg/kgBW i.p+andrographolide 30 mg/kgBW orally (Dox+And30), doxorubicin 16 mg/kgBW i.p+andrographolide 60 mg/kgBW orally (Dox+And60). Blood was collected via cardiac puncture and cardiac organs were weighed after four-weeks administration. Total LDH and CK-MB measured spectrophotometrically. LDH and CK-MB levels significantly elevated, and signs of acute toxicity in Dox group compared with Normal group. Co-treatment with andrographolide at 30 mg/kgBW and 60 mg/kgBW reduced signs of toxicity and significantly attenuated LDH and CK-MB levels compared with Dox group (P<0.05 and P<0.01). However, body weight and relative cardiac weight were not significantly different in all groups after co-treatment with andrographolide. In conclusion, andrographolide lowered LDH and CK-MB levels, therefore has a protective potency in alleviating toxic effects of doxorubicin.

Abstract

Improvement of canine reproduction has been developed by artificial insemination (AI) for effective breeding. -tocopherol widely recognized as an antioxidant that has been used to improve the quality of the spermatozoa. This study evaluated the effect of -tocopherol supplementation in dog chilled semen quality. Beagle dog breed semen samples were obtained from male Beagles using massage technique. The semen was then diluted with tris-skim milk extender supplemented with -tocopherol of various concentration, 0% as control group (T-0), 0.1% (T-0.1), 0.2% (T-0.2), and 0.3% (T-0.3), then were stored at 4 C and evaluated daily for four days, particularly for variables namely sperm motility, sperm viability, and plasma membrane integrity. Results showed that the quality of spermatozoa was higher when spermatozoa preserved in extender containing of 0.1% or 0.2% -tocopherol compared to those of control and 0.3% -tocopherol group (P0.05). The highest quality found in group with 0.2% -tocopherol. In conclusion, supplementation -tocopherol 0.2% in dog chilled semen extender is effective for improving and maintaining chilled dog sperm quality for 4 days of storage.
